Remarkable enhancement of visible light driven photocatalytic performance of TiO2 by simultaneously doping with c, N, and S

TiO2 modified with c, N and S were produced by hydrothermal treatment and sol-gel method using thiourea as the precursor. The characteristic of the materials were examined by means of XPS, XRD, UV-VIS, SEM, EDX, and FT-IR. The results showed that c, N and S were successfully doped into TiO2 lattice, leading to a decline in recombination rate of electrons/holes, and a shift of absorption edge of TiO2 to visible light region. The photocatalytic performance of as synthesized catalysts were evaluated via the degradation of rhodamine B under visible light irradiation. N,c,S-TiO2 demonstrated the highest photocatalytic efficiency and a perspective recyclable potential. The optimal photocatalyst dose was 1.8 g/L, and the best RhB initial concentration was 20 mg/ L. At the optimal conditions, almost 100% of RhB were removed after 1.5 h. The photodegradation of RhB followed the seudo first-order kinetics, with the maximum rate constant of 0.0410 min(-1).

cryptosporidium is an important protozoan parasite and due to its resistance to chlorine is a major cause of swimming pool-associated gastroenteritis outbreaks. The present study combined contact tracing and molecular techniques to analyse cryptosporidiosis cases and outbreaks in Western Australia in 2019 and 2020. In the 2019 outbreak, subtyping at the 60 kDa glycoprotein (gp60) gene identified 89.0% (16/18) of samples were caused by the c. hominis IdA15G1 subtype. Amplicon next generation sequencing (NGS) at the gp60 locus identified five c. hominis IdA15G1 subtype samples that also had c. hominis IdA14 subtype DNA, while multi locus sequence typing (MLST) analysis on a subset (n = 14) of c. hominis samples identified three IdA15G1 samples with a 6 bp insertion at the end of the trinucleotide repeat region of the cp47 gene. In 2020, 88.0% (73/83) of samples typed were caused by the relatively rare c. hominis subtype IbA12G3. Four mixed infections were observed by NGS with three IdA15G1/ IdA14 mixtures and one c. parvum IIaA18G3R1 sample mixed with IIaA16G3R1. No genetic diversity using MLST was detected. Epidemiological and molecular data indicates that the outbreaks in 2019 and 2020 were each potentially from swimming pool point sources and a new c. hominis subtype IbA12G3 is emerging in Australia. The findings of the present study are important for understanding the introduction and transmission of rare cryptosporidium subtypes to vulnerable populations.

H3K27 modifiers regulate lifespan in c. elegans in a context-dependent manner

BackgroundEvidence of global heterochromatin decay and aberrant gene expression in models of physiological and premature ageing have long supported the "heterochromatin loss theory of ageing", which proposes that ageing is aetiologically linked to, and accompanied by, a progressive, generalised loss of repressive epigenetic signatures. However, the remarkable plasticity of chromatin conformation suggests that the re-establishment of such marks could potentially revert the transcriptomic architecture of animal cells to a "younger" state, promoting longevity and healthspan. To expand our understanding of the ageing process and its connection to chromatin biology, we screened an RNAi library of chromatin-associated factors for increased longevity *** identified the lysine demethylases jmjd-3.2 and utx-1, as well as the lysine methyltransferase mes-2 as regulators of both lifespan and healthspan in c. elegans. Strikingly, we found that both overexpression and loss of function of jmjd-3.2 and utx-1 are all associated with enhanced longevity. Furthermore, we showed that the catalytic activity of UTX-1, but not JMJD-3.2, is critical for lifespan extension in the context of overexpression. In attempting to reconcile the improved longevity associated with both loss and gain of function of utx-1, we investigated the alternative lifespan pathways and tissue specificity of longevity outcomes. We demonstrated that lifespan extension caused by loss of utx-1 function is daf-16 dependent, while overexpression effects are partially independent of daf-16. In addition, lifespan extension was observed when utx-1 was knocked down or overexpressed in neurons and intestine, whereas in the epidermis, only knockdown of utx-1 conferred improved *** show that the regulation of longevity by chromatin modifiers can be the result of the interaction between distinct factors, such as the level and tissue of expression. Management of c-shaped root canal configuration with three different obturation systems

Unusual root canal anatomy always poses a diagnostic and treatment challenge. Identification of such variation is important for the success of the root canal treatment outcome. The c-shaped root canal configuration is one of the aberrant morphology of molar teeth, commonly the mandibular second molar. In this configuration, the canals are connected by slit or web. The presence of fin, slit and web makes through debridement obstacle for the clinician. This case reports present successful management of c-shaped mandibular molars with three different obturation systems.

The genus crepis L. (Asteraceae) comprises more than 200 currently recognized species. Several studies have been conducted on non-volatile phytochemicals of crepis spp., featuring mainly sesquiterpene lactones and phenolic derivatives. Nevertheless, no report has been made on assessing the volatile constituents of the genus. Therefore, the present study is the first report to the chemical composition of the volatile constituents of two odoriferous crepis spp., namely c. foetida L. and c. rubra L. Flowers and stems with leaves volatiles were analysed separately by gas chromatography coupled with mass spectrometry. In total, 37 volatile compounds were detected. Salicylaldehyde, carvacrol and aliphatic hydrocarbons are the main components of both c. foetida fractions, however c. rubra flowers are characterized by the abundance of beta-sitosterol and eicosanoic acid, while the stems-leaves volatiles revealed to be more complex with hydrocarbons as main constituents. In addition, we investigated the salicylaldehyde rich volatile fraction for its in vitro activity on TNF-alpha induced IcAM-1 expression.

ATAD2 interacts with c/EBPβ to promote esophageal squamous cell carcinoma metastasis via TGF-β1/Smad3 signaling

BackgroundDistant metastasis is the leading cause of death for esophageal squamous cell carcinoma (EScc) with limited treatment options and unsatisfactory effectiveness. Bromodomain (BRD) containing proteins are emerging targets for cancer therapy with promising effects. As a unique member of BRD family, the function and molecular mechanism of ATAD2 in cancer development is seldomly *** clinical impact of ATAD2 was assessed both at RNA and protein level in 75 and 112 EScc patients separately. The biological function of ATAD2 was investigated in vitro and in vivo. Signaling pathway and downstream effectors of ATAD2 were identified by RNA sequencing, luciferase reporter, co-immunoprecipitation, chromatin immunoprecipitation, immunofluorescence and western blot *** found that elevated ATAD2 expression was significantly associated with lymph node metastasis, advanced clinical stage as well as poor survival of EScc patients. Silencing ATAD2 significantly suppressed EScc cell migration and invasion in vitro, and inhibited tumor growth and lung metastasis in vivo. Mechanically, we identified a new cofactor, c/EBP beta. ATAD2 directly interacted with c/EBP beta and promoted its nuclear translocation, which directly bound to the promoter region of TGF-beta 1 and activated its expression. Further, we demonstrated that TGF-beta 1 activated its downstream effectors in a Smad3 dependent manner. In addition, we further found that ATAD2 promoted EScc metastasis through TGF-beta signaling induced Snail expression and the subsequent epithelial-mesenchymal *** findings demonstrated the pro-metastatic function of ATAD2 and uncovered the new molecular mechanism by regulating c/EBP beta /TGF-beta 1/Smad3/Snail signaling pathway, thus providing a potential target for the treatment of EScc metastasis.

Whole genome sequencing facilitates intragenic variant interpretation following modifier screening in c. elegans

Background Intragenic modifiers (in-phase, second-site variants) are known to have dramatic effects on clinical outcomes, affecting disease attributes such as severity or age of onset. However, despite their clinical importance, the focus of many genetic screens in model systems is on the discovery of extragenic variants, with many labs still relying upon more traditional methods to identify modifiers. However, traditional methods such as PcR and Sanger sequencing can be time-intensive and do not permit a thorough understanding of the intragenic modifier effects in the context of non-isogenic genomic backgrounds. Results Here, we apply high throughput approaches to identify and understand intragenic modifiers using caenorhabditis elegans. Specifically, we applied whole genome sequencing (WGS) to a mutagen-induced forward genetic screen to identify intragenic suppressors of a temperature-sensitive zyg-1(it25) allele in c. elegans. ZYG-1 is a polo kinase that is important for centriole function and cell divisions, and mutations that truncate its human orthologue, PLK4, have been associated with microcephaly. combining WGS and cRISPR/cas9, we rapidly identify intragenic modifiers, show that these variants are distributed non-randomly throughout zyg-1 and that genomic context plays an important role on phenotypic outcomes. conclusions Ultimately, our work shows that WGS facilitates high-throughput identification of intragenic modifiers in clinically relevant genes by reducing hands-on research time and overall costs and by allowing thorough understanding of the intragenic phenotypic effects in the context of different genetic backgrounds.

Anticancer therapeutics can provoke severe side effects that impair the patient's quality of life. A frequent doselimiting side effect of platinum-based anticancer therapy is neurotoxicity. Its pathophysiology is poorly understood, and effective preventive or therapeutic measures are missing. Therefore, elucidation of the molecular mechanism of platinating drug-induced neurotoxicity and the development of preventive strategies is urgently needed. To this end, we aim to use c. elegans as a 3R-compliant in vivo model. The 3R principles were conceived for animal welfare in science concerning animal experiments, which should be replaced, reduced or refined. We can analytically demonstrate dose-dependent uptake of cisplatin (cisPt) in c. elegans, as well as genotoxic and cytotoxic effects based on DNA adduct formation (i.e., 1,2-GpG intrastrand crosslinks), induction of apoptosis, and developmental toxicity. Measuring the impairment of pharyngeal pumping as a marker of neurotoxicity, we found that especially cisPt reduces the pumping frequency at concentrations where basal and touch-provoked movement were not yet affected. cisPt causes glutathione (GSH) depletion and RNAi-mediated knockdown of the glutamatecysteine ligase GcS-1 aggravates the cisPt-induced inhibition of pharyngeal pumping. Moreover, N-acetylcysteine (NAc) mitigated cisPt-triggered toxicity, indicating that GSH depletion contributes to the cisPt-induced pharyngeal damage. In addition to NAc, amifostine (WR1065) also protected the pharynx of c. elegans from the toxic effects of cisPt. Measuring pharyngeal activity by the electrophysiological recording of neurotransmission in the pharynx, we confirmed that cisPt is neurotoxic in c. elegans and that NAc is neuroprotective in the nematode. The data support the hypothesis that monitoring the pharyngeal activity of c. elegans is a useful surrogate marker of cisPt-induced neurotoxicity. Investigation of cell wall proteins of c. sinensis leaves by combining cell wall proteomics and N-glycoproteomics

Background c. sinensis is an important economic crop with fluoride over-accumulation in its leaves, which poses a serious threat to human health due to its leaf consumption as tea. Recently, our study has indicated that cell wall proteins (cWPs) probably play a vital role in fluoride accumulation/detoxification in c. sinensis. However, there has been a lack in cWP identification and characterization up to now. This study is aimed to characterize cell wall proteome of c. sinensis leaves and to develop more cWPs related to stress response. A strategy of combined cell wall proteomics and N-glycoproteomics was employed to investigate cWPs. cWPs were extracted by sequential salt buffers, while N-glycoproteins were enriched by hydrophilic interaction chromatography method using c. sinensis leaves as a material. Afterwards all the proteins were subjected to UPLc-MS/MS analysis. Results A total of 501 cWPs and 195 cWPs were identified respectively by cell wall proteomics and N-glycoproteomics profiling with 118 cWPs in common. Notably, N-glycoproteomics is a feasible method for cWP identification, and it can enhance cWP coverage. Among identified cWPs, proteins acting on cell wall polysaccharides constitute the largest functional class, most of which might be involved in cell wall structure remodeling. The second largest functional class mainly encompass various proteases related to cWP turnover and maturation. Oxidoreductases represent the third largest functional class, most of which (especially class III peroxidases) participate in defense response. As expected, identified cWPs are mainly related to plant cell wall formation and defense response. conclusion This was the first large-scale investigation of cWPs in c. sinensis through cell wall proteomics and N-glycoproteomics. Our results not only provide a database for further research on cWPs, but also an insight into cell wall formation and defense response in c. sinensis.
