A new approach to the synthesis of Emitasvir (DAG181), a small molecule hepatitis C virus (HCV) nonstructural protein 5A (NS5A) inhibitor, is described. Enantioselective enzymatic desymmetrization with hydrolases in the synthetical approach can significantly avoid the loss of chiral raw materials in the resolution process. The synthesis route is further optimized and scaled-up to establish a new process that is applied to the preparation of kilogram scales of target active pharmaceutical ingredients (APIs). Compared with our initial process, the overall yield of target API was dramatically improved from 17 to 40%.

The perceptible bitterness of Zanthoxylum schinifolium Sieb. et Zucc is an important factor limiting its con-sumption. Metabolomics was used to identify potentially bitter metabolites during the development of Z. schinifolium, and dynamic changes in 17 key bitter components were characterized. The key bitter components in Z. schinifolium at different developmental stages were mainly synthesized through the "phenylpropanoid biosynthesis", "flavonoid biosynthesis", and "flavone and flavonol biosynthesis" pathways. In combination with transcriptomic data, the expression of related genes regulates the accumulation process of these bitter compo-nents;based on this, a biosynthetic pathway of bitter components in Z. schinifolium was constructed. In addition, quercetin-3-galactoside, isoquercitin, quercitrin, kaempferol, and isorhamnetin were identified as the main sources of bitterness in mature Z. schinifolium based on an evaluation of taste activity value. These findings provide valuable information for understanding mechanisms underlying Z. schinifolium bitterness and promoting the further development of high-quality Z. schinifolium.

Bud endodormancy represents a pivotal and intricate biological process influenced by both genetic and epigenetic factors, the exact mechanism of which remains elusive. Hydrogen peroxide (H2O2) functions as a signalling molecule in the regulation of dormancy, with peroxidase (POD) playing a crucial role in governing H2O2 levels. Our prior transcriptomic and metabolomic investigations into diverse pear dormancy phases posited that POD predominantly oversees pear bud dormancy. In this study, we utilised qRT-PCR to screen the most significantly expressed gene, Pyrus pyrifolia POD4-like (PpPOD4-like), from seven POD genes. Subsequently, H2O2 test kits, overexpression methods, and subcellular localisation techniques were employed to assess changes in H2O2 content, POD activity, PpPOD4-like expression, and its cellular positioning during pear bud dormancy. Subcellular localisation experiments revealed that PpPOD4-like is situated on the cell membranes. Notably, H2O2 content exhibited a rapid increase during endodormancy and decreased swiftly after ecodormancy. The fluctuation pattern of POD activity aligned with that of H2O2 content. Additionally, PpPOD4-like expression was markedly upregulated, displaying an overall upward trajectory. Our findings indicate that PpPOD4-like modulates H2O2 levels by regulating POD activity, thereby actively participating in the intricate regulation of pear dormancy processes.

Multiple pre-harvest applications of antagonist Pseudomonas fluorescens ZX induce resistance against blue and green molds in postharvest citrus fruit

The pre-harvest applications of antagonists are promising strategies to minimize various postharvest diseases. Given this, we sought to evaluate the capability of Pseudomonas fluorescens ZX in inducing resistance of postharvest citrus fruit against blue and green molds, caused by Penicillium italicum and Penicillium digitatum, respectively. Results showed that pre-treatment with P. fluorescens ZX significantly inhibited blue and green molds, generally providing stronger protection with increasing number of sprays. More specifically, three applications of the antagonist reduced disease incidence by about 47% and 41%, reduced lesion diameter by around 40% and 28%, and reduced disease index by approximately 69% and 57%, when exposed to blue and green molds, respectively. Mechanistically, these protective effects might be linked to the increased activities of superoxide dismutase, ascorbate peroxidase, peroxidase, and polyphenoloxidase, along with substantial response of phenolic acids (syringic, p-coumaric, ferulic, and sinapinic acids) and flavonoids (rutin, neohesperidin, hesperetin, sinensetin and tangeretin). P. fluorescens ZX survived well on fruit surface, and more importantly, pretreatments exhibited no negative effects on fruit quality;on the contrary, it partly improved fruit quality during postharvest storage. Collectively, these results indicated that multiple pre-harvest applications of P. fluorescens ZX are useful for control of postharvest diseases.

Lipid droplets (LDs) are dynamic organelles that store neutral lipids when energy is in excess and serve as an energy reservoir during energy deprivation. Altered hepatic lipid metabolism is a critical factor influencing the development of liver disease, such as viral hepatitis, fatty liver disease, and hepatocellular carcinoma. Perilipin 2 (PLIN2) is a protein associated with the metabolism of intracellular LDs and is closely related to the clinical outcome of liver disease. While the impact of PLIN2 on the pathogenesis of liver disease is gradually being recognized, the mechanism of action remains unclear. In this review, we highlight recent advances in the understanding of PLIN2's role in the pathogenesis of liver disease through LD biogenesis, LD contact sites, LD dynamics, and lipophagy. Furthermore, we discuss the current opportunities for PLIN2-targeted therapy for liver disease.

Synthesis and bioactivity of phenyl substituted furan and oxazole carboxylic acid derivatives as potential PDE4 inhibitors

In this present study, a series of 5-phenyl-2-furan and 4-phenyl-2-oxazole derivatives were designed and synthesized as phosphodiesterase type 4 (PDE4) inhibitors. In vitro results showed that the synthesized compounds exhibited considerable inhibitory activity against PDE4B and blockade of LPS-induced TNF-alpha release. Among the designed compounds, Compound 5j exhibited lower IC50 value (1.4 mu M) against PDE4 than parent rolipram (2.0 mu M) in in vitro enzyme assay, which also displayed good in vivo activity in animal models of asthma/COPD and sepsis induced by LPS. Docking results suggested that introduction of methoxy group at para-position of phenyl ring, demonstrated good interaction with metal binding pocket domain of PDE4B, which was helpful to enhance inhibitory activity. 山东某工业园污水处理厂主要处理工业废水和少量生活污水,因进水水量水质变化较大,进水和出水均超标较多。为了确保出水达标排放,新建调节池、事故池、芬顿池、臭氧接触池等对污水处理厂进行技术改造。技术改造后,CODcr、BOD5、SS、NH3-N、TN、TP去除率分别提升42.87%、92.71%、87.27%、52.34%、60.41%和63.63%,出水稳定达到《城镇污水处理厂污染物排放标准》(GB 18918-2002)一级A标准。

Effects of inulins with various molecular weights and added concentrations on the structural properties and thermal stability of heat-induced gliadin and glutenin gels

This study investigated the influence of inulins with different molecular weights (Mw) and added concentrations on the structure and thermal stability of gliadin and glutenin gels. All inulins reduced hydrophobicity index (H0) and hindered disulfide bond (S-S) formation of both glutenin and gliadin. The H0 of glutenin reduced by 72.6%, 64.8% and 77.1% after the addition of 400 g/kg low, middle and high molecular weight inulins (L-, M-, and Hinulin, respectively), whereas the H0 of gliadin decreased by 11.7%, 31.9% and 48.6%. Furthermore, inulin increased random coils and decreased alpha-helix and beta-sheet prevalence for gliadin, while it increased alpha-helix and beta-turn prevalence and decreased beta-sheets and random coils for glutenin. Inulin also triggered S-S configuration of these two proteins to vary from g-g-g to t-g-g or t-g-t. H-inulin deepened gel electrophoresis bands of the 30-35 kDa gliadin molecules, and all inulins reduced band strength for the 30-35 kDa glutenin subunits. Inulin limited interactions between glutenin/gliadin and water, resulting in more fragile and open network structure. Moreover, the thermal stability of heat-induced inulin-gliadin gel was higher than that of inulin-glutenin gel. The impact of inulin on glutenin was stronger than on gliadin, and the effect was Mw-dependent (H-inulin > Minulin > L-inulin).

The Role of MbEGS1 and MbEGS2 in Methyleugenol Biosynthesis by Melaleuca bracteata

Many aromatic plant volatile compounds contain methyleugenol, which is an attractant for insect pollination and has antibacterial, antioxidant, and other properties. The essential oil of Melaleuca bracteata leaves contains 90.46% methyleugenol, which is an ideal material for studying the biosynthetic pathway of methyleugenol, where eugenol synthase (EGS) is one of the key enzymes involved in the synthesis of methyleugenol. We recently reported two eugenol synthase genes (MbEGS1 and MbEGS2) present in M. bracteata, where MbEGS1 and MbEGS2 were mainly expressed in flowers, followed by leaves, and had the lowest expression levels in stems. In this study, the functions of MbEGS1 and MbEGS2 in the biosynthesis of methyleugenol were investigated using transient gene expression technology and virus-induced gene silencing (VIGS) technology in M. bracteata. Here, in the MbEGSs genes overexpression group, the transcription levels of the MbEGS1 gene and MbEGS2 gene were increased 13.46 times and 12.47 times, respectively, while the methyleugenol levels increased 18.68% and 16.48%. We further verified the function of the MbEGSs genes by using VIGS, as the transcript levels of the MbEGS1 and MbEGS2 genes were downregulated by 79.48% and 90.35%, respectively, and the methyleugenol content in M. bracteata decreased by 28.04% and 19.45%, respectively. The results indicated that the MbEGS1 and MbEGS2 genes were involved in the biosynthesis of methyleugenol, and the transcript levels of the MbEGS1 and MbEGS2 genes correlated with the methyleugenol content in M. bracteata. © 2022, The Authors. 为将矿井水的氟化物从6.2 mg/L降低到1 mg/L以下,对化学沉淀、羟基磷灰石吸附、离子交换等工艺进行了实验研究及工程经济技术分析。实验结果表明:复配除氟剂沉淀、羟基磷灰石吸附、树脂离子交换工艺能够将氟离子从6.2 mg/L降低到1 mg/L以下。但钙铝药剂难以稳定将氟离子降低到1 mg/L以下。复配除氟剂沉淀、羟基磷灰石吸附及树脂离子交换除氟工艺的处理成本分别为1.14、0.91、1.59元/t。3种工艺中,投资费用最高的是树脂离子交换工艺,最低的是化学沉淀工艺。
