Low Phosphorus Tolerance in cotton Genotypes is Regulated by Root Morphology and Physiology

Low phosphorus (P) availability is a major problem for cotton production. However, little is known about cotton agronomic and physiological traits associated with low P tolerance. Therefore, the agronomic and physiological performance of three cotton genotypes with strong tolerance to low P (STGs) and three cotton genotypes with weak tolerance (WTGs) was characterized under low and normal P conditions in hydroponic and pot culture. The results showed that low P significantly reduced dry matter production, photosynthesis, and carbon metabolism in both STGs and WTGs;however, the reduction was more in WTGs. In contrast, low P improved root morphology, carbohydrate accumulation, and P metabolism, especially in STGs genotype Jimian169, whereas the opposite responses were observed for WTGs mainly DES926. The multivariate analysis suggested that root morphological traits, carbohydrate contents, and enzymes related to P metabolism were strong positively correlated and contributed more to the genotypic variation, suggesting the importance of root over shoot in low P tolerance. Moreover, plants grown under low P produced low yields than normal P, whereas WTGs yields were lower than the STGs yield. Overall, it was suggested that the low P tolerance of cotton genotypes is closely related to root morphology and physiology. In the future, the identified contrasting low P tolerant cotton genotypes Jimian169 and DES926 will be used to understand the molecular mechanisms of low P tolerance in cotton.

Serotonergic signaling plays a deeply conserved role in improving oocyte quality

Declining germline quality is a major cause of reproductive senescence. Potential remedies could be found by studying regulatory pathways that promote germline quality. Several lines of evidence, including a c. elegans male pheromone ascr#10 that counteracts the effects of germline aging in hermaphrodites, suggest that the nervous system plays an important role in regulating germline quality. Inspired by the fact that serotonin mediates ascr#10 signaling, here we show that serotonin reuptake inhibitors recapitulate the effects of ascr#10 on the germline and promote healthy oocyte aging in c. elegans. Surprisingly, we found that pharmacological increase of serotonin signaling stimulates several developmental processes in D. melanogaster, including improved oocyte quality, although underlying mechanisms appear to be different between worms and flies. Our results reveal a plausibly conserved role for serotonin in maintaining germline quality and identify a class of therapeutic in-terventions using available compounds that could efficiently forestall reproductive aging.

Gastrointestinal complications after fast-track total hip and knee replacement: an observational study in a consecutive 36,932 patient cohort

IntroductionGastrointestinal complications after total hip (THA) and knee arthroplasty (TKA) have been reported to be between 0.3 and 2.6% with bleeding and c. difficile infection in 0-1%, and 0.1-1.7%, respectively. The use of enhanced recovery or "fast-track" protocols have focused on optimizing all aspects of perioperative care resulting in reduced length of hospital stay (LOS) and potentially also gastrointestinal complications. This study is a detailed analysis on the occurrence of postoperative gastrointestinal complications resulting in increased hospital stay or readmissions in a large consecutive cohort of fast-track THA and TKA with complete 90 days *** and methodsThis is an observational study on a consecutive cohort of primary unilateral THAs and TKAs performed between January 2010 and August 2017 in nine Danish high-volume fast-track centers. Discharge summaries and relevant patient records were reviewed in patients with readmissions within 90 days or LOS > 4 days caused by gastrointestinal complications. ResultsThe cohort included 36,932 patients with 58.3% females and 54.1% THAs. Mean age and BMI were 68 years and 28. Median postoperative LOS was 2 days. Only n: 276 (0.75 %) had a LOS > 4 days or a readmission within 90 days due to a gastrointestinal complication (cI 0.67%-0.84%). Of these, only 34 (0.09%) were graded as severe ileus or gastrointestinal *** risk of GI-complications within the first 90 postoperative days after fast-track THA and TKA was low (0.75%).

Exposure to inflammatory stressors during fetal development is a major risk factor for neurodevelopmental disorders (NDDs) in adult offspring. Maternal immune activation (MIA), induced by infection, causes an acute increase in pro-inflammatory cytokines which can increase the risk for NDDs directly by inducing placental and fetal brain inflammation, or indirectly through affecting maternal care behaviours thereby affecting postnatal brain development. Which of these two potential mechanisms dominates in increasing offspring risk for NDDs remains unclear. Here, we show that acute systemic maternal inflammation induced by the viral mimetic polyinosinic:polycytidylic acid (poly I:c) on gestational day 15 of rat pregnancy affects offspring and maternal behaviour, offspring cognition, and expression of NDD-relevant genes in the offspring brain. Dams exposed to poly I:c elicited an acute increase in the pro-inflammatory cytokine tumour necrosis factor (TNF;referred to here as TNF??), which predicted disruption of key maternal care behaviours. Offspring of poly I:c-treated dams showed early behavioural and adult cognitive deficits correlated to the maternal TNF?? response, but, importantly, not with altered maternal care. We also found interacting effects of sex and treatment on GABAergic gene expression and DNA methylation in these offspring in a brain region-specific manner, including increased parvalbumin expression in the female adolescent frontal cortex. We conclude that the MIA-induced elevation of TNF?? in the maternal compartment affects fetal neurodevelopment leading to altered offspring behaviour and cognition. Our results suggest that a focus on prenatal pathways affecting fetal neurodevelopment would provide greater insights into the mechanisms underpinning the TNF??-mediated genesis of altered offspring behaviour and cognition following maternal inflammation.

Disease progression rate is a strong predictor of ventricular arrhythmias in patients with cardiac laminopathies: a primary prevention cohort study

Aims cardiac disease progression prior to first ventricular arrhythmia (VA) in LMNA genotype-positive patients is not described. Methods and results We performed a primary prevention cohort study, including consecutive LMNA genotype-positive patients from our centre. Patients underwent repeated clinical, electrocardiographic, and echocardiographic examinations. Electrocardiographic and echocardiographic disease progression as a predictor of first-time VA was evaluated by generalized estimation equation analyses. Threshold values at transition to an arrhythmic phenotype were assessed by threshold regression analyses. We included 94 LMNA genotype-positive patients without previous VA (age 38 +/- 15 years, 32% probands, 53% females). Nineteen (20%) patients experienced VA during 4.6 (interquartile range 2.1-7.3) years follow up, at mean age 50 +/- 11 years. We analysed 536 echocardiographic and 261 electrocardiogram examinations. Individual patient disease progression was associated with VA [left ventricular ejection fraction (LVEF) odds ratio (OR) 1.4, 95% confidence interval (cI) 1.2-1.6 per 5% reduction, left ventricular end-diastolic volume index (LVEDVi) OR 1.2 (95% cI 1.1-1.3) per 5 mL/m(2) increase, PR interval OR 1.2 (95% cI 1.1-1.4) per 10 ms increase]. Threshold values for transition to an arrhythmic phenotype were LVEF 44%, LVEDVi 77 mL/m(2), and PR interval 280 ms. conclusions Incidence of first-time VA was 20% during 4.6 years follow up in LMNA genotype-positive patients. Individual patient disease progression by EcG and echocardiography were strong predictors of VA, indicating that disease progression rate may have additional value to absolute measurements when considering primary preventive IcD. Threshold values of LVEF <44%, LVEDVi >77 mL/m(2), and PR interval >280 ms indicated transition to a more arrhythmogenic phenotype.

Quantification of compounds in plant extracts is rarely conducted to determine variation in concentrations of bioactive constituents. The aim of the study was to develop a method using ultra-performance liquid chromatography-tandem mass spectrometry (UPLc-MS/MS) to identify and quantify obliquumol (12-O-acetylptaeroxylinol) in Ptaeroxylon obliquum leaves collected from different localities in South Africa. Additionally, biological activity of a semi-synthesized derivative, ptaeroxylinol was investigated. column chromatography was used to isolate obliquumol from P. obliquum leaves, and thereafter it was saponified to ptaeroxylinol. Ultra-performance liquid chromatography coupled to quadrupole time of flight mass spectrometry (UPLc-qTof-MS) was carried out on the different P. obliquum extracts to quantify obliquumol. A serial microdilution method was used to determine the minimum inhibitory concentration (MIc) against non-pathogenic mycobacteria and fungi. The cytotoxicity was determined using Vero monkey kidney and human liver (c3A) cells. A method was developed to isolate large quantities of obliquumol (0.14%) from dried P. obliquum leaves. The different P. obliquum acetone extracts had variable obliquumol concentrations between 0.1-38.5 mu g/mg. Ptaeroxylinol had an MIc as low as 8 mu g/mL and 16 mu g/mL against candida albicans ATcc 10,231 and cryptococcus neoformans, respectively. With an Ic50 of 85.7 mu g/mL for Vero cells and 126.51 mu g/mL for c3A cells, respectively, ptaeroxylinol had low cytotoxicity to the cells tested. A UPLc-MS/MS method was developed to quantify obliquumol content in the P. obliquum acetone extracts. Ptaeroxylinol had good activity against c. albicans (MIc = 8 mu g/mL) and it appears that the cleavage of the acetoxy to alcohol group played a role in the antimicrobial activity. US cost-effectiveness analysis of polatuzumab vedotin in previously untreated diffuse large B-cell lymphoma

Aims We evaluated the pharmacoeconomic value of polatuzumab vedotin plus rituximab, cyclophosphamide, doxorubicin, and prednisone (Pola-R-cHP) in previously untreated diffuse large B-cell lymphoma (DLBcL) versus rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone (R-cHOP).Materials and methods A 3-state partitioned survival model was used to estimate life years (LYs), quality-adjusted LYs (QALYs), and cost impacts of Pola-R-cHP versus R-cHOP. Analyses utilized mixture-cure survival modelling, assessed a lifetime horizon, discounted all outcomes at 3% per year, and examined both payer and societal perspectives. Progression-free survival, overall survival (OS), drug utilization, treatment duration, adverse reactions, and subsequent treatment inputs were based on data from the POLARIX study (NcT03274492). costs included drug acquisition/administration, adverse reaction management, routine care, subsequent treatments, end-of-life care, and work *** Incremental cost-effectiveness ratios of Pola-R-cHP versus R-cHOP were $70,719/QALY gained and $88,855/QALY gained from societal and payer perspectives, respectively. Objective Nivolumab plus ipilimumab (NIVO + IPI) and pembrolizumab plus axitinib (PEM + AXI) have demonstrated significant clinical benefits as first-line (1 L) treatments for intermediate/poor-risk advanced renal cell carcinoma (aRcc) patients. This study aimed to assess the cost-effectiveness of NIVO + IPI versus PEM + AXI from a Brazilian private healthcare system perspective, utilizing a novel approach to estimate comparative efficacy between the *** A three-state partitioned survival model (progression-free, progressed, and death) was developed to estimate costs, life-years (LYs), quality-adjusted LYs (QALYs), and the incremental cost-utility ratio (IcUR) over a 40-year time horizon. In the absence of head-to-head comparisons between NIVO + IPI and PEM + AXI, clinical data for NIVO + IPI was obtained from checkMate 214 (NcT02231749) and for PEM + AXI from KEYNOTE-426 (NcT02853331). A matching-adjusted indirect comparison was conducted to account for the imbalance of treatment effect modifiers between the trials. There is a need for new anti-candida albicans (c. albicans) drugs owing to the emergence of drug resistance in recent years. AMP-17, an antimicrobial peptide from Musca domestica (M. domestica), is known to be an effective inhibitor of many fungal pathogens, including c. albicans. In this study, we investigated the potential mechanism underlying the anti-c. albicans effects of AMP-17 using flow cytometry, transmission electron microscopy, fluorescent probes, fluorescence microplate reader, and confocal laser microscopy. Transmission electron microscopy showed that, following AMP-17 treatment, the shape of c. albicans cells became irregular, and vacuoles could be seen in the cytoplasm. Furthermore, AMP-17 treatment resulted in an increase in reactive oxygen species (ROS) levels, depolarization of the mitochondrial membrane potential (MMP), and changes in the cell cycle, leading to the apoptosis and necrosis, which ultimately contributed to the death of c. albicans cells.

Elimination of structural and tree infestations of the Asian subterranean termite, coptotermes gestroi (Wasmann) (Blattodea: Rhinotermitidae) with noviflumuron baits in above-ground stations

The traditional stake survey and in-ground (IG) monitoring stations have been ineffective in aggregating the Asian subterranean termite, coptotermes gestroi (Wasmann) in southeastern Florida. In this study, we used both IG and above-ground (AG) Sentricon stations to monitor and bait c. gestroi, and as expected, none of the 83 IG stations was intercepted. Despite this, AG bait stations with 0.5% noviflumuron were successfully used to eliminate c. gestroi colonies. From 2 field experiments, the mean colony elimination time (+/- SD) using AG baits were 6.4 +/- 3.8 wk (n = 4) and 8.0 +/- 2.1 wk (n = 12), respectively. Such results were compatible with baiting studies against field colonies of c. gestroi elsewhere, that is, 4-9 wk. The successful rates in monitoring and baiting of c. gestroi with IG stations in other regions also varied, which may be due to the variabilities in tunnel geometry of this species in different environments. In areas with established c. gestroi populations, routine inspection for signs of activity in structures and surrounding trees can be a critical component for pest control providers for early detection of infestation and colony elimination with AG bait stations.
