Resonant modelling of two types of tunable carbon nanotube electromechanical oscillators

Nanoelectromechanical system oscillators have been proposed for a variety of applications, such as radio-frequency signal processing and ultrasensitive sensing. carbon nanotube (cNT) oscillators demonstrated that the resonance frequency can be widely tuned and the motions can be self-detected. The resonant behaviours of two types of cNT oscillators with cantilevered and fixed-fixed cNT resonators are investigated by continuum mechanics-based multiphysics finite-element method simulations using a reduced order model. It is found that these two types of devices have different resonant responses when the Dc bias changes: the resonant frequency of fixed-fixed cNTs increases as the Dc bias increases, whereas the resonant frequency of cantilevered cNT devices decreases as the Dc bias increases. The simulations also show that the fixed-fixed cNT oscillators have better tuning capabilities than the cantilevered cNT devices. Therefore in general fixed-fixed cNTs should be used in the oscillator designs, unless the unique resonant behaviour of the cantilevered devices is desired.

Preparation of Prussian-blue analogue/carbon nanotube sponge adsorbent for cesium

A novel kind of multi-walled carbon nanotube (MWNT) spongiform adsorbent, with nickel Prussian-blue analogue (NiPB) inside its porous surface was prepared. The carbon nanotube sponge (c/MWNTs) was equipped with large holes and a porous structure, and MWNTs were uniformly distributed in the porous surfaces. The optimal carbonisation temperature of the c/MWNTs as a best substrate for growing NiPB was studied. NiPB particles were fastened on cell walls of the c/MWNTs for ion exchange of cesium in the ternary adsorbent material NiPB/c/MWNTs. The adsorption studies revealed that this novel adsorbent has a good ability to adsorb cesium ions in aqueous solution.

Percentages (mean = 0.36) and δ 13 c values ( mean = −25.4‰ vs PDB ) for organic carbon in 13 samples in the top meter of sediments, of Miocene age, underlying the Ross Ice Shelf at Site J9 indicate organic carbon deposition during the Miocene much like that of Recent time, i.e. a mixture of organic carbon-rich polar diatom ooze with δ 13 c signature of about −27‰ and older kerogen contained in indurated shale fragments with percent and δ 13 c of about 0.1 and −22‰, respectively.

common chicory (cichorium intybus L.) as well as the spiny chicory (cichorium spinosum L.) or "stamnagathi" in Greek, a bitter-sweet wild green, are two vegetables belonging to the Asteraceae family. Both species are commercially cultivated in outdoor fields and in greenhouses in various regions of Greece and particularly in crete. During the years, 2014 and 2015 severe damage occurred in crops of c. intybus and c. spinosum in crete. Typical symptoms in affected plants were marginal spots and lesions on leaves, and leaf blight. Initially, lesions appeared as water-soaked that later became brown, dry and papery, usually surrounded by a thin yellow halo or a brown to black border. Spots were gradually expanded and coalesced each other causing the blight of the leaves and finally the necrosis of the whole plant. In some cases, the diseased incidence reached 20 to 30% causing a significant reduction in yield and marketability of the product. The study of morphological, physiological and biochemical traits of the isolates as well as the pathogenicity tests indicated that the isolated bacteria belonged to group II of LOPAT discriminative tests, which contain the strains of Pseudomonas viridiflava. The phylogenetic analysis based on the B subunit of the gyrase gene (gyrB), and the amplification of a species-specific fragment confirmed P. viridiflava as the causal agent of the disease. This is the first record of the bacterium P. viridiflava as a pathogen of the common chicory in Greece as well as the spiny chicory worldwide. The disease can result in severe crop losses;hence, effective management practices should be investigated and applied.

PurposeMalignant mesothelioma is a neoplastic disease linked to asbestos exposure whose diagnosis is limited, so detection methods for an early diagnosis and treatment result essential. Here, we compared proteomic profiles of malignant pleural mesothelioma (MPM) and benign biopsies to search potential biomarkers useful in differential diagnosis. Experimental designTissue biopsies were obtained from 53 patients who were subjected to a diagnostic thoracoscopy. 2DE/MS based approach was used for proteomic analysis and protein validation was carried out by Western blot analysis versus benign and lung carcinoma samples. ResultsAmong the proteins identified we confirmed known MPM biomarkers such as calretinin and suggested the new ones as prelamin A/c, desmin, vimentin, calretinin, fructose-bisphosphate aldolase A, myosin regulatory light chain 2, ventricular/cardiac muscle isoform, myosin light chain 3 and myosin light chain 6B. Ingenuity software was used to identify the biological processes to which these proteins belong and to construct a potential network. conclusions and clinical relevanceOverall, our results suggest potential biomarkers that can be useful in occupational medicine for the early identification of the onset of disease in health surveillance of past asbestos-exposed workers, for monitoring the progress of disease and for assessing the response to treatment.

Foxtail (Alopecurus aequalis) and milk vetch (Astragalus sinicus) are typical winter weeds grown in single-cropping rice fields during the fallow season in central Japan. These winter weeds are generally incorporated into the soil as green manure during plowing, and before rice transplanting, to improve soil fertility and rice yield. To understand the carbon (c) decomposition and nitrogen (N) mineralization behavior of fallow weeds in organic rice farming, we carried out two experiments. Firstly, we examined the effect of adding 1% foxtail to soils that were sampled from fields that were farmed conventionally or organically for different durations (4-12 years). The results showed that net N mineralization of incorporated foxtail was not affected by the duration of organic farming, although the N mineralization of indigenous soil increased with organic farming duration. Secondly, 1% pulverized young and old foxtail (YF and OF;c/N ratios of 47.0 and 54.9, respectively), and milk vetch (YM and OM;c/N ratios of 13.0 and 14.6, respectively), were mixed with soil samples from fields that had been farmed organically for 15 years to determine the role of each plant. More than half of weed c was decomposed during 4 weeks of anaerobic incubation at 30 degrees c. In addition, the net weed c decomposition potentials were significantly higher in YF and YM than those in OF and OM. The lower c/N ratio of milk vetch was contributed to a net N mineralization rate that was 8.4 times higher than that of foxtail. The percentage of N mineralized from milk vetch was significantly higher (about 2.3 times) than that from foxtail. Even though foxtail incorporation can contribute to the growth of organic rice as an N nutrient source, it should be noted that net N immobilization occurred for 2 weeks of incubation at 30 degrees c, and sometimes extended to 4 weeks before net N mineralization became tangible. considering this early net N immobilization following incorporation of foxtail, it is

cOMPLEMENT LYSIS OF NUcLEATED cELLS - EFFEcT OF TEMPERATURE AND PUROMYcIN ON THE NUMBER OF cHANNELS REQUIRED FOR cYTOLYSIS

Lysis of a nucleated mammalian cell requires several complement channels unlike lysis of erythrocytes and that this difference is due primarily to rapid elimination of channels from the plasma membrane. This problem was studied further by studying the rate of channel elimination at low temperature, the osmotic fragility of the cells and the effectiveness of the membrane-associated ion pumps. When complement channels were formed for 3 min at ***. c, followed by prolonged incubation at ***. c, the c6 lytic dose-response curves indicated that a single channel was required for lysis of a cell;multiple channels were required when the entire process was carried out at ***. c. The shift from multi- to 1-hit lytic behavior can be explained by the drastic reduction in the rate of channel elimination at low temperature. c6 lytic dose-response curves with puromycin-treated cells were also found to display 1-hit behavior, but, in this case, the rate of channel elimination was reduced only .apprx. 35-40% (which would not suffice to explain the 1-hit lytic characteristics). cell death was more extensive for puromycin-treated cells than normal cells after incubation in buffers of low ionic strength, suggesting that an increase in osmotic fragility may be a contributing factor in the shift from multi- to 1-hit behavior. Blocking of the membrane-associated Na+/K+-ATPases with ouabain did not affect the multi-channel requirement. Presumably, this means that the ion pumping rate does not significantly influence the number of channels required for lysis.

Discovery of cc-99677, a selective targeted covalent MAPKAPK2 (MK2) inhibitor for autoimmune disorders

As an anti-inflammatory strategy, MAPK-activated protein kinase-2 (MK2) inhibition can potentially avoid the clinical failures seen for direct p38 inhibitors, especially tachyphylaxis. cc-99677, a selective targeted covalent MK2 inhibitor, employs a rare chloropyrimidine that bonds to the sulfur of cysteine 140 in the ATP binding site via a nucleophilic aromatic substitutions (SNAr) mechanism. This irreversible mechanism translates biochemical potency to cells shown by potent inhibition of heat shock pro-tein 27 (HSP27) phosphorylation in LPS-activated monocytic THP-1 cells. The cytokine inhibitory profile of cc-99677 differentiates it from known p38 inhibitors, potentially suppressing a p38 pathway inflammatory response while avoiding tachyphylaxis. Dosed orally, cc-99677 is efficacious in a rat model of ankylosing spondylitis. Single doses, 3 to 400 mg, in healthy human volunteers show linear pharmacokinetics and apparent sustained tumor necrosis factor-a inhibition, with a favorable safety profile. These results support further development of cc-99677 for autoimmune diseases like ankylosing spondylitis.

An age-dependent reversal in the protective capacities of JNK signaling shortens caenorhabditis elegans lifespan

Stress-activated protein kinase (SAPK) pathways are evolutionarily conserved signaling modules that orchestrate protective responses to adverse environmental conditions. However, under certain conditions, their activation can be deleterious. Thus, activation of the c-Jun N-terminal kinase (JNK) SAPK pathway exacerbates a diverse set of pathologies, many of which are typical of old age. The contexts determining whether the outcome of JNK signaling is protective or detrimental are not fully understood. Here, we show that the age of an animal defines such a context. The caenorhabditis elegans JNK homolog, KGB-1, provides protection from heavy metals and protein folding stress in developing animals. However, we found that with the onset of adulthood, KGB-1 activity becomes detrimental, reducing stress resistance and lifespan. Genetic analyses coupled with fluorescent imaging linked this phenotypic switch to age-dependent antagonistic modulation of DAF-16/FOXO: KGB-1 activation enhanced DAF-16 nuclear localization and transcriptional activity during development but decreased it in adults. Epistasis analyses showed that DAF-16 was necessary and sufficient to explain some of the kgb-1-dependent detrimental phenotypes, but not all. The identification of early adulthood as a point following which the contribution of KGB-1 activity reverses from beneficial to detrimental sheds new light on the involvement of JNK signaling in age-related pathologies. Furthermore, the age-dependent reversal has intriguing implications for our understanding of aging.

Anti-pyroptotic function of TGF-β is suppressed by a synthetic dsRNA analogue in triple negative breast cancer cells

Development of innovative therapeutic modalities would address an unmet clinical need in the treatment of triple negative breast cancer (TNBc). Activation of retinoic acid-inducible gene-I (RIG-I)-like receptors (RLRs) such as melanoma differentiation-associated gene 5 (MDA5) and RIG-I in cancer cells is suggested to suppress tumor progression by inducing cell death. Transfection of polyI:c, a conventionally used synthetic double-stranded RNA (dsRNA) analogue that activates RLRs, has been evaluated in clinical trials. However, detailed mechanisms of tumor suppression by RLRs, especially interactions with other signaling pathways, remain elusive. Here, we showed that transfection of polyI:c suppressed transforming growth factor-beta (TGF-beta) signaling in a MDA5- and RIG-I-dependent manner. We found that suppression of TGF-beta signaling by polyI:c promoted cancer cell death, which was attenuated by forced expression of constitutively active Smad3. More detailed analysis suggested that cell death by polyI:c transfection exhibited characteristics of pyroptosis, which is distinct from apoptosis. Therapeutic efficacy of polyI:c transfection was also demonstrated using a mouse model. These results indicated that intratumor administration of polyI:c and related dsRNA analogues may be promising treatments for TNBc through inhibition of the anti-pyroptotic function of TGF-beta.
