SUBUNIT COMPOSITION AND COLD STABILITY Of THE PEA COTYLEDON MITOCHONDRIAL f1-ATPASE

The f1-ATPase isolated from pea cotyledon sumitochondrial particles contained six types of subunit with molecular weights of 57000 (.alpha.), 55000 (.beta.), 36500 (.gamma.), 26500 (.delta.), 22500 (.delta.'') and 8000 (.epsilon.). The same polypeptide composition was observed even when the purification was carried out at ***.C in the presence of proteolytic inhibitors, suggesting that the sixth subunit was not a proteolytic product formed during the isolation procedure. The six-subunit f1-ATPase exhibited considerable cold stability: it retained 65% of its activity after 24 h of incubation at ***.C and was more than 90% active after 48 h of incubation at ***.C. The 26.5 kDa protein could be dissociated from the remaining f1-ATPase by centrifugation in a linear sucrose gradient containing (NH4)2SO4 and deoxycholate. The resulting five-subunit f1-ATPase was considerably less stable at ***.C than the six-subunit enzyme. Several features suggest the possibility of a functional and structural relationship between the 26.5 kDa a protein of the pea cotyledon mitochondria and the mammalian oligomycin-sensitivity-conferring protein.

Isolated, nucleotide-depleted bovine-heart f1-ATPase exhibits a break in Arrhenius plot with a 2.7-fold increase in activation energy of ATP hydrolysis below ***.C. Analysis of intrinsic tyrosine fluorescence and of the circular dichroism of f1-ATPase showed an abrupt and reversible conformational change occurring at the break temperature, characteristic of a structural tightening at low temperature. Analysis of catalytic nucleotide binding sites using fluorescent ADP analog, 3''-O-(1-naphthoyl)adenosine diphosphate did not show any significant change in affinity of nucleotide binding around the transition temperature but the bound fluorophore exerted a more restricted motion and slower rotation at temperature below the break, indicating a change in the mobility of groups in the close neighbourhood. It is concluded that, as a result of temperature, two kinetically distinct states of f1-ATPase are induced, due to a change in enzyme conformation, which influences directly the properties of catalytic nucleotide binding sites.

New compounds from the aerial parts of Justicia gendarussa Burm.f. and their antioxidant and anti-inflammatory activities

Three new compounds (1-3), together with six known compounds (4-9), were isolated from the ethyl acetate fraction of ethanol extract of the aerial parts of Justicia gendarussa Burm.f. All of these known compounds were isolated from the plant for the first time. Their structures including absolute configurations were elucidated on the basis of HR-ESI-MS, NMR spectroscopic analyses, single crystal X-ray diffraction and comparison with the literatures. Compounds 1-9 were tested for their antioxidant and anti-inflammatory activities. Compounds 5 and 8 showed the antioxidant activities with IC50 values of 28.61 +/- 1.56 and 22.55 +/- 1.38 mu M, respectively. Compounds 2 and 3 showed anti-inflammatory activity in LPS-induced RAW 264.7 macrophages with the IC50 values of 20.95 +/- 1.11 and 16.50 +/- 1.04 mu M, respectively.

Preincubation of coupled submitochondrial particles with low concentrations of triorganotin compounds results in complete inhibition of the oligomycin-sensitive ATPase activity without any significant effect on the rate of succinate-driven ATP synthesis. The residual ATP synthetic activity is inhibited by oligomycin and uncouplers. The differential inhibition of ATP synthesis and hydrolysis by the triorganotin compounds examined suggests that the 2 processes are not ''mirror images'' of each other, but that they occur through different routes and that the f1f0-ATPase is at least bifunctional.

Antegrade Superficial femoral Artery Approach Using Manual Compression and 4-f Delivery System for Infrainguinal Occlusive Disease

Background: This study sought to assess the feasibility and safety of antegrade superficial femoral artery (SfA) duplex-guided puncture using a 4-f catheter sheath and manual compression for the treatment of infrainguinal occlusive lesions. Methods: from March 2017 to March 2019, all patients who underwent endovascular treatment employing an antegrade SfA approach for femoropopliteal or below-the-knee lesions were included in this study. follow-up included clinical and duplex ultrasound examinations at 1 month. The primary endpoint was the frequency of major complications at the puncture site. Results: In all, 71 procedures were performed in 70 patients (46 men;mean age: 79.1 +/- 11.5 years), most of whom presented with critical limb-threatening ischemia (n = 59;83%). Of these 71 procedures, 35 (49.3%) were performed for isolated femoropopliteal lesions and 24 (33.8%) for multilevel lesions. The primary endpoint was observed in 4 patients (5.6%). There were 3 observed hematomas (4.2%): 2 required surgical treatment, and 1 necessitated transfusion without intervention. At 30 days, we reported 1 pseudoaneurysm (1.4%). Conclusions: Our results showed that antegrade SfA puncture is feasible and safe with duplex ultrasound guidance and use of a 4-f catheter sheath.

The γ subunit of the f 1 moiety of the bovine mitochondrial H + -ATP synthase is shown to function as a component of the gate. Addition of purified γ subunit to f o -liposomes inhibits transmembrane proton conduction. This inhibition can be removed by the bifunctional thiol reagent diamide. Immunoblot analysis shows that the diamide effect is likely due to disulphide bridging of the γ subunit with the PVP protein of the f o sector.

The photoaffinity label 2-azido-2′,4,4′-trinitro-6- sec. butyldiphenylether inhibits photosynthetic electron transport in the thylakoid membrane as well as plastohydroquinone-plastocyanin-oxidoreductase activity in the isolated chloroplast cytochrome b 6 - f complex. In the latter, the 14 C-labeled inhibitor upon UV-irradiation binds covalently only to the cytochrome b 6 and the Rieske fe-S peptide subunit.

The subunit structure of the f 1 -ATPase from the thermophilic bacterium PS3 was probed by comparing electron microscopic images of the subunit complexes reconstituted to contain different subunit compositions. The following structural features were observed. (1) The α 3 β 3 complex has a hexagonal, apparently symmetrical arrangement of masses with a central cavity. (2) The projections of the α 3 β 3 δ complexes are similar to those of the α 3 β 3 complexes. (3) In contrast, the α 3 β 3 λ complex has an additional mass in the centre which is similar to that found in the native enzyme (α 3 β 3 γδϵ). from these observations, it is concluded that the central mass in the f 1 -ATPase is comprised mostly of the γ subunit.

MEMBRANE-BOUND f420H2-DEPENDENT HETERODISULfIDE REDUCTASE IN METHANOGENIC BACTERIUM STRAIN GOL AND METHANOLOBUS-TINDARIUS

Washed membrane or cytoplasmic fractions of the methanogenic bacterium strain Göl catalyzed the oxidation of coenzyme f 420 H 2 with a variety of electron acceptors. The f 420 H 2 -oxidizing activity of the cytoplasmic fraction could be assigned to a NADP + :f 420 oxidoreductase. The membrane fraction but not the cytoplasmic fraction catalyzed the oxidation of f 420 H 2 with the concomitant reduction of the heterodisulfide of 2-mercapto-ethanesulfonate and 7-mercaptoheptanoylthreonine phosphate (CoM-S-S-HTP) at a rate of 100 nmol min-mg protein according to the following equation: f 420 H 2 +CoM-S-S-HTP → f 420 + CoM + HTP-SH. The activity depended linearly on the membrane protein up to a concentration of 60 μg ml The physiological electron acceptor CoM-S-S-HTP could not be replaced by the corresponding homodisulfides CoM-S-S-CoM and HTP-S-S-HTP or by NADP + . A membrane-bound f 420 H 2 -dependent CoM-S-S-HTP reductase was also detected in Methanolobus tindarius exhibiting a specific activity of 75 nmol min m ̇ g protein. The absence of a f 420 -dependent hydrogenase in this organism excludes the involvement of this enzyme in electron transfer from H 420 H 2 to CoM-S-S-HTP.

EXPLORATION Of SIGMA-SUBUNIT INTERACTIONS IN BEEf-HEART MITOCHONDRIAL f1-ATPASE BY MONOCLONAL-ANTIBODIES

Three monoclonal antibodies (mAbs) recognizing distinct epitopes on the δ-subunit of beef heart mitochondrial f 1 -ATPase were studied for their reactivity towards the δ-subunit both in isolated f 1 and in the f 0 -f 1 complex of submitochondrial particles. Two of the antibodies termed mAb δ 195 and mAb δ239 had free access to δ in f 1 and the f 0 -f 1 complex. Partial hindrance was observed for the third antibody mAb δ22. By a double antibinding assay, it was found that the binding sites for mAb δ195 and mAb δ239 were close to each other and possibly overlapping. Mapping studies conducted with the isolated δ-subunit showed that mAb δ195 and mAb δ239 interacted with the N-terminal portion of δ extending from Ala-1 to Met-16, whereas mAb δ22 interacted with the fragment spanning Ser-17-Glu-68. It was concluded that the Ala-1-Met-16 segment of the δ-subunit in f 1 and the f 0 -f 1 complex is freely accessible from the outside, whereas the Ser-17-Glu-68 segment of δ is partially hidden, possibly as a result of interactions with other subunits.
