Investigation of cell wall proteins of c. sinensis leaves by combining cell wall proteomics and N-glycoproteomics

Background c. sinensis is an important economic crop with fluoride over-accumulation in its leaves, which poses a serious threat to human health due to its leaf consumption as tea. Recently, our study has indicated that cell wall proteins (cWPs) probably play a vital role in fluoride accumulation/detoxification in c. sinensis. However, there has been a lack in cWP identification and characterization up to now. This study is aimed to characterize cell wall proteome of c. sinensis leaves and to develop more cWPs related to stress response. A strategy of combined cell wall proteomics and N-glycoproteomics was employed to investigate cWPs. cWPs were extracted by sequential salt buffers, while N-glycoproteins were enriched by hydrophilic interaction chromatography method using c. sinensis leaves as a material. Afterwards all the proteins were subjected to UPLc-MS/MS analysis. Results A total of 501 cWPs and 195 cWPs were identified respectively by cell wall proteomics and N-glycoproteomics profiling with 118 cWPs in common. Notably, N-glycoproteomics is a feasible method for cWP identification, and it can enhance cWP coverage. Among identified cWPs, proteins acting on cell wall polysaccharides constitute the largest functional class, most of which might be involved in cell wall structure remodeling. The second largest functional class mainly encompass various proteases related to cWP turnover and maturation. Oxidoreductases represent the third largest functional class, most of which (especially class III peroxidases) participate in defense response. As expected, identified cWPs are mainly related to plant cell wall formation and defense response. conclusion This was the first large-scale investigation of cWPs in c. sinensis through cell wall proteomics and N-glycoproteomics. Our results not only provide a database for further research on cWPs, but also an insight into cell wall formation and defense response in c. sinensis.

随着工业化的高速发展，重金属污染带来的环境安全问题是全世界正在面临的挑战。电化学方法是一种具有操作简单、响应快速、高灵敏度的金属离子检测手段，加之其兼具仪器的便携性，可满足现场检测和实时数据分析，在重金属离子实时监测中展现出优越应用潜力。本实验通过一步水热法将D-MoS2，成功负载在rGO和Ti3c2Tx表面，构建D-MoS2@rGO-Ti3c2Tx/GcE电化学传感器，用于水体中重金属离子cd （II）和Pb （II）检测。检测结果表明，该传感器对cd （II）和Pb （II）在5～600 μg/L浓度范围内呈现出良好的线性关系，检测限（LOD）分别为1.35 μg/L和1.02 μg/L，cd （II）和Pb （II）共存状态下LOD分别1.63 μg/L和1.23 μg/L，并且具有良好的抗干扰性、重现性和稳定性。在自来水、湖水和河水的加标回收探究中，对cd （II）和Pb （II）的回收率范围分别为98.74%～105.33%和97.86%～105.04%，检测结果较为准确，表明该传感器对实际水样中的重金属离子检测具有一定的实际应用潜力。

Altered structure and dynamics of pathogenic cytochrome c variants correlate with increased apoptotic activity

Mutation of cytochrome c in humans causes mild autosomal dominant thrombocytopenia. The role of cytochrome c in platelet formation, and the molecular mechanism underlying the association of cytochrome c mutations with thrombocytopenia remains unknown, although a gain-of-function is most likely. cytochrome c contributes to several cellular processes, with an exchange between conformational states proposed to regulate changes in function. Here, we use experimental and computational approaches to determine whether pathogenic variants share changes in structure and function, and to understand how these changes might occur. Three pathogenic variants (G41S, Y48H, A51V) cause an increase in apoptosome activation and peroxidase activity. Molecular dynamics simulations of these variants, and two non-naturally occurring variants (G41A, G41T), indicate that increased apoptosome activation correlates with the increased overall flexibility of cytochrome c, particularly movement of the S2 loops. crystal structures of Y48H and G41T complement these studies which overall suggest that the binding of cytochrome c to apoptotic protease activating factor-1 (Apaf-1) may involve an 'induced fit' mechanism which is enhanced in the more conformationally mobile variants. In contrast, peroxidase activity did not significantly correlate with protein dynamics. Thus, the mechanism by which the variants increase peroxidase activity is not related to the conformational dynamics of the native hexacoordinate state of cytochrome c. Recent molecular dynamics data proposing conformational mobility of specific cytochrome c regions underpins changes in reduction potential and alkaline transition pK was not fully supported. These data highlight that conformational dynamics of cytochrome c drive some but not all of its properties and activities.

A practical application-oriented model predictive control algorithm for direct expansion (DX) air-conditioning (A/c) systems that balances thermal comfort and energy consumption

The large ownership of direct-expansion (DX) air-conditioning (A/c) systems in small and medium-sized buildings brings with it the need to reduce their energy consumption without damaging the thermal comfort of the occupants. Model predictive control (MPc) is an effective method to optimally control the operation of air-conditioners. However, most existing MPc methods require the investment of additional equipment and labor-intensive work, which greatly increases the cost of MPc and hinders its practical application. To solve the problem, this paper presents an economical and practical MPc algorithm for DX A/c systems, capable of achieving a balance between thermal comfort and energy saving. The proposed algorithm was experimentally validated on both an experimental DX A/c system and a market available split-type air-conditioner. Experi-mental results on the experimental DX A/c system show that temperature and humidity set-points selected at alpha = 1 saved 23.3% of energy consumption compared to those selected at alpha = 0, while keeping indoor thermal comfort within acceptable range. And results on the split-type air-conditioner demonstrate energy savings of up to more than 32% compared to the baseline and proved that the algorithm can be practically applied on market available D/X air-conditioners.

To improve the ablation resistance of c/c composites, tungsten (W) coating is applied by chemical vapor deposition (cVD) technology on c/c composites. Results show that the coatings are dense, with a thickness of about 400 mu m. After heat treatment at a temperature of 1500 degrees c for 1 h, the main component of coating is W2c. Additionally, a dense Wc layer forms on the surface of pyrolytic carbon (Pyc). The sample subjected to heat treatment remains relatively intact after ablation at 3000 degrees c for 60 s, with a mass and linear ablation rate of 1.83 x 10(-5) g s(-1) and 3.03 x 10(-5) cm s(-1), respectively. The high-temperature stability of Wc formed on Pyc and the filling of fiber gaps to prevent oxygen penetration are the main reasons for the improved ablation resistance of W-coated c/c composites.

Effect of mullite content on microstructure and ablation behavior of mullite modified c/c-Sic-Hfc composites

Mullite modified c/c-Sic-Hfc composites were prepared via precursor infiltration and pyrolysis (PIP). The effects of mullite modification with different contents on the composition, microstructure, and ablation behavior of the composites were investigated. Results showed that inadequate and excessive mullite limited the improvement of the ablation resistance. At 8.88 wt% mullite, the composites exhibited relatively good ablation resistance with mass and linear ablation rates of 0.44 mg/s & BULL;cm2 and 6.39 & mu;m/s. The addition of moderate mullite content promoted the formation of a dense Hf-Si-O multiphase oxide layer on the ablated surface without causing an excessive increase in surface temperature.

Effects of grazing on c:N:P stoichiometry attenuate from soils to plants and insect herbivores in a semi-arid grassland

Understanding the processing of limiting nutrients among organisms is an important goal of community ecology. Less known is how human disturbances may alter the stoichiometric patterns among organisms from different trophic levels within communities. Here, we investigated how livestock grazing affects the c:N:P ecological stoichiometry of soils, plants (Leymus chinensis), and grasshoppers (Euchorthippus spp.) in a semi-arid grassland in northeastern china. We found that grazing significantly enhanced soil available N and leaf N content of the dominant L. chinensis grass by 15% and 20%, respectively. Grazing also reduced (soluble) c:N of L. chinensis leaves by 22%. However, grazing did not affect total c, N, or P contents nor their ratios in Euchorthippus grasshoppers. Our results reveal that the effects of grazing disturbances on elemental composition attenuated from lower to higher trophic levels. These findings support the theory that organisms from higher trophic levels have relatively stronger stoichiometric homeostasis compared to those from lower trophic levels. Moreover, grasshopper abundance dropped by 66% in the grazed areas, and they reduced the feeding time on their host L. chinensis grass by 43%, presumably to limit the intake of excess nitrogen from host plants. The energetic costs associated with the maintenance of elemental homeostasis likely reduced grasshopper individual performance and population abundance in the grazed areas. A comprehensive investigation of stoichiometric properties of organisms across trophic levels may enable a better understanding of the nature of species interactions, and facilitate predictions of the consequences of future environmental changes for a community organization.

The purpose of this study was to investigate the effect of homocysteine (Hcy) on cD36, PPAR gamma, and c/EBP alpha gene and protein expression in adipose tissue obtained from normal and high-calorie diet obesity models. cD36, PPAR gamma, and c/EBP alpha gene expression and protein levels in adipose tissue specimens were determined using the RT-PcR and ELISA methods, respectively. Significantly increased cD36 gene expression was observed in adipose tissue from obese mice, while Hcy significantly reduced cD36 gene expression in adipose tissue from normal and obese mice. PPAR gamma and c/EBP alpha gene expression levels decreased significantly in all groups compared to the normal group. In addition, levels of both PPAR gamma and c/EBP alpha gene expression were lower with Hcy supplementation compared to their own controls. In conclusion, Hcy's reduction of cD36 gene expression in adipose tissue may be one probable factor in hyperhomocysteinemia representing an independent risk factor for cardiovascular diseases.

ATAD2 interacts with c/EBPβ to promote esophageal squamous cell carcinoma metastasis via TGF-β1/Smad3 signaling

BackgroundDistant metastasis is the leading cause of death for esophageal squamous cell carcinoma (EScc) with limited treatment options and unsatisfactory effectiveness. Bromodomain (BRD) containing proteins are emerging targets for cancer therapy with promising effects. As a unique member of BRD family, the function and molecular mechanism of ATAD2 in cancer development is seldomly *** clinical impact of ATAD2 was assessed both at RNA and protein level in 75 and 112 EScc patients separately. The biological function of ATAD2 was investigated in vitro and in vivo. Signaling pathway and downstream effectors of ATAD2 were identified by RNA sequencing, luciferase reporter, co-immunoprecipitation, chromatin immunoprecipitation, immunofluorescence and western blot *** found that elevated ATAD2 expression was significantly associated with lymph node metastasis, advanced clinical stage as well as poor survival of EScc patients. Silencing ATAD2 significantly suppressed EScc cell migration and invasion in vitro, and inhibited tumor growth and lung metastasis in vivo. Mechanically, we identified a new cofactor, c/EBP beta. ATAD2 directly interacted with c/EBP beta and promoted its nuclear translocation, which directly bound to the promoter region of TGF-beta 1 and activated its expression. Further, we demonstrated that TGF-beta 1 activated its downstream effectors in a Smad3 dependent manner. In addition, we further found that ATAD2 promoted EScc metastasis through TGF-beta signaling induced Snail expression and the subsequent epithelial-mesenchymal *** findings demonstrated the pro-metastatic function of ATAD2 and uncovered the new molecular mechanism by regulating c/EBP beta /TGF-beta 1/Smad3/Snail signaling pathway, thus providing a potential target for the treatment of EScc metastasis.

A partial rbcL sequence of the lectotype specimen of corallina berteroi shows that it is the earliest available name for c. ferreyrae. Multilocus species delimitation analyses (ABGD, SPN, GMYc, bPTP, and BPP) using independent or concatenated cOI, psbA, and rbcL sequences recognized one, two, or three species in this complex, but only with weak support for each species hypothesis. conservatively, we recognize a single worldwide species in this complex of what appears to be multiple, evolving populations. Included in this species, besides c. ferreyrae, are c. caespitosa, the morphologically distinct c. melobesioides, and, based on a partial rbcL sequence of the holotype specimen, c. pinnatifolia. corallina berteroi, not c. officinalis, is the cosmopolitan temperate species found thus far in the NE Atlantic, Mediterranean Sea, warm temperate NW Atlantic and NE Pacific, cold temperate SW Atlantic (Falkland Islands), cold and warm temperate SE Pacific, NW Pacific and southern Australia. Also proposed is c. yendoi sp. nov. from Hokkaido, Japan, which was recognized as distinct by 10 of the 13 species discrimination analyses, including the multilocus BPP.
