HS-SPME-GC-MS combined with machine learning methods for screening volatile quality indicators in Hypericum perforatum L.

Hypericum perforatum L. (HPL), a natural product with high medicinal value, exhibits diverse bioactivities. The efficacy of H. perforatum varies with different parts of the plant, and the content of active ingredients also varies significantly. Elucidation of the volatile compounds in different medicinal parts of HPL and screening of suitable active compounds as indicators for quality control are essential for its quality improvement. In this study, HS-SPME-GC-MS was used to characterize the volatile compounds in H. perforatum collected from Xinjiang, China. Subsequently, an OPLS-DA model was established to visualize differences among three distinct parts of H. perforatum. Then, network pharmacology was used to analyze the pharmacological activity of the identified differential metabolites. Finally, three classifiers (support vector machine, random forest, and K-nearest neighbor) were used to assess the qualification of the identified quality markers. A total of 159 volatile compounds were identified by combining the MS database and retention indices for rigorous evaluation to remove redundant information. And 67 differential metabolites were screened by the OPLS-DA model. Furthermore, network pharmacological analysis revealed that 48 compounds were associated with 1159 target genes. Among these, 18 highly active compounds were selected as potential markers. All three classifiers demonstrated good performance across different variables. Ultimately, eight compounds were selected as markers for laboratory quality control of H. perforatum.

Performance study of semi-active seat suspension added by quasi-zero stiffness structure under various vibratory roller models

To enhance the vibratory roller's comfort, three different types of seat suspension including the seat's semi-active suspension system (SASS) controlled by the FLC-PID controller, seat's passive suspension system (PSS) added by the quasi-zero stiffness structure (QZSS), and seat's SASS added by the QZSS are proposed. Three different vehicle dynamic models including a one-dimensional (1D) model, two-dimensional (2D) model, and three-dimensional (3D) model are established to evaluate the performance and stability of the seat's suspension systems under different operating conditions. The root mean square (RMS) acceleration and power spectral density (PSD) acceleration of the seat have been selected as the evaluation indexes. The mathematical model and numerical simulation method are then tested via the experiment to enhance the reliability of the research results. The research result shows that the seat's vibration characteristic using 1D, 2D, and 3D models is the same. However, the seat's RMS acceleration and maximum PSD acceleration using the 1D model are increased by 30.6% and 61.7% in comparison with the 3D model, whereas the seat's acceleration and PSD acceleration using the 3D model are like the measured results. Thus, the use of the vehicle model greatly influences the research result. By adding the QZSS into the seat's SASS, the seat's RMS and maximum PSD accelerations are greatly reduced in comparison with all seat's PSS, seat's SASS without the QZSS, and seat's PSS added by QZSS. Therefore, the seat's SASS added by the QZSS should be applied to further enhance the vibratory roller's comfort.

A nomogram for predicting ischaemic muscle sequelae after revascularization in patients with traumatic femoral-popliteal artery injuries: a retrospective cohort study

PurposeThis study aimed to investigate the incidence and associated risk factors of ischaemic muscle sequelae in patients with traumatic femoropopliteal artery injuries following revascularization, as well as to develop a nomogram to predict the risk of ischaemic muscle *** from patients with acute traumatic femoropopliteal artery injuries between January 2008 and December 2022 were collected. All patients with successful limb salvage were divided into two groups based on the occurrence of ischaemic muscle sequelae: the ischemic muscle sequelae group (IG) and the non-ischaemic muscle sequelae group (NG). Univariate and multivariate logistic regression analyses were used to identify potential predictive factors associated with ischaemic muscle sequelae. A predictive nomogram was constructed and internally *** the 102 patients, 30 cases (29.41%) developed ischaemic muscle sequelae. Independent predictors of ischaemic muscle sequelae were identified as crush injury, HCT, and CKMB. A nomogram was constructed based on these three parameters. The area under the receiver operating characteristic (ROC) curve of the predictive model was 0.894, indicating excellent discrimination. The calibration curve demonstrated a high degree of consistency between the predicted probabilities and the observed outcomes. Additionally, the decision curve analysis (DCA) showed that the nomogram model had good predictive *** study demonstrated that crush injury, HCT, and CKMB were independent predictors of ischaemic muscle sequelae in patients with acute traumatic femoropopliteal artery injuries following revascularization. The nomogram integrating clinical factors and blood markers can assist physicians in conveniently predicting the risk of ischaemic muscle sequelae in patients.

To address the issue of final delivery route planning in the community group purchase model, this study takes into full consideration logistics vehicles of different energy types. With the goal of minimizing the sum of vehicle operating costs, delivery timeliness costs, goods loss costs, and carbon emissions costs, a multi-objective optimization model for community group purchase final delivery route planning is constructed. An improved genetic algorithm with a hill-climbing algorithm is utilized to enhance adaptive genetic operators, preventing the algorithm from getting stuck in local optima and improving the solution efficiency. Finally, a case study simulation is conducted to validate the feasibility of the model and algorithm. Experimental results indicate that currently, among the three types of vehicles, fuel logistics vehicles still have an advantage in terms of vehicle usage cost. Electric logistics vehicles exhibit the poorest performance with the highest cost per hundred kilometers, but their sole advantage lies in their high energy release efficiency, enabling optimal low-carbon vehicle performance. Battery-swapping logistics vehicles perform the best in terms of carbon emissions, combining the advantages of both fuel-based and electric logistics vehicles. Therefore, battery-swapping logistics vehicles are a favorable choice for replacing fuel-based logistics vehicles in the future, offering promising prospects for future development. © 2025 – IOS Press. Background: Associations between exposure to single metals and cognitive impairment or related outcomes have been reported in many previous studies. However, co-exposure to more than one metal is common situation. In recent years, studies on the effects of exposure to multiple metals on cognitive impairment or related outcomes have increased, but remain very limited, with a focus on populations with occupational exposure to metals, children, and adolescents. The potential relationships between exposure to metal mixtures and risk of cognitive impairment in adults remain to be clarified. Objective: To determine the associations between blood metal mixtures and cognitive impairment risk. Methods: Inductively coupled plasma mass spectrometry (ICP-MS) was utilized to detect the blood levels of lead (Pb), iron (Fe), copper (Cu), calcium (Ca), magnesium (Mg), and zinc (Zn). Multivariable logistic regression and Bayesian kernel machine regression (BKMR) models were employed to assess the relationships of exposure to these blood metal mixtures with the risk of cognitive impairment. Results: It was found that four metals (Pb, Fe, Cu, and Mg) were positively correlated with cognitive impairment in each single metal model. The association of Pb and Cu remained significant after adjusting for these six metals, with the odds ratios (95% confidence intervals) in the highest quartiles of 9.51 (4.41-20.54, p-trend <0.01) and 4.87 (2.17-10.95, p-trend <0.01), respectively. The BKMR models indicated that co-exposure levels of Ca, Cu, Fe, Mg, Pb, and Zn were related to increased risk of cognitive impairment at >= 25th percentile compared with median, and Pb and Cu mainly contributed to the joint effect. In addition, the interaction effects of Mg and Pb/Pb and Cu on the risk of cognitive impairment were observed. Significance: Co-exposure of six metals (Pb, Fe, Cu, Ca, Mg, and Zn) increased the risk of cognitive impairment in Chinese adults, with Pb and Cu likely to have greater impact.

CD97 inhibits osteoclast differentiation via Rap1a/ERK pathway under compression

Acceleration of tooth movement during orthodontic treatment is challenging, with osteoclast-mediated bone resorption on the compressive side being the rate-limiting step. Recent studies have demonstrated that mechanoreceptors on the surface of monocytes/macrophages, especially adhesion G protein-coupled receptors (aGPCRs), play important roles in force ***, its role in the regulation of osteoclast differentiation remains unclear. Herein, through single-cell analysis, we revealed that CD97, a novel mechanosensitive aGPCR, was expressed in macrophages. Compression upregulated CD97 expression and inhibited osteoclast differentiation;while knockdown of CD97 partially rescued osteoclast differentiation. It suggests that CD97 may be an important mechanosensitive receptor during osteoclast differentiation. RNA sequencing analysis showed that the Rap1a/ERK signalling pathway mediates the effects of CD97 on osteoclast differentiation under compression. Consistently, we clarified that administration of the Rap1a inhibitor GGTI298 increased osteoclast activity, thereby accelerating tooth movement. In conclusion,our results indicate that CD97 suppresses osteoclast differentiation through the Rap1a/ERK signalling pathway under orthodontic compressive force.

A time series context self-supervised learning for soft measurement of the f-CaO content

The content of free calcium oxide (f-CaO) in cement clinker is an important index for cement quality. In the production of cement clinker, the number of unlabeled samples is excessive and there is an interplay between the variables in time. A time series context self-supervised learning (TS-CSSL) is proposed. This method constructs pretextual tasks based on the temporal relationships between different variables from a large amount of unlabeled time series data. Considering the process of cement production and the residence time of materials in each piece of equipment, the method designs the segmentation of periods for different variables in the context-based self-supervised pretextual task. On this basis, a soft sensor for f-CaO content was implemented. After the experimental validation, the evaluation metrics root means square errors ( RMSE of the TS-CSSL model decreased by 2.41% and R2 improved by 10.93% compared to the random initialization model. Compared to the model with 8 sets of temporal relationships, the TS-CSSL model showed a decrease in RMSE of 5.33% and an increase in R2 of 21.9%. The experimental results demonstrate that the feature representation learned by the model can be used in a CNN framework and the effectiveness of the proposed self-supervised assistance task.

Clinical value of predicting relapse within 3 months in alcohol-dependent patients using fNIRS in verbal fluency task

To investigate the hemodynamic differences in various brain regions between alcohol dependence (AlcD) patients and healthy controls during a verbal fluency task (VFT) using functional near-infrared spectroscopy (fNIRS), and to further explore the clinical predictive value of fNIRS before therapy for the outcome of relapse in AlcD patients after 3 months. A retrospective survey was conducted on 123 AlcD patients and 149 healthy controls during the same period. Baseline assessment of fNIRS was performed to analyze the hemodynamic differences between the two groups in different brain regions. During hospitalization, AlcD patients underwent a 3-week benzodiazepine substitution therapy, gradually tapering off the medication to achieve alcohol withdrawal treatment goals. Three months after discharge, we conducted follow-up phone calls to assess the relapse status of the patients. Compared to the control group, the AlcD group had significantly lower integral values in the frontal and bilateral temporal lobes, as well as lower beta-values in all channels of the frontal lobe except for Ch13, and in all channels of the bilateral temporal lobes (p < 0.005), with no significant difference in the parietal lobe channel(p > 0.05). ROC (Receiver Operating Characteristic Curve) analysis for predicting relapse within 3 months showed that the area under the curve for all channels was highest (0.951, sensitivity 0.924, specificity 0.886). Patients with AlcD exhibit functional impairments in the frontal and temporal lobes. fNIRS channels in the frontal and parietal lobes based on VFT have good clinical predictive value for relapse within 3 months after pharmacotherapy in AlcD and can be applied in clinical practice.

The rapid development of edge computing and the Industrial Internet of Things have facilitated near real-time optimization of compute-intensive industrial tasks. Mobile edge computing (MEC) and device-to-device (D2D) offloading are promising ways to achieve near-real-time optimization. In this article, We propose a D2D-assisted MEC computing offloading framework by using deep reinforcement Learning (DRL) with entropy and attention mechanism (DMOEA). DMOEA considers interactions among related entities, including horizontal device-to-device collaboration and vertical device-to-edge offloading. Then, a DRL-based model with multi-actor single-critic structure is designed to solve the offloading strategy. In addition, to further improve efficiency, an attention mechanism is introduced to adapt dynamic changes in network and enhance the exploration ability. The experimental results show that the proposed framework can obtain a fast convergence rate and small oscillation amplitude and also can effectively reduce latency.
