SOME EFFERENT CONNECTIONS OF THE ROSTRAL HYPOTHALAMUS IN THE SQUIRREL-MONKEY (SAIMIRI-SCIUREUS) AND CAT

The efferent connections of the ventromedial nucleus and of the anterior hypothalamic area were studied autoradiographically in the squirrel monkey and cat, and some observations were also made on the efferent connections of the periventricular, paraventricular and accessory supraoptic nuclei. As in the rat, the ventromedial nucleus was found in the monkey and cat, to have rather widespread ascending and decending projections. The ascending projections follow 3 routes: periventricularly to the periventricular nuclei of the hypothalamus and preoptic area, and to the anterior paraventricular nucleus of the thalamus;through the medial hypothalamus and preoptic area to the adjoining nuclei and through the medial forebrain bundle to the lateral hypothalamic and preoptic areas, to the medial part of the bed nucleus of the stria terminalis, and the ventral part of the lateral septal nucleus. Descending projections from the nucleus also follow 3 main pathways: in the periventricular system as far caudally as the locus coeruleus;through the medial forebrain bundle to the fibrous capsule of the mammillary body, the supramammillary region, the ventral tegmental area and the central tegmental fields;and through the ventral supraoptic commissure to the basal nucleus of Meynert, to the central tegmental fields, and to the peripeduncular nucleus of both sides. As shown elsewhere (Saper et al., 1978) the anterior hypothalamic area projects primarily upon the surrounding hypothalamic and preoptic cell groups, including the arcuate, the periventricular, the dorsomedial and ventromedial nuclei, and the dorsal premammillary region bilaterally. The paraventricular and accessory supraoptic nuclei both contribute fibers to the posterior lobe of the pituitary by way of the paraventriculohypophysial tract which passes through the external zone of the median eminence in the monkey and through its internal lamina, in the cat. ROSTRAL PROJECTIONS FROM HYPOTHALAMIC ARCUATE NUCLEUS

The response of neurones in the medial basal hypothalamus to electrical stimulation in the suprachiasmatic/preoptic region has been studied in rats anaesthetised with urethane. Neurones in the hypothalamic arcuate, periventricular, ventromedial and dorsal premammillary nuclei were antidromically activated by the stimulation showing that such cells send processes in rostral direction outside the median eminence. Tuberoinfundibular and rostrally projecting units were found interspersed in the "hypophysiotrophic" area.

Brain sites of action of endogenous interleukin-1 in the febrile response to localized inflammation in the rat

1. Interleukin (IL)-1 is a potent endogenous pyrogen which causes fever when injected into a number of brain sites. However, the brain sites at which endogenous IL-1 acts to influence body temperature remain equivocal. The aim of this study was to determine the effect of local administration of the interleukin-1 receptor antagonist (IL-1ra) into specific sites in the hypothalamus, and other brain regions known to contain receptors for IL-1, on the febrile response of rats to peripheral injection of lipopolysaccharide (LPS) into a subcutaneous air pouch (intrapouch, I.P.O.) that does not lead to LPS appearance in the circulation. 2. Injection of LPS (100 mu g kg(-1), I.P.O.) induced a rise in body temperature which commenced 1.5 h after injection and was maximal at 3 h (38.9 +/- 0.2 degrees C, compared with 37.0 +/- 0.1 degrees C at 0 h, n = 6, P < 0.001). Intracerebrorventricular (I.C.V.) IL-1ra (500 mu g in 5 mu l) significantly attenuated LPS fever (IL-1ra, 37.7 +/- 0.2 degrees C;saline, 38.9 +/- 0.2 degrees C;n = 6, P < 0.001). Unilateral microinjection of IL-1ra, (50 mu g in 0.5 mu l at 0 + 1 h) into the anterior hypothalamus (AH), paraventricular hypothalamic nucleus (PVH), peri-subfornical organ, subfornical organ (SPO) or hippocampus (dentate gyms and cA3 region) also significantly reduced the fever induced by LPS. 3. The same dose of IL-1ra had no effect on fever when administered into the ventromedial hypothalamus (VMH), organum vasculosum lamina terminalis (OVLT), CA1 field of the hippocampus, striatum or cortex. 4. These data indicate that the action of endogenous IL-1, in the brain during fever is site specific, acting at the AH, PVH, SFO and hippocampus, but not the VMH, OVLT and striatum or cortex.

AFFERENT AND EFFERENT SUB-CORTICAL PROJECTIONS OF BEHAVIORALLY DEFINED SECTORS OF PREFRONTAL GRANULAR CORTEX

Although the functional significance of the midprincipalis region is well known, the afferent and efferent connections of this zone, in comparison tot the anterior and posterior portions of the cortex lining the principal sulcus, are poorly understood. In 3 rhesus monkeys the retrograde tracer HRP [horseradish peroxidase] and the anterograde tracers, 3H- proline, lysine and leucine, were injected into the sulcal cortex lining the principal sulcus. The cortex forming the banks of the principal sulcus was divided into anterior, middle and posterior sectors with 1 animal used for each zone. As expected from previous studies, the heaviest afferents to the cortex forming the principal sulcus were from the parvocellular portions of the medial dorsal nucleus. The medial pulvinar nucleus and the nucleus limitans projected to only the anterior and posterior portions of the cortex lining the principal sulcus. Projections were seen to all 3 sectors from the anterior, midline, intralaminar and lateral thalamic nuclei. Although cells were seen in the hypothalamus following injections in all 3 sectors of the cortex lining the principal sulcus, the heaviest hypothalamic projections were noted after injections into the mid-sector of the cortex. These HRP-positive cells were in the dorsal and lateral hypothalamic area, dorsal medial nucleus and in the lateral mammillary nucleus. These findings link the midprincipalis region with the prefrontolimbic circuit, and suggest that the midprincipalis region, n. [nucleus] medialis dorsalis, the mammillary bodies and perhaps the cingulate gyrus constitute part of an anatomical circuit concerned with memory processes.

COBALT IONTOPHORESIS TECHNIQUES FOR TRACING AFFERENT AND EFFERENT CONNECTIONS IN VERTEBRATE CNS

In recent years several dye and cobalt iontophoresis techniques have been successfully used by invertebrate neurophysiologists for the localization of neuron somata and their processes. The cobalt iontophoresis technique has now been extended for use in the tracing of nerve fiber pathways and the localization of neuron somata in vertebrates. The brain and spinal cord of an animal are removed following perfusion with saline, and placed in a dish of cold saline. A suction electrode, filled with 300 mM cobalt chloride, is then placed over the cut end of the nerve trunk. Cobalt ions are then iontophoresed (by means of a voltage divider) within the nerve fibers, along their course. Following iontophoresis, the brain is bathed in an ammonium sulfide solution to precipitate the cobalt as black cobalt sulfide. The brain is then processed for histological procedures. A wide variety of vertebrates has been used, including amphibians, reptiles, aves and mammals, with uniform success. The cobalt iontophoresis technique presently in use has a wide range of applicability for neuroanatomical studies.

The dorsomedial hypothalamic nucleus harbors leptin sensitive neurons and is intrinsically connected to hypothalamic nuclei involved in feeding behavior. However, It also receives ascending input from the visceroceptive neurons of the brainstem. We have identified a unique glucagon-like-peptide-2 containing neuronal pathway connecting the nucleus of the solitary tract with the dorsomedial hypothalamic nucleus. A glucagon-like-peptide-2 fiber plexus targets neurons expressing its receptor within the dorsomedial hypothalamic nucleus. Pharmacological and behavioral studies confirmed that glucagon-like-peptide-2 signaling is a specific transmitter inhibiting rodent feeding behavior and with potential long-term effects on body weight homeostasis. The glucagon-like-peptide-1 receptor antagonist, Exendin (9-39) is also a functional antagonist of centrally applied glucagon-like-peptide-2.

The organization of feline entopeduncular nucleus projections: Anatomical studies

The organization of entopeduncular nucleus (EPN) projections was studied in cats using autoradiographic and horseradish peroxidase (HRP) techniques. In autoradiographic studies, EPN axons were found to terminate in a J‐shaped region in the dorsal and medial part of the ventral anterior nucleus (VA) and the rostral portion of the adjacent ventral lateral nucleus (VL). EPN axons also terminated in the rostral portion of the centrum medianum (CM), the ventrolateral portion of the lateral habenular nucleus (LHB), and the pedunculopontine nucleus (PP). The VA included the largest terminal field although the LHB had the greatest density of terminals. Regardless of the region of EPN into which amino acids were injected, the terminal fields were the same: there was no localization within the EPN of the cells projecting to one region. HRP‐containing cells were distributed throughout the EPN following injections into the VA, LHB, or PP, although many more cells were labeled following injections into either VA or LHB than PP. EPN cells containing HRP following injections into either VA or LHB were not morphologically different from those not containing HRP in the same respective animals. Following HRP injections into stria medullaris, only cells in the rostral part of the EPN were labeled, providing evidence that rostrally and caudally located EPN neurons have different paths to LHB. Although there may be a rostrocaudal organization of pathways to LHB, individual regions of the nucleus project to the same areas. Copyright © 1979 The Wistar Institute Press
