The DREB/CBF transcription factors play important roles during low temperature, drought, and high-salt stress in higher plants. In this paper, we transferred TsCBF1 gene from a dicotyledonous halophyte Thellungiella halophila into the monocotyledonous crop maize (Zea mays L.). PCR, Southern blot, and RT-PCR analysis indicated that the TsCBF1 gene had been integrated into the genome of transgenic plants and was expressed in their progeny. After 14 days of drought stress treatments, transgenic plants showed improved drought tolerance with higher Relative Water Content (RWC), higher solute accumulation, and less cell damage compared with wild-type (WT) plants. Most importantly, they showed shorter anthesis-silking interval (ASI) and produced much higher grain yield than WT under drought stress. The results indicate that the TsCBF1 gene conferred enhanced drought tolerance to maize plants and may have utility for improving tolerance to other abiotic stresses as well.

We examined the growth, survival and immune response of the scallop, Chlamys farreri, during a 1-year period in deep water of Haizhou Bay. Scallops were cultured using two methods: (1) in lantern nets at a 5 m depth and (2) in a bottom culture system (sleeves) on the seabed at about a 25 m depth. Shell heights, meat dry weight and immune activities in the haemolymph (superoxide dismutase and myeloperoxidase) were measured bimonthly or quarterly from July 2007 to June 2008. Survival was measured at the end of the study and environmental parameters in the experimental layers were monitored during the experiment. The growth and immune activities of scallops were lower when the water temperature was high, which was consistent with the main mortality occurring in summer. The growth and immunity of scallops were higher in the suspended culture than in the bottom culture during the experiment, with the exception of shell growth during the last study period. Survival of scallops in the suspended culture (54.6 +/- 12.3%) was significantly lower than that in the bottom culture (86.8 +/- 3.5%) at the end of this study. We conclude from our results that the high mortality of C. farreri can be prevented by culturing them in a bottom culture system before November of the first year, and then transferring them to a suspended culture to improve scallop production.

The purpose of this study was to investigate the efficacy and feasibility of unmanipulated haploidentical PBSCT for the treatment of acute leukemia (AL). This study compares the clinical outcomes of high-risk AL patients who received PBSCs harvested from family members sharing at least one common haplotype to outcomes of high-risk AL patients who received a mixture of G-CSF-primed BM (G-BM) and peripheral blood (G-PB) harvests. The results show that PBSCT achieved inferior cumulative myeloid engraftment at 30 days after transplant (89.9+/-10.1% vs 100%;P=0.04), with lower cumulative incidence of grade II-IV acute GVHD (aGVHD) (37.1+/-16.5% vs 63.2+/-6%;P=0.058) compared with G-BM/G-PB transplant. However, both transplant protocols had similar rates of 2-year relapse (29.6+/-17.1% vs 34.0+/-5.7%;P=0.954), and PBSCT produced a higher incidence of 2-year non-leukemic mortality (62.5+/-14.8% vs 35.1+/-5.1%;P=0.014), as well as lower rates of overall (26.8+/-12.3% vs 43.2+/-5.0%;P=0.052) and disease-free survival (26.8+/-12.3% vs 42.4+/-5.0%;P=0.071) compared with G-BM/G-PB transplant. These results suggest that haploidentical HSCT is an option for patients with AL who urgently need a graft and do not have matched sibling donors. PBSCT is potentially inferior to G-BM/G-PB transplant, and improvements should be made before PBSCT becomes a routine in unmanipulated mismatched/haploidentical transplant settings. The relationship between plant species coexistence and soil microbial communities under heavy metal pollution has attracted much attention in ecology. However, whether plant species coexistence could offset the impacts of heavy metal combined pollution on soil microbial community structure and soil enzymes activities is not well studied. The modified ecological dose model and PCR-RAPD method were used to assess the effects of two plant species coexistence on soil microbial community and enzymes activities subjected to Cd and Ph combined stress. The results indicated that monoculture and mixed culture would increased microbe populations under Cd and Pb combined stress, and the order of sensitivity of microbial community responding to heavy metal stress was: actinomycetes 〉 bacteria 〉 fungi. The respirations were significantly higher in planted soil than that in unplanted soil. The plant species coexistence could enhance soil enzyme activities under Cd and Pb combined. Furthermore, planted soil would be helpful to enhance soil genetic polymorphisms, but Cd and Pb pollution would cause a decrease on soil genetic polymorphisms. Mixed culture would increase the ecological dose 50% (ED50) values, and the ED50 values for soil enzyme activities decreased with increasing culture time. The dehydrogenase was most sensitive to metal addition and easily loses activity under low dose of heavy metal. However, it was difficult to fully inhibit the phoshpatase activity, and urease responded similarly with phosphatase.

A semi-works production method which the extraction and isolation of 7S and 11S was improved. The method of isoelectric point and cooling precipitation was developed for extraction and isolation of 7S and 11S fractions from low-temperature defatted soybean draff, based on the indices of the extraction yield which was determined by the Kjeldahl method and purity which was determined by SDS-PAGE. Compared the optimum parameters of five kinds of extraction methods that phosphate buffer's effect is better than others, its extraction yield is 89.55%. The optimal isolation parameters of 7S are as follows: pH 4.7, sedimentation time 1 h, the stirring rate of acid 40r/min, its purity and yield are 73.25% and 50.6% respectively. The optimal isolation parameters of 11S are as follows: pH 6.2, cooling precipitation 12h, temperature 4 degrees C, the stirring rate of acid 30r/min, its purity and yield were 76.37% and 41.2% respectively. With the improvements, the extraction yield of 11S and 7S fractions were significantly increased. They may be widely used as a source of highly functional soybean protein in more food products.

Ninety strains of Lactobacillus fermentum which were isolated from traditional dairy products of minority nationalities (53 strains from Tibet, 1 from Inner Mongolia, 7 from Yun Nan and 29 from Mongolia), were evaluated for probiotic potential with the resistance to low pH, simulated gastrointestinal juice, and bile salts. The results showed that 35 strains grew well at acid condition (pH 3.0). Eleven strains were further screened out from 35 strains with high tolerance to the Simulated gastric juice (pH 2.5, 3 h of incubation). While in Simulated gastric juice (pH 2.0), only F6 (isolated from traditional dairy products in Inner Mongolia) could survive to a rate of 53.7%. Moreover, among 11 Lactobacillus strains, L. fermentum F6 showed good tolerance to bile salt, while L. fermentum IMAU60151, IMAU60083, IMAU20080 and IMAU60120 were poorly presented. Further autoaggregation properties, coaggregation with pathogens and antimicrobial activity of the 11 strains were evaluated. The abilities of coaggregation with pathogens were strain-specific and independent on incubate time. All of the selected strains showed various antimicrobial activities against five food-borne pathogens. F6 possessed the highest ability of inhibiting pathogens. All selected strains showed high autoaggregation percentage after being incubated at room temperature for 20 h. Among the selected strains, F6 showed the best probiotic potential with high tolerance to simulated gastrointestinal juice and bile salts, and broad antibacterial activity. Nanoscratch experiments on four bulk metallic glasses using a pyramidal diamond tip (Berkovich) indenter have been carried out under different loads and scratching velocities. There exists a general correlation between the hardness and wear resistance, i.e., a lower hardness produces a lower wear resistance. Scratch velocity appears to have little effect on the scratch depths of the four alloys studied. However, faster scratch velocity leads to finer shear bands. Compared with ribbon sample with the same chemical composition, Ti-based bulk glassy sample that experienced slower cooling rate during solidification, and thus contained less free volume, exhibits better wear resistance. The Spodoptera litura nucleopolyhedrovirus (SpltNPV) antiapoptotic gene Splt-p49 is able to rescue replication of a p35-null Autographa californica nucleopolyhedrovirus (AcMNPV) in AcMNPV-permissive Sf9 cells. In this study, an AcMNPV p35 knockout bacmid was generated through ET homologous recombination in Escherichia coli and designated as vAc(P35-KO). The Splt-p49 gene was transposed into the polyhedrin locus of vAc(P35-KO) to investigate if Splt-p49 has any antiapoptotic activity in the context of AcMNPV infection of AcMNPV-nonpermissive SpLi-221 cells. Our results demonstrated that Splt-p49 could not inhibit the apoptosis induced by AcMNPV infection of SpLi-221 cells when it was under the control of its native promoter. Western blot analysis showed that Splt-P49 was poorly expressed. When the expression of Splt-P49 was under the control of Drosophila hsp70 promoter, the expression of Splt-P49 was advanced, and a higher level of Splt-P49 was detected. As a result, the apoptosis of SpLi-221 cells was inhibited;however, budded-virus production did not improve in comparison with that in AcMNPV-infected SpLi-221 cells. These data indicated that there are other barrier(s) to AcMNPV replication in the nonpermissive SpLi-221 cells besides apoptosis.

The formation of Ni-based B-Fe-Ni-Si-Ta bulk metallic glasses is explored in this work. The alloy compositions are determined by using a combination of the cluster line approach, the minor alloying strategy and the substitutions of similar elements. The basic B-Ni-Si ternary compositions are first determined by the intersection points of different cluster lines, B-Ni cluster to Si and Ni-Si cluster to B, and then are minor-alloyed with 4 at.% Ta. Finally Fe is added to partially substitute for Ni to further improve the glass-forming abilities. Bulk metallic glasses with diameters of 3 mm are formed at compositions [Br-17.7(Ni1-xFex)(70.5)](Si7.8Ta4), x=0.35-0.45, which also exhibit good mechanical and corrosion-resistant properties. Furthermore, these compositions can be expressed using our cluster-plus-glue-atom model as [B-2(Ni,Fe)(8)](1)(Si,Ta)(1.33). BMGs with compositions satisfying [B-2(Ni,Fe)(8)](Si, Ta)(1) are also obtained. (C) 2009 Published by Elsevier Ltd.
