Overexpression of CD151 as an Adverse Marker for Intrahepatic Cholangiocarcinoma Patients

BACKGROUND: Previous studies have indicated that CD151, a hydrophobic protein, forms a functional complex with the proto-oncogene that encodes an N-methyl-N'-nitro-N-nitroso-guanidine (MET) protein (c-Met), and CD151 overexpression reportedly is involved in metastasis/invasion of several tumors. The objective of the current study was to investigate the expression and role of CD151 and/or c-Met in intrahepatic cholangiocarcinoma (ICC). METHODS: Sixty ICC tissues with matched nontumorous tissues and 20 normal liver tissues were used to analyze CD151 expression at the level of messenger RNA (mRNA) and protein. Then, the expression of CD151 in an ICC cell line was interrupted using a specific lentiviral-mediated small hairpin RNA (shRNA)-CD151, and the role of CD151 in the proliferation, metastasis, and invasion of ICC cells was assessed. The expression of CD151/c-Met was examined further by immunohistochemistry in a tissue microarray (TMA) that included 140 samples of ICC, and the prognostic role of CD151 and/or c-Met in ICC was evaluated in Kaplan-Meier and Cox regression analyses. RESULTS: The expression of CD151 in ICC tissues was much higher than that in nontumorous samples and normal liver;and, after the down-regulation of CD151, HCCC-9810 cells had decreased capability for metastasis/invasion in vitro. CD151 overexpression was correlated significantly with larger tumors, poor differentiation, multiple nodular, microvascular/bile duct invasion, and lymphatic metastasis (P<.05). The postoperative 2-year and 5-year overall survival (OS) rates for patients with low CD151 expression (<50% tumor staining) and/or low c-Met expression (<20% tumor staining) were higher than the rates for patients with high CD151 expression (>= 50% tumor staining) and/or high c-Met expression (>= 20% tumor staining). Composite metamaterial enabled excellent performance of microstrip antenna array

This paper reports that the split ring resonators and complementary split ring resonators are compounded to construct a novel compact composite metamaterial. The composite metamaterial exhibits a unique property of blocking electromagnetic wave propagating in two directions near the resonant frequency. An example of two-element microstrip antenna array demonstrates that the developed metamaterial enables array performance that is an improvement in comparison with the traditional one, including mutual coupling suppression of 9.07 dB, remarkable side lobe suppression and gain improvement of 2.14 dB. The mechanism of performance enhancement is analysed based on the electric field and Poynting vector distributions in array. The present work not only is a meaningful exploration of new type composite metamaterial design, but also opens up possibilities for extensive metamaterial applications to antenna engineer.

In vivo ectopic chondrogenesis of BMSCs directed by mature chondrocytes

In vivo niche plays an important role in determining the fate of exogenously implanted stem cells Due to the lack of a proper chondrogenic niche stable ectopic chondrogenesis of mesenchymal stem cells (MSCs) in subcutaneous environments remains a great challenge The clinical application of MSC-regenerated cartilage in repairing defects in subcutaneous cartilage such as nasal or auricular cartilage is thus severely limited The creation of a chondrogenic niche in subcutaneous environments is the key to solving this problem The current study demonstrates that bone marrow stromal cells (BMSCs) could form cartilage-like tissue in a subcutaneous environment when co-transplanted with articular chondrocytes indicating that chondrocytes could create a chondrogenic niche to direct chondrogenesis of BMSCs Then a series of in vitro co-culture models revealed that It was the secretion of soluble factors by chondrocytes but not cell cell contact that provided the chondrogenic signals The subsequent studies further demonstrated that multiple factors currently used for chondroinduction (including TGF-beta 1 IGF-1 and BMP-2) were present in the supernatant of chondrocyte-engineered constructs Furthermore all of these factors were required for initiating chondrogenic differentiation and fulfilled their roles in a coordinated way These results suggest that paracrine signaling of soluble chondrogenic factors provided by chondrocytes was an important mechanism in directing the in vivo ectopic chondrogenesis of BMSCs The multiple co-culture systems established in this study provide new methods for directing committed differentiation of stem cells as well as new in vitro models for studying differentiation mechanism of stem cells determined by a tissue-specific niche (C) 2010 Elsevier Ltd All rights reserved

Behavior detection of malicious software is better than signature-based detection method when used to find unknown malicious software. The paper presents a classification method of malicious software behavior detection with hybrid set based feed forward neural network. We choose malicious software detection database for test with 57345 records from National Anti-Computer Intrusion and Anti-Virus research Center. According to the definition of selected data set relations and transfer functions, the weighted path length trees of malicious software detection data are calculated for neural network input vectors. After repeat training, different malicious software detection methods can be classified by the method with the about 83.9 percent right classification.

Decomposition of sodium aluminate solution is the key step for alumina production;however,approximately only 55% alumina yield is obtained by seeded ***,it is difficult to obtain sodium aluminate solution with high caustic *** this study,a method was explored to recover alumina from spent Bayer liquor by deep decomposition with methanol.A variety of conditions,including reaction temperature,reaction time,methanol amount and seed coefficient were elaborately *** results showed that the appropriate conditions were 1:1 in volume ratio of methanol to spent Bayer liquor,more than 1.0 seed coefficient and in a 40℃water bath for 24 *** characterizing through XRD,the crystal products were found to be Al(OH)*** this method,the molar ratio of Na2O to Al2O3 of the spent liquor can be increased from about 3.0 to 10.0 due to the recovery of alumina,which is beneficial for the treatment of red mud.

Bioassay of the Herbicidal Activity of AAC-Toxin Produced by Alternaria alternata Isolated from Ageratina adenophora

Tenuazonic acid (TeA), a naturally occurring product of Alternaria alternata, a pathogen to croftonweed, was discovered to be a novel natural photosystem II (PSII) inhibitor. However, herbicidal activity of AAC-toxin, a metabolite of this fungus containing TeA as the main active ingredient, has not been evaluated systematically. In this study, we conducted activity-evaluation experiments in the laboratory, greenhouse, and field trials to assess the herbicidal potential of this fungal metabolite. AAC-toxin had high herbicidal activity on all species tested: croftonweed, large crabgrass, barnyardgrass, redroot pigweed, and eclipta. The AAC-toxin caused brown, leaf spot symptoms and leaf necrosis, subsequently killing the seedlings. When AAC-toxin was applied POST at 83 ml ai/ha, more than 95% of large crabgrass, barnyardgrass, and redroot pigweed plants were controlled 2 d after treatment in field trials. It can be concluded that AAC-toxin has broad-spectrum, rapid, and high herbicidal activity similar to that of paraquat and may have the potential to be developed as a microbe-based herbicide.

Experimental studies on temperature rise within a hydrogen cylinder during refueling

In this research, experiments were performed to investigate the thermal behaviors such as temperature rise and distributions inside 35 MPa, 150 L hydrogen storage cylinders during its refueling. The main factors affecting the temperature rise in the fast fill process such as the mass filling rate and initial pressure in the cylinder were considered. The experimental results show that the mass filling rate is a constant when the ratio of the pressure in the tank to the cylinder is higher than 1.7, and the mass filling rate decreases when the ratio is lower than 1.7;the temperature inside the cylinder increases nonlinearly in the filling process and the maximum value of temperature rise at the interface of the cylinder exists in the caudal region;the temperature rise reaches a larger value with a lower initial pressure in the cylinder or a higher mass filling rate. P>In this study, we extended in silico mapping for single trait to analyse data from multiple environments by calculating intraclass correlations and to mapping pleiotropic QTL for multiple traits by defining new statistic to measure the correlation between multiple traits and the marker. Data sets include phenotypes of eight agronomic traits obtained from six different ecologic environments and years, and genotypic information from 477 polymorphic markers on 14 ancestral lines in the pedigree of 'Suinong14'. With in silico mapping, a total of 39 markers distributed on 14 linkage groups are detected as QTL responsible for eight agronomic traits and 10 QTL are identified as having pleiotropic effects. Tracing transmission of functional QTL in the pedigree indicated that certain QTL, such as Sat_036 on linkage group D1a, Satt182 on linkage group L, and Satt726 on linkage group B2 may be responsible for the contribution of exotic germplasm to the improved cultivars.

Three kinds of TCC (transmission cross coefficient) calculation algorithms used for partially coherent imaging simulation, including the integration algorithm, the analytical algorithm, and the matrix-based fast algorithm, are reviewed for their rigorous formulations and numerical implementations. The accuracy and speed achievable using these algorithms are compared by simulations conducted on several mainstream illumination sources commonly used in current lithographic tools. Simulation results demonstrate that the integration algorithm is quite accurate but time consuming, while the matrix-based fast algorithm is efficient but its accuracy is heavily dependent on simulation resolution. The analytical algorithm is both efficient and accurate but not suitable for arbitrary optical systems. It is therefore concluded that each TCC calculation algorithm has its pros and cons with a compromise necessary to achieve a balance between accuracy and speed. The observations are useful in fast lithographic simulation for aerial image modeling, optical proximity correction (OPC), source mask optimization (SMO), and critical dimension (CD) prediction.

Identification and characterization of a novel immunoglobulin Z isotype in zebrafish: Implications for a distinct B cell receptor in lower vertebrates

It was generally believed that teleost fish possess only few immunoglobulin isotypes such as IgM and IgD. The newly discovered IgZ/IgZ-Iike molecules in zebrafish and several other fish species greatly enriched our knowledge of immunoglobulin family in lower vertebrates. In the present study, we report a second IgZ-like isotype in zebrafish, which was designated as IgZ-2, adding a novel member to the immunoglobulin family in teleost fish. The IgZ-2 heavy chain encoding gene exhibited 76.5% nucleotide sequence identity to the previously reported IgZ. The putative IgZ-2 protein consists of four constant regions in its extracellular region, a transmembrane domain, and a short cytoplasmic tail. A number of conserved domains or residues such as the four Ig domains, cysteines required for Ig fold, hydrophobic and hydrophilic residues consistent with the CART domain, and Thr, Set, Tyr residues known to be essential for association with the B cell co-receptors CD79A/B, were identified. Phylogenetic analysis showed that IgZ-2 grouped with IgZ and other known teleost IgZ-Iike sequences. The IgZ-2 transcripts were widely expressed in immune-related tissues, and could be significantly up-regulated by in vivo stimulation with LIPS in various tissues including head kidney, spleen, intestine, skin, and gill. However, hardly IgZ-2 transcripts were detected during embryonic development until 2 weeks after fertilization. Double immunofluorescence staining showed that IgZ-2 and IgM were co-localized on B cell surfaces. Flow cytometric analysis showed the percentage of IgZ-2-positive cells could be dramatically up-regulated by interleukin-4 (IL-4), a cytokine known to activate the proliferation and differentiation of B cells. These observations indicated that the newly cloned IgZ-2 could be a novel B cell receptor. Our results will add new insights into the immunoglobulin class diversity of teleost fish, and also help us understand better the evolutionary history of adaptive immunity f
