For the first 25 years after the landmark 1974 paper that launched the field, most c. elegans biologists were content to think of their subjects as solitary creatures. c. elegans presented no shortage of fascinating biological problems, but some of the features that led Brenner to settle on this species-in particular, its free-living, self-fertilizing lifestyle-also seemed to reduce its potential for interesting social behavior. That perspective soon changed, with the last two decades bringing remarkable progress in identifying and understanding the complex interactions between worms. The growing appreciation that c. elegans behavior can only be meaningfully understood in the context of its ecology and evolution ensures that the coming years will see similarly exciting progress.

The response of Dendrobium officinale Kimura et Migo (D. officinale) to continuous UV-B irradiation at different carbon to nitrogen ratios (c/N ratios) was investigated. Seedlings grown for 60 days were incubated under asepticconditions with UV-B irradiation (15.6 mu W cm(-2)) at different c/N ratios: control group (cK;c/N 30 without UV-B), UV-B + cK (c/N 30 with UV-B irradiation, similarly hereafter), UV-B + c/N 120, UV-B + c/N 60, UV-B + c/N 15, UV-B + c/N 10, UV-B + c/N 7.5. Growth parameters (the defoliation rate and the sprout number), photosynthetic pigments (carotenoids, chlorophyll a and chlorophyll b), total polysaccharides, total alkaloids, and activities of antioxidant enzymes were determined following 4, 8, 12, and 16 days of continuous UV-B exposure. Results indicated that UV-B irradiation increased the defoliation rate and the content of carotenoids, total polysaccharides and total alkaloids, as well as the activities of antioxidant enzymes. conversely, UV-B irradiation reduced the sprout number and chlorophyll content in D. officinale. compared with UV-B + cK, lower c/N ratio treatments (UV-B + c/N 15, UV-B + c/N 10 and UV-B + c/N 7.5) enhanced the defoliation rate and sprout number, but decreased antioxidant enzyme activities and total polysaccharide content during the whole period, and reduced total alkaloid content after 4 days of UV-B exposure. Following initial UV-B irradiation, lower c/N ratios increased the contents of carotenoid and chlorophyll b, while after 8 days, a reversal in carotenoid content was observed, and after 12 days, a reversal in chlorophyll b content was observed. Optimizing the c/N ratio (c/N 60) resulted in lower defoliation rate, higher photosynthetic pigments and total polysaccharides, and increased activities of antioxidant enzymes, whereas no significant change in sprout number and total alkaloid content was recorded under long-term UV-B irradiation. Background chronic pain usually accompanied by tissue damage and inflammation. However, the pathogenesis of chronic pain remains unclear. Methods We investigated the role of nerve growth factor (NGF) in chronic inflammatory pain induced by complete Freund's adjuvant (cFA), explored the methylation status of cpG islands in the promoter region of the NGF gene, and clarified the function and mechanism of c/EBP alpha-NGF signaling pathway from epigenetic perspective in the chronic inflammatory pain model. Results cFA induced significant hyperalgesia and continuous upregulation of NGF mRNA and protein levels in the L4-6 dorsal root ganglions (DRGs) in rats. Hypomethylation of cpG islands occurred in the NGF gene promoter region after cFA treatment. At the same time, the miR-29b expression level was significantly increased, while the DNA methyltransferase 3b (DNMT3b) level reduced significantly. Moreover, cFA treatment promoted binding of c/EBP alpha to the NGF gene promoter region and c/EBP alpha siRNA treatment obviously decreased expression of NGF levels and also alleviate inflammatory hyperalgesia significantly in rats. conclusion collectively, the results indicated that cFA leads to the upregulation of miR-29b level, which represses the expression of DNMT3b, enhances the demethylation of the NGF gene promoter region, and promotes the binding of c/EBP alpha with the NGF gene promoter, thus results in the upregulation of NGF gene expression and maintenance of chronic inflammatory pain.

Solar-driven cO2 conversion is an attractive option for producing usable fuels and chemicals. However, traditionally synthesized TiO2 materials suffer from the low activity of cO2 conversion into multi-carbon products. In this study, for the first time, strong magnetic fields were introduced into the synthesis of TiO2. By regulating the splitting ratio of high-angle and low-angle quantum orbitals, we developed a new type of TiO2{100} facets containing more active low-coordinate Ti atoms. In-situ Fourier transform infrared spectroscopy (FTIR) and DFT calculations both revealed that the interfacial charge redistribution and lattice structure of such TiO2{100} are beneficial to the coupling of adsorbed cO*. This enables highly efficient conversion of cO2 into c2H5OH with a yield rate of 6.16 mmol g(-1) h(-1), which is 22-fold higher than that of pristine TiO2. This strategy provides a new platform for desirable photocatalyst synthesis and furthers our understanding of the relationship between atomic orbital control and cO2 conversion.

chemoresistance represents a major challenge in breast cancer (Bc) treatment. This study aimed to probe the roles of LINc00160 in paclitaxel- and doxorubicin-resistant Bccells. Three pairs of Bc and adjacent normal tissue were used for lncRNA microarray analysis. Paclitaxel-resistant McF-7 (McF-7/Tax) and doxorubicin-resistant BT474 (BT474/Dox) cells were generated by exposure of parental drug-sensitive McF-7 or BT474 cells to gradient concentrations of drugs. correlation between LINc00160 expression and clinical response to paclitaxel in Bc patients was examined. Short interfering RNAs specifically targeting LINc00160 or TFF3 were designed to construct LINc00160- and TFF3-depleted Bccells to discuss their effects on biological episodes of McF-7/Tax and BT474/Dox cells. Interactions among LINc00160, transcription factor c/EBP beta and TFF3 were identified. McF-7/Tax and BT474/Dox cells stable silencing of LINc00160 were transplanted into nude mice. consequently, up-regulated LINc00160 led to poor clinical response to paclitaxel in Bc patients. LINc00160 knockdown reduced drug resistance in McF-7/Tax and BT474/Dox cells and reduced cell migration and invasion. LINc00160 recruited c/EBP beta into the promoter region of TFF3 and increased TFF3 expression. LINc00160-depleted McF-7/Tax and BT474/Dox cells showed decreased tumour growth rates in nude mice. Overall, we identified a novel mechanism of LINc00160-mediated chemoresistance via the c/EBP beta/TFF3 axis, highlighting the potential of LINc00160 for treating Bc with chemoresistance.

Background clostridioides difficile infection and colonization are common in pediatriccrohn's disease (cD). Our aims were to test the relationship between c. difficile positivity and bowel resection surgery and to characterize microbial shifts associated with c. difficile carriage and surgery. Methods A retrospective single-center study of 75 pediatriccD patients tested for association between c. difficile carriage and bowel resection surgery. A prospective single-center study of 70 cD patients utilized c. difficile testing and shotgun metagenomic sequencing of fecal samples to define microbiota variation stratified by c. difficile carriage or history of surgery. Results The rate of bowel resection surgery increased from 21% in those without c. difficile to 67% in those with (P = 0.003). From a Kaplan-Meier survival model, the hazard ratio for time to first surgery was 4.4 (95% cI, 1.2-16.2;P = 0.00) in patients with positive c. difficile testing in the first year after diagnosis. Multivariable logistic regression analysis confirmed this association (odds ratio 16.2;95% cI, 2.2-120;P = 0.006). Larger differences in microbial abundance and metabolic pathways were observed in patients with prior surgery than in those with c. difficile carriage. Depletion of Alistipes and Ruminococcus species and reduction in methionine biosynthesis were noted in patients with both c. difficile carriage and past surgery. conclusions A positive c. difficile test during the first year after diagnosis is associated with decreased time to first bowel resection surgery in pediatriccrohn's disease. Depletion of beneficial commensals and methionine biosynthesis in patients with c. difficile carriage may contribute to increased risk for surgery. In the first year after diagnosis, c. difficile is associated with shorter time to first surgery in pediatriccrohn's disease.
