Umbelliferone modulates the quorum sensing and biofilm of Gram - ve bacteria: in vitro and in silico investigations

In last two decades, the world has seen an exponential increase in the antimicrobial resistance (AMR), making the issue a serious threat to human health. The mortality caused by AMR is one of the leading causes of human death worldwide. Till the end of the twentieth century, a tremendous success in the discovery of new antibiotics was seen, but in last two decades, there is negligible progress in this direction. The increase in AMR combined with slow progress of antibiotic drug discovery has created an urgent demand to search for newer methods of intervention to combat infectious diseases. One of such approach is to look for biofilm and quorum sensing (QS) inhibitors. Plants are excellent source of wide class compounds that can be harnessed to look for the compounds with such properties. This study proves a broad-spectrum biofilm and QS inhibitory potential of umbelliferone. More than 85% reduction in violacein production chromobacterium violaceum 12472 was found. All tested virulent traits of Pseudomonas aeruginosa PAO1 and Serratia marcescens MTcc 97 were remarkably inhibited that ranged from 56.62% to 86.24%. Umbelliferone also successfully prevented the biofilm of test bacteria at least by 67.68%. Umbelliferone interacted at the active site of many proteins of QS circuit, which led to the mitigation of virulent traits. The stable nature of complexes of umbelliferone with proteins further strengthens in vitro results. cutting performance and wear mechanism of zirconia toughened alumina ceramic cutting tools formed by vat photopolymerization-based 3D printing

Zirconia toughened alumina (ZTA) cutting tools are widely used for machining hard and wear-resistant mate-rials. However, the processing of ZTA ceramic cutting tools with complex shapes using traditional methods is difficult and time-consuming. Until now, to the best of our knowledge, commercial ceramic cutting tools do not contain grooves because it is very difficult to process grooves on the ceramic cutting tool, resulting from the feature of high hardness and brittle attribute of ceramics. This study investigated high-performance, complex-shaped zirconia-toughened alumina (ZTA) ceramic cutting tools equipped with a chip breaker for the first time. The investigated samples were prepared using vat photopolymerization based 3D printing combined with a hot isostatic sintering process. The relative density of 99.34%, Vickers hardness of 17.98 & PLUSMN;0.20 GPa, bending strength of 779 & PLUSMN;47 MPa and fracture toughness of 5.41 & PLUSMN;0.29 MPa m1/2 of the ZTA cutting tools were obtained. The effects of three cutting parameters, namely the cutting speed vc, feed rate f, and back cutting depth ap, on the cutting performances of the cutting tools were investigated, and the wear mechanisms of the cutting tools were examined. We believe that our study makes a significant contribution to the literature because this study provides an in-depth investigation of the properties and wear mechanisms of ZTA ceramic cutting tools with chip breaker grooves. The findings of this study provide insight into the preparation of high-performance, complex-shaped ZTA ceramic cutting tools. Further, the preparation methods investigated in this study and the evaluation of the properties of the fabricated specimens in comparison with a commercially available cutting tool illustrate the potential advantages that may be obtained via the further development of these materials.

Greater Neighborhood Disadvantage Is Associated with Alterations in Fetal Functional Brain Network Structure

Objective To determine the association between neighborhood disadvantage (ND) and functional brain development of in utero fetuses. Study design We conducted an observational study using Social Vulnerability Index (SVI) scores to assess the impact of ND on a prospectively recruited sample of healthy pregnant women from Washington, Dc. Using 79 functional magnetic resonance imaging scans from 68 healthy pregnancies at a mean gestational age of 33.12 weeks, we characterized the overall functional brain network structure using a graph metric approach. We used linear mixed effects models to assess the relationship between SVI and gestational age on 5 graph metrics, adjusting for multiple scans. Results Exposure to greater ND was associated with less well integrated functional brain networks, as observed by longer characteristic path lengths and diminished global efficiency (GE), as well as diminished small world propensity (SWP). Across gestational ages, however, the association between SVI and network integration diminished to a negligible relationship in the third trimester. conversely, SWP was significant across pregnancy, but the relationship changed such that there was a negative association with SWP earlier in the second trimester that inverted around the transition to the third trimester to a positive association. conclusions These data directly connect ND and altered functional brain maturation in fetuses. Our results suggest that, even before birth, proximity to environmental stressors in the wider neighborhood environment are associated with altered brain development. (J Pediatr 2024;274:114201).

Value of next generation sequencing (NGS) testing in advanced cancer patients

ObjectiveThe availability of targeted therapies for oncology patients is increasing. Available genomic tests to identify treatment-eligible patients include single gene tests and gene panel tests, including the whole-exome, whole-transcriptome OncoExTra test. We assessed the costs and clinical benefits of test *** Microsoft Excel-based model was developed to evaluate test choice in patients with advanced/metastatic non-small cell lung cancer (NScLc), breast, prostate, and colorectal cancer. Treatment pathways were based on NccN guidelines and medical expert opinion. Inputs were derived from published literature. Annual economic results and lifetime clinical results with OncoExTra testing were projected per-tested-patient and compared with single gene testing and no testing. Separately, results were estimated for a US health plan without the OncoExTra test and with its use in 5% of *** with no genomic testing, OncoExTra test use increased costs by $4,915 per patient;however, 82%-92% of individuals across tumour types were identified as eligible for targeted therapy or a clinical trial. compared with single gene testing, OncoExTra test use decreased costs by $9,966 per-patient-tested while increasing use of approved or investigational targeted therapies by 20%. When considering a hypothetical health plan with 1 million members, 858 patients were eligible for genomic testing. AimThis study aimed to examine the validity of EQ-5D-5L among HFrEF patients in Malaysia, and to explore the measurement equivalence of three main language *** surveyed HFrEF patients from two hospitals in Malaysia, using Malay, English or chinese versions of EQ-5D-5L. EQ-5D-5L dimensional scores were converted to utility scores using the Malaysian value set. A confirmatory factor analysis longitudinal model was constructed. The utility and visual analog scale (VAS) scores were evaluated for validity (convergent, known-group, responsiveness), and measurement equivalence of the three language ***200 HFrEF patients (mean age = 61 years), predominantly male (74%) of Malay ethnicity (55%), completed the admission and discharge EQ-5D-5L questionnaire in Malay (49%), English (26%) or chinese (25%) languages. 173 patients (86.5%) were followed up at 1-month post-discharge (1MPD). The standardized factor loadings and average variance extracted were >= 0.5 while composite reliability was >= 0.7, suggesting convergent validity. Patients with older age and higher New York Heart Association (NYHA) class reported significantly lower utility and VAS scores. The change in utility and VAS scores between admission and discharge was large, while the change between discharge and 1MPD was minimal. The minimal clinically important difference for utility and VAS scores was +/- 0.19 and +/- 11.01, respectively. Malay and English questionnaire were equivalent while the equivalence of Malay and chinese questionnaire was *** study only sampled HFrEF patients from two teaching hospitals, thus limiting the generalizability of results to the entire heart failure ***-5D-5L is a valid questionnaire to measure health-related quality of life and estimate utility values among HFrEF patients in Malaysia. Eurasian Grey and White wolf ancestors-800,000 years evolution, adaptation, pathologies and European dog origins

The oldest known wolf appears 800,000 years ago (Marine Isotope Stage 21) in Eurasia with the unspecialized short-legged old Mammoth steppe wolf canis lupus bohemica nov. spec. From this species, about 600,000-420,000 years ago (MIS 15-11), the interglacial canis lupus mosbachensis (Soergel, 1925) short-legged Mosbach grey wolf subspecies roamed Eurasia. In the late Middle Pleistocene, there are two lineages, the southern interglacial grey and northern glacial White wolves in Eurasia. Since 320,000 (MIS 8), the short-legged White wolf canis lupus spelaeus (Goldfuss, 1823) was the glacial Mammoth steppe-adapted wolf. Parallel to the "cave wolf" (found in the German Zoolithen cave), the warm climate grey wolf canis lupus brevis Kuzmina and Sablin, 1994 existed. c. l. spelaeus relates to the Holocene (MIS 1) extant Holarctic Greenland canis lupus arctos and Siberian canis lupus albus (Kerr, 1792). The Late Palaeolithic (MIS 2) "Gravettian Goyet dogs" fall into the DNA pool of c. l. spelaeus and are identified herein as pathological bite trauma individuals, which braincase shortened during the healing process. European prehistoric Neolithic dogs seem to have been imported from central Asia with the Bandkeramik people (approx. 7000 BP) first, which have the stepped frontals originating from grey wolves.

The present work reports the controlled fermentation of heat-shocked, unsalted and inoculated green olives. The effects of heat-shock (60, 70 and 80 degrees c three times for 5 min), inoculation with the oleuropeinolytic strain of L. plantarum FSO175 (L.p-FSO175) and the addition of cell-Free Supernatant of c. pelliculosa L18 (cFS of c.p-L18) on the fermentation process of unsalted green olives were examined. The results showed a drastic reduction in the initial indigenous Enterobacteria, and an improvement in the acidification of heat-shocked olives at 70 and 80 degrees c, when compared to 60 degrees c. The inoculation with L.p-FSO175 and addition of cFS of c.p-L18 en-hanced the fermentation and preservation of unsalted green olives, indicated by a significant decrease in pH, increase in free acidity and total disappearance of Enterobacteria. The heat-shock treatment at high temperature (80 degrees c), inoculation with L.p-FSO175 and addition of cFS of c.p-L18 led to the best reduction in bitterness, and favorable color changes (L, a, and b) in fermented olives. This sequential method led to more appreciated sensory characteristics (mainly bitterness and color) of fermented olives, lower spoilage incidence in olives, and reduced fermentation time to 50 days, and therefore may be suitable to control the fermentation of unsalted green olives of the Moroccan picholine variety.

Assessing the economics of biologic and small molecule therapies for the treatment of moderate to severe ulcerative colitis in Japan: a cost per responder analysis of upadacitinib

AimPatients with moderately to severely active ulcerative colitis have an increasing number of advanced therapy options including several biologics and Janus kinase inhibitors. Though data on efficacy and safety of these advanced therapies are available, less is known about the potential economic implications of their utilization in Japan. We evaluated the relative value of these advanced therapies in Japan using a locally developed cost per responder *** model was developed using relevant clinical endpoints and treatment costs to calculate cost per responder of all advanced therapies used for moderately to severely active ulcerative colitis treatment in Japan. cost per responder was assessed in biologic-naive and biologic-exposed populations, respectively. The model incorporated induction and maintenance therapy pathways as patients progressed through based on efficacy rates (clinical response, clinical remission and endoscopic improvement). Total costs for induction and maintenance included: drug acquisition, drug administration and serious adverse event management (as necessary) for responders, with additional rescue treatment cost only for *** showed lower cost per clinical response and cost per clinical remission across both biologic-naive and biologic-exposed populations with only one exemption in cost per clinical remission in biologic-naive population. Electrical and sensing properties of indium-doped barium cerate

Systematic analysis of electrical characteristics of Bace0.75In0.25O3-,5 (BcI25) sintered sample was performed in a dry and a wet argon atmosphere in the 250 degrees c-700 degrees c temperature range. The water vapor sensing properties of BcI25 porous film and its response and recovery times were investigated under different conditions of tem-perature and water vapor concentration. The 30 mu m thick film obtained from the powder calcined at 1050 degrees c exhibited sensitivity comparable to that of the sintered sample with a significantly shorter response and recovery times. While the sensitivity of the film gradually decreased with a decrease in partial pressure of water vapor (p (H2O)), a noticeable sensitivity was still observed at p(H2O) of 200 Pa. Decrease in conductivity depended logarithmically on the partial pressure of water with the slope of 0.52 that is close to the theoretical value. After several cycles, the reusability test proved an almost unchanged ratio between the impedance value in the dry and the wet Ar atmosphere (p(H2O) = 2.34 kPa), which implied that BcI25, having good stability and sensitivity, is a promising high-temperature humidity sensor.

This study aims to develop a high-performance Pt/c catalyst for the cathode and anode parts of a modified Proton exchange membrane (PEM) fuel cell in nitrogen and argon gases at different microwave radiation powers. The pure and modified forms of the synthesized Pt/c catalysts were characterised using X-ray diffraction (XRD) and scanning electron microscopy (SEM). The XRD results revealed that the crystal structure of the modified Pt/c catalyst was perfect and that the particle size of the catalytic material grew significantly as a consequence of the modification. Furthermore, the durability of PEM fuel cell electrocatalysts was investigated using the cyclic voltammetry (cV) technique. The experimental results indicated that microwave radiation had a significant effect on the particle size of the Pt/c catalyst. Additionally, the modified Pt/c catalyst exhibited a decreased activation potential loss and increased the cathode and anode power density values by 34% and 47%, respectively, compared to the Pt/c catalyst.
