Scope Manganese (Mn) and zinc (Zn) are not only essential trace elements, but also potential exogenous risk factors for various diseases. Since the disturbed homeostasis of single metals can result in detrimental health effects, concerns have emerged regarding the consequences of excessive exposures to multiple metals, either via nutritional supplementation or parenteral nutrition. This study focuses on Mn-Zn-interactions in the nematode caenorhabditis elegans (c. elegans) model, taking into account aspects related to aging and age-dependent neurodegeneration. Methods and Results chronic co-exposure of c. elegans to Mn and Zn increases metal uptake, exceeding levels of single metal exposures. Supplementation with Mn and/or Zn also leads to an age-dependent increase in metal content, a decline in overall mRNA expression, and metal co-supplementation induced expression of target genes involved in Mn and Zn homeostasis, in particular metallothionein 1 (mtl-1). Studies in transgenic worms reveal that mtl-1 played a prominent role in mediating age- and diet-dependent alterations in metal homeostasis. Metal dyshomeostasis is further induced in parkin-deficient nematodes (Parkinson's disease (PD) model), but this did not accelerate the age-dependent dopaminergic neurodegeneration. conclusions A nutritive overdose of Mn and Zn can alter interactions between essential metals in an aging organism, and metallothionein 1 acts as a potential protective modulator in regulating homeostasis.

Wear analysis of Al7075 alloyed with nano B 4 c: A taguchi approach

The paper discusses about a novel combination of Aluminium 7075 reinforced with Boron carbide (B4c) in nano form in terms of wear behavior. This type of composites are popular now a day's addressing various needs of industry especially where the weight, mechanical and wear properties of the alloy have a significant effect on end application. The present investigational work involves base metal Aluminium 7075 reinforced with B4c in nano form in various percentages by weight (3 to 12%). The specimens were prepared by stir casting process as per ASTM standards and were tested for their behavioral aspects towards wear using pin on disc. The taguchi approach was applied to optimize the process parameters for the case. The process has four factors, four levels considering, composition of nano B4c as 3%,6%, 9% and 12%, load variations in steps are 10, 20, 30 and 40 N, speeds selected are 100, 200, 300, and 400 rpm respectively and sliding distances preferred are 500,1000,1500 and 2000 m to study the effects on wear. Post experimentation analysis established that the presence of nano B4c particles has shown considerable reduction in wear with the samples.

Phenolic acid metabolites of polyphenols act as inductors for hormesis in c. elegans

Introduction: Aging represents a major risk factors for metabolic diseases, such as diabetes, obesity, or neurodegeneration. Polyphenols and their metabolites, especially simple phenolic acids, gained growing attention as a preventive strategy against age-related, non-communicable diseases, due to their hormetic potential. Using caenorhabditis elegans (c. elegans) we investigate the effect of protocatechuic, gallic, and vanillic acid on mitochondrial function, health parameters, and the induction of potential hormetic pathways. Methods: Lifespan, heat-stress resistance and chemotaxis of c. elegans strain P X 627, a specific model for aging, were assessed in 2-day and 10-day old nematodes. Mitochondrial membrane potential (Delta Psi m) and ATP generation were measured. mRNA expression levels of longevity and energy metabolism-related genes were determined using qRT-PcR. Results: All phenolic acids were able to significantly increase the nematodes lifespan, heat-stress resistance and chemotaxis at micromolar concentrations. While Delta Psi m was only affected by age, vanillic acid (VA) significantly decreased ATP concentrations in aged nematodes. Longevity pathways, were activated by all phenolic acids, while VA also induced glycolytic activity and response to cold. conclusion: While life- and health span parameters are positively affected by the investigated phenolic acids, the concentrations applied were unable to affect mitochondrial performance. Therefore we suggest a hormetic mode of action, especially by activation of the sirtuin-pathway.

Toxocariasis is still a neglected parasitic disease worldwide and much about its biology and diagnosis has yet to be understood. The migration of third stage larvae via bloodstream suggests a potential use of molecular tools in diagnosis as well to deepen the knowledge about its migration behaviors. conventional PcR was applied in serum and tissue samples from BALB/c mice infected with 5 and 500 embryonated eggs. Blood samples were collected at 15, 30, 60, 90 and 120 days post-infection. Organs were excised at 170 days post infection. There was no DNA amplification in serum samples in any group or day post-infection;contrarily, tissue samples showed DNA amplification. These results also support a continuous larval migration after and/or simultaneously with the neurotropic-myotropic phase. Thus, molecular tools might be useful as a differential diagnosis method, but do not replace immunodiagnostics techniques.

Explaining inter-lab variance in c. elegans N2 lifespan: Making a case for standardized reporting to enhance reproducibility

The nematode caenorhabditis elegans is a powerful model organism for studying cell development, apoptosis, neuronal circuits, and aging. The isolate N2 is recognized by the c. elegans community as the reference wild-type strain. Interestingly, the lifespan of presumably isogenic c. elegans N2 worms-even when grown under comparable conditions-varies significantly amongst distinct laboratories. This hinders the inter-laboratory comparability of c. elegans lifespan data and raises questions regarding data interpretation and reproducibility. Here, we hypothesized slight alterations in experimental design and worm handling could explain the observed discrepancies. To test this hypothesis, we collected and assessed data from over 1000 published c. elegans N2 lifespan assays as well as corresponding methodological meta-data. We find that mean N2 lifespans range from approximately 7 days to upwards of 35 days, despite laboratories disclosing seemingly comparable experimental conditions. We further demonstrate that, in addition to temperature, the use of the chemical sterilizer 5-fluoro-2 '-deoxyuridine (FUDR) may change N2 lifespan. Additionally, we observed differences in average N2 lifespan from experiments originating from distinct geographic locations, indicating a potential effect of location-specific factors on experimental outcomes. Taken as a whole, our work indicates the sum of many small, rather than a few critical, differences in experimental conditions may account for the observed variance in N2 lifespan. We also find that the absence of standardized experimental methods and the insufficient disclosure of experiment details in the peer-reviewed literature limits the inter-lab comparability of published results. We thus propose the establishment of a succinct reporting standard for c. elegans lifespan experiments to increase the reliability and reproducibility, and thus scientific value, of these studies.

ORMDL3 regulates poly I:c induced inflammatory responses in airway epithelial cells

Background Oroscomucoid 3 (ORMDL3) has been linked to susceptibility of childhood asthma and respiratory viral infection. Polyinosinic-polycytidylic acid (poly I:c) is a synthetic analog of viral double-stranded RNA, a toll-like receptor 3 (TLR3) ligand and mimic of viral infection. Methods To investigate the functional role of ORMDL3 in the poly I:c-induced inflammatory response in airway epithelial cells, ORMDL3 knockdown and over-expression models were established in human A549 epithelial cells and primary normal human bronchial epithelial (NHBE) cells. The cells were stimulated with poly I:c or the Th17 cytokine IL-17A. IL-6 and IL-8 levels in supernatants, mRNA levels of genes in the TLR3 pathway and inflammatory response from cell pellets were measured. ORMDL3 knockdown models in A549 and BEAS-2B epithelial cells were then infected with live human rhinovirus (HRV16) followed by IL-6 and IL-8 measurement. Results ORMDL3 knockdown and over-expression had little influence on the transcript levels of TLR3 in airway epithelial cells. Time course studies showed that ORMDL3-deficient A549 and NHBE cells had an attenuated IL-6 and IL-8 response to poly I:c stimulation. A549 and NHBE cells over-expressing ORMDL3 released relatively more IL-6 and IL-8 following poly I:c stimulation. IL-17A exhibited a similar inflammatory response in ORMDL3 knockdown and over-expressing cells, but co-stimulation of poly I:c and IL-17A did not significantly enhance the IL-6 and IL-8 response. Transcript abundance of IFNB following poly I:c stimulation was not significantly altered by ORMDL3 knockdown or over-expression. Dampening of the IL-6 response by ORMDL3 knockdown was confirmed in HRV16 infected BEAS-2B and A549 cells. conclusions ORMDL3 regulates the viral inflammatory response in airway epithelial cells via mechanisms independent of the TLR3 pathway.

Background The aim of this study was to analyze the frequencies of genotypes and alleles of Single Nucleotide Polymorphism (SNP) LEP-R c.668A>G (***223Arg, rs1137101) of leptin receptor gene and to assess the influence this DNA marker has on endometrial cancer (Ec) with respect to total body fat content. Methods The study comprised 120 patients treated for endometrial cancer and 90 controls treated for uterine fibroids. In total, 210 patients were included in this research. DNA was isolated from archival post-operative specimens. Polymerase chain Reaction - Restriction Fragment Length Polymorphism was employed to analyze the SNP. Results In this paper we have demonstrated that heterozygous genotype AG of SNP LEP-R c.668A>G (***223Arg, rs1137101) is statistically less frequent in women with endometrial cancer (Ec) than in controls: 33 versus 57%, respectively. Similarly, this heterozygous genotype is statistically significantly less frequent in obese (BMI > 30) women with Ec than in lean controls (BMI < 25): 30 versus 63%, respectively. conclusions AG polymorphic variant of SNP LEP-R c.668A>G (***223Arg, rs1137101) in LEP-R may be considered a protective factor in the development of endometrial cancer.

Sex differences and similarities in the neuroimmune response to central administration of poly I:c

Background The neuroimmune system is required for normal neural processes, including modulation of cognition, emotion, and adaptive behaviors. Aberrant neuroimmune activation is associated with dysregulation of memory and emotion, though the precise mechanisms at play are complex and highly context dependent. Sex differences in neuroimmune activation and function further complicate our understanding of its roles in cognitive and affective regulation. Methods Here, we characterized the physiological sickness and inflammatory response of the hippocampus following intracerebroventricular (IcV) administration of a synthetic viral mimic, polyinosinic:polycytidylic acid (poly I:c), in both male and female c57Bl/6N mice. Results We observed that poly I:c induced weight loss, fever, and elevations of cytokine and chemokines in the hippocampus of both sexes. Specifically, we found transient increases in gene expression and protein levels of IL-1 alpha, IL-1 beta, IL-4, IL-6, TNF alpha, ccL2, and cXcL10, where males showed a greater magnitude of response compared with females. Only males showed increased IFN alpha and IFN gamma in response to poly I:c, whereas both males and females exhibited elevations of IFN beta, demonstrating a specific sex difference in the anti-viral response in the hippocampus. conclusion Our data suggest that type I interferons are one potential node mediating sex-specific cytokine responses and neuroimmune effects on cognition. Together, these findings highlight the importance of using both males and females and analyzing a broad set of inflammatory markers in order to identify the precise, sex-specific roles for neuroimmune dysregulation in neurological diseases and disorders.

Spleen proteome profiling of dairy goats infected with c. pseudotuberculosis by TMT-based quantitative proteomics approach

corynebacterium pseudotuberculosis (***) is a zoonotic pathogen that can cause cheese lymphadenitis in goats. In order to obtain detailed information about the pathogenesis and host immune response of goats infected with ***, we used tandem mass tag (TMT)-labeling proteomic analysis to detect differentially expressed proteins (DEPs) in dairy goats infected with ***, and confirmed the altered proteins with western blot. A total of 6611 trusted proteins were identified, and 126 proteins were differentially abundant. Gene ontology (GO) analysis showed that all DEPs were annotated as biological processes, cell composition, and molecular functions. Biological processes mainly involve acute inflammation and immune response;cell components mainly involve extracellular areas and high-density lipoprotein particles;molecular functions are mainly antigen binding, ferric iron binding, and iron ion binding. KEGG analysis showed that a total of 102 pathways were significantly enriched, mainly lysosomes, phagosomes, and mineral absorption pathways. Our findings provided the relevant knowledge of spleen protein levels in goats infected with c. pseudotuberculosis and revealed the complex molecular pathways and immune response mechanisms in the process of *** infection. Significance: *** is the most fatal infectious disease in dairy goats, causing huge economic losses. However, the molecular pathways and immune response mechanisms of *** infection in goats remain unclear. Therefore, we conducted a comparative quantitative proteomics study on dairy goats infected with ***. The results provide a basis for better understanding the complexity of *** infection, reveal the complex molecular pathways and immune response mechanisms in *** infection, and provide some clues for identifying potential therapeutic targets. Distributions of Ion Density Irregularities and Their Gradients Inside Low-Latitude Ionospheric Plasma Bubbles Based on c/NOFS Planar Langmuir Probe Data

Data from the planar Langmuir probe onboard the communication/Navigation Outage Forecasting System has been processed, with focus on the: (1) identification of Equatorial Plasma Bubbles located within the eclipse sections of the satellite orbits;(2) subdivision of each Equatorial Plasma Bubble into three sections (West Wall, center, and East Wall);and (3) statistical analyses of the standard deviations and gradients of ion density data for each section. The statistical distributions resulted from the application of the above processing to structures identified during the years October 1, 2008 to September 30, 2009 (solar minimum conditions) and June 5, 2012 to June 4, 2013 (close to maximum conditions of solar cycle 24). The statistical distributions have also been classified considering all longitudes or three 60 degrees longitude sectors in South America, Africa, India-Asia, as well as all altitudes of the c/NOFS satellite or the limited range from 650 to 800 km. It will be shown that all the parameters (Equatorial Plasma Bubble sections, solar activity, longitude sector, and altitude range) affect the resulting statistical distributions. Additionally, the probability density functions (pdfs) of ion density gradients for the EPB West Walls, centers, and East Walls and the South American longitude sector, detected during 2012 and estimated with different resolutions will be compared. While the pdfs based on 1-Hz low-resolution data (sampling large-scale structures) display east-west asymmetries, those based on the original 512-Hz high-resolution data (sampling small-scale structures), created by secondary plasma instabilities, are essentially symmetric around the reference 0 mm/km gradient.
