Inhibition of growth and lung metastasis of breast cancer by tumor-homing triple-bioresponsive nanotherapeutics

Lung metastasis of breast cancer is a leading cause of cancer-related death in women. Herein, we attempted to simultaneously inhibit the growth and lung metastasis of breast cancer by delivering quercetin (QU) using LyP-1-functionalized regenerated silk fibroin-based nanoparticles (NPs). The generated LyP-1-QU-NPs had a desirable diameter (203.2 nm) and a negatively charged surface (-12.7 mV). Interestingly, these NPs exhibited intrinsic responsibilities when triggered by various stimulating factors in the tumor microenvironment (acidic pH, reactive oxygen species, and glutathione). In vitro experiments revealed that the introduction of LyP-1 to the NP surface could significantly increase their cellular uptake efficiencies by 4 T1 cells, and facilitate their accumulation in mitochondria. Moreover, LyP-1-QU-NPs showed the strongest mitochondrial damage effect among all the treatment groups. We also found that LyP-1-QU-NPs not only exhibited excellent pro-apoptotic activities but also presented strong inhibitory effects on cell mobility (migration and invasion) through anti-glycolysis and pro-autophagy. Mice experiments confirmed that LyP-1-QU-NPs could efficiently inhibit the in situ growth of breast tumors and further restrict their lung metastasis. Collectively, our results demonstrate that LyP-1-QU-NPs, which integrates the functions of tumor cell targeting, mitochondria targeting, bioresponsive drug release, proapoptosis, and anti-mobility, can be developed as a promising nanotherapeutic for the effective treatment of breast cancer and its lung metastasis.

Low atomic number silicon nitride films for transmission electron microscopy

Low stress silicon (Si)-rich silicon nitride (SiNx) films are usually used as membranes of microcapsules for transmission electron microscopy (TEM). Their relatively high atomic number has a negative impact on resolution, which is a key factor that affects imaging performance. To improve resolution, reducing Si content in films is an effective approach. However, the non-Si-rich SiNx films have large tensile stress and are ease to crack. In this study, ion implantation is utilized to adjust the stress of nearly stoichiometric (NS) SiNx films. The effects of ion implantation and thermal annealing on film properties are carefully analyzed. It is found that the NS SiNx films implanted with nitrogen (N) and annealed at low anneal temperature have both low atomic number and low stress, and their properties are very close to low stress Si-rich SiNx films. Moreover, a simple microcapsule is fabricated using the low atomic number SiNx films, and performs a TEM observation of water-soluble polypeptides growth. Polypeptides much smaller than 100 nm in size are observed.

Radiation colitis is the leading cause of diarrhea and hematochezia in pelvic radiotherapy patients. This work advances the pathogenesis of radiation colitis from the perspective of ferroptosis. An oral Pickering emulsion is stabilized with halloysite clay nanotubes to alleviate radiation colitis by inhibiting ferroptosis. Ceria nanozyme grown in situ on nanotubes can scavenge reactive oxygen species, and deferiprone was loaded into the lumen of nanotubes to relieve iron stress. These two strategies effectively inhibit lipid peroxidation and rescue ferroptosis in the intestinal microenvironment. The clay nanotubes play a critical role as either a medicine to alleviate colitis, a nanocarrier that targets the inflamed colon by electrostatic adsorption, or an interfacial stabilizer for emulsions. This ferroptosis-based strategy was effective in vitro and in vivo, providing a prospective candidate for radiotherapy protection via rational regulation of specific oxidative stress. Radiation colitis is a major side effect for pelvic radiotherapy patients, but there are limited available treatments. Here, the authors use a halloysite clay based material for the alleviation of radiation colitis in mice by inhibiting ferroptosis.

This paper presents a closed-loop hierarchical energy management control strategy for a group of connected hybrid electric vehicles (HEVs) considering the feedback of propulsion and recuperation efficiencies. The higher level controller utilizes model predictive control (MPC) incorporating signal phase and timing (SPAT) information for the prediction of the optimal velocity profile over a finite time horizon, with periodically updated efficiencies from the lower level controller. The adaptive equivalent consumption minimization strategy (A-ECMS)-based lower level controller then makes use of the optimal velocity profile for the energy management control of the HEVs. Meanwhile, the lower level controller calculates and feeds back the average propulsion and recuperation efficiencies of the current time window to the higher level controller, which updates the efficiencies with the average values and corrects the fuel consumption model for the calculation of the velocity profile of the next time window. Simulation results validate the control effectiveness of the proposed control strategy.

A targeting method for robot-assisted percutaneous needle placement under fluoroscopy guidance

Minimally invasive procedures are rapidly growing in popularity thanks to advancements in medical robots, visual navigation and space registration techniques. This paper presents a precise and efficient targeting method for robot-assisted percutaneous needle placement under C-arm fluoroscopy. In this method, a special end-effector was constructed to perform fluoroscopy calibration and robot to image-space registration simultaneously. In addition, formulations were given to compute the movement of robot targeting and evaluate targeting accuracy using only one X-ray image. With these techniques, radiation exposure and operation time were reduced significantly compared to other commonly used methods. A pre-clinical experiment showed that the maximum angle error was 0.94 degrees and the maximum position error of a target located 80mm below the end-effector was 1.31mm. And evaluation of the system in a robot-assisted pedicle screws placement surgery has justified the accuracy and reliability of proposed method in clinical applications.

Effects of P27/Bmdacapo, in the CIP/KIP family, on cell proliferation, growth and development in the silkworm (Bombyx mori)

We investigated changes in expression of the CIP/KIP family-related genes and the cycle-dependent factors Pena, Cdk4 and Cdk2 during the growth and development of mice, Drosophila and silkworms. When the organism was in a period of rapid development, the related genes of the CIP/KIP family had low expression level and the cell cycle-dependent genes were highly expressed. In mammals, the CIP/KIP family includes three genes, p21, p27/Dacapo and p57. However, only one gene, P27/Dacapo, exists in the CIP/KIP family in silkworm' and the orthologous gene in the silkworm is named Bmdacapo. Down-regulation of Bmdacapo in silkworm embryos caused overdevelopment of the embryos and indicated that Bmdacapo can inhibit silkworm growth and development. Up-regulation of Bmdacapo in silkworm cells inhibited cell proliferation, whereas down-regulation of Bmdacapo promoted cell proliferation. In order to explore the mechanism of Bmdacapo regulated silkworm development and cell proliferation, the effect of Bmdacapo on cell cycle changes was examined. The results demonstrate that Bmdacapo was able to induce G1/S phase arrest in the cell cycle. In silkworm cells, Bmdacapo inhibits the expression of Pcna, CDK4 and CDK2, which affects the cell cycle and ultimately inhibits cell proliferation. This regulatory mechanism is particularly different from mammals.

In cellular networks the primary issue in network design is Base Station Deployment. This paper proposes an unprecedented procedure for the difficulties faced in cell planning We are using metaheuristic algorithms for the 4G-LTE networks. In this proposition, we focus on satisfying coverage constraint and cell capacity constrainttogether by developing a practical optimisation problem. We begin by implementing coverage dimensioning and capacity dimensioning to calculate the amount of base stations needed in the beginning. Thereupon, we execute a Particle Swarm Optimization algorithm and Grey Wolf Optimizer to locate the optimal BS position. The base station positions fulfil the constraints in the area considered which is split into various sub areas with unique user densities. We have implemented these algorithms using Matlab software to evaluate the performance of the offered scheme. © 2019 IEEE.

Complement factor B/C2 family (Bf/C2F) proteins are core complement system components in vertebrates that are absent in invertebrates and have been lost by numerous species, raising evolutionary questions. At least 3 duplication events have occurred from Cnidaria (ancestor) to mammals. Type II Bf/C2 genes appeared during separation of Proterostomia and Deuterostomes. The second event occurred during separation of vertebrates and invertebrates, yielding type II-2 Bf/C2. The third event occurred when jawed and jawless fish were separated, eventually producing Bf and C2 genes. Herein, we report the second mollusc Sinonovacula constricta Bf/C2-type gene (Sept). ScBf is similar to Ruditapes decussatus Bf-like because both lack the first complement control protein module at the N terminus present in mammalian Bf/C2 proteins. Uniquely, the Ser protease (SP) module at the C terminus of ScBf is similar to 50 aa longer than in other complement factor B/C2-type (Bf/C2T) proteins, and is Glu-rich. Bf/C2T proteins in molluscs lack the catalytic Ser in the SP module. Surprisingly, ScBf regulates rabbit erythrocyte agglutination, during which it is localized on the erythrocyte surface. Thus, ScBf may mediate the agglutination cascade and may be an upstream regulator of this process. Our findings provide new insight into the origin of the Bf/C2F.

Survival, growth and physiology of marine bivalve (Sinonovacula constricta) in long-term low-salt culture

In this study, we investigated the possibility of rearing and breeding the razor clam (Sinonovacula constricta) in inland low salinity water or freshwater. Long-term low salinity (LS) rearing was performed for 3 months to determine the effects of LS on the survival rate, growth rate, and the activities of critical enzymes in juvenile S. constricta (JSC). The survival rate in the LS group was only 15.67% at the end of the LS rearing test. In the first month, the survival rate in the LS group was significantly lower than that in the control group (P < 0.001). The growth rate (shell length growth rate and weight gain rate) was significantly lower in the LS group than the control group in the first month (P < 0.001 for length and weight). However, the growth rates in the two groups differed little during the second and third months. The oxygen consumption and ammonia excretion rates by JSCs were significantly higher in the LS group than the control group during the first month, but they decreased gradually during the following 2 months. The Na+/K+ ATPase and superoxide dismutase activities were significantly higher in the LS group than the control group during the first month, but they then decreased gradually until there were no significant differences between the groups. However, the aspartate aminotransferase activity was higher in the LS group than the control group during all 3 months. Most of the JSCs died due to LS but the survival of some JSCs suggests the possibility of breeding LS resistant or freshwater S. constricta.
