8-Azido-ATP is a suitable photoaffinity label for beef-heart mitochondrial ATPase (f1). 8-Azido-ATP is hydrolyzed slowly by f1 in the dark. Photolysis at 350 nm in the presence off1 leads to inhibition of the ATPase activity. The presence of ATP during illumination prevents the inhibition. Illumination off1 in the absence of 8-azido-ATP causes no inhibition. Added Mg2+ is not necessary for the binding of the 8-azido-ATP to f1. 8-Azido-ATP binds specifically to the .beta. subunits off1. The ATPase activity is completely inhibited when 2 mol of 8-azido-ATP are bound/mol f1.

The transduction of energy from the oxidation of substrates by the electron transport chain or from the hydrolysis of ATP by the Mg2+-ATPase was measured in everted membrane vesicles of E. coli using the energy-dependent quenching of quinacrine fluorescence and the active transport of Ca. Treatment of everted membranes derived from a wild-type strain with the chaotropic agents quanidine-HCl and urea caused a loss of energy-linked functions and an increase in the permeability of the membrane to protons, as measured by the loss of respiratory-linked proton uptake. The coupling of energy to the quenching of quinacrine fluorescence and Ca transport could be restored by treatment of the membranes with N,N''-dicyclohyexylcarbodiimide. Chaotrope-treated membranes lacked Mg2+-ATPase activity. Binding of crude soluble Mg2+-ATPase to treated membranes restored energy-linked functions. Membranes prepared from a wild-type strain grown under anaerobic conditions in the presence of nitrate retained respiration-linked quenching of quinacrine fluorescence and active Ca transport after treatment with chaotropic agents. Everted membrane vesicles prepared from an Mg2+-ATPase deficient strain lacked respiratory-driven functions when the cells were grown aerobically but were not distinguishable from membranes of the wild-type when both were grown under anaerobic conditions in the presence of nitrate. Apparently, chaotropic agents solubilize a portion of the Mg2+-ATPase, causing an increase in the permeability of the membrane to protons;growth under anaerobic conditions in the presence of nitrate may prevent the increase in proton permeability caused by genetic or chemical removal of the catalytic portion of the Mg2+-ATPase.

Theoretical aspects of the kinetics of interaction of enzymes with lipid substrates are presented. Rate equations were written and used to simulate v [velocity] vs. S [substrate] curves for interaction of enzymes with monomers (i.e., a molecular solution) or micelles (aggregated form) of the soluble, amphiphilic lipids. The rate equations were written assuming separate kinetic parameters for the interaction of the enzyme with these 2 forms. Although the rate equations are based on the kinetic theory of Michaelis and Menten, most of the simulated v vs. S curves were not hyperbolic. A procedure is suggested for determining the kinetic parameters with the aid of a graphic method.

Protein synthesis in rabbit reticulocytes and their lysates is regulated by heme. In heme-deficient reticulocyte lysates, protein synthesis proceeds at the initial rate for several minutes and then declines abruptly. Inhibition of protein synthesis is due to the activation of a heme-regulated translational inhibitor (HRI) which blocks the initiation of protein synthesis. Addition of the isolated HRI to hemin-supplemented lysates causes inhibition of initiation similar to that observed in heme-deficiency. HRI has been shown to be a protein kinase that specifically phosphorylates the Met-tRNA(f) binding factor (eIf-2). We have isolated an inhibitor (LI) of protein chain initiation from rat liver which displays properties similar to those of HRI: (i) the chromatographic behavior of LI on DEAE-Sephadex, DEAE-cellulose, and phosphocellulose is similar to that of HRI; (ii) both LI and HRI inhibit protein chain initiation in rabbit reticulocyte lysates with the same kinetics of inhibition-i.e., an initial period of synthesis for several minutes at the control rate followed by an abrupt decline in the rate of initiation; (iii) both inhibitions are prevented or reversed by eIf-2; (iv) GTP (2 mM) prevents, and ATP (2 mM) potentiates, the inhibition of protein synthesis induced by either inhibitor; (v) LI is associated with a protein kinase that also phosphorylates the 38,000-dalton subunit of elf-2. These findings indicate that a mechanism for the regulation of protein synthesis similar to that found in rabbit reticulocytes may be present in rat liver.
