computerized microimage analysis of age-related changes of the human sinoatrial node

This paper reports the age-related changes in the histological features of the sinoatrial node (SAN). The hearts of 106 persons dying at different ages (2 days-83 years), without clinical or pathological evidence of cardiac disease, were taken and divided into seven age-groups. Serial sections with a thickness of 2 mu m, parallel to the long axis of the SAN. were prepared. Sections with the maximal area of the SAN were stained with hematoxylin/eosin (H & E) and Betty's method. The largest area of SAN were calculated by a computerized microimage analysis system. The age-related changes of the human SAN were discussed.

Thrombocytopenia with absent radius (TAR) syndrome is infrequently (7%) associated with mental retardation. In those cases, the mental deficiency is presumed to be a consequence of intracranial hemorrhage due to the thrombocytopenia, We report on 2 infants with TAR syndrome. One had developmental delay with evidence of cerebral dysgenesis by magnetic resonance imaging (MRI). Such findings have not been noted in the literature, but may not have been investigated in most cases. The other infant with TAR syndrome, who has had normal psychomotor development, has a normal brain on MRI scan. Detailed neuroimaging studies, preferably MRI, should be considered in the evaluation of patients with TAR syndrome, especially when there are documented signs of developmental delay, with or without a history of intracranial hemorrhage. Bone marrow angiogenesis in multiple myeloma and its correlation with clinicopathological factors

Increased angiogenesis has been found to be an adverse prognostic factor in solid tumors but evidences show that angiogenesis also plays an important role in hematological malignancies including multiple myeloma (MM). In this report we studied the various angiogenesis parameters like microvessel density (MVD) and total vascular area (TVA), on bone marrow biopsies in 50 newly diagnosed cases of MM. The aim was to study bone marrow angiogenesis in MM using light microscopy (MVD-A) and computerized image analyzer (MVD-B and TVA) and correlate it with clinical features, laboratory findings, histological features, and response to treatment on follow-up. Bone marrow biopsies of test cases (n = 50) were immunohistochemically stained with CD34 for visualization of microvessels. MVD-A (range 8-80;mean 50.4;SD 17.5), MVD-B (5.2-33.2;mean 16.3;SD 5.1), and TVA in percentage (range 0.42-7.20;mean 2.8;SD 1.5) were measured. Ten age- and sex-matched controls were studied and their parameters were taken as grade I. There was a significant correlation between these angiogenesis parameters (MVD-A vs MVD-B, Pearson's correlation coefficient (pcc) = 0.724;MVD-A vs TVA, pcc = 0.370;MVD-B vs TVA, pcc = 0.406). The angiogenesis was significantly higher in cases as compared to controls. Patients with residual disease had a higher MVD as compared to the complete responders. High tumor burden and diffuse pattern of infiltration were also associated with grade III MVD and TVA. Hence, it can be concluded that angiogenesis correlates with other histological features associated with prognosis and is also a good predictor for complete response in patients with multiple myeloma.

The relative proportion, ultrastructure and DNA-content of the binucleate cells in the celiac superior mesenteric ganglion of the guinea pig was studied using light and electron microscopy as well as computerized image analysis of Feulgen stained cells. The number of mono - versus binucleate cells was found to vary with stage of development with about 40% of the cells being binucleate in adult animals and 50% in late prenatal stage. No difference in ultrastructure was observed between the nuclei of the two cell types. The binucleate cells contain twice the amount of DNA found in the mononucleate cells.

A morphometric and microdensitometric characterization of the dopamine neurons of the mediobasal hypothalamus and their relationships with several other chemically identified systems, including putative tyrosine hydroxylase-positive/dopamine-negative neurons, was carried out after visualization of dopamine content by both immunocytochemistry and the Falck-Hillarp technique. Quantitative assessment of co-existence demonstrated that more than 95% of dopamine-immunoreactive neurons also contained tyrosine hydroxylase immunoreactivity and more than 90% of growth hormone-releasing factor-immunoreactive neurons also contained tyrosine hydroxylase immunoreactivity. Morphometric and densitometric analysis of dopamine, tyrosine hydroxylase and growth hormone-releasing factor-immunoreactive neurons in the arcuate nucleus showed that dopamine/tyrosine hydroxylase-containing and growth hormone-releasing factor/tyrosine hydroxylase-containing neuronal populations are two largely segregated cell groups with specific localization in the arcuate region, rostrocaudal extension and tyrosine hydroxylase-immunoreactivity content. Morphometric characteristics of dopamine-immunoreactive neurons were shown to be equivalent to those of catecholamine fluorescent cell bodies in the arcuate region. In addition, a cell group lacking detectable catecholamine fluorescence in normal animals but accumulating L-DOPA after peripheral loading was identified and characterized from a morphometric standpoint in the ventral premammillary nucleus. Quantitative analysis of nerve terminal co-distribution in the median eminence revealed significant correlations between dopamine and other transmitter or neurohormone systems, such as gamma-aminobutyric acid, galanin, luteinizing hormone-releasing hormone, in specific subregions of the palissade zone. Optic disc measurements with the 78 diopter lens, Zeiss 4-mirror contact lens and computerized image analysing system

We evaluated the 78 diopter (D) lens and Zeiss 4-mirror lens for the measurement of vertical and horizontal optic disc diameters during slit-lamp biomicroscopy and compared the results with the measurements made with the computerized image analysing system (imagenet 640, Topcon, Japan) in 30 eyes of 30 patients. The 78D lens and Zeiss 4-mirror lens measurements both correlated well with the computerized image analysis measurements (r=0.881 for vertical and r=0.895 for horizontal disc diameter measurements with the 78D lens and r=0.883 for vertical and r=0.891 for horizontal disc diameter measurements with the Zeiss 4-mirror lens). The 780 lens measurements overestimated vertical disc diameter by 5.3% and horizontal disc diameter by 4.4%. The Zeiss 4-mirror lens underestimated the vertical disc diameter by 2.4% and horizontal disc diameter by 2.2%. Thus either lens can be used for a quick estimation of the optic disc size, since the results correlate well with the computerized image analysis measurements.

Human sperm morphometry and smoking, caffeine, and alcohol consumption

The purpose of this study is to determine whether sperm nuclear size, shape, and chromatin texture parameters are associated with lifestyle exposures including smoking, caffeine, and alcohol consumption. Eighty-six healthy male volunteers (ages 18-35), recruited through newspaper advertisements, provided a semen, blood, and urine sample and completed a questionnaire concerning demographic and lifestyle exposures. Sperm nuclear size, shape, and chromatin texture parameters were measured using computerized image analysis. Results indicated no associations between the sperm nuclear morphometric parameters and age, smoking, or alcohol consumption. There was weak evidence for an association with caffeine intake. In conclusion, the lifestyle factors smoking, caffeine intake, and alcohol consumption do not appear to significantly affect sperm nuclear size, shape, or chromatin texture in this study population. Semi-automatic stereotactic coordinate identification algorithm for routine localization of Deep Brain Stimulation electrodes

Deep Brain Stimulation (DBS) is a routine therapy for movement disorders, and has several emerging indications. We present a novel protocol to define the stereotactic coordinates of metallic DBS implants that may be routinely employed for validating therapeutic anatomical targets. Patients were referred for troubleshooting or new DBS implantation. A volumetric MRI of the brain obtained prior to or during this protocol was formatted to the Anterior Commissure-Posterior Commissure (AC-PC) coordinate system. Patients underwent a CT scan of the brain in an extended Hounsfield unit (EHU) mode. A semi-automatic detection algorithm based on a Normalized Mutual Information (NMI) co-registration method was implemented to measure the AC-PC coordinates of each DBS contact. This algorithm was validated using manual DBS contact identification. Fifty MRI-CT image pairs were available in 39 patients with a total of 336 DBS electrodes. The median and mean Euclidean distance errors for automatic identification of electrode locations were 0.20 mm and 0.22 mm, respectively. This method is an accurate method of localization of active DBS contacts within the sub-cortical region. As the investigational indications of DBS expand, this method may be used for verification of final implant coordinates, critical for understanding clinical benefit and comparing efficacy between subjects. CORRELATION OF STRUCTURE AND FUNCTION IN GLAUCOMA - QUANTITATIVE MEASUREMENTS OF DISK AND FIELD

The relationship between quantitative structural measurements of the optic nerve head and quantitative measurements of the visual field in glaucoma was studied. computerized videographic image analysis (Rodenstock Analyzer) was used to obtain cup-disc ratio, disc rim area, and cup volume in 50 glaucoma suspects and 37 glaucoma patients. The visual field indices, mean defect and loss standard deviation, were calculated from Octopus Program 32. There were statistically significant linear correlations between each of the optic nerve structural parameters and visual field mean defect and loss standard deviation. The strongest correlation was between disc rim area and visual field mean defect (r = −0.49), which are both global measures of glaucomatous damage. The correlations were not strong enough to readily allow the recognition of early nerve damage by the parameters analyzed thus far. Closer correlations may be evident if structural parameters can be found that more accurately reflect the number of surviving axons in the optic nerve head.

Preoperative measurements of macular hole with scanning laser ophthalmoscopy. Correlation with functional outcome

Purpose: To measure the dimensions of macular holes before surgery and compare them with the functional outcome. Methods: A prospective study of ten consecutive patients scheduled for macular hole surgery. Macular holes were imaged with scanning laser ophthalmoscopy with different laser wavelengths. computerized image analyses were used to measure the size of the hole and the surrounding rim. Results: Eyes were referred to three different groups preoperatively according to visual function. Measurements of the longest diameter and the area of the macular hole were found to correlate well with visual function tested before and after surgery. All holes closed after one operation. Eighty per cent of eyes improved two or more lines on the distance visual chart and all patients but one could read a newspaper six months after surgery. Conclusion: Preoperative size of macular hole can be used as a predictor of the functional outcome of macular hole surgery.
