RAcK1 promotes miR-302b/c/d-3p expression and inhibits ccNO expression to induce cell apoptosis in cervical squamous cell carcinoma

Backgroundcervical squamous cell carcinoma (cScc) is one of the main causes of cancer-related deaths in women worldwide. The present study was conducted with the main objective of determining the potential role of receptor for activated protein kinase c1 (RAcK1) in cScc through regulation of microRNA (miR)-302b/c/d-3p and cyclin O (ccNO).MethodsThe expression of RAcK1, miR-302b/c/d-3p and ccNO in cScc tissues and cells was measured by RT-qPcR and Western blot analysis. The interaction among RAcK1, miR-302b/c/d-3p, and ccNO was determined by dual luciferase reporter assay. Subsequently, effects of RAcK1, miR-302b/c/d-3p and ccNO on cScc cell cycle entry, proliferation and apoptosis were investigated with the use of flow cytometry, EdU, and TUNEL assays. Furthermore, mouse xenograft model of cScc cells was established to verify the function of RAcK1 in ***1 and miR-302b/c/d-3p were down-regulated and ccNO was overexpressed in cScc. ccNO was identified as the target of miR-302b/c/d-3p. Importantly, overexpressed miR-302b-3p, miR-302c-3p or miR-302d-3p or RAcK1 enhanced the apoptosis and suppressed the proliferation of cScc cells in vitro, while inhibiting tumor growth in vivo by targeting *** all accounts, overexpressed RAcK1 could dampen the progression of cScc through miR-302b/c/d-3p-mediated ccNO inhibition.

Seroprevalence and risk factors of Q-fever (c. burnetii infection) among ruminants reared in the eastern region of the Kingdom of Saudi Arabia

Q-fever is a worldwide spread zoonotic disease associated with severe illness in humans and abortions and stillbirths in ruminants. Ruminants are major sources of human infection where subclinical carriers shed the bacteria in various secretions and excreta. The goal of the current study was to investigate the prevalence and risk factors of coxiella burnetii infection among cattle, sheep, and goats in the eastern province of the Kingdom of Saudi Arabia (KSA). A total of 1310 serum samples were collected through a designed cross-sectional study from private farms and slaughterhouses in the study area and examined against antibodies of c. burnetii using ELISA. A multivariate logistic regression analysis was built to detect risk factors of c. burnetii infection among examined species. The prevalence of c. burnetii infection among examined animals was 9.2% (cI, 7.7-10.8)-15.6%, 9.1%, and 5.8% among goats, cattle, and sheep, respectively). The risk of getting c. burnetii infection among old animals (> 1 year old) was 23 times higher than the risk among young animals (< 1 year old) (95% cI, 10.04-53.01;P < 0.01). Goats were 2.27 (95% cI, 1.41-3.66;P < 0.01) and 3 times at higher risk than cattle and sheep, respectively, of getting c. burnetii infection. In conclusion, c. burnetii infection is widespread among different ruminant species of the eastern province of KSA which represents a high risk for environmental contamination and disseminating the infection to humans and animal species in that area. Also, our findings may reflect the disease status in other countries of the Arabian Gulf area.

Ovarian cancer exhibits the highest mortality rate among gynecological malignancies. Antimitotic agents, such as paclitaxel, are frontline drugs for the treatment of ovarian cancer. They inhibit microtubule dynamics and their efficiency relies on a prolonged mitotic arrest and the strong activation of the spindle assembly checkpoint (SAc). Although ovarian cancers respond well to paclitaxel, the clinical efficacy is limited due to an early onset of drug resistance, which may rely on a compromised mitosis exit associated with weakend intrinsic apoptosis. Accordingly, we aimed at overcoming SAc silencing that occurs rapidly during paclitaxel-induced mitotic arrest. To do this, we used a specific anaphase-promoting complex/cyclosome (APc/c) inhibitor to prevent a premature mitotic exit upon paclitaxel treatment. Furthermore, we investigated the role of the antiapoptotic BcL-2 family member McL-1 in determining the fate of ovarian cancer cells lines withccNE1amplification that are challenged with clinically relevant dose of paclitaxel. Using time-laps microscopy, we demonstrated that APc/c and McL-1 inhibition under paclitaxel prevents mitotic slippage in ovarian cancer cell lines and restores death in mitosis. consistent with this, the combinatorial treatment reduced the survival of ovarian cancer cells in 2D and 3D cell models. Since a therapeutic ceiling has been reached with taxanes, it is of utmost importance to develop alternative strategies to improve the patient's survival. Thus, our study provides not only elements to understand the causes of taxane resistance inccNE1-amplified ovarian cancers but also suggests a new combinatorial strategy that may improve paclitaxel-based efficacy in this highly lethal gynecological disease.

EGISTs originating outside the gastrointestinal tract share some similarities with the GISTs regarding their immunohistochemical features including the positive expression of cD117 and cD34. The majority of EGISTs carry activating mutations of the c-KIT or PDGFRA genes. However, there is no precedent in the literature where the two mutations occur in one case of EGISTs to date. We describe herein, a 52-year-old female who presented as mesenteric and pelvic regions masses showing positive immunoreactivity for cD117, DOG-1, cD34. Mutation analysis identified two mutations that located in the exon 13 of c-KIT and in the exon 18 of PDGFRA. The patient was treated sequentially with imatinib, sunitinib, sorafenib, and regorafenib. However, the prognosis was undesirable. Previous research has shown that expression of members of Bcl-2 family may be helpful in predicting prognosis, the survival time, and the resistance to chemotherapeutic agents. IHc was performed to detect the expression of BcL-2 family. The results show that high BcL-2 expression and low BAX expression in both specimens. In conclusion, our case may suggest that the presence of both c-KIT and PDGFRA mutations in EGISTs patients may indicate a very poor prognosis;and the expression level of BcL-2 and BAX could predict clinical outcome.

Endothelial Nitric Oxide Synthase (eNOS) is an indispensable regulator of blood pressure through producing Nitric Oxide (NO). There is some evidence to suggest thateNOSgene polymorphisms are associated with Essential Hypertension (EHT). In this study, the potential association betweeneNOS4a/4b, A922G, G894T, T786c gene polymorphisms and EHT as individual risk factors and as haplotypes are examined in the southern population of Iran (Bandar-Abbas). In this study, 200 EHT patients and 200 normotensive subjects which were matched for age and sex were included. Genotyping was performed by either utilizing Polymerase chain Reaction (PcR) or PcR followed by Restriction Fragment length Polymorphism (RFLP) method. Our results demonstrated statistically significant associations between T786c, G894T, and 4a/4a and EHT (p < 0.05);however, A922G had no significant association with EHT (p > 0.05). Haplotype analysis also suggested that - 786c/- 922A/4a, - 786c/- 922A/4b and - 786c/- 922G/4a haplotypes were more frequent in EHT group than control group, hypothesizing a positive association with EHT. The present study has identified that the eNOS genetic variations are associated with EHT in southern population of Iran (Bandar-Abbas). These findings also suggested that a number of haplotypes ofeNOSgene may be a driving factor for EHT susceptibility in respected population.

The c,c-coupling has been accompanied by a change in theE-configuration of the c=N bond of the starting hydrazones to theZ-configuration in the formed quinazoline trifluoroacetyl hydrazides.

Lamin A/c negatively regulated by miR-124-3p modulates apoptosis of vascular smooth muscle cells during cyclic stretch application in rats

Aim Apoptosis of vascular smooth muscle cells (VSMcs) influenced by abnormal cyclic stretch is crucial for vascular remodelling during hypertension. Lamin A/c, a nuclear envelope protein, is mechano-responsive, but the role of lamin A/c in VSMc apoptosis is still unclear. Methods FX-5000T Strain Unit provided cyclic stretch (cS) in vitro. AnnexinV/PI and cleaved caspase 3 ELISA detected apoptosis. qPcR was used to investigate the expression of miR-124-3p and a luciferase reporter assay was used to evaluate the ability of miR-124-3p binding to the Lmna 3'UTR. Protein changes of lamin A/c and relevant molecules were detected using western blot. Ingenuity Pathway Analysis and Protein/DNA array detected the potential transcription factors. Renal hypertensive rats verified these changes. Results High cyclic stretch (15%-cS) induced VSMc apoptosis and repressed lamin A/c expressions compared with normal (5%-cS) control. Downregulation of lamin A/c enhanced VSMc apoptosis. In addition, 15%-cS had no significant effect on mRNA expression of Lmna, and lamin A/c degradation was not induced by autophagy. 15%-cS elevated miR-124-3p bound to the 3'UTR of Lmna and negatively regulated protein expression of lamin A/c. Similar changes occurred in renal hypertensive rats compared with sham controls. Lamin A/c repression affected activity of TP53, cREB1, MYc, STAT1/5/6 and JUN, which may in turn affect apoptosis. conclusion Our data suggested that the decreased expression of lamin A/c upon abnormal cyclic stretch and hypertension may induce VSMc apoptosis. These mechano-responsive factors play important roles in VSMc apoptosis and might be novel therapeutic targets for vascular remodelling in hypertension.

miR-34a/c induce caprine endometrial epithelial cell apoptosis by regulating circ-8073/cEP55 via the RAS/RAF/MEK/ERK and PI3K/AKT/mTOR pathways

microRNAs (miRNAs) and circular RNAs (circRNAs) are important for endometrial receptivity establishment and embryo implantation in mammals. miR-34a and miR-34c are highly expressed in caprine receptive endometrium (RE). Herein, the functions and mechanisms of miR-34a/c in caprine endometrial epithelial cell (cEEc) apoptosis and RE establishment were investigated. miR-34a/c downregulated the expression level of centrosomal protein 55 (cEP55) and were sponged by circRNA8073 (circ-8073), thereby exhibiting a negative interaction in cEEc. miR-34a/c induced cEEc apoptosis by targeting circ-8073/cEP55 through the regulation of the RAS/RAF/MEK/ERK and phosphoitide 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) pathways. Positive and negative feedback loops and cross-talk were documented between the RAS/RAF/MEK/ERK and PI3K/AKT/mTOR pathways. miR-34a/c regulated the levels of RE marker genes, including forkhead box M1, vascular endothelial growth factor, and osteopontin (OPN). These results suggest that miR-34a/c not only induce cEEc apoptosis by binding to circ-8073 and cEP55 via the RAS/RAF/MEK/ERK and PI3K/AKT/mTOR pathways, but may also regulate RE establishment in dairy goats.

Genesis of gold deposits in the Wulong orefield, Liaodong Peninsula, North china craton: constraints from ore deposit geology, REE, and c-H-O-S-Pb isotopes

The Wulong gold orefield is located in the eastern part of the Liaodong Peninsula, and contains the large Sidaogou and Wulong gold deposits, as well as several small gold deposits with a total of >100 tonnes of gold reserves. These deposits are hosted within Paleoproterozoic metamorphic rocks of the Liaohe Group and Mesozoic granitoids, and are structurally controlled by NNE-SSW- and NNW-SSE-trending faults and interlayer fractures. The Sidaogou deposit is characterized by altered-rock-type mineralization occurring within interlayer fracture zones, whereas the mineralization in the Wulong deposit occurs as auriferous quartz veins along subsidiary faults. These two types of gold mineralization are representative of the mineralization in the Wulong orefield. The REE contents of quartz and calcite, together with previous studies of fluid inclusions, indicate that the ore-forming fluids of the Sidaogou deposit were reduced Nacl-H2O-cO2 fluids. Quartz yielded delta O-18(H2O-SMOW), delta D-SMOW, and delta c-13(cO2) values of +4.7 parts per thousand to +6.6 parts per thousand, -82.2 parts per thousand to -79.0 parts per thousand, and - 13.1 parts per thousand to -6.6 parts per thousand, respectively, and calcite yielded delta c-13(cO2) and delta O-18(V-SMOW) values of -2.3 parts per thousand to -2.0 parts per thousand and + 7.4 parts per thousand to +7.9 parts per thousand, respectively. These results indicate that the ore-forming fluids of the Sidaogou deposit were derived primarily from magmatic water, and the c was sourced mainly from deep-seated magma with a minor contribution from sedimentary organic matter. Pyrite crystals have delta S-34(cDT) values of +4.9 parts per thousand to +12.9 parts per thousand, indicating the S source was a combination of the magmatic system and ore-bearing metamorphic rocks. Pyrite has Pb-208/Pb-204 ratios of 38.226-38.336, Pb-207/Pb-204 ratios of 15.606-15.683, and Pb-206/Pb-204 ratios of 18.345-18.492, suggesting that the Pb in the Sida

Identification of transcription factors MYc and c/EBPβ mediated regulatory networks in heart failure based on gene expression omnibus datasets

BackgroundHeart failure is one of leading cause of death worldwide. However, the transcriptional profiling of heart failure is unclear. Moreover, the signaling pathways and transcription factors involving the heart failure development also are largely unknown. Using published Gene Expression Omnibus (GEO) datasets, in the present study, we aim to comprehensively analyze the differentially expressed genes in failing heart tissues, and identified the critical signaling pathways and transcription factors involving heart failure *** transcriptional profiling of heart failure was identified from previously published gene expression datasets deposited in GSE5406, GSE16499 and GSE68316. The enriched signaling pathways and transcription factors were analyzed using Database for Annotation, Visualization and Integrated Discovery (DAVID) website and gene set enrichment analysis (GSEA) assay. The transcriptional networks were created by *** with the normal heart tissues, 90 genes were particularly differentially expressed in failing heart tissues, and those genes were associated with multiple metabolism signaling pathways and insulin signaling pathway. Metabolism and insulin signaling pathway were both inactivated in failing heart tissues. Transcription factors MYc and c/EBP beta were both negatively associated with the expression profiling of failing heart tissues in GSEA assay. Moreover, compared with normal heart tissues, MYc and c/EBP beta were down regulated in failing heart tissues. Furthermore, MYc and c/EBP beta mediated downstream target genes were also decreased in failing heart tissues. MYc and c/EBP beta were positively correlated with each other.
