SOME IMMUNOLOGICAL INVESTIGATIONS ON ANTITHROMBIN-III BUDAPEST

Immunoelectrophoretic studies were carried out on plasma from a patient with an abnormal antithrombin III molecule (antithrombin III ''Budapest''). One-dimensional immunoelectrophoresis using a commercial antibody [Ab] to antithrombin III (Ab to .alpha.2-AtIII) showed 2 precipitin peaks of .alpha.2-AtIII antigen [Ag]. Two-dimensional immunoelectrophoresis showed 2 precipitin peaks, 1 with normal electrophoretic mobility, the other with increased electrophoretic mobility. .alpha.2-AtIII Ag with normal mobility was identical to normal .alpha.2-AtIII and the .alpha.2-AtIII Ag with increased electrophoretic mobility was antigenically deficient and appeared to be present in higher concentration than the normal .alpha.2-AtIII Ag. Although 1-dimensional immunoelectrophoresis on plasma from the patient''s son showed only 1 peak, 2-dimensional immunoelectrophoresis revealed that the son had 2 populations of .alpha.2-AtIII, 1 with normal mobility, the other with increased mobility. The .alpha.2-AtIII Ag with normal mobility appeared to be present in higher concentration than the .alpha.2-AtIII Ag with increased mobility. One- and 2-dimensional immunoelectrophoresis using a different commerical Ab to .alpha.2-AtIII showed only 1 precipitin peak using the patient''s plasma. The precipitin peak observed following 2-dimensional immunoelectrophoresis was asymmetric and showed increased mobility.

PRECIPITIN RESPONSES OF INFECTED MICE TO EXOANTIGENS AND CELLULAR ANTIGENS OF TRYPANOSOMA-MUSCULI

Plasma were collected from mice immunosuppressed with 650 R from a Co-60 .gamma.-radiation source and infected with T. musculi. Trypanosomes were collected from immunosuppressed mice and from nonirradiated, infected animals. Rabbit antiserum was prepared against trypanosomes from nonirradiated mice and employed in immunodiffusion analyses to detect trypanosome exoantigens (ExAg) in plasma of irradiated, infected mice and cellular antigens (CAg) in extracts of parasites collected from immunosuppressed and nonirradiated hosts. The rabbit antiserum formed at least 3 precipitin lines with plasma from irradiated, infected mice and 8-9 precipitin lines with extracts of parasites obtained from immunosuppressed and untreated mice. Of the precipitin reactions 2 were against mouse plasma antigens (PAg). Lower levels of PAg appeared present in extracts of trypanosomes isolated from the irradiated mice than in those from nonirradiated animals. Mice synthesized antibodies against 1 ExAg demonstrable in immunodiffusion tests by 14 days after T. musculi infection. A single precipitin reaction was also seen after 21 days. One of 2 precipitin lines were formed with ExAg after 42 days of infection. Two to 3 precipitin lines formed between the ExAg and mouse antisera collected 98, 175 and 341 days after injection of the T. musculi. Similar immunodiffusion reactions were detected with CAg present in extracts of T. musculi isolated from irradiated and those from nonirradiated mice and the mouse antisera. One of 2 precipitin lines were found between CAg and antisera from mice infected for 14 days. Two precipitating antigen-antibody systems were seen with antisera collected after 21, 42 and 98 days and 2-3 precipitin reactions were formed between CAg antisera collected from mice 175 and 341 days after infection. Absorption and immunodiffusion analyses conducted with rabbit and mouse antisera indicated parasite ExAg in plasma of irradiated, T. musculi infected mice were present in preparat

A case with the characteristics of allergic bronchopulmonary aspergillosis was described. The species of Aspergillus involved, A. ochraceus, was not previously found in this disorder. The organism had antigenic properties distinct from 5 other Aspergillus spp. most commonly associated with allergic bronchopulmonary aspergillosis. The patient had immediate skin test, immunodiffusion and radioallergosorbent reactivity to the species. Of 112 serum samples from others suspected of having allergic bronchopulmonary aspergillosis, 4% had precipitins to A. ochraceus only. It may be necessary in some cases to prepare and test extracts of the patient''s Aspergillus isolate in order to confirm the diagnosis.

The haemagglitinating activity of different anti-DNP antibody populations when dinitrophenylated sheep and human red cells are used as agglutinogen

By means of serial precipitations, using 1/20th of the antigen (DNP - BSA) dose necessary for optimal antibody precipitation, it is possible to separate precipitating antibodies from anti-DNP sera corresponding to different antibody species. If the remaining serum is passed through a column of immunoadsorbent, a population of non-precipitating antibodies, capable of co-precipitating with the precipitating antibodies is retained. In the serum which has passed through the immunoadsorbent there remains a third population of antibodies lacking in precipitating and co-precipitating activity. Its quantification can only be achieved by saturation at high free hapten concentration. The average Ko and a values are as follows: precipitating antibodies. Ko = 1.14 × 10⁵ L/M, a = 0.72;non-precipitating (co-precipitating) antibodies isolated by immunoadsorption, Ko = 3.16 × 10⁴ L/M, a = 0.51;and non-precipitating antibodies isolated from the serum lacking in precipitating and co-precipitating antibodies, Ko = 1.05 × 10³ L/M, a = 0.69. By means of passive haemagglutination and the use of dinitrophenylated sheep and human red cells as agglutinogen it is possible to differentiate the three populations of antibodies. The precipitating antibodies agglutinate both types of red cells;the non-precipitating antibodies separable by immunoadsorption only agglutinate sheep red cells;while the third population of antibodies is totally lacking in haemagglutinating activity. This suggests that haemagglutination detects only those antibodies capable of firm union with antigen at two combination sites, at least in the case of red cells without exposed Fc receptors (human red cells), while if the red cells possess such receptors (sheep red cells) haemagglutination takes place even with antibodies capable of a firm union to the antigen with at least one of its combination sites (non-precipitating antibodies retained by immunoadsorption or co-precipitating antibodies
