The aim of this study is to investigate some utility tunnel fire laws based on theoretical and experimental analysis of ceiling temperature distribution. Comparing to the previous model with empirical parameters, a theoretical model is developed to include some physical parameters by using dimensional analysis method, which can fit complex tunnel fire scenes. Full-scale tunnel fire experiments with different ventilation speeds and fire sources were conducted to study the temperature distribution laws of tunnel fires and support the model. The results show that increasement of ventilation speed can reduce fire severity under the certain condition. Finally, according to compare the corresponding experimental data as well as the results obtained from the other known models, the excellent prediction ability and validity of the model are verified. The model can provide a useful method to investigate and predict the ceiling temperature distribution in utility tunnel fires, which can also describe the relationship between the maximum ceiling smoke temperature rise, heat relation rate and ventilation speed.

Metabolic flux, or the rate of metabolic reactions, is one of the most fundamental metrics describing the status of metabolism in living organisms. However, measuring fluxes across the entire metabolic network remains nearly impossible, especially in multicellular organisms. Computational methods based on flux balance analysis have been used with genome-scale metabolic network models to predict network-level flux wiring1, 2, 3, 4, 5-6. However, such approaches have limited power because of the lack of experimental constraints. Here, we introduce a strategy that infers whole-animal metabolic flux wiring from transcriptional phenotypes in the nematode Caenorhabditis elegans. Using a large-scale Worm Perturb-Seq (WPS) dataset forroughly 900 metabolic genes7, we show that the transcriptional response to metabolic gene perturbations can be integrated with the metabolic network model to infer a highly constrained, semi-quantitative flux distribution. We discover several features of adult C. elegans metabolism, including cyclic flux through the pentose phosphate pathway, lack of de novo purine synthesis flux and the primary use of amino acids and bacterial rNA as a tricarboxylic acid cycle carbon source, all of which we validate by stable isotope tracing. Our strategy for inferring metabolic wiring based on transcriptional phenotypes should be applicable to a variety of systems, including human cells.

Chiral 1-tetrahydronaphthamides are the core structures of many bioactive molecules, yet their efficient asymmetric synthesis from a simple feedstock remains a challenge. Herein, we present a one-pot synthesis strategy that combines electrochemical reduction andruthenium-catalyzed asymmetric hydrogenation to achieve the enantioselective reduction of 1-naphthalenamides to chiral 1-tetrahydronaphthamides. The protocol provides a practical platform for selectively constructing high-value chiral tetrahydronaphthenes from readily available naphthalene feedstock, thereby expanding the scope of asymmetric hydrogenation. The synthetic utility of this protocol is furtherdemonstrated through the synthesis of bioactive molecules.

This paper presents a novel optical performance monitoring (OPM) scheme based on reservoir computing (rC) andresnet network for monitoring the optical signal-to-noise ratio (OSNr) of few-mode transmission channels, without the need for any demodulation process. By using 300 rC nodes, the number of floating-point operations (FLOPs) is reduced by 75%, with almost no change in prediction accuracy compared to a network without rC. In a system ranging from 0 to 30 dB, 40 simulation runs yield an OSNr prediction accuracy band around 0.9900. We also investigate the prediction accuracy when using different spectral information, with results showing that frequency-domain information effectively captures the characteristics of both signal and noise. Additionally, we examine the impact of different mode combinations on OSNr prediction accuracy and find that the prediction performance is nearly unaffected by the mode combination.

PurposeVarious ternary components, such as lactose fines and magnesium stearate (MgSt), are frequently incorporated into carrier-baseddry powder inhalations (dPIs) to enhance the fine particle fraction (FPF). However, the specific effects of a combination of lactose fines and MgSt on the performance of dPI formulations have not yet been *** salbutamol sulfate (SS), which has a weak drug-carrier adhesion, as a model drug, this study investigated the impact of lactose fines content on the powder properties and aerosolization performance of 0.5% MgSt containing dPI formulations. The influence of the mixing order of MgSt and lactose fines on dPI formulation performance was also examined. Finally, an attempt was made to predict the FPF using powderrheological *** addition of 1-10% lactose fines to 0.5% MgSt containing SS-dPI formulations significantly improved the FPF, but the addition of MgSt had a negative impact on FPF according to the mixing order of the carrier-MgSt-lactose fines-drug. The mixing order of the MgSt and lactose fines significantly affected the deposition of SS. The FPF of the formulation mixed with 5% lactose fines followed by 0.5% MgSt was significantly higher (46.55%) than that of the formulation mixed with 0.5% MgSt followed by 5% lactose fines (33.86%) and the formulation with only 5% lactose fines (39.53%). Good flowability (24.44%) and strong adhesion (75.56%) were the key factors affecting the in vitro drug deposition in this *** the development of dPIs, ternary components should be selectively added in a specific mixing order based on drug-carrier adhesion to achieve optimal lung deposition.

Neuroinflammation plays a vital role in neural development as well as the pathogenesis and progression of neurodegenerative diseases. It is recognized as a promising therapeutic target for neurological disorders. Although multiple anti-inflammatory drugs have been examined for their therapeutic potential in neurodegenerative diseases, the development of new model systems to evaluate the safety and efficiency of anti-inflammatory agents is still greatly needed. Here, we presented a human cortical organoid-on-a-chip for modeling neuroinflammation and exploring the neuroprotective activity of cerium oxide (CeO2) nanozyme, a novel antioxidant, on neuroinflammation-induced neurotoxicity. hiPSC-derived cortical organoids, resembling the key characteristics of early human brain development, were generated on micropillar arrays. We showed that lipopolysaccharide (LPS) treatment induced inflammatory responses and neurological impairments in cortical organoids. Notably, the adverse responses caused by LPS administration could be effectively attenuated by treatment with nanozyme at a proper concentration, indicating that the CeO2 nanozyme exerts anti-neuroinflammatory and neuroprotective effects and could serve as a potential candidate to alleviate neuroinflammation-associateddiseases. Together, the human cortical organoid-on-a-chip model presents a biomimetic system to investigate the complicated impacts of neuroinflammation on neural development and exhibits high potential in neurological study anddrug evaluation.

In bioinformation and medicine, an enormous amount of high-dimensional multi-population data is collected. For the inference of several-samples mean problem, traditional tests do not perform well and many new theories mainly focus on normal distribution and low correlation assumptions. Motivated by the weighted sign test, we propose two projection-based tests which are robust against the choice of correlation matrix. One test utilizes Scheffe's transformation to generate a group of new samples andderives the optimal projection direction. The other test is adaptive to projection direction and is generalized to the assumption of the whole elliptical distribution and independent component model. Further the theoretical properties are deduced and numerical experiments are carried out to examine the finite sample performance. They show that our tests outperform others under certain circumstances.
