Multifunctional Janus hydrogel targeting mitochondrial regulation and inflammatory pathways promotes infected burn wound repair

Infected burn wound repair is hindered by bacterial infection, inflammatory imbalance, and oxidative stress. Conventional antibiotic treatments face challenges from drug-resistant bacteria and biofilm formation, limiting their ability to improve the wound microenvironment. To address this change and provide a more comprehensive solution, we developed a Janus-structured smart hydrogel (P//GH + Ni + ZnO) that integrates NiCo2O4 nano-enzymes and ZnO nanoparticles. The combination of these two components provides synergistic antimicrobial, immunomodulatory, and mitochondrial protective effects, enhancing the therapeutic effect. In vitro, the hydrogel effectively inhibited drug-resistant bacteria, reduced ROS levels, stabilized mitochondrial membrane potential, and protected cellular function via PINK1/Parkin-mediated mitochondrial autophagy. Additionally, it promoted cell migration, angiogenesis, and tissue regeneration by optimizing the wound microenvironment. Building on the positive in vitro results, in vivo studies further confirmed its ability to accelerate wound healing, enhance M2 macrophage polarization, reduce TNF-α expression, and promote angiogenesis and tissue repair through TNF and VEGF signaling pathways. In summary, this Janus hydrogel significantly improved infected burn wound healing through its intelligent antimicrobial, immunoregulatory, and mitochondrial protective effects, offering a promising biomaterial solution for complex wound management and advancing the application of smart hydrogels in tissue repair. © 2025 Elsevier B.V.

Lateral transportation of soil heavy metals in rainfall events could significantly increase the scope of pollution. Therefore, it is necessary to develop a model with high accuracy to simulate the migration quantity of heavy metals. A model for heavy metal migration simulation was developed based on the SWAT(Soil and Water Assessment Tool) model. This model took into consideration the influence of soil p H value, soil particle size, runoff volume, sediment amount,concentration of water-soluble heavy metals dissolved in runoff and insoluble absorbed to the soil particles. This model was reasonable in Huanjiang watershed, Guangxi Zhuang Autonomous Region, south china, covering an area of 273 km^2. The optimal drainage area threshold was determined by analyzing the effects of watershed subdivision on the simulation results to ensure the simulation accuracy. The main conclusions of this paper were:(1) watershed subdivision could affect simulation migration quantity of heavy metals;(2) the quantity of heavy metals transported by sediment accounted for 97%–99% of the total migration quantity in the study watershed. Therefore, sediment played the most important role in heavy metal migration;(3) the optimal drainage area threshold percentage to ensure high simulation accuracy was determined to be 2.01% of the total watershed;(4) with the optimal threshold percentage, this model could simulate the migration quantity of As, Pb and Cd accurately at the total watershed and subwatershed level. The results of this paper were useful for identifying the key regions with heavy metal migration.

Light focusing in multiple scattering circumstances is important in biomedical imaging, manipulation, and therapy. Until now, many traditional photorefractive crystals have been used to generate an optical phase conjugated wavefront in an analogue time-reversed optical focusing technology. However, owing to erasure of a volume hologram during a reading procedure, the optical energy gain can never reach unity, limiting its application in delivering more energy into a target area. This paper studies vision-based road detection for a robot's path following in off-road environments. We define the problem as detecting the region in front of the robot that is mechanically traversable (i.e., mechanical traversability), that is apt to be chosen by a human to drive through (i.e., human selection), and that extends for a distance to show the road's direction, shape, or even network of the intersection ahead (i.e., far-field capability). An algorithm framework is designed that contains two parts: inference and learning. In inference, the problem is formulated as a consecutive road type classification and road region segmentation to address the diversity of terrain surfaces. In model learning, the robot is first driven by a human being, with image samples on the track of the robot being collected that meet the prerequisites of both mechanical traversability and human selection. Evaluation measures are defined to examine the three requirements of mechanical traversability, human selection, and far-field capability. The performances of the above aspects are demonstrated on a data set using LiDAR, track and manual references, which will be released together with this publication.

Stabilizing diode laser to 1 Hz-level Allan deviation with atomic spectroscopy for Rb four-level active optical frequency standard

We achieve a compact ultra-stable 420 nm blue diode laser system by immediately stabilizing the laser on the hyperfine transition line of Rb atom. The Allan deviation of the residual error signal reaches 1 Hz-level Allan deviation within 1 s averaging time, and the fractional frequency Allan deviation is 1.4x10(-15)/tau, which shows the best result of frequency-stabilized lasers based on the atomic spectroscopy without Pound-Drever-Hall (PDH) system. The signal-to-noise ratio of the atomic spectroscopy is evaluated to be 3,000,000 from the Allan deviation formula, which is the highest record, to the best of our knowledge. The frequency noise suppression characterization is demonstrated and the maximal noise suppression can be near 40 dB at 6 Hz. As a good candidate of pumping source, the ultra-stable 420 nm diode laser is successfully used in our Rb four-level active optical frequency standard system. The method can be easily extended to other wavelengths ultra-stable lasers with a Allan deviation of 10(-15) level retaining an atomic reference with low cost and low complexity while in the absence of an expensive and complicated PDH system.

Increased productivity of l-2-aminobutyric acid and total turnover number of NAD⁺/NADH in a one-pot system through enhanced thermostability of l-threonine deaminase

ObjectiveTo strengthen NADH regeneration in the biosynthesis of l-2-aminobutyric acid (l-ABA).Resultsl-Threonine deaminase (l-TD) from Escherichia coli K12 was modified by directed evolution and rational design to improve its endurance to heat treatment. The half-life of mutant G323D/F510L/T344A at 42 degrees C increased from 10 to 210min, a 20-fold increase compared to the wild-type l-TD, and the temperature at which the activity of the enzyme decreased by 50% in 15min increased from 39 to 53 degrees C. The mutant together with thermostable l-leucine dehydrogenase from Bacillus sphaericus DSM730 and formate dehydrogenase from Candida boidinii constituted a one-pot system for l-ABA biosynthesis. Employing preheat treatment in the one-pot system, the biosynthesis of l-ABA and total turnover number of NAD(+)/NADH were 0.993M and 16,469, in contrast to 0.635M and 10,531 with wild-type l-TD, *** using the engineered l-TD during endured preheat treatment, the one-pot system has achieved a higher productivity of l-ABA and total turnover number of coenzyme.

Hyperspectral ghost imaging camera based on a flat-field grating

A spectral camera based on ghost imaging via sparsity constraints (GISC) acquires a three-dimensional (3D) spatial-spectral data cube of the target through a two-dimensional (2D) detector in a single snapshot. However, the spectral and spatial resolution are interrelated because both of them are modulated by the same spatial random phase modulator. In this paper, we theoretically and experimentally demonstrate a system by equipping the GISC spectral camera with a flat-field grating to disperse the light fields before the spatial random phase modulator, hence consequently decoupling the spatial and spectral resolution. By theoretical derivation of the imaging process we obtain the spectral resolution 1nm and spatial resolution 50 mu m about the new system which are verified by the experiment. The new system can not only modulate the spatial and spectral resolution separately, but also provide a possibility of optimizing the light field fluctuations of different wavelengths according to the imaging scene. FACT (Facilitates Chromatin Transactions) is a heterodimeric protein complex involved in RNA poly-merase H transcription elongation, playing essential roles in chromatin remodeling during transcription, replication, and DNA damage repair. The FACT subunit hSpt16 is essential for nucleosome reorganization. The N-terminal domain of hSpt16 (hSpt16-NTD) was recently described as a histone (H3-H4)(2)-binding domain;however, its mode of interaction remains unknown. In this study, we solved the structure of hSpt16-NTD437 at 2.19 angstrom and found that a long-disordered region (hSpt16-LDR), after the main body of hSpt16-NTD, is a novel histone-binding motif. Furthermore, hSpt16-LDR interaction with (H3-H4)(2) is H3 N-terminal tail-independent. Therefore, Spt16-NTD is a histone H3-H4-specific binding domain with a distinct mechanism of interaction between histones and histone chaperones. (C) 2018 Elsevier Inc. All rights reserved.

Improving the stability of wheat gliadin nanoparticles - Effect of gum arabic addition

Wheat gliadin nanoparticles were prepared by antisolvent precipitation, and gum arabic (GA) was added to improve the stability of the nanoparticles. Effect of pH, gliadin/GA ratio and gliadin concentration on the gliadin-GA particles were studied by turbidimetric measurement. Spinodal lines for the GA-gliadin-solvent system were built under different pH. The influences of gliadin/GA ratio and their concentrations were represented in the diagram, thus the conditions for preparing stable composite were defined. The stability of GA-coated gliadin nanoparticles (Gliadin/GA = 1:3) was further studied compared with un-coated gliadin particles. GA-coated nanoparticles had a relatively good stability at pH 4.0-7.0 and remained a relatively low particle size with the elevated ionic strengths. They also had good thermal stability at 80 degrees C. Moreover, the interactions between gliadin particles and GA under different pH were further investigated. Hydrogen bonding was found to be the predominant force at pH 5.0, while hydrophobic force took charge of the formation of complex at pH 7.0. The findings are of great importance for extending the current knowledge about gliadin nanoparticles coupled with polysaccharides, thus providing valuable information for the development of gliadin-polysaccharide nanoparticles as potential nano-delivery systems.
