The reduction of ruthenium(III) complexes with triazolopyrimidine ligands by ascorbic acid and mechanistic insight into their action in anticancer therapy

Kinetic studies of the reduction of two ruthenium(III) complexes, mer-[RuCl3(dmso)(H2O)(tmtp)]center dot 2H(2)O (1) and mer,cis-[RuCl3(dbtp)(2)(dmso)] (2) (where dmso - dimethylsulfoxide, tmtp - 5,6,7-trimethyl-1,2,4-triazolo[1,5-a]pyrimidine and dbtp -5,7-ditertbutyl-1,2,4-triazolo[1,5-a]pyrimidine), by ascorbic acid were performed as a function of antioxidant concentration in acetate buffer within the pH range of 2.9-5. The rapid reduction of the ruthenium(III) complexes (1) and (2) resulted in the formation of the mer-[RuCl3(dmso)(H2O)(tmtp)](-) and mer,cis-[RuCl3(dbtp)(2)(dmso)]- ions and was followed by successive dissociation of the chloride ligands. The second-order rate constant (k(1)) for the reduction of the mer-[RuCl3(dmso)(H2O)(tmtp)]center dot 2H(2)O complex and the first-order rate constant for the hydrolysis of its reduced form were found to be 134 +/- 2 M-1 s(-1) and (3.8 +/- 0.9) x 10(-2) s(-1) at 25 degrees C and pH = 2.9, respectively. Similarly, the fast process assigned to the reduction of the mer,cis-[RuCl3(dbtp)(2)(dmso)] complex and the subsequent, slower process attributed to the hydrolysis of the cis-[RuCl3(dbtp)(2)(dmso)](-) ion were characterized by rate constants of 145.5 +/- 0.8 M-1 s(-1) and (9 +/- 2) x 10(-3) s(-1) at 25 degrees C and pH = 2.9, respectively. Obtained data indicated that the reduction of the ruthenium(III) complexes strongly depends on pH and accelerates with increasing pH. The kinetic data indicates that the redox process followed an inner-sphere electron-transfer mechanism at pH higher than 3.

Background Abdominal aortic aneurysm (AAA) is a serious vascular disease for which there is no effective drug treatment. The incidence of AAA increases significantly as a subject ages, and the molecular mechanism of AAA formation remains elusive. In the present study, we investigated the role of syndecan-4 (SDC4), an important component of focal adhesions, in AAA formation and its association with phenotypic changes in vascular smooth muscle cells (VSMCs). Methods and results The protein expression levels of SDC4 were significantly decreased in human AAA tissue and those of an AAA mouse model. Moreover, SDC4 knockout (KO) in mice accelerated the formation and rupture of AAAs induced by angiotensin II (Ang II) and calcium chloride (CaCl2) Mechanistically, the decrease in SDC4 led to the transformation of cultured VSMCs from a contractile to a secretory phenotype. The RhoA-f/G-actin-myocardin-related transcription factor-A (MRTf-A) signalling pathway was shown to be involved in SDC4-dependent VSMC alteration. Sphingosine-1-phosphate (S1P), a G-protein-coupled receptor, attenuated the AAA formation in SDC4-KO and wild-type (WT) mice in response to Ang II and CaCl2 stimulation. Conclusion We herein demonstrated that silencing SDC4 was associated with increased AAA formation and phenotypic changes in VSMCs via the RhoA-f/G-actin-MRTf-A pathway. These findings indicated that a reduction in SDC4 expression was an important pathological alteration and potential therapeutic target for AAA formation.

分析不同梨品种及其杂交后代的腐烂病抗性,旨在为进一步挖掘抗病相关基因,推动梨树抗病育种提供借鉴。采用室内人工接种腐烂病菌的方法对7个梨品种进行进行腐烂病抗性鉴定评价,并于2019—2022年连续4 a对6个梨杂交组合后代进行田间腐烂病病级调查。结果表明,7个品种中,‘黄冠’和‘八月红’病害级别为1级,而‘红香酥’的病害级别达到4级,其他品种为3级。2019—2022年,这6个杂交组合的发病率为67.24%～100%,其中最抗病的组合为‘锦丰’×‘黄冠’,抗病单株比例达到92.02%,6个杂交组合的f1代抗感分离比差异也较大,为0.01:1～1.79:1,各组合的变异系数与超低亲率分别为23.95%～67.99%、0～8.65%。综上,初步将梨腐烂病抗性鉴定为多基因控制的数量性状,并且认为梨腐烂病的抗性遗传会更加趋向于感病性状。

The effect of slag variability in the attempted manufacture of AYf (alite-ye'elimite-ferrite) cement clinker at both laboratory and pilot scale

The production of AYf (alite-ye'elimite-ferrite) clinker was tested at laboratory and semi-industrial scale using by-products from the metallurgical industry: AOD slag; ladle slag; and fayalitic slag. Alite could be produced with ye'elimite using fluorine originating from AOD (argon oxygen decarburisation) slag as a mineraliser. After a successful laboratory demonstration, the clinker production was scaled to a semi-industrial trial. It was discovered that the reason for the absence of alite formation in a semi-industrial demonstration was that the AOD slag from the specific batch did not perform the designed mineralisation effect for alite formation. This study demonstrates that alite-ye'elimite can be produced at 1260 °C at laboratory scale by using fluorine mineralisation originating from an industrial by-product – in this case, AOD slag. However, the utilisation of by-products for delicate reactions requires detailed determination of the properties of the slag, as the variability from the same source yields different clinker chemistries and mineral phases.

Mugwort (Artemisia vulgaris L.) essential oils rich in germacrene D, and their toxic activity

The chemical composition of essential oils of Artemisia vulgaris L. plants collected in Lithuanian has been reported. The oils were obtained by hydro-distillation from air-dried aerial parts. Up to 75 compounds (an average 88.7- 99.3% of the total oil content) were identified by GC-MS and GC (fID). Germacrene D (10.6 +/- 2.5 - 30.5 +/- 2.0%) was revealed as the first principal constituent in the investigated oils. To the best of our knowledge, mugwort essential oils rich in germacrene D are quite rare. 1,8-Cineole (6.9 +/- 1.9 - 9.8 +/- 1.1%, in 4 oils), beta-pinene (6.7 +/- 1.2 and 8.4 +/- 2.3%), sabinene (8.5 +/- 1.8%), cis-thujone (12.9 +/- 2.5%), beta-caryophyllene (5.5 +/- 1.3 - 16.7 +/- 4.2%, in 5 samples), caryophyllene oxide (6.4 +/- 1.1 - 9.3 +/- 1.3%, in 4 oils), alpha-humulene (5.5 +/- 1.5%) and davanone (6.5 +/- 2.5%) were found to be the second or third major compounds. The toxic activity of five mugwort essential oil samples was evaluated using a brine shrimp (Artemia sp.) bioassay. LC50 mean values (10.25-19.11 mu g/mL) revealed that the oils containing appreciable amounts of germacrene D were notably toxic.

Intensification of plasma process for the decomposition of p-xylene as a tar model compound to minimize the unwanted byproduct formation

In this study, the decomposition of p-xylene is studied in different carrier gases (N 2 , H 2, and CO 2 ) across a range of plasma powers (5 to 40 W). At 5 to 10 W, the decomposition of p-xylene was maximum in N 2 carrier gas, whereas it was minimum in CO 2 carrier gas. However, nearly complete decomposition of p-xylene >98 % occurred in all carrier gases at 20 W. The effect of carrier gases and plasma input power on the decomposition of p-xylene disappears at higher power (20–40 W) due to the maximum decomposition of p-xylene. However, the yield of gaseous products significantly varies at all levels of power and the nature of carrier gas. The yield of H 2 and CO (produced only in CO 2 ) continuously increases with increasing power (5 to 40 W) in N 2 and CO 2 gases. However, the yield of lower hydrocarbons remains unaffected with power and it was below 2 % in both carrier gases. In H 2 carrier gas, the yield of lower hydrocarbons (LHC) increases significantly to 18.5 % with the increase in power to 40 W. A significant amount of carbon was missing in all carrier gases because of the formation of solid residues (unwanted byproducts). Therefore, the synergistic effect of temperature was studied to investigate the effect on product distribution. It was noted that the LHC’s yield increased to 82 % from 18.5 % with increasing the temperature up to 300 °C in H 2 carrier gas with the maximum yield of CH 4 (58 %). further, increase in temperature up to 400 °C, the yield of CH 4 increased to 75 %, whereas the total yield remained constant. The effect of temperature on the yield of lower hydrocarbons was not significant in CO 2 and N 2 carrier gas as the maximum yield of lower hydrocarbons reached to only 10 % in N 2 carrier gas.

Optimizing membrane performance for efficient water treatment is crucial for sustainable development and environmental protection, aligning with UN SDGs. This study involves experimental design, statistical reliability of small data, and explainable machine learning (ML) using SHAP (Shapley Additive Explanations). The research uses ML models and statistical tests to ensure data reliability and stationarity and investigate various membranes’ fouling and separation efficiency (MX-CM, PDMX-CM, and SPDMX-CM). Stationarity tests, including the Augmented Dickey–fuller (ADf) and Phillips–Perron (PP) tests, revealed that MX-CM is stationary at level (I(0)), while PDMX-CM and SPDMX-CM required first differencing (I(1)) to achieve stationarity. SHAP analysis showed that in the fouling study, higher values of PDMX-CM and MX-CM positively influenced model predictions, with SHAP values of +0.09 for Cycle, −0.06 for PDMX-CM, and −0.06 for MX-CM. In the separation efficiency study, Cycle had a neutral impact (0.00), PDMX-CM had a slight positive effect, and MX-CM had a slight negative impact. These findings highlight the importance of ensuring data stationarity and utilizing SHAP for model explainability in predicting membrane performance. Accurate preprocessing and model interpretation enhance decision-making and optimization in membrane fouling and separation efficiency studies, ensuring robust and reliable ML models.

A theoretic study of a distance-based regression model

The distance-based regression model has many applications in analysis of multivariate response regression in various ?elds, such as ecology, genomics, genetics, human microbiomics, and neuroimaging. It yields a pseudo f test statistic that assesses the relation between the distance(dissimilarity) of the subjects and the predictors of interest. Despite its popularity in recent decades, the statistical properties of the pseudo f test statistic have not been revealed to our knowledge. This study derives the asymptotic properties of the pseudo f test statistic using spectral decomposition under the matrix normal assumption, when the utilized dissimilarity measure is the Euclidean or Mahalanobis distance. The pseudo f test statistic with the Euclidean distance has the same distribution as the quotient of two Chi-squared-type mixtures. The denominator and numerator of the quotient are approximated using a random variable of the form ξχ_d^2+ η, and the approximate error bound is given. The pseudo f test statistic with the Mahalanobis distance follows an f *** simulation studies, the approximated distribution well matched the "exact" distribution obtained by the permutation procedure. The obtained distribution was further validated on H1N1 in?uenza data, aging human brain data, and embryonic imprint data.

Tripterygium wilfordii Hook f (TwHf) and its extracts have long been used for the treatment of rheumatoid arthritis, autoimmune diseases, and kidney disease due to their anti-inflammatory, immunoregulatory, and other pharmacological effects. However, the clinical immunoregulatory effects of TwHf and its extracts remain unclear, so we reviewed their effects for use in clinical practice. This review provides a comprehensive summary of the recent literature on the immunoregulatory effects of TwHf and its extracts in clinical studies. TwHf and its extracts affect the proliferation and activation of T and B cells;ratio of T cell subsets;inflammatory response of monocytes, macrophages, and immunoglobulins;and secretion of many cytokines. Together, these effects dictate immune function in a variety of diseases. TwHf and its extracts can be used alone or in combination with existing therapies against many immune disorders through immunomodulation.
