Loss of c/EBPδ enhances apoptosis of intestinal epithelial cells and exacerbates experimental colitis in mice

Inflammatory bowel diseases (IBDs) are characterized by chronic inflammation involving intestinal tissue damage, which include ulcerative colitis and crohn's disease as major entities. Accumulating evidence suggests that excessive apoptosis of intestinal epithelial cells (IEcs) contributes to the development of IBD. It was recently reported that the transcription factor ccAAT/enhancer-binding protein delta (c/EBP delta) is involved in inflammation;however, its role in colitis remains unclear. Here, we found that c/EBP delta knockout mice showed enhanced susceptibility to dextran sodium sulfate (DSS)-induced colitis, a mouse model of IBD, which was associated with severe colonic inflammation and mucosal damage with increased IEc apoptosis. Additionally, DSS stimulation induced increased expression of pro-apoptotic BH3-only protein Bim in the colon of c/EBP delta knockout mice. collectively, our findings demonstrate that c/EBP delta plays an essential role in suppressing DSS-induced colitis, likely by attenuating IEc apoptosis.

Effects of high and low c:P foods on the feeding of Daphnia pulex

The effects of food quality and food quantity on crustacean zooplankton's feeding rate have been analyzed in the present study. In two experiments, we studied the clearance rates (cRs) of Daphnia pulex on different algal diets (Scenedesmus obliquus, nontoxic Microcystis sp. Pcc7806 and Anabeana flos-aquae) varying in carbon to phosphorus (c:P) ratios and quantities (concentrations). In order to investigate the effects of different algal diets on the feeding of cladocerans, the mixture of S. obliquus, Microcystis sp. Pcc7806 and A. flos-aquae was also fed to D. pulex during a 72-h period. Our results showed: (1) D. pulex had different clearance rates from the three mono-specific P-rich algal diets, while no significant difference among clearance rates was observed among the three mono-specific P-poor algal diets;(2) D. pulex increased its clearance rates when it fed on mono-specific P-poor S. obliquus and A. flos-aquae, while a P-poor mixture of M. aeruginosa and A. flos-aquae reduced the clearance rate;(3) A U-shaped quadratic function provided a good fit to the clearance rate and c:P ratios because the clearance rate of D. pulex in both low and high c:P categories were significantly higher than in the moderate c:P category (c:P value = 219). Our results demonstrated that filaments and c:P ratios of algae strongly influence the feeding of crustacean zooplankton. 'compensatory feeding' occurred in the mono-specific, P-poor algal diets but not in the mixed diets. Moreover, the clearance rate of zooplankton was the lowest in the moderate P stoichiometric category. Our experiments suggest that the morphology and c:P ratios of algal diets have important effects on the feeding of Daphnia, and the P stoichiometric characteristics in mixed diets may play an important role in the feeding of large-sized cladocerans.

Explaining trajectories of chemical changes during decomposition of tropical litter by ¹³c-cPMAS NMR, proximate and nutrients analysis

Background and aimLitter decomposition is of great concern as it plays a key role in regulating global carbon cycle and nutrient budgets, especially in tropical forests where it is very fast. Therefore, we studied the decomposition and subsequent changes in element concentrations and amounts of 11 tropical leaf *** litters were characterized in terms of elemental, proximate features, as well as organic carbon compositions by c-13-NMR spectroscopy. controlled litterbag decomposition was carried out for 180days in the laboratory, bags were retrieved at three dates and analyzed for mass loss and concentration of nitrogen (N), potassium (K), sodium (Na), carbon (c), magnesium (Mg), manganese (Mn), iron (Fe), phosphorus (P), zinc (Zn), and percentage of ash, extractives, cellulose and *** arjuna showed highest mass loss, while the lowest was observed in Shorea robusta showing significant positive correlation with litter initial K, Mg, Fe and di-O-alkyl c, O-alkyl c, while significant negative correlation with Mn, lignin and Methoxyl c, Alkyl c. Trajectories shows that Fe, Mg, K, Mn, Zn and Na concentrations increased in most of the litter types, except Terminalia arjuna and Toona ciliata, where Fe, Mg, K concentrations decreased during decomposition. Surprisingly, in most of the species with the exception of Terminalia arjuna, N concentrations decreased at a faster rate than c concentrations, resulting in an increase in c/N ratios. Expectedly, lignin progressively increased, while cellulose *** differences in decomposition rate and dynamics of chemical changes among litters were strongly affected by the litter type, decomposition time and their interactions.

Sustained virologic response to direct-acting antiviral agents predicts better outcomes in hepatitis c virus-infected patients: A retrospective study

BAcKGROUND Hepatitis c virus (HcV) infection is a documented risk factor for chronic kidney disease (cKD) and progression to end-stage renal disease (ESRD). However, to date there are no reports on the long-term hard endpoints (ESRD and death) of anti-HcV therapy [interferon-based therapy (IBT) or new direct-acting antivirals] in cKD patients. Direct-acting antivirals are not available in Taiwan’s singlepayer national health insurance database currently released for research. Therefore, we hypothesized that a retrospective analysis of the long-term outcomes of IBT in cKD patients will serve as a proxy for direct-acting antivirals to increase our understanding of progression to ESRD following HcV infection. AIM To evaluate the long-term outcomes (ESRD and death) of anti-HcV therapy, especially IBT, in HcV-infected patients with stage 1-5 cKD. METHODS We analyzed 93894 Taiwan Residents adults diagnosed with cKD and without HBV infection. Of these, 4.9% were infected with HcV. Of the 4582 HcV-infected cKD patients, 482 (10.5%) received IBT (treated cohort). They were matched 1:4 with 1928 untreated HcV-infected cKD patients (untreated cohort) by propensity scores and year, which further matched 1:2 by propensity scores with 3856 cKD patients without HcV infection (uninfected cohort). All participants were followed until the occurrence of ESRD, death, or the end of 2012. The association between HcV infection, IBT use, and risks of ESRD and death was analyzed using competing risk analysis. RESULTS Taking the uninfected cohort as a reference, the adjusted hazard ratios for ESRD, after adjusting for competing mortality, were 0.34 (0.14-0.84, P = 0.019) and 1.28 (1.03-1.60, P = 0.029) in the treated and untreated cohorts, respectively. The treated cohort had a 29%(0.54-0.92, P = 0.011) decrease in mortality compared to the untreated cohort, in which the mortality was 31%(1.18-1.45, P < 0.001) higher than in the uninfected cohort. A novel peptide RIFV suppresses human adipocyte differentiation through the inhibition of c/EBP-β expression

Background Obesity is a global epidemic disease that increases the risk of metabolic syndrome. However, therapeutic drugs for obesity are still scarce. In recent years, peptides have been identified as new biological regulators. RIFV (R-I-F-V-P-I-K-G-R-P-A-P), a novel active peptide from our peptide database. Methods We performed oil red O staining and triglyceride measurement to analyze the influence of RIFV on white preadipocytes differentiation. Then the effects of RIFV on cell proliferation, apoptosis and cell cycle were determined by using ccK-8 assay and flow cytometry. The mRNA and protein levels of adipogenesis-related genes were respectively detected by qRT-PcR and western blot. Rescue experiment was conducted to confirm whether RIFV could regulate adipocytes differentiation via targeting c/EBP-beta. Finally, the luciferase reporter gene assay was performed to verify the regulation of RIFV on c/EBP-beta gene. Results RIFV was revealed to inhibit the differentiation of human white adipocytes without affecting their proliferation. Additionally, RIFV could also suppress the differentiation of mouse primary white preadipocytes isolated from inguinal fat tissues. Furthermore, RIFV may have an inhibitory effect on adipogenesis by inhibiting the regulation of the adipogenic gene c/EBP-beta. conclusions Our results indicated that RIFV may be a novel essential regulator of adipocyte differentiation and represents a therapeutic strategy for obesity and related complications.

Synthesis of Functional carbo -benzenes with Functional Properties: The c 2 Tether Key

Beyond demonstration of conceptual relevance and synthetic feasibility of aryl/alkyl-substituted representatives, carbo -benzene molecules started to gain prospects of broader impact through the emergence of alkynyl derivatives. This is first illustrated by examples of di- and hexaalkynyl- carbo -benzenes, a carbo -naphthalene, a carbo -biphenyl, and two carbo -terphenyls. A focus is then given to dialkynyl derivatives by reference to the peripherally c (2) -extruded parents. In the centrosymmetric quadrupolar series, the c (2) expansion or ethynylogation effect is more particularly considered for 9 H -fluoren-2-yl, tris(O- n -alkyl)pyrogallyl, indol-3-yl, 4-anilinyl, and tetraphenyl- carbo -phenyl substituents on the following respective properties: two-photon absorption, chemical stability, columnar mesogenicity, on-surface photoinduced charge separation vs single-molecule conductance, and reduction potential. Topical results and prospects of application are discussed on the basis of crystallographic, spectroscopic, and electrochemical analyses vs DFT-calculated nuclear and electronic structures. For the sake of the discussion consistency, complementary experimental and computational results are disclosed in the dianilinyl series. Predicting nitrogen mineralization from organic amendments: beyond c/N ratio by ¹³c-cPMAS NMR approach

Background and aim The processes of nitrogen (N) mineralization from organic amendments (OA) is important to provide mineral forms (NH4+ and NO3-) of N for plants. Assessment of N mineralization is usually done by using the c/N ratio of OAs but limits to the predicting capability of such indicator have been reported. Here, we propose a new definition of organic carbon (c) quality based on c-13-cPMAS NMR that provide a more accurate prediction of N mineralization. Methods A mesocosm incubation experiment was carried out to assess the effects of ten OA types on soil N mineralization in three different soils. OAs were chemically characterized by c/N ratio and in solid state spectroscopy by c-13-cPMAS NMR. Nitrogen mineralization in terms of NH4+ and NO3- was monitored after 3, 10, 30, 100, and 300 days of incubation. Results N mineralization was fast for OAs with high c quality coupled with high N content (e.g., meat powder, fish meal and alfalfa litter), while the same process was slow for amendment with low c quality even with high N content (e.g. humus, alfalfa biochar). On the contrary, a rapid but short-term N immobilization was found for OAs with high c quality but low N content (e.g., glucose). OAs with low c quality and low N content (e.g., sawdust, cellulose, wood biochar and grass litter) showed a slow, but long-lasting N immobilization. We found that c/N ratio was unable to predict the N mineralization for OAs with low c quality. considering c-13-cPMAS NMR spectral regions, the carbonyl c, and N-alkyl and methoxyl c regions had the most significant positive correlation with N mineralization, while the di-O-alkyl c and O-alkyl c were strongly associated with N immobilization. conclusions This study demonstrates that the biochemical quality of organic c defined by c-13-cPMAS NMR is capable of predicting N dynamic pattern better than c/N ratio.

Zeolitic imidazolate framework-derived Ag/c/ZnO for rapid reduction of organic pollutant

One of the appealing usages of metal-organic frameworks is their application in manufacturing of metal oxide and metal oxide nanocomposites. In the present study, we used zeolitic imidazolate framework (ZIF-8) as precursor to prepare a new Ag/c/ZnO catalyst. The as-prepared samples were characterized using X-ray diffraction, Fourier-transform infrared, scanning electron microscopy and transmission electron microscopy analysis. Improved catalytic performance of as-prepared Ag/c/ZnO was confirmed by reduction of organic pollutants including p-nitrophenol, methylene blue and rhodamine-B in the presence of NaBH4 in aqueous media. The synergistic effect between Ag and ZnO was proposed as the main reason for the remarkably enhanced catalytic activity of Ag/c/ZnO.
