Directed actin polymerization is the driving force for epithelial cell-cell adhesion

We have found that epithelial cells engage in a process of cadherin-mediated intercellular adhesion that utilizes calcium and actin polymerization in unexpected ways. Calcium stimulates filopodia, which penetrate and embed into neighboring cells. E-cadherin complexes cluster at filopodia tips, generating a two-rowed zipper of embedded puncta. Opposing cell surfaces are clamped by desmosomes, while vinculin, zyxin, VASP, and Mena are recruited to adhesion zippers by a mechanism that requires alpha-catenin. Actin reorganizes and polymerizes to merge puncta into a single row and seal cell borders. In keratinocytes either null for alpha-catenin or blocked in VASP/Mena function, filopodia embed, but actin reorganization/polymerization is prevented, and membranes cannot seal. Taken together, a dynamic mechanism for intercellular adhesion is unveiled involving calcium-activated filopodia penetration and VASP/Mena-dependent actin reorganization/polymerization.

Maize calreticulin localizes preferentially to plasmodesmata in root apex

Using a polyclonal antibody raised against calreticulin purified and sequenced from maize, we performed an immunocytological study to characterize putative domain-specific subcellular distributions of endoplasmic reticulum (ER)-resident calreticulin in meristematic cells of maize root tip. At the light microscopy level, calreticulin was immunolocalized preferentially at cellular peripheries, in addition to nuclear envelopes and cytoplasmic structures. Punctate labelling at the longitudinal walls and continuous labelling at the transverse walls was characteristic. Immunogold electron microscopy revealed plasmodesmata as the most prominently labelled cell periphery structure. In order to further probe the ER-domain-specific distribution of maize calreticulin at plasmodesmata, root apices were exposed to mannitol-induced osmotic stress. Plasmolysis was associated with prominent accumulations of calreticulin at callose-enriched plasmodesmata and pit fields while the contracting protoplasts were depleted of calreticulin. In contrast, other ER-resident proteins recognized by HDEL peptide and Sip antibodies localized exclusively to contracted protoplasts. This finding reveals that, in plasmolysed cells, calreticulin enriched ER domains at plasmodesmata and pit fields are depleted of other ER-resident proteins containing the HDEL retention peptide.

Pathogenesis of African horse sickness:: Ultrastructural study of the capillaries in experimental infection

African horse sickness (AHS) was induced in five horses by inoculation, to determine the ultrastructural changes in endothelial cells of capillaries in the myocardium, lung, spleen and liver. The animals developed cardiac and mixed forms of the disease. Alterations detected in the endothelial cells of the vessels of infected animals included: the presence of structures associated with viral infection, hypertrophy, degenerative changes, appearance of cytoplasmic projections, changes in permeability, alteration of intercellular junctions, loss of endothelium, subendothelial deposition of cell debris and fibrin, and vascular repair. In association with these changes, oedema, haemorrhages and microthromboses were detected, particularly in the myocardium and lung. This study showed that infection of, and changes to, the capillary endothelial cells of the organs under study was independent of the form in which the disease manifested itself but was dependent on the organ and blood vessel tl;pe Thus, different levels of viral tropism were observed for the endothelial cells of the vessels in different organs. Viral infection was commonest in the endothelial cells of myocardial vessels, followed by those in the lung, whereas in the spleen and liver, endothelial cell infection was rare and, in the case of the liver, limited to the interstitial capillaries. CONFRONTING SUBSURFACE CISTERNAE AND DESMOSOMES IN HELA MONOLAYER-CULTURES

Confronting subsurface cisternae (CSC) and desmosomes are abundant at sites of parallel apposition of plasma membranes in confluent epitheloid monolayers of HeLa cells. Both structures are absent in young and subconfluent cultures, where stable intercellular contacts do not occur. They first appear when the plane of intercellular contact approaches towards the vertical; they disappear when cells separate spontaneously at superconfluency or by treatment with EDTA. The structure of CSC, lacking ribosomes at their side facing the plasma membrane, might fit the production of glycoproteins involved in intercellular adhesion.

The ultrastructure of a well studied insect chemosensory unit is presented. Two separate lumina are present in this chemosensory unit, the trichogen and sensillar lumina. The fluid within the trichogen lumen exclusively bathes the dendritic terminals and may be involved with the reception and/or modulation of environmental stimuli. Cytoplasmic extensions of the trichogen cell which line the trichogen lumen may be involved in the production of the cuticular sheath. The sensillar lumen is bordered by the tormogen and a sleeve cell and is continuous with the unoccupied channel of the setal shaft. Functions for the various cellular components of the blowfly chemoreceptor sensillum are offered.

The present paper desoribes ultrastructural aspect of lymphatic transport of dermal melanosomes in blue nevus. At least two processes may be responsible for the transport across the lymphatic endothelial wallin vivo; one is the passage of melanophages through the intercellular junctions and the another of free melanosomes. Phagocytosis of melanosomes by lymphatic endothelial cells is discussed.

The retrocorneal fibrous membrane in 8 cases of the vitreous touch syndrome was characterized by homogeneous granular material, fine filaments arranged either singly or in bundles, multiple lamellae of basement membrane like material and banded figures of 110 nm periodicity. Seven cases demonstrated an intact endothelial monolayer while only one showed cells in a markedly thickened fibrous layer. These findings support the pathogenic mechanism of endothelial fibrous metaplasia and secretory activity as the origin of the retrocorneal fibrous membrane in the vitreous touch syndrome.

STRUCTURAL MODIFICATIONS OF LUTEIN CELL GAP JUNCTIONS DURING PREGNANCY IN RAT AND MOUSE

AbstractBy use of lanthanum tracer and freeze‐fracture procedures it was found that granulosa‐lutein cells of the pregnant mouse and rat ovaries are connected by gap junctions and septate‐like zones of contact. Lutein cell gap junctions enlarge and become partially internalized by the end of the first week of gestation. Expansion of the gap junction domain appears to be due initially to intercalation of particles along borders of small gap junctions devoid of smaller non‐junctional particles. The number of gap junction lined processes appearing at the cell border increases concomitantly with hypertrophy of the lutein cell during the second week of pregnancy. Strands of particulate or grooved membrane emanate from the margin of larger gap junctions undergoing interiorization. Most large gap junctions are intimately associated with elements of the smooth endoplasmic reticulum. Spherical gap junctional profiles assume a deeper location in the lutein cell and may form concentric arrays by term while true surface gap junctions appear to fragment in the post‐partum corpus luteum. The modifications observed are interpreted with respect to biogenesis of the gap junction and the hormonal control of lutein cell
