a new hybrid controller for the vertical motion of a two-mass hopping robot is proposed and applied to a triple articulated leg robot of quadruped robots. This work starts by investigating the dynamics of a two-mass model which differs the common-used SLIP model in that the contact forces between the lower leg and the ground is taken into consideration and directly further controlled. This hybrid control strategy mainly consists of three controllers, that is, the leg length controller in flight phase;the direct contact force controller and the simple switch controller in stance phase. Combine the three controllers we can eliminate deviation of the system states trajectory from the desired reference limit cycles. These controllers are then applied on the articulated leg robot by mapping the equivalent actuating force, originally modeled in the two-mass system, into the joint torques based on the dynamics of the proposed legged robot. We conducted several simulations to test the effectiveness of the proposed hybrid controller and the expected hopping motion of the articulated legged robot.

Hemolytic reactions in the hemolymph of bivalve Sinonovacula constricta show complement-like activity

The complement-like hemolysis method was used to determine the total complement-like activity of the plasma of Sinonovacula constricta. In this study, the effects of both physical and chemical conditions on complement hemolysis of S. constricta were measured. Physical conditions included proportion (S. constricta plasma: 2% rabbit red blood cells), temperature, time, and incubation, while the chemical factors consisted of Lipopolysaccharide (LPS), Flagellin (FLA), Zymosan, Peptidoglycan (PGN), Phenylmethanesulfonyl fluoride (PMSF), Methylamine, and Poly (I: C). The results showed that LPS, flagellin, Zymosan and PGN could activate complement-like activity of S. constricta plasma and cause hemolysis. PMSF and methylamine inhibited complementlike activity, resulting in the disappearance of hemolysis. Poly (I: C) had no effect on plasma complement-like activity. When the reaction temperature was less than 50 degrees C, hemolytic activity would increase following an increase in temperature. The ratio of plasma to rabbit blood cells had a great impact on the rate of hemolysis. Additionally, incubation with low speed oscillation could improve the hemolysis rate. It is indicated that the hemolytic reactions in the hemolymph of bivalve S. constricta show complement-like activity. The results contribute to further research on immune function of complement in bivalve.

Efficient CRISPR-Cas9 mediated multiplex genome editing in yeasts

Background: The thermotolerant methylotrophic yeast Ogataea polymorpha has been regarded as an important organism for basic research and biotechnological applications. It is generally recognized as an efficient and safe cell factory in fermentative productions of chemicals, biofuels and other bio-products. However, it is difficult to genetically engineer for the deficiency of an efficient and versatile genome editing technology. Results: In this study, we developed a CRISPR-Cas9-assisted multiplex genome editing (CMGE) approach including multiplex genes knock-outs, multi-locus (ML) and multi-copy (MC) integration methods in yeasts. Based on CMGE, various genome modifications, including gene deletion, integration, and precise point mutation, were performed in O. polymorpha. Using the CMGE-ML integration method, three genes TAL from Herpetosiphon aurantiacus, 4CL from Arabidopsis thaliana and STS from Vitis vinifera of resveratrol biosynthetic pathway were simultaneously integrated at three different loci, firstly achieving the biosynthesis of resveratrol in O. polymorpha. Using the CMGE-MC method,similar to 10 copies of the fusion expression cassette P-ScTEF1-TAL-P-ScTPI1-4CL-P-ScTEF2-STS were integrated into the genome. Resveratrol production was increased similar to 20 fold compared to the one copy integrant and reached 97.23 +/- 4.84 mg/L. Moreover, the biosynthesis of human serum albumin and cadaverine were achieved in O. polymorpha using CMGE-MC to integrate genes HSA and cadA, respectively. In addition, the CMGE-MC method was successfully developed in Saccharomyces cerevisiae. Conclusions: An efficient and versatile multiplex genome editing method was developed in yeasts. The method would provide an efficient toolkit for genetic engineering and synthetic biology researches of O. polymorpha and other yeast species.

Self-assembly of natural protein and imidazole molecules on gold nanoparticles: Applications in wound healing against multi-drug resistant bacteria

Developing highly active and green antibacterial agents for pathogens, especially multidrug-resistant superbugs, is vital for solving the problem of serious antibiotic resistance. Herein, we report a unique system of gold nano particles coated with chicken egg white (CEW) and 2-mercapto-1-methylimidazole (MMT) as a novel antibacterial agent. The CEW was used to prepare the gold nanoparticles as a commercially available reducing and stabilizing agent, and then the MMT self-assembled on the surface of nanoparticles. The resulting Au@CEW/MMT was found to be a highly efficient antibacterial agent, and the activity is mainly attributed to the synergistic effects of MMT and Au@CEW in undermining the bacterial membrane. Meanwhile, the studies of antibacterial activities and biocompatibility of Au@CEW/MMT with different ratios of MMT conjugation to Au@CEW confirmed that Au@CEW/MMT3 (MMT:HAuCl4 = 1:50) can maintain a balance between antibacterial properties and biocompatibility. Furthermore, in an in-vivo study using the rabbit model, gauze loaded with Au@CEW/MMT3 can effectively accelerate the healing of wounds infected with methicillin-resistant S. aureus and promote the formation of collagen. Therefore, this work illustrated a promising material with broad-spectrum antibacterial activities for preclinical applications in treating wound infections. (C) 2018 Elsevier B.V. CeO₂-ZrO₂-Al₂O₃ Modified by Selective Doping with SrO for Improved Pd-Only Three-Way Catalyst

Composite CeO2-ZrO2-Al2O3 (CZA) modified by selective doping with SrO was prepared by simple coprecipitation method. The characterization results reveal that the structural property of the modified sample is fundamentally different from that of undoped CZA, the doped SrO can selectively combine with Al2O3 and act as a stabilizer, meanwhile homogeneous CZ solid solution is formed which appears insusceptible to Al2O3. Consequently more lattice defects are created in CZAS, which facilitate the improvement of oxygen mobility. The UV-Vis DRS and XPS results demonstrate that after doping with SrO, the supported catalyst Pd/CZAS possesses higher surface Pd and Ce3+ concentrations, which consequently leads to improved reducibility and catalytic performance. Furthermore, the synergistic effect between CeO2-ZrO2 and SrO-Al2O3 helps improve the thermal stability of Pd/CZAS. As a result, after aging treatment, Pd/CZASa still maintains improved structural, textural, morphological and reduction properties along with enhanced three-way catalytic performance.

Alanine mother liquor as a nitrogen source for docosahexaenoic acid production by Schizochytrium sp B4D1

Alanine mother liquor, a type of industrial waste from alanine fermentation, was used as a nitrogen source to produce docosahexaenoic acid (DHA) by Schizochytrium sp. B4D1. The results indicated that yeast extract could trigger the utilization of the alanine mother liquor. Additionally, the alanine can be quenched during the culture, which aids in DHA accumulation. The medium components were optimized via response surface methodology as follows: 99.98-g/L glucose, 0.05-g/L yeast extract and a 183.17 dilution factor of the alanine mother liquid (v/v, with an alanine content of 0.72 g/L) and 17.98% inoculum concentration (v/v). Hermansky-Pudlak syndrome (HPS) is a human autosomal recessive disorder that is characterized by oculocutaneous albinism and a deficiency of the platelet storage pool resulting from defective biogenesis of lysosome-related organelles (LROs). To date, 10 HPS genes have been identified, three of which belong to the octamer complex BLOC-1 (biogenesis of lysosome-related organelles complex 1). One subunit of the BLOC-1 complex, BLOS1, also participates in the BLOC-1-related complex (BORC). Due to lethality at the early embryo stage in BLOS1 knockout mice, the function of BLOS1 in the above two complexes and whether it has a novel function are unclear. Here, we generated three zebrafish mutant lines with a BLOC-1 deficiency, in which melanin and silver pigment formation was attenuated as a result of mutation of bloc1s1, bloc1s2, and dtnbp1a, suggesting that they function in the same complex. In addition, mutations of bloc1s1 and bloc1s2 caused an accumulation of clusters of lysosomal vesicles at the posterior part of the tectum, representing a BORC-specific function in zebrafish. Moreover, bloc1s1 is highly expressed in the swimbladder during postembryonic stages and is required for positively regulating the expression of the genes, which is known to govern surfactant production and lung development in mammals. Our study identified BLOS1 as a crucial regulator of the surfactant system. Thus, the zebrafish swimbladder might be an easy system to screen and study genetic modifiers that control surfactant production and homeostasis.

An Automated Calibration Method of Ultrasonic Probe Based on Coherent Point Drift Algorithm

Ultrasound-based navigation, as a non-invasive and non-radiation image guiding system, is becoming a research focus in minimally invasive navigation surgery, and calibration between an ultrasonic probe and a 3D vision device is one of the key technologies for ultrasonic registration-based navigation. In this paper, a phantom model was designed as a benchmark for calibration. Both iterative closet point (ICP) and coherent point drift (CPD) algorithms are chosen as point cloud registration methods to implement calibration between ultrasonic scanned points and original phantom points to set up the relationship between the ultrasonic probe and the 3D vision device. Because of large topological difference between the ultrasound scanned points and the points from model, ICP algorithm cannot complete the registration, but the CPD algorithm could implement the registration automatically. The average errors of the center point position for each cavity were 1.50, 1.31, and 1.19 mm, respectively, and the average errors of the axis for each cavity were 0.85 degrees, 0.61 degrees, and 0.99 degrees, respectively. Experiment results showed that the average error of calibration by this method satisfies acquirements of most orthopedic surgeries, and the fully automatic implementation of ultrasonic image processing and subsequent calculation is suitable for on-line calibration and verification in surgery.

A transparent acrylic centrifugal pump model with vaned diffusers which is suitable for PIV (Particle Image Velocimetry) measurement has been built. The internal flow field inside the pump under different flow-rate conditions for different NPSH (Net Positive Suction Head) has been measured using the PIV technique with fluorescent particles. The RNG k- turbulence model has been employed to calculate the three-dimensional unsteady turbulent flows in the pump and the predicted results are examined and certified by the PIV measurement data. The predicted results of cavitation performance and internal flow velocity fields agree with the PIV data. It can be concluded that the cavitation greatly affects the flow in the impeller channel and even jammed the impeller passage with successive decrease in cavitation allowance. And simultaneously, reverse flows happen at the outlet of the impeller and inside the diffuse blades, and even the low-velocity retarding area emerges at the outlet of volute passage near the wall.
