Novel peptides that inhibit the propagation of Newcastle disease virus

A disulfide constrained random heptapeptide library displayed on filamentous bacteriophage M13 was applied to select specific ligands that interact with Newcastle disease virus (NDV). A fusion phage carrying the amino acid sequence TLTTKLY was selected from the panning procedure. An antibody competition assay showed that the selected phage was capable of competing with the polyclonal antibodies raised against NDV for binding sites on the virus. Determination of the binding affinity of this phage with NDV by an equilibrium binding assay in solution revealed two different dissociation constants, suggesting that there could be two distinct binding sites for the phage on NDV. Synthetic peptides with the sequence CTLTTKLYC, either in linear or cyclic conformations inhibited the binding of phage bearing the same sequence to NDV. These peptides also inhibited the hemolytic activity of the virus as well as its propagation in embryonated chicken eggs.

Kasumigamide, an antialgal peptide from the cyanobacterium Microcystis aeruginosa

Kasumigamide (1), a novel antialgal tetrapeptide containing an N-terminal alpha-hydroxy acid, was isolated from the freshwater cyanobacterium Microcystis aeruginosa (NIES-87). Its structure was elucidated by two-dimensional H-1-H-1 and H-1-C-13 NMR correlation experiments and confirmed by mass spectral and amino acid analyses. The absolute stereochemistry of 1 was determined by chemical studies. This peptide showed an antialgal activity against the green alga Chlamydomonas neglecta (NIES-439).

Preparation of indium-115-labeled diethylenetriaminetetraacetic acid monoacetamide peptides purified by 8-hydroxyquinoline

In this communication we describe a novel procedure for the preparation and purification of diethylenetriaminepentaacetic (DTPA)-acylated and (115)In(3+)- labeled oligopeptides using 8-hydroxyquinoline for the removal and quantification of nonbound indium ions. First the N(alpha)- or N(alpha),N(epsilon)-DTPA oligopeptides containing C-terminal KDEL signal motif were produced by solid-phase synthesis. For this the free carboxyl group of DTPA dianhydride was activated in situ for a short period of time yielding a major product. Reversed-phase HPLC-purified DTPA oligopeptides were labeled with (115)In(3+) in aqueous buffer solution at pH 3.8. For the removal as well as for the detection of uncoordinated (115)In(3+) ions we have utilized the (115)In(3+) complex-forming ability of 8-hydroxyquinoline in chloroform. Following an optimized extraction procedure the free indium ion content was measured by spectrophotometry in the organic phase. Data obtained by this method and verified by total-reflection X-ray fluorescence spectroscopy and thin-layer chromatography demonstrated that free (115)In(3+) could be efficiently removed and sensitively detected in the presence of DTPA oligopeptide chelator. No release of (115)In(3+) from its DTPA complex was observed. This method could be useful for the preparation of indium complexes of peptides and perhaps proteins containing a DTPA moiety and nonradioactive isotope ligand. (C) 2000 Academic Press.

Separation of isomeric peptides using electrospray ionization/high-resolution ion mobility spectrometry

In this paper, the first examples of baseline separation of isomeric macromolecules by electrospray ionization/ion mobility spectrometry (ESI/IMS) at atmospheric pressure are presented. The behavior of a number of different isomeric peptides in the IMS was investigated using nitrogen as a drift gas. The IMS was coupled to a quadrupole mass spectrometer, which was used for identification and selective detection of the electrosprayed ions. The mobility data were used to determine their average collision cross sections. The gas-phase ions of isomeric peptides were found to have different collision cross sections. In all cases, doubly charged ions exhibited significantly (8-20%) larger collision cross sections than the respective singly charged species. The analysis of mixtures of the isomeric peptides clearly demonstrated the capability of IMS to separate gas-phase peptide ions due to small differences in their conformational structures, which cannot be determined by mass spectrometry, An actual resolving power of 80 was achieved for two doubly charged reversed sequenced pentapeptides, Baseline separation was provided for ions differing by only 2.5% in their measured collision cross sections;partial separation was shown for isomeric ions exhibiting differences as small as 1.1%.

An endogenous pentapeptide acting as a sodium channel blocker in inflammatory autoimmune disorders of the central nervous system

Reversible blockade of sodium channels by endogenous substances has been claimed to account for the fast exacerbations and relapses commonly seen in demyelinating autoimmune diseases(1-4). Evidence has been provided that in the cerebrospinal fluid of patients with multiple sclerosis(5) or Guillain-Barre syndrome(6), a sodium-channel-blocking factor exists that has properties of local anesthetic agents(7,8). This factor could contribute to the nerve conduction block and paresis seen in these disorders(9-11). We describe here a previously unknown endogenous substance in human cerebrospinal fluid with distinct channel-blocking properties even at very tow (0.00001 M) concentrations. The pentapeptide with the sequence Gln-Tyr-Asn-Ala-Asp exerted its blocking action by shifting the steady-state inactivation curve of the sodium channels to more-negative potentials, as most local anesthetics do. In the cerebrospinal fluid of healthy individuals, its concentration was about 3 mu M, whereas in patients with multiple sclerosis and Guillain-Barre syndrome, it increased 300-1,400%. At these concentrations, the peptide's blocking efficacy was higher than that of 50 mu M lidocaine. At a concentration of 10 mu M, lidocaine is able to 'unmask' subclinical lesions in multiple sclerosis(12);thus, the endogenous pentapeptide may well contribute to the fast changes of symptoms. Furthermore, it may become valuable as a marker of disease activity.

EFFECT OF DIRECT CONTROL OF ELECTROOSMOSIS ON PEPTIDE AND PROTEIN SEPARATIONS IN CAPILLARY ELECTROPHORESIS

The separations of peptide and protein mixtures in capillary zone electrophoresis (CZE) at various solution conditions were studied with the direct control of electroosmosis. The zeta-potential at the aqueous/capillary interface and the resulted electroosmosis in the presence of an electric field were directly controlled by using an additional electric field applied from outside of the capillary. The controlled electroosmotic flow affected the migration time arid zone resolution of peptide and protein mixtures. The changes in the magnitude and polarity of the zeta-potential caused the various degrees of peptide and protein adsorption onto the capillary through the electrostatic interactions. The separation efficiencies of peptide and protein mixtures were enhanced due to the reduction In peptide and protein adsorption at the capillary wall. The direct manipulations of the separation efficiency and resolution of peptide and protein mixtures in CZE were demonstrated by simply controlling the zeta-potential and the electroosmotic flow with the application of an external electric field.

A substance that competes for opiate receptor binding was isolated from calf brain and identified as 2 pentapeptides, H-Tyr-Gly-Gly-Phe-Met-OH (methionine enkephalin) and H-Tyr-Gly-Gly-Phe-Leu-OH (leucine enkephalin), with about 4 times more leucine enkephalin than methionine enkephalin. Na and Mg effects on opiate receptor interactions show that both peptides are agonists, whereas leucine enkephalin may be a purer agonist than methionine enkephalin.

DIFFERENTIATION OF FACTOR C-LHIH AND SYNTHETIC CONTRACEPTIVE POLYPEPTIDE, H-THR-PRO-ARG-LYS-OH

The contraceptive polypeptide designated by Kent and shown by him to be H-Thr-Pro-Arg-Lys-OH has been synthesized by a solid-phase procedure, and the tetrapeptide was purified and characterized. Factor C-LHIH, partially purified from porcine hypothalami, which inhibits the luteinizing hormone from basal release and from the synthetic luteinizing hormone releasing hormone (LHRH), and the synthetic H-Thr-Pro-Agr-Lys-OH are different by cation exchange and by other high pressure liquid chromatographic techniques. At high dosage, H-Thr-Pro-Arg-Lys-OH released FSH and to a lesser extent LH, but the tetrapeptide showed no antagonist activity against synthetic LHRH, even at very high levels.
