本研究制备并鉴定了鼠抗粉尘螨变应原Der f 5的单克隆抗体(monoclonal antibody,McAb)。将质粒pET28a(+)-Der f 5转化ArcticExpress(DE3)感受态细胞,取单个菌落接种培养,加入IPTG诱导表达,经纯化获得Der f 5重组蛋白。利用Der f 5重组蛋白为免疫原,免疫BALB/c小鼠,取免疫小鼠脾脏细胞与其骨髓瘤细胞融合。通过间接ELISA法筛选特异性分泌抗体的杂交瘤细胞。用杂交瘤细胞株诱生小鼠腹水,应用蛋白A亲和层析法进行抗体纯化。通过间接ELISA、Western blotting鉴定该单克隆抗体的特性和交叉性。最终获得3株稳定分泌鼠抗粉尘螨变应原Der f 5的高效价单克隆抗体。研究分析表明该3株单抗均能识别重组Der f 5蛋白和天然的粉尘螨提取物。本实验成功制备了3株鼠抗粉尘螨变应原Der f 5的单克隆抗体,为建立粉尘螨主要变应原Der f 5检测及纯化方法奠定了基础。

Integrative approach reveals new insights into photosynthetic and redox protection in ex vitro tobacco plantlets acclimatization to increasing light intensity

Integrative mechanisms involving photosynthetic and antioxidant protection regulated by light intensity during in vitro and ex vitro plantlets acclimatization are poorly understood. Tobacco plantlets grown under in vitro and ex vitro environments were exposed to different light regimes to evaluate the role of photosynthesis and antioxidant protection. In vitro plantlets displayed a narrow photosynthetic capacity to cope with light as revealed by low net CO 2 assimilation ( P N ), decreased actual quantum efficiency of photosystem II (ΦPSII) associated with non-induction of non-photochemical quenching (NPQ). In contrast, acclimated ex vitro plants showed strong stimulation in P N whereas ΦPSII and NPQ remained at high levels. In vitro plantlets exposed to moderate light suffered strong oxidative stress associated with increased activities of superoxide dismutases, catalases and ascorbate peroxidases, revealing an ineffective antioxidant system. In contrast, ex vitro plants presented lower oxidative damage in parallel to unchanged enzymatic activities, indicating an efficient antioxidant steady state. The levels of reduced ascorbate (ASC) and glutathione (GSH) were also increased only in ex vitro plants in response to excess light. An integrative study based on correlation networks and principal component analyses (PCA) corroborate that the two plant groups indeed displayed contrasting acclimation processes. In conclusion, during in vitro to ex vitro transition, tobacco plantlets exposed to increasing light display physiological adjustments involving photosynthesis and improvement of the enzymatic and non-enzymatic antioxidant systems. These findings highlight the importance of integrative approaches to understand ex vitr o acclimatization to environmental stimuli.

Decoding the Translations of Political Terms in the Nineteenth-Century Chinese-English Dictionaries - Lobscheid and his Chinese-English Dictionary

German missionary Wilhelm Lobscheid ((sic), 1822-1893) played a vital role in English-Chinese translations. His English-Chinese Dictionary ((sic), 1866-1869) was the first comprehensive bilingual dictionary published in Hong Kong with both Cantonese and Mandarin pronunciations. Regarding his dictionary as the bridge between two languages, political terms previously nonexisting in the Chinese diction are shaped and shared during China's crucial political transformation from the Qing Empire to the Republic of China. With selected examples, this paper emphasizes Lobschied's contribution in sharing political knowledge through his dictionary.

Molten steel level stability in continuous casting slab mold under electromagnetic stirring

fluctuation of molten steel level in continuous casting process is a critical issue. The analytic function for the liquid steel surface stability index( f) of the continuous casting slab mold under the action of electromagnetic stirring was derived. The effects on the f value of the electromagnetic field,casting mold,submerged entry nozzle,and casting speed were observed,and the stability of the process parameters of a practical continuous casting system was evaluated. The main results generated are as follows:①Based on the free flow jet theory,a formula is established for the level stability index of continuous casting slab mold under electromagnetic stirring.②According to the results of calculation,the various parameters in decreasing order of the magnitude of their influence are magnetic flux density,nozzle submersion depth,casting speed,slab width,stirring current frequency,and nozzle side-hole inclination angle.③for practical continuous casting with a slab thickness of 230 mm,the calculated f values are between 3 and 5 for different matches between slab width and casting speed,but the parameter optimization space is much larger for wider slab widths and lower casting speeds.

The influence of Spencer's ideas is now generating a good deal of analysis. The aim of this paper is to highlight the importance of Spencer's work on economic thought. It also analyses the way in which this work was interpreted by the Italian economists. In particular, it investigates the influence of Spencer's theory of evolution on the thought of Pantaleoni (1857-1924) and Nitti (1868-1953). These two representative Italian scholars were on opposite sides for their economic methodology and the issue of government intervention in the economy. The paper clarifies whether their two divergent visions on social change could both be in accordance with Spencer ideas.

He was born in Klang Selangor British Malaya and was educated in Harvard University in the early 1960s. He majored in History and minored in Philosophy and International Relations and has been teaching in colleges and universities since late 1960s. He had taught mainly courses on social thought political sociology and social history of Modern China and involved himself in Southeast Asian Studies with emphasis on Chinese in Southeast Asia. In 1977–1978 he taught History of the US and its Foreign Policy in Malone College Canton Ohio. Entering 1980s he became a Visiting Scholar in institutions in China Japan India and South Korea and was offered an Honorary Doctor by the Far Eastern Research Institute of the Russian Academy of Sciences. He has written and printed more than 20 books in Chinese. His many presentations about the four Chinese socioeconomies and their international implications have proved to be provocative and creative. His new book entitled On the Future History of the Chinese Civilization: 22nd World and Its Chinese Civilizations is expected to be published in early 2019.

Sociocultural development and international relation are virtually incessant interconnectivities at different levels of human society which lead to the creation of different civilizations and its interactions. Therefore, the so-called “PAH-BRI” is believed to be a possible new form of socioeconomic cooperation. If so, the current trade disputes and future Sino-American relationship could be reconstructed effectively within the “‘ f -shaped’ PAH-BRI” formula. furthermore, entering the “#3.0”, it is expected that any form of the “PAH-BRI”-like projects is not easy for any single power or its bloc to manipulate mainly for its own interests, ethno-centrism and egoism. Any promising arrangement between any two parties should always leave a room for the third party to come or to drop out.

O cabelo apresenta estabilidade elevada quanto à deterioração, que ocorre desde a coleta até a análise, o que pode permitir um amplo período de investigação entre alguns meses até anos. Nesse contexto, amostras de cabelo humano e sintético foram analisadas utilizando as técnicas de fluorescência de raios X por dispersão de comprimento de onda (WDXRf) e Espectroscopia de emissão óptica com plasma induzido por laser (LIBS). A ideia dessa tese de doutorado foi identificar variações na composição química que pudessem diferenciar estas amostras. Além disso, foi efetuado nas amostras um procedimento de modificação estética comum como a descoloração dos cabelos. Assim, o objetivo foi avaliar o cabelo antes e após este procedimento para abranger uma ampla representatividade destas amostras. A partir das técnicas espectroanalíticas foi possível obter perfis espectrais e correlacioná-los com as amostras antes e após o procedimento de descoloração dos cabelos. Outro fato importante foi que os resultados mostraram a complementaridade entre as técnicas, permitindo estabelecer uma impressão digital para os diferentes tipos de cabelo. Sutureless Versus Conventional Pulmonary Vein Repair: A Magnetic Resonance Pilot Study

Background. Two different surgical techniques are used to repair anomalous pulmonary venous connection or pulmonary vein (PV) stenosis: the classic repair (CR) and the sutureless repair (SR). The purpose of this study was to compare the prevalence of PV stenosis between the two surgical approaches. Methods. Patients were prospectively recruited irrespective of symptoms or previous imaging findings. Cardiac magnetic resonance imaging and echocardiography were performed in a blinded fashion on the same day. Results. Twenty-five patients (13 male) after PV repair completed the study. Twelve patients had undergone CR and 13 SR (in 1 patient as a reoperation after CR). The median age at operation was 2 months (range: 1 day to 5 years) and was similar for both groups;the median age at the time of cardiac magnetic resonance was 9 years range: 6 to 17 years) and 9 years (range: 6 to 14 years) for the CR and SR, respectively. four patients had PV stenosis. All 4 patients had had total anomalous pulmonary venous connection, 1 patient had undergone repair with the CR and 2 with a primary SR;1 patient had first undergone a CR, followed by a SR for stenosis. Echocardiography provided complete visualization of all PVs in only 11 patients (44%). Notable stenosis of at least one PV was missed by echocardiography in 2 patients. Conclusions. This pilot study indicates that not only CR but also SR may be burdened by a risk of postoperative PV stenosis. Magnetic resonance imaging should be used routinely for the postoperative monitoring for the development of PV obstruction. (C) 2018 by The Society of Thoracic Surgeons

Membrane separation technology contributes significantly to solving the global water crisis due to the advances in the materials of desalination and water treatment membranes. Nevertheless, improving the flux, rejection, and fouling resistance of polymeric membranes remain important requirements to further increase their reliability and decrease the cost associated with their frequently required cleaning. In this thesis, graphene oxide (GO) was functionalized with amine (NH2) and carboxyl (COOH) functional groups to enhance its hydrophilicity and antifouling characteristics. Subsequently, nanocomposites polysulfone (PS) and different concentrations of functionalized graphene oxide (f-GO) were formulated and used to fabricate mixed matrix ultrafiltration membranes via phase inversion process. The developed membranes were characterized and tested for separation of water-oil emulsion. An array of characterization techniques was utilized to confirm GO functionalization including XPS, EDX, Raman, XRD, SEM, and TEM. The morphology of the fabricated membranes was also characterized by AfM and SEM to study the porosity, roughness and channeling structure of f-GO membranes. The mechanical properties of the membranes were analyzed using dynamic mechanical analysis (DMA) and the effect of f-GO on the membrane hydrophilicity was probed by contact angle measurements. Our results confirmed the functionalization of GO to f-GO leading to significant increase in hydrophilicity as indicated by ~ 13° decrease in water contact angle. Moreover, Young’s modulus of f-GO membranes displayed enhancement of up to 93% and 88% for GO/NH2 and GO/COOH, respectively. The concentration of f-GO dictates the porosity, roughness and water flux, which all increase at small f-GO concentrations (0.05 – 0.2 wt. %), but then decrease with higher loading (0.4 and 0.8 wt. %). An increase of 97% and 44% of the pure water permeability was attained for 0.2%GO/COOH and 0.2%GO/NH2 and high oil rejection of 97.92

The eclipse of Daniel's narrative: The limits of historical knowledge in the theological reading of Daniel

This article uses Hans frei's famous image of the eclipse' of biblical narrative to explore the link between situating the book of Daniel historically and grasping its theological point(s). The critical/conservative stand-off over the book of Daniel is rehearsed by way of key agenda-setting Victorian voices, and it is then argued that frei's perspective allows the reader to move on from assessing descriptive accuracy towards focusing on ascriptive purpose(s). Various examples of how such an ascriptive approach might clarify Daniel are considered, including specific attention to the complexities of Daniel 11's problematic relationship to what did and did not happen to Antiochus Epiphanes.
