Prostaglandin synthases: Molecular characterization and involvement in prostaglandin biosynthesis

Prostaglandins (PGs) belong to a subclass of eicosanoids and are classified based on the structures of the cyclopentane ring and their number of double bonds in their hydrocarbon structures. PGs are important lipid mediators that are involved in inflammatory response. The biosynthesis of diverse PGs from unsaturated c20 fatty acids containing at least three double bonds such as dihomo-gamma-linoleic acid (20:3(Delta 8Z,11Z,14Z)), arachidonic acid (20:4(Delta 5Z,8Z,11Z,14Z)), and eicosapentaenoic acid (20:5(Delta 5Z,8Z,11Z,14Z,17Z)) is enables by various PG synthases, including prostaglandin H synthase (PGHS), 15-hydroxyprostaglandin dehydrogenase (15-HPGD), PGES, PGDS, PGFS, PGIS, and thromboxane A synthase (TXAS). This review summarizes the biochemical properties, reaction mechanism, and active site details of PG synthases. Because PGs are involved in the immune system, an understanding of PG synthases is important in the design of new anti-inflammatory drugs. The biosynthesis of PGs in various organisms, such as mammals, corals, florideae (a class of red algae), yeast, and fungi, is also introduced. The expression of PG synthases in the microbial systems for the synthesis of PGs is discussed. Now, the biosynthesis of PGs from glucose or glycerol is possible using metabolically engineered cells expressing both unsaturated fatty acid-producing enzymes and PG synthases.

前清蛋白、心肌酶、c 反应蛋白检测在重症手足口病合并心肌损伤患儿诊治中的意义

目的：探讨前清蛋白（PA）、心肌酶、c 反应蛋白（cRP）检测在重症手足口病（HFMD）合并心肌损伤患儿诊治中的应用价值。方法选取183例手足口病患儿作为观察组，其中重症患儿54例，普通患儿129例，另选取100例同期健康体检儿童作为对照组，检测所有研究对象血清 PA、α-丁酸脱氮酶（α-HBDH），肌酸激酶（cK），肌酸激酶同工酶（cK-MB）、cRP 的水平并进行比较。结果观察组血清 PA 水平为（194．37±42．86）mg／L，对照组为（267．15±52．04）mg／L，观察组血清 PA 水平明显低于对照组（t ＝5．942， P ＜0．05）；观察组血清α-HBDH、cK、cK-MB 及 cRP 分别为（261．58±61．40）U ／L、（124．80±67．51）U ／L、（38．79±16．26）U ／L 和（18．06±5．27）mg／L，对照组血清α-HBDH、cK、cK-MB 及 cRP 分别为（112．93±28．56）U ／L、（103．94±60．86）U ／L、（15．17±9．34）U ／L 和（4．82±0．95）mg／L，观察组α-HBDH、cK、cK-MB及 cRP 均明显高于对照组（t ＝8．356、5．087、9．446、10．755，均 P ＜0．05）。重症组血清 PA 水平为（144．92±40．36）mg／L，普通组为（230．89±46．27）mg／L，重症组血清 PA 水平明显低于普通组（t ＝7．035，P ＜0．05）；重症组血清α-HBDH、cK、cK-MB 及 cRP 分别为（301．58±71．40）U ／L、（145．31±67．84）U ／L、（43．66±18．95）U ／L 和（21．86±6．55）mg／L，普通组血清α-HBDH、cK、cK-MB 及 cRP 分别为（162．93±28．56）U ／L、（116．05±61．37）U ／L、（24．75±12．04）U ／L 和（7．25±3．82）mg／L，重症组患儿血清α-HBDH、cK、cK-MB 及cRP 均明显高于普通组（t ＝9．440、6．204、11．038、13．118，均 P ＜0．05）。重症 HFMD 合并心肌损伤患儿血清PA 水平为（131．02±36．54）mg／L，重症组为（149．73±42．81）mg／L，重症 HFMD 合并心肌损伤组血清 PA 水平明显低于重症组（t ＝7．801，P ＜0．05）；重症 HFMD 合并心肌损伤患儿血清α-HBDH、cK、cK-MB 及 cRP 分别为（327．84±74．95）U ／L、（152．92±68．75）U ／L、（45．17±20．68）U ／L 和（23．07±7．46）mg／L，重症组血清α-HBDH、cK、cK-MB 及 cRP 分别为（286．30±70．16）U ／L、（134．80±66．31）U ／L、（41．08±17．23）U ／L 和（20．95±6．20）mg／L，重症 HFMD 合并心肌损伤患儿血清α-HBDH、cK、cK-MB 及 cRP 均明显高于重症HFMD 组（t ＝4．394、5．938、4．077、4．165，均 P ＜0．05）。结论前清蛋白、心肌酶、cRP 检测利于手足口病患儿病情严重程度的判断，且利于患儿心肌损伤的早期诊断。

Mechanical Ventilation: State of the Art

Mechanical ventilation is the most used short-term life support technique worldwide and is applied daily for a diverse spectrum of indications, from scheduled surgical procedures to acute organ failure. This state-of-the-art review provides an update on the basic physiology of respiratory mechanics, the working principles, and the main ventilatory settings, as well as the potential complications of mechanical ventilation. Specific ventilatory approaches in particular situations such as acute respiratory distress syndrome and chronic obstructive pulmonary disease are detailed along with protective ventilation in patients with normal lungs. We also highlight recent data on patient-ventilator dyssynchrony, humidified high-flow oxygen through nasal cannula, extracorporeal life support, and the weaning phase. Finally, we discuss the future of mechanical ventilation, addressing avenues for improvement. (c) 2017 Mayo Foundation for Medical Education and Research.

Genetic and Morphological Differentiation of the Semiterrestrial crab Armases angustipes (Brachyura: Sesarmidae) along the Brazilian coast

The genetic and morphometric population structures of the semiterrestrial crab Armases angustipes from along the Brazilian coast were examined. The influence of the central South Equatorial current on larval dispersal of A. angustipes also was evaluated. Six populations were sampled from estuarine areas in Sao Luis do Maranhao, Maranhao;Natal, Rio Grande do Norte;Maceio, Alagoas;Ilheus, Bahia;Aracruz, Espirito Santo;and Guaratuba, Parana. Patterns of genetic differentiation were assessed using DNA sequence data corresponding to parts of the mitochondrial cytochrome c oxidase subunit 1. Geometric morphometric techniques were used to evaluate morphological variation in shape and size of the carapace and right cheliped propodus. Our results revealed low genetic variability and lack of phylogeographic structure;geometric morphometrics showed statistically significant morphological differentiation and geographic structuring. Our data indicate the absence of possible barriers to gene flow for this mobile species, and no clear correlation of morphological or genetic variation with ocean currents and/or geographic distance. Our results also suggest that historical geological and climatological events and/or possible bottleneck effects influenced the current low genetic variability among the populations of A. angustipes.

Tensile and compression Behaviour of Boron carbide Reinforced 6061Al MMc’s processed through conventional Melt Stirring

Metal matrix composites have large application in various fields like aerospace and automobile applications due to higher strength to weight ratio owing to their excellent combination properties. considering this fact, the present work is taken up with objective of evaluating tensile and compressive behavior of 6061Al-B 4 c composite at room temperature. The composite is prepared via melt stirring method with 9wt% of boron carbide particulates of size 8 microns at processing temperature of 750°c. Microstructural studies of prepared composites were carried out using SEM analysis. Hardness measurements of the composite were carried out using micro-Vickers hardness tester with a load of 250gm and dwell time of 30sec. Tensile and compressive behaviors were studied using universal testing machine at room temperature condition. Microstructural studies revealed fairly uniform distribution of B 4 c particles in 6061Al matrix. Al-B 4 c composite has exhibited better tensile and compressive strength in comparison with matrix alone, which increases its durability and load carrying capacity. Therefore, 6061Al-B 4 c system could be used as replacement of steel and other ferrous materials in structural applications.

Rao's theorem the distribution of the random vector determines the distributions of the random variables up to a change of location. We prove an analogue of this theorem for independent random variables with values in a locally compact Abelian group. We also prove an analogue of a similar ***'s theorem for independent random variables with values in an -adic solenoid. In so doing coefficients of linear forms are continuous endomorphisms of the group.

目的分析cys c、Hcy、Hb A1c、hs-cRP在糖尿病早期肾损伤中的应用价值。方法将我院2012年4月－2014年9月收治的62例糖尿病早期肾损伤患者作为研究组;将同期62例健康体检者作为对照组,检测两组Hb A1c、hs-cRP、cys c、Hcy等指标水平。统计两组研究对象Hb A1c、hs-cRP、cys c、Hcy水平,比较研究组相关指标联合检测阳性率及单独检测阳性率。结果研究组Hb A1c、hs-cRP、cys c、Hcy等指标水平均高于对照组,两组差异显著（P〈0.05）;研究组相关指标单独检测阳性率显著低于联合检测阳性率,差异显著（P〈0.05）。结论 Hb A1c、hs-cRP、cys c、Hcy均可作为糖尿病早期肾损伤的检测指标,且四项指标联合检测可有效提高其诊断的阳性率及准确率,在糖尿病早期肾损伤患者的早期诊断与治疗中具有重要意义。

AIM To uncover novel genetic markers that could contribute to predicting hepatocellular carcinoma(Hcc)susceptibility in caucasians. METHODS The present retrospective case-control study compared genotype frequencies between a cohort of Hcc cases and two,independent,Hcc-free,age/sex-matched control *** Hcc cohort comprised 192 homogeneous patients that had undergone orthotopic liver *** first control group comprised167 patients that were matched to the Hcc cohort for the percentage of hepatitis B(HBV)and/or hepatitis c(HcV)infections.A second control group included192 virus-free,healthy individuals that were used to evaluate the generalizability of the identified predictive *** cases and controls were *** three study populations were characterized with a panel of 31 markers derived from 21 genes that encoded key proteins involved in hepatocarcinogenesis-related *** study end-point was to assess the association between genetic variants and Hcc onset. RESULTS Five genetic markers were identified as risk factors for Hcc in high-risk patients infected with HBV/*** to a dominant model,reduced Hcc risk was associated with three polymorphisms:ERcc1rs3212986(OR=0.46,95%cI:0.30-0.71,P=0.0005),GST-P1 rs1138272(OR=0.41,95%cI:0.21-0.81,P=0.0097),and cYP17A1 rs743572(OR=0.50,95%cI:0.31-0.79,P=0.0032).conversely,according to a recessive model,increased Hcc risk was associated with two polymorphisms:XRcc3 rs1799794(OR=3.70,95%cI:1.02-13.39,P=0.0461)and ABcB1 rs1128503(OR=2.06,95%cI:1.18-3.61,P=0.0111).These associations remained significant in a subgroup analysis,where patients were stratified according to viral status(HBV-or HcV-positive serology).Two variants exhibited a serology-specific effect:ABcB1 rs1128503(OR=4.18,95%cI:1.55-11.29,P=0.0048)showed an effect in the HBV-positive subgroup;and ERcc1 rs3212986(OR=0.33,95%cI:0.18-0.60,P=0.0003)showed an effect in the HcV-positive *** the five markers identified,E

Pegylated interferon α/ribavirin therapy enhances bone mineral density in children with chronic genotype 4 HcV infection

Background:The impact of chronic hepatitis c (cHc) on bone mineral density (BMD) has been well studied in adults with a relative paucity of data in children,especially concerning effect of treatment with pegylated interferon (PEG-IFN) plus ribavirin (RV).In the current work,we assessed prospectively changes in BMD in children with cHc before,during,and after ***:Forty-six consecutive children with noncirrhotic genotype 4 cHc were subjected to dual-energy X-ray absorptiometry at baseline,24 weeks,48 weeks of therapy and 24 weeks after ***,bone mineral content (BMc),and Z score of lumbar spine (L2-L4) were *** pubertal stage,viral load,liver function tests,serum calcium,phosphorus,alkaline phosphatase,parathyroid hormone,and liver histopathology were assessed in all included ***:Thirty (65.2%) patients had normal BMD,10 (21.7%) were at risk for low BMD,and 6 (13.1%) had low BMD for chronological *** with low BMD were significantly older (P=0.001),with higher frequency of delayed puberty than other groups (P=0.002).Baseline densitometric parameters (BMD & BMc) were significantly positively correlated with patients' age,weight,height,body mass index and hemoglobin level;while they were insignificantly correlated with basal viral load,histopathology activity index and fibrosis *** parameters improved significantly on PEG-IFN plus RV treatment,this improvement was found to be sustainable 24 weeks after ***:Low BMD is detectable in a proportion of cHc *** therapy leads to a sustainable increase in BMD.

血清超敏c反应蛋白与老年人缺血性脑卒中的相关性分析

目的：探讨血清超敏 c 反应蛋白与老年人缺血性脑卒中的相关性。方法选择急性脑梗死患者120例为脑梗死组，选择同期体检的健康老年人100例为对照组。对两组血清中的超敏 c 反应蛋白水平进行检测，对不同脑梗死面积、不同神经功能缺损评分患者的血清超敏 c 反应蛋白水平进行分析。结果脑梗死组血清超敏 c 反应蛋白水平为（10．17±3．13）mg／L，显著高于对照组[（0．81±0．24）mg／L]，差异有统计学意义（t ＝29．82，P ＜0．05）；随着脑梗死患者梗死灶容积的增大和神经功能缺损评分的增加，患者血清超敏 c反应蛋白的水平逐渐升高（P ＝0．01）。结论老年缺血性脑卒中患者血清中超敏 c 反应蛋白水平高于健康人，且梗死灶容积越大、神经功能缺损越严重，血清超敏 c 反应蛋白水平越高，对于缺血性脑卒中的早期诊断有一定的借鉴意义。
