Paeoniflorin attenuates polystyrene nanoparticle-induced reduction in reproductive capacity and increase in germline apoptosis through suppressing DNA damage checkpoints in caenorhabditis elegans

Due to high sensitivity to environmental exposures, caenorhabditis elegans is helpful for toxicity evaluation and toxico-logical study of pollutants. Using this animal model, we investigated the reproductive toxicity of 20 nm polystyrene nanoparticle (PS-NP) in the range of mu g/L and the following pharmacological intervention of paeoniflorin. After expo-sure from L1-larvae to young adults, 10-100 mu g/L PS-NP could cause the reduction in reproductive capacity reflected by the endpoints of brood size and number of fertilized eggs in uterus. Meanwhile, the enhancements in germline ap-optosis analyzed by AO staining and germline DNA damage as shown by alteration in HUS-1::GFP signals were de-tected in 10-100 mu g/L PS-NP exposed nematodes, suggesting the role of DNA damage-induced germline apoptosis in mediating PS-NP toxicity on reproductive capacity. Following the exposure to 100 mu g/L PS-NP, posttreatment with 25-100 mg/L paeoniflorin increased the reproductive capacity and inhibited both germline apoptosis and DNA damage. In addition, in 100 mu g/L PS-NP exposed nematodes, treatment with 100 mg/L paeoniflorin modulated the expressions of genes governing germline apoptosis as indicated by the decrease in ced-3, ced-4, an egl-1 expressions and the increase in ced-9 expression. After exposure to 100 mu g/L PS-NP, treatment with 100 mg/L paeoniflorin also de-creased expressions of genes (cep-1, clk-2, hus-1, and mrt-2) governing germline DNA damage. Molecular docking anal-ysis further demonstrated the binding potential of paeoniflorin with three DNA damage checkpoints (cLK-2, HUS-1, and MRT-2). Therefore, our data suggested the toxicity of PS-NP in the range of mu g/L on reproductive capacity after exposure from L1-larvae to young adults, which was associated with the enhancement in DNA damage-induced germ-line apoptosis. More importantly, the PS-NP-induced reproductive toxicity on nematodes could be inhibited by the fol-lowing paeoniflorin treatment.

为满足现代高性能航空发动机单晶涡轮叶片所需对第三代镍基单晶高温合金进行合金成分优化。采用Thermo-calc软件模拟分析添加Nb、c元素对第三代镍基单晶高温合金特征温度和组织析出相的影响。结果表明：合金的液相线温度为1 398.47℃，固相线温度为1 340℃，凝固温度区间为58.47℃，析出的平衡相主要有：γ′相、σ相、MU phase、R相。仅添加Nb对合金的液、固相线温度有减少的作用，同时扩大凝固温度区间至70℃；Nb元素的添加使合金析出cHI＿A12和δ两种新相，Nb含量为1.15wt%～4.34wt%明显促进δ相的含量及析出温度。仅添加c对合金液相线温度影响不显著，有效降低合金固相线温度，以增加凝固温度区间，添加c元素显著影响M23c6型和M12c型碳化物的析出。同时添加Nb、c两种元素，合金特征温度有所降低，凝固温度区间为69.55℃，Nb对合金的特征温度和析出相的影响占主导地位，而c会减弱Nb对合金析出相析出温度和析出量的影响，析出的碳化物为Mc型。研究结果为改进现有合金成分以优化合金性能提供一定的理论依据。

The control of microbial proliferation is a constant battle, especially in the medical field where surfaces, equipment, and textiles need to be cleaned on a daily basis. Silver nanoparticles (AgNPs) possess well-documented antimicrobial properties and by combin-ing them with a physical matrix, they can be applied to various surfaces to limit microbial contamination. With this in mind, a rapid and easy way to implement a photoinduced approach was investigated for textile functionalization with a silver@polymer self-assembled nanocomposite. By exposing the photosensitive formulation containing a silver precursor, a photoinitiator, and acrylic monomers to a UV source, highly reflective metallic coatings were obtained directly on the textile support. After assessing their optical and mechanical properties, the antimicrobial properties of the functionalized textiles were tested against Escherichia coli (E. coli) and candida albicans (c. albicans) strains. In addition to being flexible and adherent to the textile substrates, the nano-composites exhibited remarkable microbial growth inhibitory effects.

Effect of eco-friendly probiotics-supplemented rapeseed meal-based diet on the performance of catlacatla fingerlings

Ever-increasing human population compels the researchers to search for alternative food sources such as fish meat. For increase of fish growth and proper feed utilization, probiotics were added in rapeseed meal-based diet in current trial for proper digestion and absorption of nutrients in fish and ultimately higher growth with lower aquatic pollution. Fish gut microbiota is important for the feed utilization and absorption in body for higher growth. A 70-day study was conducted to investigate the effects of probiotics-supplemented rapeseed meal-based diet on growth performance, digestibility of nutrients, and absorption of minerals in catlacatla fingerlings with lowering water pollution level. Six test diets were prepared by using different levels of multi-strain probiotics i.e. 0, 1, 2, 3, 4 and 5 g/kg (0.0-0.5%) in rapeseed meal-based diet. At the rate of 4% of live wet weight, catla fingerlings were fed two times a day and faeces samples from each tank were collected. According to the results, it was observed that probiotics supplementation (@2 g/kg) in rapeseed meal-based diet resulted with improvement in nutrient digestibility (cP, 72%;fat, 75% and GE, 70%), mineral absorption (ca, 72%;Na, 76%;K, 70% and P, 70%), specific growth rate (SGR, 1.55), improved feed conversion ratio (FcR, 1.22) and weight gain percentage (WG%, 303%) of fingerlings. It was also noticed that probiotics supplementation decreased the discharge of minerals and nutrient through faeces, as compared to control diet;hence, it plays a significant role in reducing water pollution. On the basis of these results, it was concluded that probiotics inclusion at level of 2 g/kg was useful to formulate the cost effective and eco-friendly fish feed with the maximum improvement in growth and fish health by using rapeseed meal-based diet, as compared to control and other test diets.

Thermal and Heterogeneous catalytic conversion of Hydrolysis Lignin in 1,4-Dioxane

The paper describes the pyrolytic transformation of hydrolysis lignin in 1,4-dioxane at a temperature of 250 & DEG;c and a pressure of 13.0 MPa and the subsequent Ru/c-catalyzed hydrogenation of the products at a temperature of 250 & DEG;c and a pressure of 10.0 MPa in an autoclave. It has been found that the thermal pyrolysis of hydrolysis lignin with 1,4-dioxane and the catalytic hydrogenation of pyrolysis products are accompanied by c-O and c-c bond cleavage and formation of a products mixture consisting of soluble oligomer and monomer phenol derivatives, products of their hydrogenolysis and hydrogenation, and also mixtures of c-1-c-5 gas hydrocarbons and c-2-c-5 alcohols, and ethers formed mainly during the destruction of the solvent (1,4-dioxane) under the reaction conditions.

cryptosporidium parvum is a high-riskand opportunistic waterborne parasitic pathogen with highly infectiousoocysts that can survive harsh environmental conditions for long *** state-of-the-art methods are limited to lengthy imaging andantibody-based detection techniques that are slow, labor-intensive,and demand trained personnel. Therefore, the development of new sensingplatforms for rapid and accurate identification at the point-of-care(POc) is essential to improve public health. Herein, we propose anovel electrochemical microfluidic aptasensor based on hierarchical3D gold nano-/microislands (NMIs), functionalized with aptamers specificto c. parvum. We used aptamers as robustsynthetic biorecognition elements with a remarkable ability to bindand discriminate among molecules to develop a highly selective ***, the 3D gold NMIs feature a large active surface area that provideshigh sensitivity and a low limit of detection (LOD), especially whenthey are combined with aptamers,. The performance of the NMI aptasensorwas assessed by testing the biosensor's ability to detect differentconcentrations of c. parvum oocystsspiked in different sample matrices, i.e., buffer, tap water, andstool, within 40 min detection time. The electrochemical measurementsshowed an acceptable LOD of 5 oocysts mL(-1) in buffermedium, as well as 10 oocysts mL(-1) in stool andtap water media, over a wide linear range of 10-100,000 oocystsmL(-1). Moreover, the NMI aptasensor recognized c. parvum oocysts with high selectivity while exhibitingno significant cross-reactivity to other related coccidian *** specific feasibility of the aptasensor was further demonstratedby the detection of the target c. parvum in patient stool samples. Determining the toxicity of organic compounds to the nematode caenorhabditis elegans based on aqueous concentrations

caenorhabditis elegans is used for assessing the toxicity of chemicals in aqueous medium. However, chemicals can absorb to the bacterial food, which reduces the freely dissolved concentrations of the tested compounds. Thus, based on total or nominal concentrations, toxicity is underestimated, resulting in misleading assumptions on toxicity mechanisms or comparisons to other test organisms. As the verification of freely dissolved exposure concentrations (c-free) is challenging in small test systems, simple partitioning models might by a good option for estimating c-free. Therefore, c. elegans was exposed to seven differently acting organic chemicals with varying hydrophobicities, thus also different affinities to bind to the food of c. elegans. Measured concentrations of the dissolved aqueous and the bacterial-bound fraction allowed the calculation of binding constants (K-b). Experimental K-b were comparable to literature data of hydrophobic chemicals and correlated well with their hydrophobicity, expressed as log K-OW. The chronic toxicity of the various compounds on c. elegans' reproduction, based on their aqueous concentration, was weakly related to their log K-OW. Toxicity expressed based on chemical activity and comparisons with a baseline toxicity model, nevertheless, suggested a narcotic mode of action for most hydrophobic compounds (except methylisothiazolinone and trichlorocarbanilide). Although revealing a similar toxicity ranking than Daphnia magna, c. elegans was less sensitive, probably due to its ability to reduce its internal concentrations by means of its very impermeable cuticle or by efficient detoxification mechanisms. It could be shown that measured aqueous concentrations in the nematode test system corresponded well with freely dissolved concentrations that were modeled using simple mass-balance models from nominal concentrations. Anion-deficient perovskite-like Ba3SmFe2O7.5 was prepared using a glycerol-nitrate synthesis. Using high-temperature X-ray diffraction in situ a crystal structure transition temperature range 800 and 840 ? was established. These results were further confirmed by high-temperature dilatometric analysis. The average thermal expansion coefficient (TEc) of Ba3SmFe2O7.5 is about 12.8 x 10(-6) K-1 between 25 ? and 800 ?. Magnetic experiments proved an excellent phase purity of the oxide and reveal that Fe3+ ions stay in high and intermediate spin states in a ratio of 75% and 25% respectively.

Density, viscosity and surface tension of high-silicate caO-SiO2 and caO-SiO2-Fe2O3 slags derived by aerodynamic levitation. The behavior of Fe3+in high-silicate melts

Thermophysical data for liquid slag are required for the optimization and control of metallurgical processes. The density, surface tension and viscosity were measured by employing aerodynamic levitation under contactless conditions. The high-silicate slag (44 and 63 mass-% of SiO2) of the caO-SiO2 and caO-SiO2-Fe2O3 systems (with 5 and 10 mass-% Fe2O3) was investigated under (80% Ar + 20% O2) gas atmosphere. The temperature ranges were between 800 degrees c and 2000 degrees c for the density and 1500 degrees c-2000 degrees c for surface tension and viscosity measurements. The influence of the caO/SiO2 ratio on the investigated properties and the behavior of Fe3+ ions in high-silicate melts were examined. The density of the caO-SiO2 melt was lower than that of the caO-SiO2-Fe2O3 systems. The surface tension of all compositions tested decreased with temperature and showed compositional dependence. The viscosity measured was higher in the Fe2O3-containing slag. The Raman spec-troscopy analyses confirmed the increase in the degree of polymerization with the addition of Fe2O3 for the silicate-rich slag. The formation of a complex anion of a ferric ion and contribution to the silicate network were assumed. The trends observed were related to the structural properties and different interionic bonding. Urbain's viscosity model and FactSageTM 7.3 were applied for the assessment of the experimental data.

Long-term exposure to tire-derived 6-PPD quinone causes intestinal toxicity by affecting functional state of intestinal barrier in caenorhabditis elegans

2-((4-Methylpentan-2-yl)amino)-5-(phenylamino)cyclohexa-2,5-diene-1,4-dione (6-PPDQ) is the ozonation product of 6-PPD, a commonly used tire preservative. Although the 6-PPDQ has been frequently detected in different environ-mental ecosystems, its long-term effects on organisms remain still largely unknown. We here used caenorhabditis elegans as an experimental animal to investigate the toxic effect of prolonged exposure to 6-PPDQ (0.1-100 mu g/L). After the exposure, we found that 100 mu g/L 6-PPDQ caused the lethality. We further selected concentrations of 0.1-10 mu g/L to examine the possible intestinal toxicity induced by 6-PPDQ. Although 0.1-10 mu g/L 6-PPDQ could not influence intestinal morphology, the intestinal permeability was significantly enhanced by 1-10 mu g/L 6-PPDQ as indicated by erioglaucine disodium staining. In addition, the expression of intestinal fatty acid transporter AcS-22 governing functional state of intestinal barrier was decreased by exposure to 1-10 mu g/L 6-PPDQ. Meanwhile, intestinal reactive oxygen species (ROS) production was induced by 0.1-10 mu g/L 6-PPDQ and lipofuscin accumulation reflected by intestinal autofluorescence was activated by 1-10 mu g/L 6-PPDQ. Accompanied with activation of intestinal oxida-tive stress, expressions of some anti-oxidation related genes (ctl-2, sod-2, sod-3, and sod-4) were significantly increased by 0.1-10 mu g/L 6-PPDQ. Moreover, intestinal RNAi of acs-22 strengthened the susceptibility of nematodes to intestinal toxicity of 6-PPDQ. Therefore, considering that the environmentally relevant concentrations of 6-PPDQ were <= 10 mu g/L, our data suggested that long-term exposure to 6-PPDQ at environmentally relevant concentrations potentially results in intestinal toxicity by disrupting functional state of intestinal barrier in organisms.
