The loop-mediated isothermal amplification (LAMP) was standardized for rapid detection of Dichelobacter nodosus and fusobacterium necrophorum. A total of 250 foot swabs were screened from sheep (200) and goats (50) from different districts of Rayalaseema, viz., Chittoor, Nellore, Kadapa, and Anantapur. Out of 250 samples 75 (30.0%) and 85 (34.0%) were positive for D. nodosus and f. necrophorum, respectively. All the 250 samples were screened individually for both the organisms by LAMP. Among them, 104 (41.6%) were found to be positive for D. nodosus and 120 (48.0%) were positive for f. necrophorum. The efficacy of LAMP in terms of sample DNA detection limit was compared with the PCR by using standard dilutions of DNA extracted from D. nodosus and f. necrophorum cultures. The detection limit was found to be higher than PCR for both the organisms. The sensitivity of LAMP is compared with PCR by targeting 16S rRNA gene of D. nodosus and lktA gene off. necrophorum. In case of D. nodosus, out of 250 samples, 75 (30.0%) were positive by PCR and 104 (41.6%) were positive by LAMP. Among 250 samples, 85 (34.0%) were positive by PCR and 120 (48.0%) were positive by LAMP in case off. necrophorum. The LAMP was found to be more sensitive than PCR in detecting the organisms with high statistical significance.

Accumulating evidence links fusobacterium nucleatum with ulcerative colitis (UC). The mechanism by which f. nucleatum promotes intestinal inflammation in UC remains poorly defined. Here, we first examined the abundance and impact off. nucleatum on disease activity in UC tissues. Next, we isolated a strain off. nucleatum from UC tissues and explored whether f. nucleatum aggravates the intestinal inflammatory response in vitro and in vivo. We also examined whether f. nucleatum infection involves the Nf-kappa B or IL-17f signaling pathways. Our data showed that f. nucleatum was enriched in 51.78% of UC tissues and was correlated with the clinical course, clinical activity and refractory behavior of UC (p < 0.05). furthermore, we demonstrated that f. nucleatum promoted intestinal epithelial damage and the expression of the inflammatory cytokines IL-1 beta, Il-6, IL-17f and TNf-alpha. Mechanistically, f. nucleatum targeted caspase activation and recruitment domain 3 (CARD3) through NOD2 to activate the IL-17f/Nf-kappa B pathway in vivo and in vitro. Thus, f. nucleatum orchestrates a molecular network involving CARD3 and IL-17f to control the UC process. Measuring and targeting f. nucleatum and its associated pathways will yield valuable insight into the prevention and treatment of UC. (c) 2019 Pathological Society of Great Britain and Ireland. Grouper iridovirus causes high mortality rates in cultured groupers, and effective treatment for grouper iridovirus infection is urgently required. Illicium verum Hook. f. is a well-known medicinal plant with a variety of biological activities. The aim of this study was to analyse the use of I. verum extracts to treat grouper iridovirus infection. The safe working concentration of each I. verum extract was identified both in vitro and in vivo as follows: I. verum aqueous extract (IVAE) <= 500 mu g/ml;I. verum ethanol extract (IVEE) <= 250 mu g/ml;shikimic acid (SKA) <= 250 mu g/ml;trans-anethole (TAT) <= 800 mu g/ml;3,4-dihydroxybenzoic acid (DDBA) <= 400 mu g/ml;and quercetin (QCE) <= 50 mu g/ml. The inhibitory activity of each I. verum extract against grouper iridovirus infection was analysed using aptamer (Q2)-based fluorescent molecular probe (Q2-AfMP) and RT-qPCR. All of the I. verum extracts displayed dose-dependent antiviral activities against grouper iridovirus. Based on the achieved per cent inhibition, IVAE, IVEE, DDBA and QCE were associated with the greatest antiviral activity (all > 90%). Together, our results indicate that I. verum extracts have effective antiviral properties, making it an excellent potential source material for the development of effective treatment for grouper iridovirus infection.
