Trichodermates A - f, New Cytotoxic Trichothecenes from the Plant Pathogenic fungus Trichoderma sp.

Trichodermates A-f (1-6, resp.), six new trichothecene polyunsaturated octadioic acid esters, and (-)-harzianum B (7) were isolated from the fermentation extract of Trichoderma sp., a plant pathogenic fungus isolated from stem rot of an unidentified tree in Thailand. The structures of 1-7 were elucidated by NMR experiments. The absolute configuration at C(12) in 3 was assigned by in situ dimolybdenum circular dichroism method, whereas that in 7 was deduced after hydrolysis of 7 to 8 via modified Mosher's method. Compounds 1 and 2 showed modest cytotoxic activities against the K562 (human myelogenous leukemia) cell line with IC50 values of 12.12 and 13.08 mu m, respectively.

Umbelliferae plants, which are widely used as traditional Chinese medicine because of their characteristics of relieving rheumatism, alleviating fever, circulating blood and easing pain. This experimental study was based on ear edema model caused by 12-O-tetracycline-propylphenol-13-acetic acid (TPA) in mice and compared with the Ibuprofen (Ib) group. Gas chromatography-mass spectrometry (GC -MS) was used to analyse the composition of the essential oils from the four studied Umbelliferae plants (Angelica sinensis (Oliv.) Diels, A. dahurica (Hoffm.) Benth. & Hook.f. ex franch. & Sav., A. pubescens Maxim and foeniculum vulgare Mill.). Biologically active components in volatile oils from the four studied Umbelliferae plants were evaluated. The expression levels of inflammatory cytokines Tumor Necrosis factor-alpha (TNf-alpha), Cyclooxygenase-2 (COX-2), Interleukin-6 (IL-6) and RelA (p65) in mouse skin were determined by immunohistochemical method. The refractive index of the four essential oils was calculated. A total of 239 compounds were identified by GC-MS from the four studied plants, and the main constituents were osthole (44.61%, APEOs), obepin (0.59%, APEOs & 86.58%, fVEOs), undecanol (8.58%, ADEOs), alpha-muurolene (7.95%, ADEOs) and cis-anethol (9.11%, ADEOs). E-ligustilide (0.14%, APEOs & 81.14%, ASEOs), (-)-spathulenol (0.08%, fVEOs & 1.21%, ASEOs), (-)-terpinen-4-ol (4.91%, fVEOs), 2-butylthiolane (5.76%, APEOs) and alpha-bisabolol (3.80%, APEOs). This study showed that all the essential oils from the four studied Umbelliferae plants contained various lactones, including ligustrongolactone, trans-anisol and imperatorin. According to the results of the TPA induction test in the mouse ear edema model, the essential oils of four Umbelliferae plants reduced the levels of inflammatory cytokines TNf-alpha, COX-2, IL-6 and p65. A computer program to calculate precise solutions (exact to the limit of computer accuracy) for the cumulative distribution function of the statistical t and f distributions is detailed. A routine that makes iterative calls of this algorithm to calculate precisely the inverse function of these distributions by successive approximation is also provided. The program provided yields precise levels of probability confidence of both fisher''s f and Student''s t tests, and the inverse calculation of critical values given p. It is written in a subset of BASIC which should run on any small computer, requiring only 1538 bytes on the Commodore 64.

Sparse LMS/f algorithms with application to adaptive system identification

Standard least mean square/fourth (LMS/f) is a classical adaptive algorithm that combined the advantages of both least mean square (LMS) and least mean fourth (LMf). The advantage of LMS is fast convergence speed while its shortcoming is suboptimal solution in low signal-to-noise ratio (SNR) environment. On the contrary, the advantage of LMf algorithm is robust in low SNR while its drawback is slow convergence speed in high SNR case. Many finite impulse response systems are modeled as sparse rather than traditionally dense. To take advantage of system sparsity, different sparse LMS algorithms with l(p)-LMS and l(0)-LMS have been proposed to improve adaptive identification performance. However, sparse LMS algorithms have the same drawback as standard LMS. Different from LMS filter, standard LMS/f filter can achieve better performance. Hence, the aim of this paper is to introduce sparse penalties to the LMS/f algorithm so that it can further improve identification performance. We propose two sparse LMS/f algorithms using two sparse constraints to improve adaptive identification performance. Two experiments are performed to show the effectiveness of the proposed algorithms by computer simulation. In the first experiment, the number of nonzero coefficients is changing, and the proposed algorithms can achieve better mean square deviation performance than sparse LMS algorithms. In the second experiment, the number of nonzero coefficient is fixed, and mean square deviation performance of sparse LMS/f algorithms is still better than that of sparse LMS algorithms. Gastrodia elata, dry tubers of which are processed into traditional medicine Tianma, is a heterotrophic plant species exclusively depending on the fungal species of Armillaria. To identify whether species of Armillaria impact the yield and medicinal quality of G. elata BL. f. elata, we experimentally tested the effect of eight Armillaria species (identified by sterility test) on the tuber production of G. elata f. elata along an altitude gradient. for the first time, we found that the taxonomic status of Armillaria had a significant effect on the tuber yield. Two species of Armillaria (Chinese Biological Species B and D) could not form an effective symbiosis with G. elata. We also showed that the altitude could affect the water content of the tubers of G. elata. Our results revealed a variable symbiotic relationship among biological species of Armillaria and G. elata f. elata and will be of value in Tianma production in China.

A series of experiments was carried out to investigate the role of some polar amino acids in the a-subunit of the ATP synthase of Escherichia coli. Site-directed mutagenesis resulted in the amino acid substitutions Ser-199 .fwdarw. Ala, Ser-202 .fwdarw. Ala, Ser-206 .fwdarw. Ala, Arg-61 .fwdarw. Gln or Asp-44 .fwdarw. Asn. None of these amino acid substitutions affected the ability of the cells to carry out oxidative phosphorylation. It was concluded therefore that the effect of the subtitution of leucine for Ser-206 reported previously (Cain, B.D. and Simoni, R.D. (1986) J. Biol. Chem. 261, 10043-10050) was due to the presence of the leucine rather than the absence of serine. Even though cells carrying the Asp-44 .fwdarw. Asn substitution were able to carry out oxidative phosphorylation, membranes from such cells remained proton-impermeable after removal of the f1-ATPase. It appears likely that the proton pore of the f0 of the ATP synthase of E. coli consists of four amino acids, namely Arg-219, Glu-210 and His-245 of the a-subunit and Asp-61 of the c-subunit.

Bovine heart submitochondrial particles depleted of f1 by treatment with urea (f1-depleted particles) were incubated with soluble f1-ATPase. The binding of f1 to the particles and the concomitant conferral of oligomycin sensitivity on the ATPase activity required the presence of cations in the incubation medium. NH4+, K+, Rb+, Cs+, Na+ and Li+ promoted reconstitution maximally at 40-74 mM, guanidinium+ and Tris+ at 20-30 mM, and Ca2+ and Mg2+ at 3-5 mM. The particles exhibited a negative .zeta.-potential, as determined by microelectrophoresis, and this was neutralized by mono- and divalent cations in the same concentration range as that needed to promote f1 binding and reconstituion of oligomycin-sensitive ATPase. The cations act by neutralizing negative charges on the membrane surface, mainly negatively charged phospholipids. These results are discussed in relation to earlier findings reported in the literature with f1-depleted thylakoid membranes and with submitochondrial particles depleted of both f1 and the coupling proteins f6 and oligomycin sensitivity-conferring protein.

Tectona grandis L.f is a timber plant that is commonly referred to as teak. Its wide use as a medicine in the various indigenous systems makes it a plant of importance. A wide gamut of phytoconstituents like alkaloids, phenolic glycosides, steroids, etc. has been reported. A renewed interest in this plant has resulted in scientific investigations by various researchers towards the isolation and identification of active constituents along with scientific proof of its biological activities. The different parts of the plant have been scientifically evaluated for their antioxidant, antipyretic, analgesic, hypoglycemic, wound healing, cytotoxic, and many more biological activities. Documentation of this scientific knowledge is of importance to have consolidated precise information encompassing the various aspects of this plant, which could provide a base for future studies. This review is a compilation of the salient reports on these investigations concerning phytochemistry, the methods used to identify and quantify the constituents, the evaluation methods of the biological activity, toxicological studies, allergies and the patent/patent applications. INTERACTION Of f1-ATPASE, fROM OX HEART-MITOCHONDRIA WITH ITS NATURALLY-OCCURRING INHIBITOR PROTEIN - STUDIES USING RADIOIODINATED INHIBITOR PROTEIN

The ox heart mitochondrial inhibitor protein may be iodinated with up to 0.8 mol 125I/mol inhibitor with no loss of inhibitory activity, with no change in binding affinity to submitochondrial particles and without alteration in the response of membrane-bound inhibitor to energization. Tryptic peptide maps reveal a single labeled peptide, consistent with modification of the single tyrosine residue of the protein. A single type of high-affinity binding site (Kd = 96 .cntdot. 10-9 M) for the inhibitor protein was measured in submitochondrial particles. The concentration of this site is proportional to the amount of membrane-bound f1, and there appears to be 1 such site per f1 molecule. The ATP hydrolytic activity of submitochondrial particles is inversely proportional to the occupancy of the high-affinity binding site for the inhibitor protein. No evidence is found for a non-inhibitory binding site on the membrane or on other mitochondrial proteins. In intact mitochondria from bovine heart, the inhibitor protein is present in an .apprx. 1:1 ratio with f1. Submitochondrial particles prepared by sonication of these mitochondria with MgATP contain about 0.75 mol inhibitor protein per mol f1, and show about 25% of the ATPase activity of inhibitor-free submitochondrial particles. Additional inhibitor protein can be bound to these particles to a level of 0.2 mol/mol f1, with consequent loss of ATPase activity. If MgATP is omitted from the medium, or inhibitors of ATP hydrolysis are present, the rate of combination between f1 and its inhibitor protein is very much reduced. The equilibrium level of binding is, however, unaltered. These results suggest the presence of a single, high-affinity, inhibitory binding site for inhibitor protein on membrane-bound f1. The energization of coupled submitochondrial particles by succinate oxidation or by ATP hydrolysis results in both the dissociation of inhibitor protein into solution, and the activation of ATP hydrolysis. Peritz'' f test was previously found to be the most robust statistical multiple comparison test able to hold all comparisons among group means to a given experimentwise error rate. A BASIC program which will perform this test quickly on a desk-top microcomputer, needing fewer than 11 kbytes [kilobyte] of RAM memory, was presented. The program was run in the author''s laboratory using an inexpensive VIC-20 computer.
