Decoding dna labels by melting curve analysis using real-time PCR

Synthetic dna has been used as an authentication code for a diverse number of applications. However, existing decoding approaches are based on either dna sequencing or the determination of dna length variations. Here, we present a simple alternative protocol for labeling different objects using a small number of short dna sequences that differ in their melting points. Code amplification and decoding can be done in two steps using quantitative PCR (qPCR). To obtain a dna barcode with high complexity, we defined 8 template groups, each having 4 different dna templates, yielding 15(8) (>2.5 billion) combinations of different individual melting temperature (T-m) values and corresponding ID codes. The reproducibility and specificity of the decoding was confirmed by using the most complex template mixture, which had 32 different products in 8 groups with different T-m values. The industrial applicability of our protocol was also demonstrated by labeling a drone with an oil-based paint containing a predefined dna code, which was then successfully decoded. The method presented here consists of a simple code system based on a small number of synthetic dna sequences and a cost-effective, rapid decoding protocol using a few qPCR reactions, enabling a wide range of authentication applications.

One of the key problems in dna computation is the design of large and reliable codes. Due to the biological and chemical restrictions, dna codes need to satisfy certain constraints. Here, we provide an algebraic procedure to construct dna codes by using homomorphism of groups. The construction procedure is used to determine mutation.

Color Image Encryption Using Angular Graph Fourier Transform

In this paper, an angular graph Fourier transform (AGFT) is introduced to encrypt color images with their intrinsic structures. The graph Fourier transform (GFT) is extended to the AGFT and proven to have the desired properties of angular transform and graph transform. In the proposed encryption method, color images are encoded by dna sequences and confused under the control of chaotic key streams firstly. Secondly, sparse decomposition based on the random walk is applied to scramble pixels spatially, and a series of sub-images are obtained. This step increases encryption efficiency. Finally, the intrinsic sub-image structure is reflected by graphs, and the signals on different subgraphs are transformed into different AGFT domains with particular angular parameters, which makes the proposed method relevant to the original image structure and enhances security. The experimental results demonstrate that the proposed algorithm can resist various potential attacks and achieve better performance than the state-of-the-art algorithms.

The conventional way of identifying dna motifs, solely based on match alignment information, is susceptible to a high number of spurious sites. A novel scoring system has been introduced by taking both match and mismatch alignment information into account. The mismatch alignment information is useful to remove spurious sites encountered in dna motif searching. As an example, a correct TATA box site in Homo sapiens gene has successfully been identified based on match and mismatch alignment information.

Immune Genetic Learning of Fuzzy Cognitive Map

This paper presents a hybrid methodology of automatically constructing fuzzy cognitive map (FCM). The method uses immune genetic algorithm to learn the connection matrix of FCM. In the algorithm, the dna coding method is used and an immune operator based on immune mechanism is constructed. The characteristics of the system and the experts' knowledge are abstracted as vaccine for restraining the degenerative phenomena during evolution so as to improve the algorithmic efficiency. Finally, an illustrative example is provided, and its results suggest that the method is capable of automatically generating FCM model.

The conventional way of identifying possible motif sequences in a dna strand is to use representative scalar weight matrix for searching good match substring alignments. However, this approach, solely based on match alignment information, is susceptible to a high number of ambiguous sites or false positives if the motif sequences are not well conserved. A significant amount of time is then required to verify these sites for the suggested motifs. Hence in this paper, the use of mismatch alignment information in addition to match alignment information for dna motif searching is proposed. The objective is to reduce the number of ambiguous false positives encountered in the dna motif searching, thereby making the process more efficient for biologists to use.

dna cryptography is an upcoming field in the cryptography area. We introduce a hybrid cryptography method based on RGB colors, where the security of data is improved by using a data encryption algorithm and that of key is enhanced by means of an algorithm based on dna steganography. Information can be encoded by using any schemes like ASCII or UNICODE. Plain text is converted to matrix form which is then passed through a number of manipulation steps. Security is further enhanced by using a strong key which is encapsulated by using dna steganography method. As the next layer of security we propose encryption using dna bases and amino acids. For this, some dna techniques like coding are used. The fundamental idea is to provide a multiphase cryptosystem by combining dna and Amino Acids concepts to other security techniques. Finally, the cipher form is converted to colors to improve the level of security.

A two stage encryption algorithm based on dna sequence has proposed in this paper. In the first stage an encryption of plain text is done by generating a random key. The plain text is again encrypted to produce the cipher text in the second stage. Moreover, this encryption algorithm is based on a symmetric key cryptography system, where we provide a shared key to encrypt as well as decrypt the intended message. To encrypt the original key two stages are maintained and sending it over a separate secure channel other than the channel through which we are transferring the cipher text. A numerical study confirms that the proposed algorithm is reliable, secure, scalable, and robust for transmitting message.

Feature extraction in image encryption is a selective encryption method that has emerged in recent years. Traditional image encryption algorithms are low in complexity, inefficient, and vulnerable to attack. In this paper, significant detection is used to extract important information from the image, and we can pre-encrypt this information. The pre encryption process uses an image block encryption algorithm based on Chen chaotic system and dna coding. Secondly, the discrete wavelet transform (DWT) is used to encrypt the reference image. Finally, the pre-encrypted image is combined with transformed image which obtained from the reference image. After that, a visually meaningful encrypted image can obtain by inverse discrete wavelet transform. Because this final image is very similar to an ordinary image, it is not vulnerable to attack. This method effectively protects the plaintext image.

Rapid technological advances and the increasing use of social media has caused a tremendous increase in the generation of digital data, a fact that imposes nowadays a great challenge for the field of digital data storage due to the short-term reliability of conventional storage devices. Hard disks, fiash, tape or even optical storage have a durability of 5 to 20 years while running data centers also require huge amounts of energy. An alternative to hard drives is the use of dna, which is life's information-storage material, as a means of digital data storage. Recent works have proven that storing digital data into dna is not only feasible but also very promising as the dna's biological properties allow the storage of a great amount of information into an extraordinary small volume for centuries or even longer with no loss of information. In this work we present an extended end-to-end storage workflow specially designed for the efficient storage of images onto synthetic dna. This workflow uses a new encoding algorithm which serves the needs of image compression while also being robust to the biological errors which may corrupt the encoding.
