Three-dimensional structure prediction of bovine AP lyase, bapi:: prediction of interaction with DNA and alterations as a result of Arg176→Ala, Asp282→Ala, and His308→Asn mutations

bapi is an apurinic/apyrimidinic lyase (AP lyase) that plays an important role in the repair of DNA damage. The present study deals with the prediction of the 3D structure of bovine AP lyase based on its sequence homology with human AP lyase. The predicted 3D model of bovine API shows remarkable similarity with human endonuclease in the overall 3D fold. However, significant differences in the model and the X-ray structure were located at some of the important sites. We have analyzed the active center of the enzyme and other sites that are involved in DNA repair. A number Of amino acids bind the bases located in the major/minor grooves of DNA. An insertion of Arg176 in the major groove and Met270 in the minor groove caps the DNA bound enzyme's active site, stabilizing the extra helical AP site conformation and effectively locking the protein onto the AP-DNA. Three bapi mutants were also modeled and analyzed as regards the changes in the structure. Substitution of Arg176 --> Ala leads to the loss of DNA binding whereas mutation of Asp282 --> Ala and His308 --> Asn leads to a decrease in the enzymatic activity. (C) 2004 Elsevier Inc. Tavaborole (TVB), a benzoboroxole derivative, was approved for treating onychomycosis. Boron's unique properties, such as variable oxidation states, and stability, require specialized analytical methods. The current RP-HPLC method was developed using eco-friendly solvents, maintaining pH, and providing a sharp peak at 5.7 min at 272 nm. A new NP-HPTLC technique was established employing a mobile phase composed of toluene and methanol, yielding an Rf value of 0.58 with a similar correlation coefficient of 0.9989 for both methods. The accuracy of TVB for both methods was evaluated at levels of 80 to 120%, providing the %RSD in the range of 0.42 to 0.86% (for HPLC) and 0.95 to 1.50% (for HPTLC). The reproducibility for the two methods was performed in triplicate (30-50 mu g/ml) for HPLC and (4000 to 6000 ng/band) for HPTLC, and %RSD was evaluated. Replicates at 20 mu g/ml for HPLC and 5000 ng/band for HPLTC show the %RSD of 1.42 and 0.65, respectively. Robustness parameters were evaluated by altering pH, wavelength, and flow rate for HPLC and mobile phase composition, plate saturation time, and development distance for HPTLC. The robustness was evaluated by altering the analyst to calculate a %RSD of 0.88 and 0.31 for HPLC and 0.12 and 0.22 for HPTLC. From the results, all the validation parameters are found within acceptable ranges. TVB showed stability in acidic and basic conditions, with degradation observed only under peroxide treatment. An attempt will be made to identify the probable and major degradants of TVB using LC-MS spectra, which was crucial considering the boron's chemical properties. The RP-HPLC method was confirmed as environmentally sustainable using the AGREE program metrics. In contrast, the NP-HPTLC method met validation standards but was not considered eco-friendly due to solvent choice. Both methods are suitable for commercial applications.

Fraud is a widespread international problem for enterprises. Organizations increasingly use self-learning classifiers to detect fraud. Such classifiers need training data to successfully distinguish normal from fraudulent behavior. However, data containing authentic fraud scenarios is often not available for researchers. Therefore, we have implemented a data generation tool, which simulates fraudulent and non-fraudulent user behavior within the purchase-to-pay business process of an ERP system. We identified fraud scenarios from literature and implemented them as automated routines using SAP's programming language ABAP. The data generated can be used to train fraud detection classifiers as well as to benchmark existing ones.

Histopathological evaluation of melanocytic tumours usually allows reliable distinction of benign melanocytic naevi from melanoma. More difficult is the histopathological classification of Spitz tumours, a heterogeneous group of tumours composed of large epithelioid or spindle-shaped melanocytes. Spitz tumours are biologically distinct from conventional melanocytic naevi and melanoma, as exemplified by their distinct patterns of genetic aberrations. Whereas common acquired naevi and melanoma often harbour BRAF mutations, NRAS mutations, or inactivation of NF1, Spitz tumours show HRAS mutations, inactivation of BAP1 (often combined with BRAF mutations), or genomic rearrangements involving the kinases ALK, ROS1, NTRK1, BRAF, RET, and MET. In Spitz naevi, which lack significant histological atypia, all of these mitogenic driver aberrations trigger rapid cell proliferation, but after an initial growth phase, various tumour suppressive mechanisms stably block further growth. In some tumours, additional genomic aberrations may abrogate various tumour suppressive mechanisms, such as cell cycle arrest, telomere shortening, or DNA damage response. The melanocytes then start to grow in a less organised fashion and may spread to regional lymph nodes, and are termed atypical Spitz tumours. Upon acquisition of even more aberrations, which often activate additional oncogenic pathways or alter cell differentiation, the neoplastic cells become entirely malignant and may colonise and take over distant organs (spitzoid melanoma). The sequential acquisition of genomic aberrations suggests that Spitz tumours represent a continuous biological spectrum, rather than a dichotomy of benign versus malignant, and that tumours with ambiguous histological features (atypical Spitz tumours) might be best classified as low-grade melanocytic tumours. This paper gives a synopsis of the SAP R/3 Business Framework. The architecture is based on an object-orientated approach using business objects and Business Components. Using this architecture together with a distribution model, distributed applications (distributed business processes) can be mapped and implemented. The spectrum of distributed applications extends from the simple integration of two components using a few interfaces to the integration of many components distributed across the globe.

Determinants of the addiction treatment drop-out rates in an addiction counseling centre: a cross-sectional study

OBJECTIVE: In the addiction treatment, treatment drop-out rates are very high. Drop-out is a crucial problem in the addiction treatment;therefore, understanding the reasons underlying this problem will help clinicians in developing new treatment strategies for treatment maintainance. In this present study, we aimed to examine the reasons for drop-out behaviour in the outpatients who presented to an addiction counseling centre in Turkey. METHODS: This study was conducted in 554 outpatients with alcohol and/ or substance use disorder who presented to the Turkish Green Crescent Counseling Center (YEDAM) between January 2016 and July 2017. The patients were evaluated retrospectively. The sociodemographic characteristics, substance use-related and psychological characteristics were extracted by using the Addiction Profile Index (bapi) and Addiction Profile Clinical Form (bapi-K) included in the YEDAMSOFT software system. The patients who came to the second, fifth, and tenth sessions and the patients who dropped-out of the treatment were analyzed in order to determine the sociodemographic, substance use-related, and psychological characteristics that might caused patients to drop-out from the treatment. RESULTS: The drop-out rates was found higher in male patients compared to the females. Among the factors that caused drop-out behaviour were low education status, negative relationship with father, adverse effects of the substance use, severe desire for the substance use, low motivation level, being under probation, and higher numbers of psychiatric treatments in the past. We found that the factors predicted the drop-out may vary depending on the treatment session when drop-out from the treatment was actualized. Workflow processing using ERP Objects

Enterprise Resources Planning (ERP) Systems, and Workflow Management Systems (WFMS) evolved parallel in the past. The main business drivers for automation came from the ERP world, but the WFMS solutions discovered their own way figuring out the necessity of such applications without ERP as well. In our paper we follow only the usage of workflows in ERP systems. The central elements of built-in workflows are the ERP objects, which embed and handle the business data providing real life meaning of business objects as well. The capabilities of built-in workflow systems of such ERP solutions are presented via two market-leaders: SAP and Microsoft Dynamics AX. Both solutions are dealing with ERP objects in sence of the workflow management. We recognized and present the weaknesses and restrictions of the built-in workflow systems on these two ERP examples. In our paper we describe the results of our analysis of the interoperability of the built-in workflow systems as well and demonstrate the required add-on functionalities to provide usable cross-system workflows in such an environment. We also mention the possibility of using a built-in workflow as a full-featured WFMS.

三星电子(北京)技术服务有限公司，是韩国三星电子集团在大陆设立的外资公司，专门负责三星电子产品市场售前和售后服务业务及服务管理软件的开发和维护。自2000年起，三星服务总部内部业务均使用SAPR/3系统进行统一管理。对下属的维修中心进行管理，公司开发了相应B2B系统。\n 随着三星公司业务的不断发展，原有两系统间采用文本方式进行数据交流的方式已经不能满足业务的需要。如何在不改变原有系统构架的情况下，如何实现两系统间时实的数据交换和共享成为三星公司系统开发的一个重要课题。\n 本论文主要讨论了在实现两系统数据时实共享的同时，维修中心在 B2B 系统中零配件申请如何在SAP R/3系统中自动生成零配件申请单的课题。考虑到系统的兼容性、硬件设备的成本、开发的困难程度以及数据安全性和稳定性的情况下，最终采用了利用SAP-DCOM组件，通过RFC调用SAPR/3系统的bapi的方式，实现了维修中心的零配件申请在SAPR/3系统中自动生成零配件定单的业务流程。\n 本课题的完成，达到了三星公司最初系统改善的目的，在不改变原有系统构架的基础上实现了系统间的时实数据共享以及申请单的自动生成。
