Unveiling esophageal cancer treatment mechanisms: network pharmacology and molecular docking of Physcion

This study investigates the effects of Physcion on esophageal cancer and its possible mechanisms of action. Potential Physcion targets were identified using databases. Transcriptomic data from 17 esophageal cancer and adjacent tissues were analyzed to find differentially expressed genes, intersecting with potential targets to select 16 key genes. Their expression and distribution were evaluated in patient sequencing data. Diagnostic potential was assessed through differential gene expression and ROC curves. Pathway enrichment analysis was performed using KEGG, and molecular docking simulations were conducted to assess Physcion's binding affinity to key genes. In vitro assays complemented these findings. A total of 161 drug targets were identified, narrowing down to 16 pivotal genes. Expression patterns were examined across cell populations, and enrichment analysis showed significant PI3K/AKT pathway involvement. Molecular docking indicated strong binding of Physcion to HSP90AA1 and MMP2. In vitro assays confirmed Physcion's dose- and time-dependent impact on esophageal cancer cells, with significant DAPI staining effects. Physcion shows promise as a therapeutic agent for esophageal cancer. The study supports its potential for clinical development and future research in esophageal cancer treatment.

The FLNA Gene in Tumour-Educated Platelets Can Be Utilised to Identify High-Risk Populations for NSCLCs

Selective screening of the population based on NSCLC risk is an effective technique for minimising overdiagnosis and overtreatment. Platelets and the components can be used as liquid-biopsy markers, potentially assessing the risk of NSCLC, which are easily deployed in clinical applications. Platelet RNA sequencing datasets were analysed to identify specific genes derived from NSCLC patients and healthy donors. Then, expressions of the selected gene were validated in a clinical trial. Not only the availability of the specific gene in differentiating NSCLC patients from healthy donors but also from patients with benign nodules was estimated respectively. Finally, the values of the specific TEP-gene in metastasis and survival prognosis were also evaluated. FLNA was selected based on the GSE datasets, of which mRNA expression levels were higher in platelets from NSCLC patients than in healthy donors and also higher than in benign patients. To discriminate the malignant patients from the healthy individuals, FLNA got an AUC for the ROC curve of 0.716. When discriminating from the benign individuals, FLNA got an AUC of 0.705. In addition, an AUC of 0.595 was found when the metastatic group was distinguished from the non-metastatic group using the relative quantitative results of FLNA, and it seemed that the high-FLNA-expression group had a poorer long-term survival rate than the low-expression group. These findings suggested that high expression of FLNA in TEPs may indicate the incidence and metastasis of NSCLC and serve as a biomarker for high-risk estimation for NSCLC.

Digital Twin Construction and Resource Allocation on Internet of Vehicles

Aiming at the problems of data synchronization and resource limitation in smart driving on Internet of Vehicles (IoV), we investigate the construction of digital twins (DTs) and resource allocation on IoVs with the aim of achieving the optimal system performance. DT is a key technology in the future of autonomous driving. By digitizing physical components, it can effectively predict, analyze, and optimize various services of intelligent driving. First, we propose an accurate DT construction scheme (SL-ADTC) based on swarm learning (SL), which uses distributed learning algorithms to predict the queuing waiting time at relay nodes and the update frequency of DTs to solve data deviation and desynchronization for constructing DTs. Second, to address the problem of insufficient local computational resources of the task vehicles, a DT-based task division cooperative processing (DT-TDCP) mechanism is proposed. Then, considering the contradiction between task delay and system energy consumption, a joint optimization problem of computational offloading decision and resource allocation is proposed to minimize the total system cost under the premise of guaranteeing the high accuracy of DTs. Regarding the problem that the state space dimension is too high in reinforcement learning and the algorithm has difficulty in convergence, a DT-assisted multiagent classification proximal policy optimization (MACPPO) algorithm is proposed. Numerical results show that the proposed schemes outperform several benchmark schemes in terms of DTs construction accuracy, service latency, and system energy consumption.

Biochar doping of synthesized mordenite improves adsorption and oxidation in As(III) removal: Experiments and DFT calculations

In this study, a composite material was synthesized through the co-pyrolysis of biochar doped with synthetic mordenite. The adsorption experiments conducted with BC@ASM on As(III) facilitated the determination of the optimal mass ratio of 20:1 (ASM: Yak dung) and a pyrolysis temperature of 500 degrees C. The adsorption properties of ASM and BC@ASM were examined through batch adsorption experiments and a range of characterization techniques. And the reaction mechanism was further elucidated by DFT calculations, revealing the essential difference in the adsorption of As(III) by ASM and BC@ASM. The adsorption kinetics of As(III) were found to align with both the pseudo-second-order and Elovich kinetic models, while the isothermal adsorption was consistent with the Freundlich model. The maximum theoretical adsorption capacities were determined to be 371.9 mg/g and 449.6 mg/g, respectively. When the initial concentration of arsenite (As(III)) is 100 mg/L, the optimal dosage of synthetic mordenite is determined to be between 6 and 8 g/L, while the optimal dosage of the composite material ranges from 5 to 6 g/L. The composite material demonstrated significant resistance to fluctuations in pH. Within the pH range of 2-12, the removal efficiency is sustained between 78.3% and 88.7%. Furthermore, the adsorption capacity exhibited minimal sensitivity to the presence of anions such as chloride (Cl-), nitrate (NO3-), bicarbonate (HCO3-), and sulfate (SO42-) in the surrounding environment. In addition, BC@ASM facilitated the formation of arsenite-tannic acid complexes, which markedly improved its adsorption capacity for arsenite. In conclusion, the composite material presents a viable approach for addressing arsenic contamination in aquatic environments, while the foundational data offers a novel perspective for the remediation of metallic pollutants.

Low-light/low-power de-icing composite coatings with thermal insulation and tunable optical radiation absorption for all-weather anti-icing of wind turbine blades

Anti-icing/de-icing strategies that utilize sunlight to increase the external surface temperature of components have become a hot research topic to ensure the efficiency of wind turbine generators. Nevertheless, the photothermal effect is difficult to realize in low-light or even no-light environments. Additionally, the heat buildup caused by the photothermal effect during the daytime in the hot summer months accelerates the thermal aging process. To address these challenges, a composite anti-icing/de-icing coating with tunable solar radiation absorption and thermal insulation was designed. The high solar absorption ratio of the coating in cold environments allows for a rapid increase in surface temperature through the photothermal effect. Reciprocally, diminished solar absorption ratio through thermochromism in hot environments helps to prevent overheating of the coating. In addition, stable and controllable electrothermal assist capability enables all-weather anti-icing/ de-icing. There is also a porous heat-insulating structure obtained by evaporation-induced phase separation, which not only effectively hinders the heat transfer from water droplets to the substrate, but also facilitates trapping of heat accumulated by photothermal/electrothermal effects. E-PPCT with heat transfer barrier, intelligent temperature control, and photothermal/electrothermal all-weather de-icing functionality will be an excellent new material to overcome the challenges of icing and surface overheating on wind turbine blades.

Optimizing processing stability with ionized polymers in magnetorheological finishing fluid

Magnetorheological finishing (MRF) is being widely utilized in Semiconductor and optical processing. The properties of the finishing fluid have a great influence on the finishing quality. This study revealed that sodium polystyrene sulfonate (PSS), when employed as a dispersant in the finishing fluid, effectively enhances the polishing performance and maintains the processing stability of the finishing fluid. In the MRF processing, the group with the addition of 0.2 % PSS achieved a reduction in the roughness (Rq) of the fused silica (FS) workpiece to 1.02 nm compared to 4.3 nm in the control group. The PSS elevated the surface potential of abrasive particles, impeding abrasive agglomeration by reinforcing electrostatic repulsion. Simultaneously, the operation life of the finishing fluid significantly extends more than 3 times. The addition of PSS resulted in a significant decrease in the oxidation of Carbonyl iron particles (CIPs). Energy spectrum analysis revealed that diamond abrasives scratched the CIPs. Scratched CIPs were prone to be oxidized to Fe3+, thus leading to the significant failure of the finishing fluid. PSS adsorb to particle surfaces, creating steric hindrance that reduces friction between abrasives and CIPs. The MRF fluid with PSS exhibited a notable reduction of Fe3+ concentration. Both mechanisms concurrently enhanced the processing stability.

Battery lumped fractional-order hysteresis thermoelectric coupling model for state of charge estimation adaptive to time-varying core temperature conditions

As electric vehicles become more common, there is increasing concern regarding their battery reliability and safety. The estimation accuracy is strongly correlated with the performance of the battery model. The lumped fractional-order hysteresis thermoelectric coupling model (LFHTCM), considering temperature and hysteresis effects, is established in this paper. Firstly, the fractional-order hysteresis sub-model is established based on the Grunwald-Letnikov (G-L) fractional calculus principle and the recursive model of hysteresis voltage. Then, the thermal sub-model is established by integrating the Bernardi heating mechanism and the heat transfer model. The fractional-order sub-model provides voltage to the thermal sub-model for heat generation calculation. The thermal sub-model provides temperature to the fractional-order model for parameter correction. Finally, the fractional-order unscented Kalman filtering (FOUKF) algorithm was developed to estimate the state of charge (SOC) of the battery. Experimental results confirmed the effectiveness of the model, with its estimation enhancing the utilization, operational optimization, and calibration of batteries in practical applications. Establishment and application of novel transient cotyledon and seed transformation systems in Tartary buckwheat

With the complete sequencing of the Tartary buckwheat (TB) genome, Studies are increasingly focusing on genes related to flavonoid metabolism and environmental adaptability. However, the absence of a mature TB genetic transformation system has limited studies on TB at the molecular level. There is a pressing need to establish effective transient expression systems for TB. This study utilized TB cotyledons and seeds as explants, and optimized transformation conditions by applying GUS driven by the CaMV35S promoter as a reporter for the transformation efficiency. Results indicated high transformation efficiency under specific conditions: an OD600 of Agrobacterium at 0.9 with 3 days of co-cultivation for cotyledons, and an OD600 of Agrobacterium at 1.5 with 2-3 days of co-cultivation during the middle filling stage for seeds. Furthermore, this system was used to demonstrate significant activation of the FtFLS1 promoter by environmental stresses such as cold, UV-B and drought stress and a significant response of the FtRDE2 promoter to cold stress, validating its utility for promoter analysis. Additionally, the system facilitated not only subcellular localization of characterized proteins including FtMYB6 and FtANX7, but also functional validation of key genes like FtMYB6 and FtAGPS1. Conclusively, the established TB cotyledon and seed transient expression systems provide robust tools for studying promoter activity, subcellular localization, and gene function, offering new and effective means for advancing TB molecular biology research.

Chitosan hydrogel microspheres loaded with Bacillus subtilis promote plant growth and reduce chromium uptake

Cr contamination can lead to reduced crop yields and threaten food security. Eliminating soil Cr contamination or improving crop resistance to Cr is challenging in terms of costly, environment and biodiversity risk. Here, we used chitosan hydrogel microspheres loaded with Bacillus subtilis to cope with plant stress caused by Cr contamination. The addition of chitosan hydrogel microspheres loaded with Bacillus subtilis increased shoot biomass by 88 % and decreased the plant Cr concentration by 17 %. The bacterial chitosan hydrogel microsphere treatment enhanced the decomposition of organic matter and facilitated the uptake of nutrients by the plants. It also significantly increased the abundance of anti-heavy metal stress functional bacteria (Proteobacteria, Actinobacteria, and Chloroflexi), strongly promoting interactions and correlations between microbes. This technology of bacterial-chitosan hydrogel microspheres presents promising opportunities for sustainable strategies for addressing heavy metal pollution and enhancing agricultural productivity.

Online Data-Driven Diagnosis for Common Electrical and Sensor Faults in Dual Three-Phase PMSM Drives

This article proposes a data-driven online diagnosis method for detecting common electrical faults and current sensor faults (CSFs) in dual three-phase permanent-magnet synchronous motor (DTP-PMSM) drives. The proposed diagnosis method fulfills feature extraction by obtaining the average values of phase currents in the positive and negative half-cycles of fault features to reduce dimensionality. By designing a one-vote veto classification rule, the proposed diagnosis method can accurately distinguish 24 fault modes and effectively prevent misdiagnosis. The classification model consists of only 24 decision functions, making it easy to implement on the mainstream industrial microcontrollers. The experiments are carried out to verify the validity of the proposed method.
