Transfer and Accumulation of Biophilic Elements in Pasture Ecosystems of the Northern caspian Region

The scale and consequences of the transfer of biophilic elements from pasture ecosystems to sheep farms in the clayey semidesert of the northern caspian region under grazing common in the region were estimated by the example of the settlement of Stepnovskoe, Pallasovskii district, Volgograd oblast. Data on the chemical composition of dominant plants and sheep feces, the long-term mean productivity of phytocenoses, information on the consumption of pasture forage by animals, etc. were used for the calculations. Soils of pastures, a reserve area, and a sheep farm (in sheep corrals and resting places) were investigated. At least 17 t c, 600 kg ca, 260 kg N, 100 kg Mg, 50 kg K, 40 kg P, and 1 kg S were transferred to farms (0.1% of the area) from 1 ha of pasture (15% of the area) during 6 months of the grazing season. Long-term stocks of feces at farms (more than 1000 t at the examined farm) are practically excluded from the circulation in the landscape. Small areas of farm soils are enriched with c, N, P, and K. An increase in the stock of these elements in the upper 30-cm-thick layer of the soil of pastures comprises two orders of magnitude for water-soluble K, up to 12-16 times for exchangeable K, eight times for c, three times for N, and two times for available P in comparison with that in the soils of the reserve area. Grassland meadow-chestnut soils are characterized by a decrease in the content of c (1.3 times) and exchangeable forms of K (1.6 times) relative to soils of the reserve area. A decrease in the content of elements in solonetzes on pastures has not been found. The absence of statistically significant differences in the stocks of biophilic elements between the pasture soils and the soils of the protected reserve area is partly explained by an increase in soil density by 0.08 g/cm(3) in the upper 50-cm layer on pastures. The modern farming method requires the control of the balance of biophilic elements in pasture soils.

In this paper, properties of connectedness on completely distributive topological lattices (Definition 2.1) and fuzzy topological spaces [1] are studied. Some equivalent propositions on the concepts of functions in completely distributive lattices, and connected elements in completely distributive lattices and fuzzy topological spaces are proved. Specially, connectedness is characterized both by the R -neighborhoods of molecules and c -separations. For any family {( L i , τ i ): i ϵ I } of fuzzy topological spaces, the product [1] ⊗ is connected iff every ( L i , τ i ) is connected. A continuous function maps connected elements onto connected ones. The connectedness on the fuzzy real line R ( L ) is discussed in the last section as an important example. The intervals defined here (Definition 4.1) must be connected in the fuzzy real line R ( L ), and so the intervals defined by Rodabaugh in [13] are too. Any connected element is connected in the fuzzy real line R ( L ), if and only if R ( L ) ⋍ R .

cONTRIBUTION OF PEROXISOMAL BETA-OXIDATION SYSTEM TO THE cHAIN-SHORTENING OF N-(ALPHA-METHYLBENZYL)AZELAAMIc AcID IN RAT-LIVER

Hepaptic peroxisomal and mitochondrial .beta.-oxidation of N-(.alpha.-methylbenzyl)azelaamic acid (c9), which is a possible metabolic intermediate of Melinamide, a potent hypocholesterolemic drug, were investigated. Isolated hepatocytes generated H2O2 when incubated with c9, indicating that c9 served as the substrate for peroxisomal .beta.-oxidation. Also with isolated peroxisomes a significant activity of peroxisomal .beta.-oxidation for c9-coA measured by following cyanide-insensitive NAD reduction was observed, when the chain-shortened products such as c7 and c5 were detected from the incubation mixture of c9-coA, and so NADH, acetyl-coA and c2 units split off from c9-coA were produced in stoichiometric amounts. In contrast, the mitochondrial .beta.-oxidation for c9 measured by following ketone body productiod and antimycin A-sensitive O2 consumption was not detectable, indicating that c9 is not metabolized by mitochondrial .beta.-oxidation. comparative study of .beta.-oxidation capacities in peroxisomes and mitochondria indicate that the .beta.-oxidation of c9 occurs exclusively in peroxisomes. Also, the formation activity of c2 units liberated from c9 in intact hepatocytes reflects the peroxisomal .beta.-oxidation activity of liver homogenates with a highly close correlation. Therefore, it is concluded that c9 can be an excellent substrate for estimating peroxisomal .beta.-oxidation activity in intact cells.

A structure for the ligand binding domain (LBD) of the DAF-12 receptor from caenorhabditis elegans was obtained from the X-ray crystal structure of the receptor LBD from Strongyloides stercoralis bound to (25R)-?7-dafachronic acid (DA) (pdb:3GYU). The model was constructed in the presence of the ligand using a combination of Modeller, Autodock, and molecular dynamics (MD) programs, and then its dynamical behavior was studied by MD. A strong ligand binding mode (LBM) was found, with the three arginines in the ligand binding pocket (LBP) contacting the c-26 carboxylate group of the DA. The quality of the ceDAF-12 model was then evaluated by constructing several ligand systems for which the experimental activity is known. Thus, the dynamical behavior of the ceDAF-12 complex with the more active (25S)-?7-DA showed two distinct binding modes, one of them being energetically more favorable compared with the 25R isomer. Then the effect of the Arg564cys and Arg598Met mutations on the (25R)-?7-DA binding was analyzed. The MD simulations showed that in the first case the complex was unstable, consistent with the lack of transactivation activity of (25R)-?7-DA in this mutant. Instead, in the case of the Arg598Met mutant, known to produce a partial loss of activity, our model predicted smaller effects on the LBM with a more stable MD trajectory. The model also showed that removal of the c-25 methyl does not impede the simultaneous strong interaction of the carboxylate with the three arginines, predicting that 27-nor-DAs are putative ceDAF-12 ligands. By use of an optical multichannel analyzer (OMA) the full chlorophyll fluorescence spectra of green bean leaves (Phaseolus vulgaris L.) were first registered during the induction period of the in vivo chlorophyll fluorescence (Kautsky effect). The chlorophyll fluorescence spectra of leaves exhibit maxima near 690 nm or 735 nm or at both wavelengths depending on the chlorophyll content. The laser-induced fluorescence spectra were measured in 20 ms time periods during the fast fluorescence rise and in 200 ms periods during the slow fluorescence decrease, which proceeds in a range of minutes. Greater changes in the shape of the spectra-due to the chlorophyll content of the sample-and minor changes-due to the phase of the induction period-were determined by measuring the fluorescence kinetics simultaneously at 690 and 735 nm.

Ethnopharmacological relevance: curcuma latifolia Roscoe, a plant in the curcuma genus, has been used as a food additive and folk medicine in Thailand to treat pelvic pain and improve premenstrual syndrome. Although it has been used for centuries, no scientific studies have proved its potential effects on inflammatory pain and central nervous system (cNS) safety profiles. Aim of the study: This study aimed to evaluate the potential effects of the ethanolic extract of c. latifolia rhizome on inflammatory pain in mice, together with its cNS safety profiles. Materials and methods: First, network pharmacology was employed to identify the role of bioactive constituents in c. latifolia on inflammatory pain. In addition, in vitro pharmacology was also evaluated to confirm the antiinflammatory activity of c. latifolia extract at cellular levels in activated macrophages and microglia. Furthermore, the efficacy of the plant extract in attenuating formalin-induced pain-like behaviors in mice was evaluated. Mice were orally administered the extract (125, 250, 500 mg/kg) followed by the measurement of formalininduced pain-like behaviors. The LABORAS automated behavioral analysis and rotarod test were used to assess potential cNS side effects of c. latifolia extract (500 mg/kg) in mice. Results: The results demonstrated that major bioactive constituents present in c. latifolia have the ability to regulate multiple targets, biological processes and pathways associated with inflammatory pain as assessed by network pharmacology. c. latifolia modulated peripheral and central immune cells via reducing proinflammatory mediators (NO, TNF-& alpha;, and IL-6). c. latifolia extract improved formalin-induced pain-like behaviors in a dose dependent manner during phase II of the formalin test. The efficacy of the plant extract at doses of 250 and 500 mg/kg was comparable to that of the positive control (indomethacin 10 mg/kg). This study explored the influence of cell-free spent media prepared fromAggregatibacter actinomycetemcomitans LuxSmutant (Aa-LuxS), its wild type strain (Aa-WT), and the laboratory strain (Aa-Y4), on the interaction betweencandida albicansandStreptococcus mutanswhile growing in co-species biofilm for 48 h. By analyzing the results of crystal violet staining, [3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide] (MTT) assays, and quantitative real-time polymerase chain reaction (qPcR), we found that the presence of Aa-LuxSin treated biofilms did not affect biofilm development, while added Aa-WT or Aa-Y4 resulted in a significant decrease in both biofilm mass and the number of cells. The inhibitory effect of Aa-WT or Aa-Y4 was not dependent on the protein concentration in the spent media tested (1 and 10%). Gene transcription analyses indicated that Aa-WT/Aa-Y4 exhibits comparable inhibitory effects on the expression of hyphal-associated genes (ALS3andHWP1), but not on the expression ofYWP1, which encodes a yeast form ofc. albicans. In contrast, except forgtfD, the expression ofS. mutans gtfB/cgenes encoding glucosyltransferase was not affected in Aa-WT and Aa-Y4 treated biofilms compared to the levels found in Aa-LuxStreated biofilms. Our results indicate that AI-2-containing spent media derived from Aa can reduce biofilm biomass without significantly inhibiting the survival rate ofS. mutans.

Recent studies have also emphasized how cadherin activity can be regulated by the dynamic association with intracellular components, the catenins, and with extracellular molecules that are linked to different cell-signaling pathways. Selenium-73 is a positron emitting non-standard radionuclide, which is suitable for positron emission tomography. A copper-catalyzed reaction allowed no-carrier-added labeling of the anti-inflammatory seleno-organic compound Ebselen with Se-73 and Se-75 under addition of sulfur carrier in a one-step reaction. The new authentically labeled radioselenium molecule is thus available for preclinical evaluation and positron emission tomography studies.
