DNA methylation plays vital roles in both prokaryotes and eukaryotes. There are three forms of DNA methylation in prokaryotes: N-6-methyladenine (6mA), N-4-methylcytosine (4mC), and 5-methylcytosine (5mC). Although many sequencing methods have been developed to sequence specific types of methylation, few technologies can be used for efficiently mapping multiple types of methylation. Here, we present NT-seq for mapping all three types of methylation simultaneously. NT-seq reliably detects all known methylation motifs in two bacterial genomes and can be used for identifying de novo methylation motifs. NT-seq provides a simple and efficient solution for detecting multiple types of DNA methylation.

Lignin exhibits a long-active UV-blocking property due to its macromolecular aromatic structure and unique semiquione-quione-hydroquione transition. However, its poor absorbance in UVA region (320-400 nm) and irregular aggregation are still the obstacles. Herein, active spirapyrane (SP-Br) is synthesized and covalently introduced into alkali lignin (AL) to construct photo-stimulated UV-absorbing enhancement sunscreen actives (AL-SPn). The introduction of SP significantly improved the absorbance of AL in UVA region and its aggregation is effectively eased. When the optimal AL-SP3 solution is exposed to UV irradiation, the color turned purple, and the absorbance in UVA region further improved as the unconjugated SP transformed into conjugated merocyanine (MC) structure. When the light is removed, the solution recovered light yellow. The antioxidant property of AL endow the reversible SP-MC transformation with good cyclic stability. As-prepared AL-SP3 based sunscreen exhibit obvious photo-stimulated enhancement effect. The sun protection factor (SPF) improved from 23 to 89 after 4 h irradiation, and can maintain the high performance for another 8 h. In addition, it exhibit low permeation and good biocompatibily in vitro and vivo, demonstrating its good potential in practical use.

顾客契合行为是顾客-企业互动中的非交易行为,以社会交换理论为理论基础,运用实证研究方法探讨旅游虚拟社区中社区质量对顾客契合行为的影响,同时探讨顾客-社区关系质量对这一影响的中介作用。通过向旅游虚拟社区成员回收396份有效问卷,运用SPSS18.0和LISREL8.72统计软件对结构方程模型进行检验。研究表明,“社区质量”包含信息质量、系统质量、服务质量和互动质量4个维度,信息质量、服务质量和互动质量都对顾客-社区关系质量有显著的正向影响,系统质量则直接影响社区导向的顾客契合行为;顾客-社区关系质量对社区导向和顾客导向的顾客契合行为都有显著的正向影响,且社区导向的顾客契合行为直接影响顾客导向的顾客契合行为。研究认为,企业管理者要有效管理虚拟社区并促进顾客契合行为,应保证信息质量的时效性、可靠性和个性化,提高服务质量的全面性和专业性;应建立平台或创造契机激励社区与成员提高互动质量,着力为用户提供符合预计甚至超出预期的使用体验,确保用户信息安全,打造良好的社区形象和口碑,来提高顾客与社区的关系质量。

Capsule structured Al/FeF3/AP energetic microspheres with enhanced combustion performance and energy release efficiency by a microexplosion reaction

The agglomeration and sintering of aluminum (Al) metal fuels lead to inadequate combustion and low energy release efficiency in the solid propellant systems. Herein, we assemble Al particles with gas generator of ferric trifluoride (FeF3) into capsule structured microspheres to detonate a microexplosion reaction by the evaporation -induced self-assembly technique, with encapsulating by ammonium perchlorate (AP) oxidant. The generated ternary Al/FeF3/AP microspheres achieved superior heat of combustion (12719.75 J g-1) and combustion temperature (1639.6 degrees C) owing to the full contact between components, reduced reaction transport distance, and the fierce shattering microexplosion combustion. Combustion mechanism analysis indicated that Al2O3 layer that hindered the oxidation reaction of Al was etched by the hydrogen fluoride (HF) from FeF3 decomposition, which caused the rapid splashing of nano Al clusters to detonate shattering microexplosion combustion. Furthermore, the ternary Al/FeF3/AP microspheres dramatically improve the combustion performance of hydroxy-terminated polybutadiene (HTPB)-based propellants and achieve a burning rate of 2.975 mm s-1, higher than most of the published HTPB/AP/Al propellants to date. The construction of Al/FeF3/AP microspheres exhibits a huge po-tential for the improvement of the propulsion power of rockets and weapons.

Potential triggers and development processes of submarine landslides in the Xisha Trough Basin, South China Sea

Both submarine landslides and submarine canyons can transfer large amounts of terrigenous quantities from the continental shelf to the deep sea. In some cases, they can have a close relationship to their spatial distribution, formation and evolution. Submarine landslides on the shelf-slope can provide early depressions for submarine canyons, representing the early stage of submarine canyons. The continuous downcutting process during the evolution of submarine canyons can steepen the gradient of the canyon sidewalls, which can lead to the occurrence of submarine landslides. However, the role of submarine canyons in the initiation of slope failure has been overlooked. In this study, high-resolution multibeam bathymetry data, submarine image data and 2-D seismic data were used to investigate two adjacent submarine landslides developed on the northern slope of the Xisha Trough and the spatiotemporal relationship of these landslides to the Central Canyon. We identified a steep downslope between the submarine landslides and the Central Canyon. The Central Canyon formation age was much earlier than those of the submarine landslides. The Central Canyon is filled with modern submarine landslide deposits. The characteristics of the multiple failure scarps indicate retrogressive failure of the submarine landslides. We suggest that the submarine landslides developed on the northern slope of the Xisha Trough Basin have finally been captured by the Central Canyon. We determined that the emptying of sediments at the end of the northern slope of the Xisha Trough Basin caused by the development of the Central Canyon caused the end of the northern slope of the Xisha Trough Basin to lose support, which may have affected the slope stability and could have even played a role in the initiation of submarine landslides. Consequently, the slope area along the Central Canyon may be susceptible to slope instability and slope failure processes.

为解决装配式剪力墙灌浆套筒构造施工难、存在安全风险的问题,提出一种竖向分布钢筋不连续并加粗两侧现浇区纵筋直径的装配式剪力墙体系。提出了该剪力墙的承载力计算式,并建立了30个有限元分析模型对其进行检验,结果表明,采用正截面承载力设计值对承载力的估计较为准确,且初始刚度与现浇剪力墙一致。为便于实际工程设计,提出了间接设计方法,即将现浇剪力墙按压弯等强和抗剪等强的配筋原则等效为装配式剪力墙,设计过程中,所有标准层剪力均按底层标准层进行等效配筋。通过对间接设计方法合理性分析,发现虽然按照“偏心受压等强替换”原则配筋的装配式剪力墙在低轴压比条件下承载力低于现浇剪力墙,但由于层间剪力较低,结构仍满足安全性要求。

Gamma-aminobutyric acid(GABA)is an important metabolite which functions in plant growth,development,and stress ***,its role in plant defense and how it is regulated are largely ***,we report a detailed analysis of GABA induction during the resistance response to Pseudomonas syringae in Arabidopsis *** searching for the mechanism underlying the pathogen-responsive mitogenactivated protein kinase(MPK)3/MPK6 signaling cascade in plant immunity,we found that activation of MPK3/MPK6 greatly induced GABA biosynthesis,which is dependent on the glutamate decarboxylase genes GAD1 and *** with Pseudomonas syringae pv tomato DC3000(Pst)and Pst-avrRpt2 expressing the civrRpt2 effector gene induced GACH and GAD4 gene expression and increased the levels of *** evidence revealed that GAD1,GAD2,and GAD4 play important roles in bothGABA biosynthesis and plant resistance in response to Pst-avrRpt2 *** gad1/2/4 triple and gad1/2/4/5 quad-ruple mutants,in which the GABA levels were extremelylow,were more susceptible to both Pst and *** loss of MPK3/MPK6,or their upstream MKK4/MKK5,or their downstream substrate WRKY33 sup-pressed the induction of GAD1 and GAD4 expression afterPst-avrRpt2 *** findings shed light on both theregulation and role of GABA in the plant immunity to abacterial pathogen.

Objective: The Asthma Quality of Life Questionnaire (AQLQ) and COPD assessment test (CAT) are used to assess the health status of asthma and chronic obstructive pulmonary disease (COPD), respectively. However, whether these questionnaires are appropriate in patients with asthma-COPD overlap (ACO) has not been reported. This study aimed to evaluate the performance of the AQLQ and CAT in subjects with ***: Subjects were enrolled from two previously described observational studies in Beijing, China. ACO was defined by a consensus definition from a roundtable discussion. All subjects completed the AQLQ, CAT, St George's Respiratory Questionnaire (SGRQ), pulmonary function tests, and the Asthma Control Questionnaire (ACQ)-5. Cross-sectional construct validity was evaluated by correlating the AQLQ and CAT with SGRQ score and other measures of asthma and COPD ***: 147 subjects with ACO were recruited. There were floor effects on non-respiratory components of the CAT, and ceiling effects on emotion domains of the AQLQ. Both questionnaires were significantly correlated with ACQ-5 score but were not correlated with FEV1% predicted or FVC% predicted. The AQLQ and CAT were strongly correlated with SGRQ score (r = -0.657 and r = 0.623, respectively). Multivariable linear regression analysis showed that the AQLQ (standardized beta-coefficient = -0.449, p < .001) had a stronger association with SGRQ score compared with CAT (standardized beta-coefficient = 0.211, p = .023).Discussion: The AQLQ and CAT were both valid for assessing the health-related quality of life in subjects with ACO, but the AQLQ performed better than CAT.

Immunogenicity and safety of an inactivated SARS-CoV-2 vaccine in people living with HIV: A cross-sectional study

We aimed to analyze the efficacy and safety of an inactivated SARS-CoV-2 vaccine in people living with HIV (PLWH). A total of 143 PLWH and 50 healthy individuals were included in this study. A commercially available magnetic chemiluminescence enzyme immunoassay kit was used to detect serum IgG and IgM antibodies against SARS-CoV-2. Serum levels of SARS-CoV-2-specific IgG were significantly higher in the control group than in the PLWH group (p = 0.001). Overall, 76% of individuals in the control group were detected with seropositivity IgG against SARS-CoV-2 compared to 58% in the PLWH group (p = 0.024). In PLWH with IgG seropositivity, CD4+ T-cell counts before antiretroviral therapy (ART) was higher (p = 0.015). Multivariable analysis indicated that CD4+ T cells at IgG detection (odds ratio [OR] = 1.004, p = 0.006) and time after vaccination (OR = 0.977, p = 0.014) were independently associated with seropositivity IgG against SARS-CoV-2 in PLWH. Neutralizing antibody (nAb) titers in PLWH against wild-type SARS-CoV-2 were similar to those in the control group (p = 0.160). The proportion of seropositive nAbs against wild-type SARS-CoV-2 was also similar (95% in the control group vs. 97% in the PLWH group, p = 0.665). Similar results were obtained when nAb was detected against the delta variants with similar titers (p = 0.355) and a similar proportion of seropositive nAbs were observed (p = 0.588). All the side effects observed in our study were mild and self-limiting. The inactivated COVID-19 vaccine appears to be safe with good immunogenicity in Chinese PLWH.

The traditional identity authentication approach of username and password has the security risk of password leakage in the transmission process. For the security of password transmission in the blockchain education architecture, an asymmetric encryption algorithm is used to encrypt the transmitted password. Then use an improved random sampling algorithm to randomly extract part of the ciphertext and transmit those selected ciphertexts. The authentication credentials are sampled ciphertext and the location index. At the same time, the authentication factor is generated through the Time-Stamp technology, and mutual authentication is realized based on the Response-Challenge protocol. Through security analysis and experimental analysis, the proposed scheme is proved to be secure.
