Evaluation of an incubation instrument-free reverse transcription recombinase polymerase amplification assay for rapid and point-of-need detection of canine distemper virus

Canine distemper, caused by Canine distemper virus (cdv), is a highly contagious and fatal systemic disease in free-living and captive carnivores worldwide. Accurate, rapid and simple detection of cdv is critical to improve disease management and prevent outbreaks. In this study, a visible and incubation instrument-free reverse transcription recombinase polymerase amplification assay combined with lateral flow strip (LFS RT-RPA) was developed to detect cdv using primers and lateral flow (LF) probe specific for the nucleocapsid (N) protein gene. The cdv LFS RT-RPA assay was performed in a closed fist using body heat for 15 min, and the products were visible to the naked eyes on the LFS within 5 min. The assay could detect cdv, and there was no cross-reaction with the other viruses tested. Using the in vitro transcribed cdv RNA as template, the analytical sensitivity was 9.4 x 10(1) copies per reaction, which was the same result as that of a real-time RT-PCR. The assay performance was further evaluated by testing 32 nasal/oropharyngeal swab samples, and cdv RNA positive rate was 62.0% (20/32) by LFS RT-RPA, which was the same result as that of the real-time RT-PCR assay. The performance of the LFS RT-RPA was comparable to real-time RT-PCR, while the LFS RT-RPA assay was much faster and easier to perform. The novel cdv LFS RT-RPA assay provides an attractive and promising tool for rapid and reliable detection of cdv in the underequipped laboratory and point-of-need facility, which is of great significance in CD control in low resource settings.

Expression of Physalis mottle tymovirus coat protein in Escherichia coli was earlier shown to self-assemble into empty capsids that were nearly identical to the capsids formed in vivo Amino acid substitutions were made at the N-terminus of wild-type Physalis mottle virus coat protein with neutralizing epitopes of Canine parvovirus containing the antigenic sites 1-2, 4 and 6-7 and T-cell epitope of the fusion protein of Canine distemper virus in various combinations to yield PhMV 1, PhMV2, PhMV3, PhMV4 and PhMV5 These constructs were cloned and expressed in E colt The chimeric proteins self-assembled into chimeric tymovirus-like particles (TVLPs) as determined by electron microscopy. Comparative analyses of canine distemper viral isolates from clinical cases of canine distemper in vaccinated dogs

Sequence and phylogenetic analyses of three isolates of canine distemper virus (cdv) isolated from three dogs with a vaccination history were compared with the same analyses of vaccine virus isolated from a vaccine used for dogs. The three dogs showed clinical signs of a recent major type of CD in Japan, including oculonasal discharge and diarrhea, and pathological findings including non-suppurative encephalitis, pneumonia, mild gastroenteritis and lymphoid depletion. Inclusion bodies were in the stomach without inflammation and encephalitis was without clinical signs. One of the highest titers of cdv in different organs of the three dogs was commonly systemic lymphatic organs, including the spleen, lymph nodes and tonsils. New isolates of cdv joined to the clades of the Asia I group that is far from the vaccine group. These results surely indicate that wild strains of cdv from dogs with a vaccination history were not reversed vaccine virus, and that the dogs showed characteristics of recent CD in Japan. Vaccination of puppies born to immune dams with a canine adenovirus-based vaccine protects against a canine distemper virus challenge

None of the currently available distemper vaccines provides a satisfactory solution for the immunization of very young carnivores in the face of maternal-derived immunity. Since mucosal immunization with replication-competent adenovirus-based vaccines has been proven effective in the face of passive immunity against the vector, it has the potential to provide a solution for the vaccination of young puppies born to canine distemper virus (cdv)-immune dams. We report the engineering and the characterization of two replication-competent canine adenovirus type 2 (CAV2)-based vaccines expressing, respectively, the cdv hemagglutinin (HA) and fusion (F) antigens. We first demonstrated that the intranasal vaccination with a mixture of both recombinant CAV2s provides an excellent level of protection in seronegative puppies, confirming the value of replication-competent adenovirus-based vectors for mucosal vaccination. In contrast, intranasal immunization with the same vaccine of puppies born to cdv- and CAV2-immune dams, failed to activate specific and protective immune responses. We hypothesized that an active CAV2 infection occurred while puppies were in close contact with the vaccinated dams in the breeding units and that the resulting active mucosal immunity interfered with the intranasal administration of CAV2-based cdv vaccine. However, when puppies born to cdv- and CAV2-immune dams were vaccinated subcutaneously with the CAV2-based cdv vaccine, significant seroconversion and solid protective immunity were triggered despite pre-existing systemic immunity to the vector. This latter result is surprising and suggests that subcutaneous vaccination with a replication-competent recombinant CAV2 may be an efficient strategy to overcome both passive and active adenovirus specific immunity in the dog. The immune system of many mammalian species is not fully developed at birth, with newborns obtaining temporary immunological protection from maternal antibodies. Little is known of the immune system of the harbour seal, and developmental aspects of its immune system have not been systematically studied. We collected blood and milk samples from nine free-ranging mother-pup pairs throughout the lactation period on Sable Island, Canada, in an effort to characterise developmental aspects of the immune system of this newborn pinniped. Pup lymphocytes responded stronger to the mitogens concanavalin A, phytohaemagglutinin, and pokeweed mitogen than the lymphocytes of their mothers. In contrast to newborn cats and dogs, newborn seal pups developed high specific antibody responses after immunisation with an inactivated rabies vaccine. Circulating levels of total IgG in newborn pups were low (3% of maternal levels), but increased rapidly after colostrum intake (to 65% of maternal levels after 15 days). A similar pattern of increase in pup serum was observed for phocine distemper virus specific antibodies which had been detected in the serum and milk of mothers, suggesting that the transfer of colostral antibodies is an important feature of temporary protection for the pup. We speculate that the relative immunocompetence of the harbour seal at birth reflects an adaptation to its relatively short nursing period and limited maternal care.

Feline immunodeficiency virus (FIV) infects domestic cats and at least 20 additional species of non-domestic felids throughout the world. Strains specific to domestic cat (FIVFca) produce AIDS-like disease progression, sequelae and pathology providing an informative model for HIV infection in humans. Less is known about the immunological and pathological influence of FIV in other felid species although multiple distinct strains of FIV circulate in natural populations. As in HIV-1 and HIV-2, multiple diverse cross-species infections may have occurred. In the Serengeti National Park, Tanzania, three divergent subtypes of lion FIV (FIVPle) are endemic, whereby 100% of adult lions are infected with one or more of these strains. Herein, the relative distribution of these subtypes in the population are surveyed and, combined with observed differences in lion mortality due to secondary infections based on FIVPle subtypes, the data suggest that FIVPle subtypes may have different patterns of pathogenicity and transmissibility among wild lion populations. A unique cell division machinery in the Archaea

In contrast to the cell division machineries of bacteria, euryarchaea, and eukaryotes, no division components have been identified in the second main archaeal phylum, Crenarchaeota. Here, we demonstrate that a three-gene operon, cdv, in the crenarchaeon Sulfolobus acidocaldarius, forms part of a unique cell division machinery. The operon is induced at the onset of genome segregation and division, and the cdv proteins then polymerize between segregating nucleoids and persist throughout cell division, forming a successively smaller structure during constriction. The cdv operon is dramatically down-regulated after UV irradiation, indicating division inhibition in response to DNA damage, reminiscent of eukaryotic checkpoint systems. The cdv genes exhibit a complementary phylogenetic range relative to FtsZ-based archaeal division systems such that, in most archaeal lineages, either one or the other system is present. Two of the cdv proteins, cdvB and cdvC, display homology to components of the eukaryotic ESCRT-III sorting complex involved in budding of luminal vesicles and HIV-1 virion release, suggesting mechanistic similarities and a common evolutionary origin.

Natural infection with canine distemper virus in hand-feeding Rhesus monkeys in China

An outbreak of canine distemper virus (cdv) in hand-feeding Rhesus monkeys in China was reported Twenty Rhesus monkeys presented blood and mucus in feces, respiratory symptoms, anorexia, acute fever, thicken of footpad and red rashes in the faces over 1-month period. cdv infection was identified by characteristic clinical signs, the specific detection of the BIT Rapid color cdv detection kit, electron microscopy and the results of sequence aligning A phylogenetic analysis further confirmed that the cdv in the Rhesus monkeys belonged to the clade of the epidemic cdv types of China All the Infected monkeys were monitored and treated with antiserum therapy. The antiserum therapy seemed more effective for adult monkeys than young monkeys. Twelve monkeys died. The high mortality might indicate that the virulence of cdv to monkeys was enhanced. This is the first report we are aware of documenting Rhesus monkeys infected with cdv in China. Urgent work should be done to prevent the possibly epidemic of cdv in non-human primate (C) 2009 Elsevier B.V. All rights reserved.

Vaccination of puppies with a lipid-formulated plasmid vaccine protects against a severe canine distemper virus challenge

We assessed whether the formulation of a DNA vaccine expressing the canine distemper virus (cdv) hemagglutinin (HA) and fusion (F) immunogens with the cationic lipid DMRIE-DOPE could induce serological responses and protection against a severe cdv challenge in the dog. Although clear protection was observed in dogs vaccinated with formulated plasmids only limited cdv specific antibody titers were observed in protected dogs before challenge, suggesting that protection could be explained by cell-mediated immunity and/or by a strong antibody-based memory response (priming) triggered by the infectious challenge. The high level of protection achieved in this study, demonstrated that formulated DNA cdv vaccines can generate in dogs a level a protection comparable to conventional cdv vaccines. Canine distemper virus (cdv, Paramyxoviridae, Morbillivirus) is the causative agent of a severe infectious disease affecting terrestrial and marine carnivores worldwide. Phylogenetic relationships and the genetic variability of the hemagglutinin (H) protein and the fusion protein signal-peptide (Fsp) allow for the classification of field strains into genetic lineages. Currently, there are nine cdv lineages worldwide, two of them co-circulating in South America. Using the Fsp-coding region, we analyzed the genetic variability of strains from Uruguay, Brazil, and Ecuador, and compared them with those described previously in South America and other geographical areas. The results revealed that the Brazilian and Uruguayan strains belong to the already described South America lineage (EU1/SA1), whereas the Ecuadorian strains cluster in a new clade, here named South America 3, which may represent the third cdv lineage described in South America.
