Apelin is an endogenous ligand of the APJ receptor (also known as Aplnr) which is a G protein-coupled receptor [1,2].Elabela (also known as Toddler,Apela),which shares little sequence similarity with Apelin (Fig.1),has been identified as a new endogenous ligand for APJ receptor [3].Elabela is encoded by AK092S78 gene from a region of the human genome which was previously classified as ‘non-coding’.Elabela is initially translated into a peptide of 54 amino acids including a secretory signal ***,its mature form only contains 32 amino acids.

Regucalcin (RGN),also known as senescence maker protein-30,is a unique calcium-binding protein which lacks of classic EF-hand motif [1].It is mainly expressed in the liver and kidney and highly conserved in the coding region among eukaryotes [2].RGN is involved in regulating a series of cellular processes,such as intracellular calcium homeostasis,calcium signal transduction,cell metabolism,function of the nuclei,and gene expression [3-5].Given that calcium exerts a profound effect on cytoskeleton,and cytoskeleton alteration is closely related to tumor formation,invasion,and metastasis [6],we speculated that RGN might play a role in cytoskeleton rearrangement and further affect cytoskeleton associated signaling pathways.

Cumulative evidences have demonstrated that most of the non-repetitive genome in higher organisms are actively transcribed and surprisingly only a small percentage (＜ 20％) of transcripts are associated with genes that encode proteins [1].One of the emerging themes in the study of non-coding transcripts is large intergenic non-coding RNAs (lincRNAs),a class of large regulatory RNAs implicated in imprinting,dosage compensation,and transcriptional regulation [2].In light of recent discoveries revealing the flexibility of lincRNAs and their abilities to act as modular scaffolds for protein-chromatin interactions and to form spatially compact arrays of complexes [3,4],many would acknowledge that most lincRNAs act as sensors and integrators of a wide variety of regulated transcriptional responses and probably epigenetic events,which may have an impact on various human diseases.

The discovery of microRNAs （miRNAs） is one of tbe most significant breakthroughs in recent decades, miRNAs are a class of small, endogenous, non-coding RNAs （19-24 nt） that negatively regulate the expression of target genes by binding to their 3＇ untranslated region and are involved in various biological processes. Three main approaches that are globally used for the detection of miRNA include direct cloning, forward genetics and computational approaches using bioinformatics.

It is carried out by the spliceosome, which catalyzes the removal of non-coding intronic sequences to assemble exons into mature mRNAs prior to export and *** in splicing lead to many human genetic diseases [1], and splicing mutations in a number of genes involved in growth control have been implicated in multiple types of cancer.

How to correct chromosomal trisomy

Global gene expression analyses show that not only chromosome 21-1inked genes but also genes on other chromosomes are disrupted in cells with trisomy 21,suggesting wide effects on the *** such comparisons are usually between different individuals and not between isogenic cells,a large portion of the changes observed could be due to variants unrelated to the *** fact,few specific genes have been implicated in the trisomy phenotypes,and the effects of abnormal gene dosage are not clear in different *** fold-increase in expression for genes on chromosome 21 is about 1.2-1.4 and only reaches the theoretical 1.5 for a minor portion of genes in trisomic cells,suggesting that dampening effects partially correct abnormal expression *** dampening and feedback mechanisms mitigate deleterious dosage effects of aneuploidy [1,2].To achieve correct gene expression,additional feed-forward mechanisms of dosage compensation evolved,for example to regulate the X *** mechanisms include X upregulation to increase expression from the single active X chromosome,and X inactivation to silence one X chromosome in mammalian females [1].

Transcriptionally silent sperm contains a variety of RNA fragments of both coding and non-coding transcripts.A recent article by Peng and colleagues reveals several new families of small RNAs enriched in sperm,which are derived from the same locus as *** finding of these short fragments of tRNA in the sperm raises once again the question of the possible function(s) of such a miniaturized form of information carried by the spermatozoon.
