A DROSOPHILA SHC GENE-PRODUCT IS IMPLICATED IN SIGNALING BY THE DER RECEPTOR TYROSINE KINASE

Antibodies to the human Shc adaptor protein were used to isolate a cDNA encoding a Drosophila Shc protein (dShc) by screening an expression library. The dshc gene, which maps to position 67B-C on the third chromosome, encodes a 45-kDa protein that is widely expressed throughout the Drosophila life cycle. In flies, the dShc protein physically associates with the activated Drosophila epidermal growth factor receptor homolog (DER) and is inducibly phosphorylated on tyrosine by DER. The 45-kDa dShc protein is closely related both in overall organization and in amino acid sequence (46% identity) to the 52-kDa mammalian Shc isoform. In addition to a C-terminal Src homology 2 (SH2) domain, dShc contains an N-terminal phosphotyrosine binding (PTB) domain, which associates in vitro with the autophosphorylated DER receptor tyrosine kinase and with phosphopeptides containing an Asn-Pro-X-pTyr motif, where pTyr stands for phosphotyrosine. A potential binding site for the dShc PTB domain is located at Tyr-1228 of DER. These results indicate that the shc gene has been conserved in evolution, as have the binding properties of the Shc PTB and SH2 domains. Despite the close relationship between the Drosophila and mammalian Shc proteins, dShc lacks the high-affinity Grb2-binding site found in mammalian Shc, suggesting that Shc proteins may have functions in addition to regulation of the Ras pathway.

The spin-orbit excited (2) Pi(3/2) manifold of (BAr)-B-11(X (2) Pi) was observed in a free jet supersonic expansion by laser fluorescence excitation in the B (2) Sigma(+)-X (2) Pi (7, 0) band. In previous work we observed transitions from the lower (2) Pi(1/2) manifold. The spin-orbit constant A was determined to equal 10.88 +/- 0.08(1 sigma) cm(-1), close to a simple estimate, 2/3 Delta E=10.17 cm(-1), where Delta E is the boron 2p P-2(3/2)-P-2(1/2) spin-orbit splitting. The near equality of experimental and theoretical values of A suggests that the boron 2p orbital is not strongly affected by the weak, nonbonding B-Ar interaction.

ENANTIODIFFERENTIATION IN JET-COOLED VAN-DER-WAALS COMPLEXES OF CHIRAL MOLECULES

Laser-induced fluorescence excitation spectroscopy in a supersonic expansion is used to evidence intermolecular chiral discrimination at the microscopic level. Measurements have been performed in (R)-(+)- and (S)-(-)-alpha-methyl-2-naphthalenemethanol (a naphthalene compound made chiral by the asymmetric carbon in the 2 position) in the presence of alcoholic solvents (methanol, 1-propanol, and (S)-(+)-chloro-1-propanol). While identical spectral shifts are observed for 1:1 complexes of (R)-(+)- and (S)-(-)-alpha-methyl-2-naphthalenemethanol with non-chiral solvents, complexation of each enantiomer with the chiral (S)-(+)-2-chloro-1-propanol molecule gives rise to different solvent shifts and allows differentiation between the two forms in the racemic mixture.

The signal recovered from the first reported experimental secure communication system via chaotic synchronization contains some inevitable noise which degrades the fidelity of the original message. By cascading the output of the receiver in the original system into an identical copy of this receiver, it had be shown by computer experiments that this noise can be significantly reduced. We discuss the heuristic laws governing the errors of the recovered signal which are observed in a new series of very careful computer experiments. This discussion is done comparing these laws to the results obtained using the linear filtering theory. Some discrepancy appears. In order to understand the origin of the discrepancy we consider another simpler model based on the Bonhoeffer-Van der Pol equation where no chaos occurs. In this case both two heuristic laws are in good agreement with the linear filtering theory.

The experiment detailed in this paper was carried out at the French company EDF. It gives both points of view, of the expert and of the client, on how the EDF CAD department made its choice of a CASE (computer-aided software engineering) method and tool. The CASE expert explains how the assessment was defined and then conducted starting from a rather wide request, and how a set of choice elements was established to help decision-making. The EDF-CAD client is an "a posteriori" analysis, involving the different steps from the identification of the need until the final choice. The conclusions show a large agreement on the main results: a maturation step, client and expert getting to know more of each other, thus establishing a mutual confidence relationship; the will to succeed to choose, remaining clear-minded, for a method and a tool will not solve all problems; an objective comparison approach and a joint assessment considering nontechnical criteria and mostly the client's needs and requirements, a continuous help on behalf of the CASE expert not ending with the decision file, but going through integration of the method and tool in client projects.< >

Detecting activations (ACTs) from unipolar electrograms (UEGs) is difficult during ventricular fibrillation (VF) due to the superposition of distant activity from multiple wavefronts. Epicardial UEGs were recorded during the first 2 s of VF from an 11/spl times/11 array of Ag-AgCl electrodes spaced 280 /spl mu/m apart on the LV apex of 5 open-chest pigs. The Rule-Based Method (RBM) used maximum negative derivatives (DERs) located at least 56 ms apart to identify RBM-ACTs while the Transmembrane Current Method (TCM) used electrograms recorded from sites with 4 orthogonal neighbors to estimate I/sub m/ at the center site. Biphasic deflections of I/sub m/ that occurred at the same time as negative DERs were used to indicate TCM-ACTs. All RBM-ACTs with DER /spl les/-1.5 mV/ms (5119 of the 5650 investigated) were assumed to represent true activations. TCM-ACTs confirmed activation for 251 RBM-ACTs with -1.5

SINGULARITY INDUCED BIFURCATION AND THE VANDERPOL OSCILLATOR

In parameter dependent differential-algebraic models (DAEs) of the form x = f and 0 = g, it has been shown recently that the generic codimension one local bifurcations are the well-known saddle node and Hopf bifurcations and a new bifurcation called the singularity induced bifurcation. The latter occurs generically when an equilibrium of the DAE system crosses the singular surface of noncausal points. In this paper, it is shown that when singularly perturbed models of the form x = f and epsilony = g are considered, the singularity induced bifurcation in the slow DAE system corresponds to oscillatory behavior in the singularly perturbed models. As an example, it is proved that the oscillations in the classical van der Pol oscillator arise when a stable equilibrium undergoes the singularity induced bifurcation in the slow DAE system, which in turn corresponds to the occurrence of supercritical Hopf bifurcations in the singularly perturbed models.

In this paper we present a temporal multisensor fusion module called Dynamic Date Manager (DDM). This module was developed for "ProLab2", the French Driving Assistance Demonstrator of the European Prometheus Program. The goal of the DDM is to maintain a representation of the dynamic environment of the vehicle. This representation is then used by an electronic copilot to inform the driver about potential risks and collisions. The demonstrator is an ordinary car equipped with a large number of exteroceptive and proprioceptive sensors. The presented module realizes a temporal multisensor fusion. Symbolic concepts are associated to numerical data in order to filter the numerical data, evaluate reliability indexes, manage the events on the perceptive systems, classify the numerical data, and limit the uncertainties. The demonstrator's environment to be estimated consists of the state of the vehicle itself and the set of surrounding obstacles.
