目的:探讨两种微波炉在电镜标本制备中的应用效果对比。方法:分别采用两种微波炉对电镜标本聚合,进行电镜标本制备的实验性探索。结果:同种功率的微波采用同种实验条件聚合电镜标本,其切片同样完整,不易破碎,标本超微结构图像清晰、染色效果理想。结论:在相同功率及实验条件下运用家用微波炉制备电镜标本,其标本制备结果与使用医用微波炉制备结果无明显差别,可将家用微波炉运用到日常工作中。

The effects of collection methods on oral fluid codeine concentrations

The use of a variety of alternative biological specimens such as oral fluid for the detection and quantitation of drugs has recently been the focus of considerable scientific research and evaluation. A disadvantage of drug testing using alternative specimens is the lack of scientific literature describing the collection and analyses of these specimens and the limited literature about the pharmacokinetics and disposition of drugs in the specimen. Common methods of oral fluid collection are spitting, draining, suction, and collection on various types of absorbent swabs. The effect(s) of collection techniques on the resultant oral fluid drug concentration has not been thoroughly evaluated. Reported is a controlled clinical study (using codeine) that was designed to determine the effects of five collection techniques and devices on oral fluid codeine concentrations. The collection techniques were control (spitting), acidic stimulation, nonacidic stimulation, and use of either the Salivette? or the Finger Collector (containing Accu-Sorb?) oral fluid collection devices. Preliminary data were collected from two subjects using the Orasure? device. The in vitro drug recovery was also evaluated for the Salivette and the Finger Collector devices. With the exception of a single time point, codeine concentrations in specimens collected by the control method (spitting) were consistently higher than concentrations in specimens collected by the other methods. The control collection concentrations averaged 3.6 times higher than concentrations in specimens collected by acidic stimulation and 1.3 to 2.0 higher than concentrations in specimens collected by nonacidic stimulation or collection using either the Salivette or the Finger Collector devices. When calculated using oral fluid codeine concentrations from the clinical study, the elimination rate constant, t1/2, AUC and the peak oral fluid concentrations demonstrated device differences. Stand-alone automated solutions can enhance laboratory operations

Clinical laboratory automation has developed over the past decade as one means of consolidating testing, reducing costs, and improving the effectiveness of laboratory testing. Most of the developments have been aimed at core clinical laboratory operations, and have primarily addressed preanalytical and analytical processing of traditional specimens arriving in blood collection or similar aliquot tubes. Much less attention has been given to specialized applications such as processing specimens for urine toxicology, and only recently have vendors attacked the problems associated with sorting and maintaining the laboratory's inventory of specimens. This report highlights selected developments in these areas, describes one approach to cost-effective custom platform development, and discusses the advantages and pitfalls to solving problems with laboratory automation. The demand for microscale analysis has stimulated the development of new solid phase formats in sample *** interest in studying complex patterns of proteins in cells and tissues has led to new developments in analytical technologies. This interest in following changes in the cellular protein phenotype, termed proteomics, has generated a need for rapid and accurate protein identification. Such methods are derived from high-resolution separation approaches, like two-dimensional gel electrophoresis followed by protease digestion. The characterization methodology of choice is no longer Edman-based sequencing, but rather the more rapid and sensitive mass spectrometry (MS) approach. Both methods, however, are compromised by dirty and contaminated samples. Additionally, in MS, much smaller sample volumes and a higher concentration of analyte are required. To meet these challenges, new sample preparation processes are being developed for microscale volumes in miniaturized formats. At a recent meeting^1, microscale methodologies and their increasing application to drug discovery, combinatorial chemistry, and protein/peptide characterization studies were described. This article reviews the development of new formats of microscale sample preparation devices.

Endobag extractor to remove masses during laparoscopy

Background: Intra-abdominal masses are removed during laparoscopy using different types of endobags. However, in many cases the specimens are larger than the trocar or the incision in the abdomen, with a potential risk of endobag rupture. Instrument: We developed an instrument to facilitate extraction of an endobag during laparoscopy without the need for a conventional minilaparotomy. The endobag extractor has three removable diverging blades that symmetrically enlarge the operative canal in the abdominal wall if spread after sharp extension of the skin incision. The full endobag can be drawn through the canal without the risk of endobag rupture because the size of the canal can be individualized, building a funnel. Experience: We removed various kinds of ovarian tumors, specimens from salpingo-oophorectomies, and other specimens in 22 cases. Conclusion: This new instrument allows easy removal of surgical specimens during laparoscopy without conventional minilaparotomy, regardless of the type of endobag used. Some historical aspects of urinals and urine receptacles

In the history of mankind the first receptacles for urine were made and employed for diagnostic purposes and developed over centuries to a sophisticated matula. In ancient Greek and Roman history, chamber pots existed and urine was collected to bleach sheets, but it was only in the late medieval and renaissance times that a real urine receptacle or urinal for daily use was developed. We give a short description of the materials used, including clay, pewter, copper, and silver, but more sophisticated receptacles made of china, such as the bourdaloue, and of glass, such as the Kuttrolf, were also developed for use during long church ceremonies. Less known are the wooden "pipes" from Turkestan, used to keep babies dry. In the long history of mankind. urinals sometimes became very original objects.

Technique for harvesting keratolimbal allografts from corneoscleral buttons

PURPOSE: To report a method for obtaining thin keratolimbal lenticules from cadaveric corneoscleral buttons for corneal epithelial stem cell allografts. METHOD: A standard silicone orbital sizing sphere and three 25-gauge needles were employed to fix the corneoscleral button to facilitate excision of keratolimbal lenticules, RESULT: Limbal stem cells in the form of keratolimbal lenticules were obtained without difficulty. CONCLUSION: With this technique, thin keratolimbal lenticules of good quality can be obtained efficiently, inexpensively, and without specialized instruments. (C) 1999 by Elsevier Science Inc. Assessment of an instrument for automated reagent and specimen dispensing in blood banking

We have evaluated a semi-automated computer controlled dispensing device in conjunction with microtube technology for the performance of routine blood grouping and antibody screening procedures. A total of 787 specimens have been tested, 78 with unexpected antibodies previously identified by manual microtube methods. All were tested in duplicate in different order;there was complete agreement in blood group (ABO, RhD) determinations and the unexpected antibodies were found in the appropriate microtube location in all duplicate sets of results. We conclude that the device accurately and reproducibly dispenses cells, sera and reagents. The savings of technologists' time over traditional manual tube methods exceed 75%, excluding the time to perform elements common to both methods. The combination of automation and microtube technology offers the opportunity for substantial savings in technologist time with accuracy in reagent and specimen dispensing.
