Functional characterization of TBL genes revealed the role of GhTBL7 and GhTBL58 in cotton fiber elongation

TBL (Trichome Birefringence Like) gene family members are involved in trichome initiation and xylan acetylation in several plant species. In our research, we identified 102 TBLs from G. hirsutum. The phylogenetic tree classified TBL genes into five groups. Collinearity analysis of TBL genes indicated 136 paralogous gene pairs in G. hirsutum. Gene duplication indicated that WGD or segmental duplication contributed to the GhTBL gene family expansion. Promoter cis-elements of GhTBLs were related to growth and development, seed-specific regulation, light, and stress responses. GhTBL genes (GhTBL7, GhTBL15, GhTBL21, GhTBL25, GhTBL45, GhTBL54, GhTBL67, GhTBL72, and GhTBL77) exhibited upregulated response under exposure to cold, heat, NaCl, and PEG. GhTBL genes exhibited high expression during fiber development stages. Two GhTBL genes (GhTBL7 and GhTBL58) showed differential expression at 10 DPA fiber, as 10 DPA is a fast fiber elongation stage and fiber elongation is a very important stage of cotton fiber development. Subcellular localization of GhTBL7 and GhTBL58 revealed that these genes reside inside the cell membrane. Promoter GUS activity of GhTBL7 and GhTBL58 exhibited deep staining in roots. To further validate the role of these genes in cotton fiber elongation, we silenced these genes and observed a significant reduction in the fiber length at 10 DPA. In conclusion, the functional study of cell membrane-associated genes (GhTBL7 and GhTBL58) showed deep staining in root tissues and potential function during cotton fiber elongation at 10 DPA fiber.

Background: It was reported that one in four parents were hesitant about vaccinating their children in China. Previous studies have revealed a declining trend in the vaccine willingness rate in China. There is a need to monitor the level of parental vaccine hesitancy toward routine childhood vaccination and hesitancy toward the COVID-19 vaccine during the ongoing COVID-19 pandemic. Objective: This study aims to assess changes in trends of parental attitudes toward routine childhood vaccines and COVID-19 vaccinations across different time periods in China. Methods: Three waves of cross-sectional surveys were conducted on parents residing in Wuxi City in Jiangsu Province, China from September to October 2020, February to March 2021, and May to June 2021. Participants were recruited from immunization clinics. Chi-square tests were used to compare the results of the three surveys, controlling for sociodemographic factors. Binary and multivariable logistic regression analysis was used to examine factors related to parental vaccine hesitancy and COVID-19 vaccine willingness. Results: Overall, 2881, 1038, and 1183 participants were included in the survey???s three waves. Using the Vaccine Hesitancy Scale, 7.8% (225/2881), 15.1% (157/1038), and 5.5% (65/1183) of parents showed hesitancy to childhood vaccination (P<.001), and 59.3% (1709/2881), 64.6% (671/1038), and 92% (1088/1183) of parents agreed to receive a COVID-19 vaccine themselves in the first, second, and third surveys, respectively (P<.001). In all three surveys, ???concerns about vaccine safety and side effects??? was the most common reason for refusal. Conclusions: There has been an increasing acceptance of COVID-19 vaccination in Wuxi City, China. Effective interventions are needed to mitigate public concerns about vaccine safety.

In this paper, based on the single chip microcomputer as the core, designed a set of automatic optical fiber lighting intelligent lighting system, automatic with light, by judging the intensity of light intensity to track the sun, each time after collecting voltage according to the two battery voltage value of different switching charge and discharge mode. On sunny days, the system collects sunlight efficiently through a daylighting device and transmits it to the room by optical fiber for lighting. At the same time, solar panels convert solar energy into electricity and store it in batteries for system operation and night lighting.

Public space lighting is important for public perception, human health and urban planning. Previous studies used nighttime light remote sensing data to evaluate light pollution and its impacts on human health, few investigated the impact on public perception. This study combines Beijing's SDGSAT-1 imagery with ground-measured and public perception data to evaluate the feasibility of SDGSAT-1 images simulating public perception. First, ground measurements were performed to collect light attributes and questionnaires were distributed to assess public safety and comfort. Next, the relationship between SDGSAT-1 image and measured illuminance was established. Lastly, public perception in Beijing was estimated. The results show that, (1) public perception is affected by panoramic illuminance, uniformity, and blue light;(2) although illuminance is positively related to public perception, it shows diminishing marginal returns;(3) a strong correlation is found between panoramic illu-minance and SDGSAT-1 image;(4) the perception map shows that most areas in Beijing can provide safe and comfortable lighting, while there are also insufficient or overlit in some places. This study explores methods for assessing public perceptions by satellite data, indicates remote sensing can provide a larger-scale and quanti-tative evaluation of public perception, which can contribute to urban lighting design and sustainable development.

Detection of surface defects using computer vision is a pivotal technology for achieving intelligent manufacturing. Leather products are one of the most widely traded goods in the world, and automatic identification, localization, and detection of surface defects in leather are indispensable for achieving intelligent manufacturing of leather products. This research paper proposes FG-DETR, a multi-scale feature fusion extraction structure for small target detection algorithms. Given the recent success of deep learning methods in various related fields and that certain defects and flaws in leather pictures in our dataset are relatively small. FG-DETR is based on DETR with some improvements as follows, Initially, DETR's ResNet50 network performs feature extraction on the original image and produces four scales of feature maps. Second, we apply a modified Feature Pyramid Network (FPN) to downsample and upsample the four scales of feature maps, followed by the fusion and output of a single feature map. A Gaussian kernel and output then process the fused feature maps as the final feature maps. Finally, the feature maps are merged with the position encoding information and combined into the Transformer encoder-decoder architecture, resulting in the detection outcomes through the Position-wise Feed-Forward Networks (FFN). In our leather dataset, the FG-DETR model is more effective in detecting small targets than DETR, as our leather images are large, and the original DETR is nearly ineffective for images with relatively small defects. The average detection accuracy of medium and large targets also improved, with a 7.6 percentage point improvement at AP50. The experimental findings demonstrate that the multi-scale feature fusion extraction structure significantly enhances the detection accuracy of DETR.

Most atomic physics setups require a uniform static magnetic field to define a quantization axis. The straight four-conductor assembly has been proven to be a useful structure for producing such a uniform static magnetic field. Here, we present an improved four-conductor assembly, where each conductor consists of one straight section and two curved sections. This hybrid structure arrangement offers more degrees of freedom than the conventional straight four-conductor assembly, enabling a larger uniform field region and better field spatial uniformity. The modified magnetic field generation structure can be used in many fields of atomic precision spectroscopy.

Process planning is the core of guiding process production and one of the most critical parts to realize intelligent manufacturing of products. In order to realize the monitoring, simulation, prediction and control of the physical space in the intelligent manufacturing mode, the digital twin technology is used to guide the process planning. This research describes a modeling method of digital twin process model (DTPM) for manufacturing process and the contents of DT data, discusses the acquisition method of real-time data and the management method of simulation data. Aiming at the data fusion between physical space and virtual space, a hierarchical model and mapping strategy for multi-source heterogeneous data in machining process are proposed to generate the DT data. Finally, the guidance and the visualization function in process planning of DTPM is analyzed, and the effectiveness of this method is verified by choosing the design process of a key component in a marine diesel engine.

TMBstable: a variant caller controls performance variation across heterogeneous sequencing samples

In cancer genomics, variant calling has advanced, but traditional mean accuracy evaluations are inadequate for biomarkers like tumor mutation burden, which vary significantly across samples, affecting immunotherapy patient selection and threshold settings. In this study, we introduce TMBstable, an innovative method that dynamically selects optimal variant calling strategies for specific genomic regions using a meta-learning framework, distinguishing it from traditional callers with uniform sample-wide strategies. The process begins with segmenting the sample into windows and extracting meta-features for clustering, followed by using a pre-trained meta-model to select suitable algorithms for each cluster, thereby addressing strategy-sample mismatches, reducing performance fluctuations and ensuring consistent performance across various samples. We evaluated TMBstable using both simulated and real non-small cell lung cancer and nasopharyngeal carcinoma samples, comparing it with advanced callers. The assessment, focusing on stability measures, such as the variance and coefficient of variation in false positive rate, false negative rate, precision and recall, involved 300 simulated and 106 real tumor samples. Benchmark results showed TMBstable's superior stability with the lowest variance and coefficient of variation across performance metrics, highlighting its effectiveness in analyzing the counting-based biomarker. SLC24A5 plays fundamental roles in regulating melanophore development in Cyprinidae fish

The mechanism of color pattern formation in animals is complicated. Oujiang color common carp (Cyprinus carpio var. color) is an excellent fish model for studying the molecular mechanism of pigmentation. Solute carrier family 24 member 5 (SLC24A5) was identified as a key gene in regulating melanophore development and melanin distribution in organisms. In this study, functional experiments were conducted on the SLC24A5 gene in Oujiang color common carp. The full-length of SLC24A5 mRNA was cloned and obtained. In Oujiang color common carp, qRT-PCR and whole-mount in situ hybridization indicated that SLC24A5 was highly expressed in tissues in the presence of melanin and initiated its expression along with the presence of melanin during embryo development. The SLC24A5 knocked-out via CRISPR/cas9 caused an obvious loss of melanophore in larva and adult zebrafish. In addition, the homozygous SLC24A5^−/− zebrafish mutant phenotype was rescued by the over expression of the SLC24A5 from Oujiang color common carp. Results suggested that SLC24A5 is essential for regulating melanophore development and that the functional and structures of SLC24A5 are conserved in Cyprinidae fish. © 2021 The Authors
