The expression levels of coding sequences for pituitary growth hormone, introduced into Escherichia coli by genetic manipulation techniques, vary markedly according to the precise sequence introduced. In order to understand the basis of this variation more fully, we have studied the relationship between the level of expression in E. coli of a series of ovine growth hormone variants and the nucleotide sequences coding for their N-terminal regions, sequence variation resulted from the introduction of deletions, or site-directed mutations, into a plasmid containing the coding sequence for ovine growth hormone preceded by the initiation codon and 25 bases derived from P-galactosidase or linker regions of plasmid pUC8. The expression levels of the variants varied from less than 0.01% to over 34% of the total cell protein, indicating that changes in the nucleotide sequence close to the initiation codon had a marked effect on expression level. The results of a comparison of closely related sequences in pairs of plasmids giving poor or good expression are consistent with the hypothesis that poor translation of growth hormone mRNAs is caused by the presence of secondary structures close to the initiation codon. Secondary structures are identified that appear to explain the variation in expression levels.

Probiotic Escherichia coli strain Nissle 1917 (O6:K5:H1) is a commensal E coli isolate that has a long tradition in medicine for the treatment of various intestinal disorders in humans. To elucidate the molecular basis of its probiotic nature, we started sequencing the genome of this organism with a whole-genome shotgun approach. A 7.8-fold coverage of the genomic sequence has been generated and is now in the finishing stage. To exploit the genome data as early as possible and to generate hypotheses for functional studies, the unfinished sequencing data were analyzed in this work using a new method [Sun, J., Zeng, A.P., 2004. IdentiCS-identification of coding sequence and in silico reconstruction of the metabolic network directly from unannotated low-coverage bacterial genome sequence. BMC Bioinformatics 5, 112] which is particularly suitable for the prediction of coding sequences (CDSs) from unannotated genome sequence. The CDSs predicted for E. coli Nissle 1917 were compared with those of all five other sequenced E. coli strains (E. coli K-12 MG 1655, E. coli K-12 W3110, E. coli CFT073, EHEC O157:H7 EDL933 and EHEC 0 1 57:H7 Sakai) published to date. Five thousand one hundred and ninety-two CDSs were predicted for E. coli Nissle 1917, of which 1065 were assigned with enzyme EC numbers. The comparison of all predicted CDSs of E. coli Nissle 1917 to the other E. coli strains revealed 108 CDSs specific for this isolate. They are organized as four big genome islands and many other smaller gene clusters. Based on CDSs with EC numbers for enzymes, the potential metabolic network of Nissle 1917 was reconstructed and compared to those of the other five E. coli strains. Overall, the comparative genomic analysis sheds light on the genomic peculiarity of the probiotic E. coli strain Nissle 1917 and is helpful for designing further functional studies long before the sequencing project is completely finished. In this paper, we propose a strategy to achieve a single-feed multi-beam metasurface antenna by combining phase gradient and coding sequence. Theoretically, the multi-beam antennas with various far-field patterns can be realized via superimposing different kinds of phase coding sequences. As three proof-of-concepts, the 0101... /0101... coding sequence, chessboard coding sequence, and 1-bit sector coding sequence are selected to design double-beam, four-beam, and eight-beam circular-polarized reflector antennas, respectively. Furthermore, the four-beam reflector antenna is fabricated and measured to demonstrate the feasibility of our strategy. The fabricated antenna can produce four perfect pencil-shaped beams in a wide frequency range from 11 to 13 GHz. At central frequency, the realized gain is about 19.9 dB and the axial ratio is lower than 3 dB. Moreover, the circular polarization state of the reflective beams is consistent with the polarization state of spot source, which differs from the reflector antennas based on the polarization-insensitive unit cells. This paper connects the multi-beam patterns with binary digits, which provide the potential for reconfiguring the multi-beam patterns by computer programs.
