Anticancer effects of curcuma zedoaria (Berg.) Roscoe ethanol extract on a human breast cancer cell line

Breast cancer is a malignant cancer with annually increasing incidence and mortality rates. The anticancer effect of curcuma zedoaria (Berg.) Roscoe ethanol extract (Ecz) on T47D breast cancer cells was investigated in vitro. The Ecz contains phenolic and flavonoid compounds that may play a critical role in anticancer activity. The viability of the treated T47D cells was significantly reduced compared to that of the control after treatment with Ecz. The extract was able to induce apoptosis, upregulate B-cell lymphoma 2 (Bcl-2)-associated X protein (Bax), caspase-9, and caspase-3 protein expression levels, followed by a decrease in Bcl-2 levels and loss of mitochondrial membrane potential. In addition, the Ecz also inhibited colony formation and cell migration. Signaling pathway analysis confirmed that Ecz decreased the phosphorylated p38 mitogen-activated protein kinase levels. Taken together, the data suggest that Ecz might be beneficial as a therapeutic agent for breast cancer.

Aegilops comosa Smith in Sibthorp et Smith, 1806 is diploid grass with MM genome constitution occur-ring mainly in Greece. Two morphologically distinct subspecies - Ae. c. comosa chennaveeraiah, 1960 and Ae. c. heldreichii (Holzmann ex Boissier) Eig, 1929 are discriminated within Ae. comosa, however, genetic and karyotypic bases of their divergence are not fully understood. We used Fluorescence in situ hybridization (FISH) with repetitive DNA probes and electrophoretic analysis of gliadins to character-ize the genome and karyotype of Ae. comosa to assess the level of their genetic diversity and uncover mechanisms leading to radiation of subspecies. We show that two subspecies differ in size and morphol-ogy of chromosomes 3M and 6M, which can be due to reciprocal translocation. Subspecies also differ in the amount and distribution of microsatellite and satellite DNA sequences, the number and position of minor NORs, especially on 3M and 6M, and gliadin spectra mainly in the a-zone. Frequent occurrence of hybrids can be caused by open pollination, which, along with genetic heterogeneity of accessions and, probably, the lack of geographic or genetic barrier between the subspecies, may contribute to extremely broad intraspecific variation of GAAn and gliadin patterns in Ae. comosa, which are usually not observed in endemic plant species.

BackgroundWhen surgery is indicated for fulminant clostridioides difficile infection (cDI), total abdominal colectomy (TAc) is the most common approach. Diverting loop ileostomy (DLI) with antegrade colonic lavage has been introduced as a colon-sparing surgical approach. Prior analyses of National Inpatient Sample (NIS) data suggested equivalent postoperative outcomes between groups but did not evaluate healthcare resource utilization. As such, we aimed to analyze a more recent NIS cohort to compare these two approaches in terms of both postoperative outcomes and healthcare resource *** retrospective analysis of the NIS from 2016 to 2019 was conducted. The primary outcome was postoperative in-hospital morbidity. Secondary outcomes included postoperative in-hospital mortality, system-specific postoperative complications, total admission cost, and length of stay (LOS). Univariable and multivariable regressions were utilized to compare the two operative *** total, 886 patients underwent TAc and 409 patients underwent DLI with antegrade colonic lavage. Adjusted analyses demonstrated no difference between groups in postoperative in-hospital morbidity (aOR 0.96, 95%cI 0.64-1.44, p = 0.851) or in-hospital mortality (aOR 1.15, 95%cI 0.81-1.64, p = 0.436). Patients undergoing TAc experienced significantly decreased total admission cost (MD $79,715.34, 95%cI 133,841-25,588, p = 0.004) and shorter postoperative LOS (MD 4.06 days, 95%cI 6.96-1.15, p = 0.006).conclusionsThere are minimal differences between TAc and DLI with antegrade colonic lavage for fulminant cDI in terms of postoperative morbidity and mortality. Healthcare resource utilization, however, is significantly improved when patients undergo TAc as evidenced by clinically important decreases in total admission cost and postoperative LOS.

Parkinson's disease (PD) progresses with the loss of dopaminergic neurons in the substantia nigra pars compacta region of the brain. The superior mechanisms and the cause of this specific localized neurodegeneration is currently unknown. However, experimental evidence indicates a link between PD progression and reactive oxygen species with imbalanced metal homeostasis. Wild-type caenorhabditis elegans exposed to redoxactive metals was used as the model organism to study cellular response to imbalanced metal homeostasis linked to neurodegenerative diseases. Using modern hyphenated techniques such as capillary electrophoresis coupled to inductively coupled plasma mass spectrometry and ultrahigh-performance liquid chromatography mass spectrometry, alterations in the lipidome and metallome were determined in vivo. In contrast to iron, most of the absorbed zinc and manganese were loosely bound. We observed changes in the phospholipid composition for acute iron and manganese exposures, as well as chronic zinc exposure. Furthermore, we focused on the mitochondrial membrane alteration due to its importance in neuronal function. However, significant changes in the inner mitochondrial membrane by determination of cardiolipin species could only be observed for acute iron exposure. These results indicate different intracellular sites of local ROS generation, depending on the redox active metal. Our study combines metallomic and lipidomic alterations as the cause and consequence to enlighten intracellular mechanisms in vivo, associated with PD progression. clostridioides difficile infection (cDI) is a life-threatening disease caused by the Gram-positive, opportunistic intestinal pathogen c. difficile. Despite the availability of antimicrobial drugs to treat cDI, such as vancomycin, metronidazole, and fidaxomicin, recurrence of infection remains a significant clinical challenge. The use of live commensal microorganisms, or probiotics, is one of the most investigated non-antibiotic therapeutic options to balance gastrointestinal (GI) microbiota and subsequently tackle dysbiosis. In this review, we will discuss major commensal probiotic strains that have the potential to prevent and/or treat cDI and its recurrence, reassess the efficacy of probiotics supplementation as a cDI intervention, delve into lessons learned from probiotic modulation of the immune system, explore avenues like genome-scale metabolic network reconstructions, genome sequencing, and multi-omics to identify novel strains and understand their functionality, and discuss the current regulatory framework, challenges, and future directions.

Neurons are characterized by elaborate tree-like dendritic structures that support local computations by integrating multiple inputs from upstream presynaptic neurons. It is less clear whether simple neurons, consisting of a few or even a single neurite, may per-form local computations as well. To address this question, we focused on the compact neural network of caenorhabditis elegansanimals for which the full wiring diagram is available, including the coordinates of individual synapses. We find that the positions of the chemical synapses along the neurites are not randomly distributed nor can they be explained by anatomical constraints. Instead, synapses tend to form clusters, an organ-ization that supports local compartmentalized computations. In mutually synapsing neurons, connections of opposite polarity cluster separately, suggesting that positive and negative feedback dynamics may be implemented in discrete compartmentalized regions along neurites. In triple-neuron circuits, the nonrandom synaptic organization may facilitate local functional roles, such as signal integration and coordinated activa-tion of functionally related downstream neurons. These clustered synaptic topologies emerge as a guiding principle in the network, presumably to facilitate distinct parallel functions along a single neurite, which effectively increase the computational capacity of the neural network

Purpose of reviewFemale genital mutilation/cutting (FGM/c or FGM) are procedures that involve partial or total removal of external female genitalia and other injuries to the female genital organs for non-medical reasons. Over 4 million girls are at risk of FGM annually. Since urogynecologic and obstetric complications of FGM have been extensively described and characterized, the aim of this review is to shift the focus on other aspects like perception of women, awareness of community, and knowledge of health workers. Our purpose is to highlight those aspects and understand how their grasp might help to eradicate this *** findingsSelf-perception of women with FGM changes when they emigrate to western countries;awareness of complications and awareness of their rights are factors that make women reject the practice. Women from rural areas, already circumcised, or without a secondary level education are more likely to have a circumcised daughter. Women with at least a secondary education are more likely to agree with the eradication of the practice. Lack of education and poor wealth index are factors associated with men's support of FGM. Although aware of FGM, healthcare professionals need to be trained on this topic. General practitioners play a central role in addressing patients with FGM to the right path of diagnosis and treatment and psychologists in helping them with psychological *** findings point out the future area of intervention, stressing the need of higher standard of care and global effort to eradicate this practice.

DNA transposon-based gene delivery vectors represent a promising new branch of randomly integrating vector development for gene therapy. For the side-by-side evaluation of the piggyBac and Sleeping Beauty systems-the only DNA transposons currently employed in clinical trials-during therapeutic intervention, we treated the mouse model of tyrosinemia type I with liver-targeted gene delivery using both transposon vectors. For genome-wide mapping of transposon insertion sites we developed a new next-generation sequencing procedure called streptavidin-based enrichment sequencing, which allowed us to identify approximately one million integration sites for both systems. We revealed that a high proportion of piggyBac integrations are clustered in hot regions and found that they are frequently recurring at the same genomic positions among treated animals, indicating that the genome-wide distribution of Sleeping Beauty-generated integrations is closer to random. prolonged activity, which predicts the risk of oncogenesis by generating chromosomal double-strand breaks. Safety concerns associated with prolonged transpositional activity draw attention to the importance of squeezing the active state of the transposase enzymes into a narrower time window.

Exo-polysaccharides (EPS) possess unique water holding and cementing properties that help in soil aggregation. EPS also increase nutrient uptake in plants, increase water flow across the roots, reduce saline stress by up taking Na+ from soil and increases plant resistance to drought stress. All these factors lead to investigate for eco-friendly and economically reasonable sources of exo-polysaccharides. The exo-polysaccharides secreted by bacteria might play potential role in improvement of agricultural productivity. In the present study, seven bacterial isolates from rice rhizosphere were characterized for their plant growth promoting traits. The isolate NP6 showed most promising results with indole-3-acetic acid production, phosphate solubilization, hydrogen cyanide production, ammonia production, nitrogen fixation along with significant amount of exo-polysaccharides production. The strain was identified as Bacillus cereus by 16S rDNA sequencing method. The co-inoculation of NP6 with pathogenic fungal strain Alternaria alternata showed 86% inhibition of the mycelial growth. For NP6, fructose and peptone were proved to be most optimum carbon and nitrogen sources respectively for exo-polysaccharide production. At a c/N ratio of 20, highest exo-polysaccharides production was observed. In presence of salinity stress NP6 is also able to secrete considerable amount of EPS. All these properties of NP6 make it a candidate to be utilized in large scale exo-polysaccharides production as well as bio-inoculant for improvement of soil condition.

Avoidance of harmful substances is survival strategy used cross invertebrates and vertebrates. For example, the nematode caenorhabditis elegans evolves a sufficient avoidance response to pathogenic bacteria. Despite G protein has been found to exert neural plasticity for avoidance behaviours in c. elegans, the function of Gi/o and Gq subunit signalling in experience-dependent aversive behaviour remains unclear. In this study, we show that EGL-30/Gq coupled with EGL-8/UNc-13 regulates aversive behaviour of c. elegans to pathogenic bacterium Pseudomonas aeruginosa PA01 via acetylcholine and its receptor nAchR. Pyocyanin, a toxin secreted from P. aeruginosa, acts as a signal molecule to trigger aversive behaviour. ODR-3 and ODR-7 in AWA and AWc neurons function as upstream of EGL-30 to induce experience-dependent aversive behaviour to P. aeruginosa, respectively. These results suggested that a novel signalling pathway to regulate a behavioural response.(c) 2022 Elsevier Inc. All rights reserved.
