DEATH AFTER PICHINDE VIRUS-INFECTION IN LARGE AND SMALL STRAIN-13 GUINEA-PIGS

The lethal events of Pichinde virus infections were studied in large (> 36-week-old), small (4- to 6-week-old), and uninfected strain 13 guinea pigs. Time to death was shorter and the rate of body weight loss greater for large than for small virus-infected guinea pigs. Severely ill large guinea pigs developed a life-threatening, nonlactate metabolic acidemia with an increased anion gap, hypokalemia, and renal failure. Small infected guinea pigs near death demonstrated a modest increase in blood and plasma volume indices and in lung and heart permeability to albumin. This group showed a severe hypoxemia, elevated blood lactate, and bicarbonate ion concentrations with a doubled respiratory rate. These findings are consistent with respiratory failure due to obstruction of small airways. Although virus-infected guinea pig endothelial cells showed a decrease in DNA and protein synthesis, no change in bradykinin-induced intracellular calcium signaling was noted.

Social networks' unnoticed influence on body image in Spanish university students

The influence of advertising on social networks and its effect on young people's self-image has been studied. This study first explored the frequency of published videos reposted on social networks using different application programming interfaces as search engines. The study also implemented self-valuation surveys assessed by experts and distributed among mental health professionals who assessed the influence of these social networks. Finally, the survey was designed regarding social networks and body assessment and distributed among Cordoba university students (N = 328). The majority of health professionals concluded that there is an elevated influence of social networks on eating disorders, assessing advertising as a risk factor with a value of 4.60 out of 5. Young people's perceptions of their body image indicate that women's degree of satisfaction is highly correlated with the frequency of their connections to social networks, with the body image those women present and with the effect of advertising. Conversely, this high correlation and influence does not exist in men.

CONSTRUCTION OF A COMPLEMENTARY DNA LIBRARY OF PARELAPHOSTRONGYLUS TENUIS AND IDENTIFICATION OF A POTENTIALLY SERO-DIAGNOSTIC RECOMBINANT ANTIGEN

Newly developed serological tests for diagnosing parelaphostrongylosis in cervids, using the excretory-secretory products (ES) of the infective larvae of larvae of Parelaphostrongylus enzyme-linked immunosorbent assays (ELISAs), demonstrable Superiority over the traditional method of, larval recovery and microscopic identification. To generate a source of ELISA antigen by genetic engineering. we created it complementary DNA (cDNA) expression library by the reverse transcription of mRNA of P. tenuis adult worms and by ligation with the vector lambda-ZAP II. The library was screened using antisera produced in mice by immunization with a somatic antigen preparation of adult worms. Seventeen clones were isolated, sequenced, and checked for similarity to other DNA sequences in GenBank. A Previously identified parasite gene encoding an asparlyl protease inhibitor (API) was isolated front the cDNA library, subcloned and expressed using the pET expression vector to produce a glutathione S transferase (GSTI-His-S-tag-P. tenuis API fusion Protein (molecular weight = 63 kDa). An ELISA utilizing the API fusion protein as the coating antigen was used to serologically diagnose all white-tailed deer (WTD, Odocoileus virginianus 10 out of 10) that had been inoculated with a range of 6-150 L3 P. tenuis, indicating that the antigen may be useful serodiagnostic antigen for P. tenuis infection in this cervid species.

Designing an API at an appropriate abstraction level for programming social robot applications

Whilst robots are increasingly being deployed as social agents, it is still difficult to program them to interact socially. To create usable tools for programming these robots, tool developers need to know what abstraction levels are appropriate for programming social robot applications. We explore this through the iterative design and evaluation of an API for programming social robots. The results show that high level primitives, with a close mapping to social interaction, are suitable for programming social robot applications. However, the abstraction level should not be so high that it takes away too much control from programmers. This has the potential to enable programmers to produce high quality social robot applications with less programming effort. (C) 2017 Published by Elsevier Ltd.

Bacterial counts on the contents of an anaerobic fixed-bed digester receiving a whey substrate were conducted using the modified roll tube technique. Average anaerobic counts after 48 hours incubation on lactate containing media were 3.12 × 10⁹ and 3.7 × 10⁹ ml^?1, respectively. These counts were between 140 and 190 times higher than aerobic counts on the same media. Seventy-four strains from both media were isolated and characterized, and the relationship between the organisms was calculated according to the similarity coefficient of Sokal and Michener [20]. The organisms were clustered using the unweighted pair group method and the results were presented in the form of a simplified dendrogram. The isolates clustered in three major groups (A, B, and C) at a similarity level of 76%. A small diffuse group of five organisms was also found. The organisms in two (A and B) of the major clusters were obligate anaerobes. Cluster A represented 34% of the isolates, cluster B represented 50%, and the facultative streptococci in cluster C, 11%. The isolates in clusters A and B could only tentatively be identified as members of the generaBacteroides andPeptostreptococcus, respectively. The isolates could not be positively identified.

Promoter sequences involved in transforming growth factor β1 gene induction in HaCaT keratinocytes after gamma irradiation

Transforming growth factor beta 1 (TGFB1) is a cytokine involved in the development of both acute and late cutaneous radiation syndromes. We previously demonstrated that ionizing radiation induces TGFB1 expression in vivo in pig skin within a few hours. The purpose of the present study was to develop an in vitro human model to identify the mechanisms of this early activation. Accordingly, human HaCaT keratinocytes were irradiated with a single dose of 20 Gy. First, radiation-induced TGFB1 overexpression was checked at both the transcriptional and transductional levels in HaCaT cells. Then electrophoretic mobility shift assays (EMSA) and transient transfection with various TGFB1 promoter constructs were used to identify the sequences involved in regulating this promoter. ENISA analysis showed the induction of nuclear protein binding activity by T irradiation to the -365 AP1 sequence (TGTCTCA), suggesting the involvement of AP1 sequences in the regulation of TGFBI transcription. In gene reporter assays, maximal TGFBI promoter activation was found for the longest construct, which contains two AP1 sequences. However, assays with constructs including deletions showed that these two AP1 sequences were not sufficient to confer TGFBI inducibility. These results showed for the first time, to our knowledge, that transcriptional regulation is involved in radiation-induced activation of TGFB1 gene expression. We assembled an internatlional collection of strains from sporadic and epidemic human infection with Streptococcus zooepidemicus (Lancefield group C) for laboratory study. Cultural and physiological characteristics of the isolates were determined, including biotyping with the API 20 STREP test kit and susceptibility testing with penicillin, erythromycin and tetracycline. The strains were examined for bacteriocin production and sensitivity and typed with a specially developed group-C streptococcal bacteriophage system incorporating a panel of 14 phages. Results of these tests gave useful discrimination between many of the strains: differences were shown between each of the major outbreak strains, including those complicated by post-streptococcal infection may be caused by S. zooepidemicus and isolates should be identified to species level;the application of a typing scheme such as this may help to distinguish epidemiological patterns of infection.
