Cascading oxidoreductases-like nanozymes for high selective and sensitive fiuorescent detection of ascorbic acid

Compared with natural enzymes, nanozymes have the advantages of high stability and low cost; however,selectivity and sensitivity are key issues that prevent their further development. In this study, we report a cascade nanozymatic system with significantly improved selectivity and sensitivity that combines more substrate-specific reactions and sensitive fiuorescence detection. Taking detection of ascorbic acid(AA)as an example, a cascade catalytic reaction system consisting of oxidase-like N-doped carbon nanocages(NC) and peroxidase-like copper oxide(Cu O) improved the reaction selectivity in transforming the substrate into the target product by more than 1200 times against the interference of uric acid. The cascade catalytic reaction system was also applicable for transfer from open reactors into a spatially confined microfiuidic device, increasing the slope of the calibration curves by approximately 1000-fold with a linear detection range of 2.5 nmol/L to 100 nmol/L and a low limit of detection of 0.77 nmol/L. This work offers a new strategy that achieves significant improvements in selectivity and sensitivity.

Research on alkali corrosion resistance and life prediction of glass fiber recycled concrete based on grey system theory

In this paper, four kinds of recycled concrete with 0 %, 0.5 %, 1.0 %, and 1.5 % glass fiber volume ratios were prepared to investigate the alkali corrosion resistance life of glass fiber recycled concrete in an alkaline environment. The alkali solution erosion test, uniaxial compression test, and scanning electron microscope (SEM) test were carried out, respectively. The results show that glass fiber can reduce the mass loss of recycled concrete in an alkaline environment, improve its compressive strength, and prolong its alkali resistance life. When the volume ratio of glass fiber was 1.0 %, the mass loss was reduced by 0.65 %, the compressive strength was increased by 11.22 %, and the alkali corrosion life was prolonged by 40 days, up to 183 days. Based on the grey degree theory GM(1,1), the compressive strength and corrosion resistance coefficient prediction models of glass fiber recycled concrete were established. The accuracy grade of the compressive strength prediction model and the corrosion resistance coefficient prediction model were level 1 (good), indicating that the accuracy of the prediction model was good, which provided a certain theoretical basis for the research in related fields of glass fiber recycled concrete in an alkaline environment.

Due to periodic occlusion caused by rotating vehicles, the antenna cannot continuously receive GNSS signal. At the same time, the Doppler frequency shift caused by rotation also brings additional influence on GNSS signal tracking by the vehicles. This paper proposes a GNSS signal tracking algorithm based on dual antennas. The algorithm realizes the continuous and stable tracking of GNSS signal by signal preprocessing of symmetric dual antenna and joint calculation. Simulation results show that the algorithm can effectively reduce the influence of rotation on GNSS tracking PLL and improve the stability of tracking process. © 2022 ACM.

Compounds extracted from Ganoderma atrum (G. atrum) is significantly beneficial to human health, have been incorporated in numerous remedies owing to their brain and kidney health benefits and antiinflammatory and diuretic properties. G. atrum a pilot of homology of medicine and food, has an important development value of functional food and health food. In this study, to develop a new ionic-liquid-based ultrasound-assisted extraction method and bio-affinity ultrafiltration coupled with LC and MS. Moreover, molecular docking was used to evaluate the activity of the AChE inhibitors, which were subsequently isolated and purified using consecutive counter-current chromatography (CCC) and semi-preparative high-performance liquid chromatography (HPLC). Lastly, the active compounds were identified using MS and nuclear magnetic resonance. Four AChE inhibitors, viz. ergosterol, ergosta-7,22-dien-3-one, ergosta-7-en-3 beta-ol, and ergosta-4,6,8(14),22-tetraen-3-one with purities of 92.23%, 90.19%, 90.52% and 96.72%, respectively, were obtained from G. atrum using consecutive CCC (ethyl acetate/methanol/water, 4.0:1.0:7.0, v/v/v) coupled with semi-preparative HPLC (4% A (0-30 min), 4-100% A (30-40 min)). Our results suggested that G. atrum extracts can be used in the potential prevention and treatment of neurodegenerative diseases. It will help to elucidate the mechanisms of action of various medicinal plants at the molecular level and provide more opportunities to discover and develop new potential therapeutic remedies from health care food resources.

Coreactant-free and Near-Infrared Electrochemiluminescence Immunoassay with n-Type Au Nanocrystals as Luminophores

The electrochemiluminescence (ECL) bioassay is prominently carried out with the involvement of the coreactant. To remove the detrimental effects of the coreactant on the ECL of luminophores, herein, a promising ECL immunoassay strategy with biocompatible nanoparticles as the luminophore is proposed, which involves directly and electrochemically oxidizing the luminophore methionine-capped Au (Met@Au) nanocrystals (NCs) without the participation of any coreactant. Met@Au NCs are a kind of n-type nanoparticles, and they can be electrochemically injected with valence band (VB) holes around +0.80 and +1.10 V (vs Ag/AgCl). The electrochemically injected exogenous VB hole can recombine with the endogenous conduction band electron of Met@Au NCs and eventually bring out two coreactant-free and near-infrared ECL processes around 0.80 V (ECL-1) and 1.10 V (ECL-2). The intensity of coreactant-free ECL is primarily determined by the electrochemical oxidation-induced hole-injection process. ECL-2 is considerably stronger than ECL-1 and can be exploited for determining the carcinoembryonic antigen (CEA) in a sandwich immunoassay procedure with a linear range from 0.1 to 50 pg/mL as well as a limit of detection of 0.03 pg/mL (S/N = 3).

煤直接液化技术是缓解石油供需矛盾、实现煤炭清洁高效利用的重要技术途径之一,油煤浆的黏温特性对其配制、输送、加压、预热及反应性能具有重要影响。本文从油煤浆黏温特性的变化规律、研究方法、变化机理、影响因素等方面的研究进展进行了综述。在升温过程中,油煤浆流变特性表现为非牛顿流体,具有剪切稀化特性;高温高压下黏度测定方法主要有4种;分析黏温特性变化机理:煤浆制备和升温阶段黏度变化主要由溶胀作用导致,高温阶段主要由煤裂解产生沥青质(尤其是前沥青烯)导致;从溶剂、煤的性质(煤阶、显微组分及粒径分布)、剪切速率、油煤浆浓度、催化剂和氢分压等方面分析了油煤浆黏温特性的影响因素。最后指出了未来油煤浆黏温特性的研究方向,为我国煤直接液化技术的进一步发展提供参考。

Kruppel like factor 10 up-regulates PDZ and LIM domain containing protein 2 via nuclear factor kappa-B pathway to inhibit proliferation and inflammatory of fibroblastoid synovial cell in rheumatoid arthritis

Rheumatoid arthritis (RA) is an autoimmune disease caused by synovitis. Two genes, KLF10 (Kruppel like factor 10) and PDZ and LIM domain containing protein 2 (PDLIM2), play key roles in cell inflammation and proliferation. However, the specific roles of the two on inflammation and proliferation of RA-fibroblastoid synovial cell (RA-FLS) have not been reported so far. RT-qPCR and Western blot detected the expressions of PDLIM2 and KLF10 in Human Rheumatoid arthritis FLSs (HFLSs-RA). Cell transfection techniques overexpressed PDLIM2 and KLF10 or inhibited the expression of KLF10. JAPAR database predicted the binding sites of PDLIM2 and KLF10, and the binding between the two was detected and verified using luciferase reporter genes and ChIP. Subsequently, CCK-8 technology, TUNEL staining, Western blot, wound healing and ELISA detected proliferation-related indicators, migration-related indications and inflammation-related indicators. Finally, western blot was used to detect the expression of NF-kappa B pathway-related proteins to further explore the *** expression of PDLIM2 was decreased in HFLSs-RA. Overexpression of PDLIM2 inhibited proliferation, migration and inflammation in HFLSs-RA. KLF10 can transcriptionally activate PDLIM2. Interfering with KLF10 reversed the inhibition effects of PDLIM2 overexpression on the proliferation, migration and inflammation, which was possibly through the NF-kappa B pathway. Overall, KLF10 can up-regulate PDLIM2 by regulating the NF-kappa B pathway to inhibit inflammation and proliferation of HFLSs-RA.

Research on Fault Feature Extraction of Rolling Bearing Based on Adaptive DFIF and CYCBD

Due to the fact that the fault features of rolling bearing is easily drowned out by strong noise, it is difficult to exploit the impact components which contain fault information, a combination method based on snake optimizer (SO), direct fast iterative filtering (DFIF) and maximum second order cyclostationary blind deconvolution (CYCBD) is proposed, namely, SO-DFIF-CYCBD, for rolling bearing fault features extraction. Firstly, the fault vibration signal is adaptively decomposed employing parameter optimized DFIF to acquire several intrinsic mode function (IMF) components. Secondly, the sensitive components are screened for reconstruction based on the kurtosis- correlation coefficient criterion. Finally, the reconstructed vibration signal is deconvoluted utilizing the CYCBD method to reinforce the fault shock components, and obvious frequency components are extracted using envelope demodulation to distinguish the fault type. The reliability of the proposed approach are verified through simulation vibration signals and actual rolling bearing signals. In addition, compared to other combination model techniques in this article, the proposed approach can extract richer fault feature information and has stronger fault feature extraction capabilities.

Surface Defect-Involved and Single-Color Electrochemiluminescence of Gold Nanoclusters for Immunoassay

Single-color electrochemiluminescence (ECL) of nanoparticles is normally achieved in a bandgap engineered route via passivating the nanoparticle surface. Herein, when linear mercaptoalkanoic acids are employed as the thiol-capping agent of unary Au nanoclusters (NCs), a single-stabilizer-capped strategy is proposed to achieve surface defect-involved and single-color ECL from the AuNCs with hydrazine (N2H4) as the coreactant. The carbon skeleton of the linear mercaptoalkanoic acids exhibits important effects on the ECL of the AuNCs, and efficient oxidative-reductive ECL is achieved with 8-mercaptooctanoic acid (MOA), 11-mercaptoundecanoic acid (MUA), and 12-mercaptododecanoic acid (MDA) capped AuNCs, respectively. The ECL of these AuNCs not only exhibits similar ECL intensity-potential profiles with the same maximum emission potential of similar to 1.20 V (vs Ag/AgCl), but also demonstrates almost identical spectral ECL profiles of the same maximum emission wavelength around 713 nm as well as the same fwhm of 64 nm. The ECL of AuNCs/N2H4 is obviously red-shifted to the photoluminescence of AuNCs, which not only provides unambiguous evidence that bandgap-engineered ECL of these AuNCs is quenched but also manifests that the capping agent of linear mercaptoalkanoic acid is promising for the achievement of surface defect-involved and single-color ECL from AuNCs. The MUA capped AuNCs can be utilized as an ECL tag for a sensitive and selective immunoassay, which exhibits a broad linear range from 0.5 mU/mL to 1 U/mL with a low limit of detection of 0.1 mU/mL (S/N = 3) with CA125 as the model analyte. This work provides a promising alternative to the traditional surface-passivating strategy for the achievement of single-color ECL from nanoparticle luminophores.

Encapsulated polymer refers to Micro/Nano-scale particles with polymer as the core, which can release the polymer to achieve thickening. Encapsulated polymer flooding is a new type of deep profile control technology suitable for further improving the oil recovery of conventional reservoirs. However, the adsorption and temporary blockage behaviors of encapsulated polymer particles in porous media significantly affect the accurate description of their targeted viscosity enhancement. On this account, we investigated how encapsulated polymer particles adsorb and temporarily block. Using PDMS material microfluidic chips, we investigated the flow behavior of encapsulated polymer in seven different pore throat structures. High-precision pressure sensors and microscopes were employed to real-time capture microscopic flow images of microcapsule particles inside single micro-channels and observe pressure fluctuation characteristics. Based on particle image velocimetry (PIV) systems, we studied the diverting fluid behavior in parallel micro-channels with different width ratios. Building on this, we conducted dynamic adsorption experiments to quantitatively characterize the adsorption and retention behavior of encapsulated polymer particles in porous media. In this study, we observed four phenomena of free flow, surface adsorption, blockage, and multi-particle aggregation slug flow during the flow of encapsulated polymer particles in a single microchannel. In parallel microchannels, the capsule particles exhibited selectivity towards flow paths, primarily entering wider throats. With the increase of the width ratio, the encapsulated polymer particles in the narrow throat are more confined and are more likely to accumulate in the throat and cause blockage. Experiments in porous media show that the adsorption and retention of capsule particles are inversely proportional to the permeability.
