目的比较分析尘螨过敏原蛋白Der p 2和Der f 2的一级、二级结构并预测抗原表位。方法从网上数据库下载Der p 2和Der f 2蛋白的序列,分析比较Der p 2和Der f 2蛋白的一级、二级结构,得出Der p 2和Der f2蛋白中可能的功能位点、信号肽位置。结果 Der p 2和Der f 2蛋白都由146个氨基酸组成,含有9个潜在可能的蛋白结合位点,二级结构主要由β-折叠片组成,在其N-末端为第1-17区段为信号肽。Der p 2蛋白含有9个线性B细胞表位、2个构象B细胞表位和6个HLA II的高亲和力区域,而Der f 2蛋白含有9个线性B细胞表位、2个构象B细胞表位和5个HLA II的高亲和力区域。结论应用生物信息学方法预测和比较过敏原蛋白Der p 2和Der f 2的结构与抗原表位信息,可为进一步研究过敏原蛋白Der p 2和Der f 2的生物学功能、疫苗研究奠定基础。

Kristeva vis-a-vis Hegel: forgiveness as Psychoanalytic Interpretation and Absolute Knowing

This article reconstructs and compares Kristeva's account of psychoanalytic interpretation as a practice of forgiveness with Hegel's account of the origin of Absolute Knowing in the forgiveness constitutive of mutual recognition. An emphasis on homologies between the memory-work of Kristevan psychoanalysis and the recollective process of Hegelian Absolute Knowing elicits deeper affinities between Kristeva and Hegel than have previously been supposed. Both Kristevan and Hegelian forgiveness operate as the healing of an originary guilt, achieved through the verbal confession and examination of the confessor's particular biographical and historical past, negating and raising up-sublating or sublimating-the contradictions of consciousness. Psychoanalysis and Absolute Knowing both enact in the present the reconciliation which religion perpetually defers to an unspecified future. While not replacing the institution of legal punishment, Kristevan and Hegelian forgiveness qua personal and social renewal and rebirth provide for the possibility of radical political transformation.

New Models Used to Determine the Dioxins Total Amount and Toxicity (TEQ) in Atmospheric Emissions from Thermal Processes

In order to reduce the calculation effort during the simulation of the emission of polychlorinated dibenzo-p-dioxins and furans (PCDD/f) during municipal solid waste incineration, minimizing the number of simulated components is mandatory. for this purpose, two new multilinear regression models capable of determining the dioxins total amount and toxicity of an atmospheric emission have been adjusted based on previously published ones. The new source of data used (almost 200 PCDD/f analyses) provides a wider range of application to the models, increasing also the diversity of the emission sources, from industrial and laboratory scale thermal processes. Only three of the 17 toxic congeners (1,2,3,6,7,8-HxCDD, 2,3,7,8-TCDf and OCDf), whose formation was found to be linearly independent, were necessary as inputs for the models. All model parameters have been statistically validated and their confidence intervals have been calculated using the Bootstrap method. The resulting coefficients of determination (R-2) for the models are 0.9711 +/- 0.0056 and 0.9583 +/- 0.0085;its root mean square errors (RMSE) are 0.2115 and 0.2424, and its mean absolute errors (MAE) are 0.1541 and 0.1733 respectively.

Research on Rice Leaf Disease Recognition Based on BP Neural Network

To solve the problem of mistake recognition among rice diseases, automatic recognition methods based on BP(back propagation) neural network were studied in this paper for blast, sheath blight and bacterial blight. Chose mobile terminal equipment as image collecting tool and built database of rice leaf images with diseases under threshold segmentation method. Characteristic parameters were extracted from color, shape and texture. furthermore, parameters were optimized using the single-factor variance analysis and the effects of BP neural network model. The optimization would simplify BP neural network model without reducing the recognition accuracy. The finally model could successfully recognize 98%, 96% and 98% of rice blast, sheath blight and white leaf blight, respectively.

Vitamin K antagonist rodenticide pharmacodynamics (PD) is studied in rodents with traditional laboratory tests. We wondered if thrombin generation test (TGT) could add value. Difethialone (10 mg/kg) was administered per os to 97 OfA-Sprague Dawley rats. PD was studied over a 72 h-period using the Calibrated Automated Thrombogram on platelet poor plasma before and after intoxication (3 female and 3 male rats for each 13 time points) and TGT parameters were compared with the prothrombin time (PT) and vitamin K dependent factor activities previously reported. following intoxication, preliminary tests evidenced rapid and full inhibition of thrombin generation triggered with 5 or 20 pM human recombinant tissue factor. To study the evolution of TGT parameters following difethialone intake, we adapted the test by complementing intoxicated rat samples with pooled normal rat plasma (3/1, v/v). Adapted TGT confirmed the known higher procoagulant basal level in females compared to males through higher endogenous thrombin potential (ETP) and peak height (PH) (p < 0.0001 and p = 0.0003, respectively). An exponential model fitted well the PH and ETP decay after intoxication. In contrast to PT, the decreases were observed immediately following VKA intake and had comparable time to halving values: 10.5 h (95% CI [8.2;13.6]) for ETP and 10.4 h (95% CI [7.8;14.1]) for PH. The decrease of fVII and fX preceded that of PH, ETP and fII while fIX decreased later on, contributing to the severe hypo-coagulability. We demonstrated that TGT performed in samples of intoxicated rats complemented with normal plasma is a reliable tool for evaluation of VKA rodenticide PD in rats.

Comparison of Double-freeze versus Modified Triple-freeze Pulmonary Cryoablation and Hemorrhage Volume Using Different Probe Sizes in an In Vivo Porcine Lung

Purpose: To determine size of ablation zone and pulmonary hemorrhage in double-freeze (Df) vs modified triple-freeze (mTf) cryoablation protocols with different probe sizes in porcine lung. Materials and Methods: In 10 healthy adult pigs, 20 pulmonary cryoablations were performed using either a 2.4-mm or a 1.7-mm probe. Either conventional Df or mTf protocol was used. Serial noncontrast CT scans were performed during ablations. Ablation iceball and hemorrhage volumes were measured and compared between protocols and probe sizes. Results: With 1.7-mm probe, greater peak iceball volume was observed with Df compared with mTf, although difference was not statistically significant (16.1 mL +/- 1.9 vs 8.8 mL +/- 3.6, P = .07). With 2.4-mm probe, Df and mTf produced similar peak iceball volumes (14.0 mL +/- 2.8 vs 14.6 mL +/- 2.7, P = .88). Midcycle hemorrhage was significantly larger with Df with the 1.7-mm probe (94.3 mL +/- 22.2 vs 19.6 mL +/- 2.1, P = .02) and with both sizes combined (93.2 mL +/- 17.5 vs. 50.9 mL +/- 12.6, P = .048). Rate of hemorrhage increase was significantly higher in Df (10.4 mL/min vs 5.1 mL/min, P = .003). End-cycle hemorrhage was visibly larger in Df compared with mTf across probe sizes, although differences were not statistically significant (P = .14 for 1.7 mm probe, P = .18 for 2.4 mm probe, and P = .07 for both probes combined). Rate of increase in hemorrhage during the last thaw period was not statistically different between Df and mTf (3.0 mL/min vs 2.8 mL/min, P = .992). Conclusions: mTf reduced rate of midcycle hemorrhage compared with Df. With mTf, midcycle hemorrhage was significantly smaller with 1.7-mm probe;although noticeably smaller with 2.4-mm probe, statistical significance was not achieved. Iceball size was not significantly different across both protocols and probe types.

Dual BRD4 and AURKA Inhibition Is Synergistic against MYCN-Amplified and Nonamplified Neuroblastoma

A majority of cases of high-risk neuroblastoma, an embryonal childhood cancer, are driven by MYC or MYCN-driven oncogenic signaling. While considered to be directly "undruggable" therapeutically, MYC and MYCN can be repressed transcriptionally by inhibition of Bromodomain-containing protein 4 (BRD4) or destabilized post-translationally by inhibition of Aurora Kinase A (AURKA). Preclinical and early-phase clinical studies of BRD4 and AURKA inhibitors, however, show limited efficacy against neuroblastoma when used alone. We report our studies on the concomitant use of the BRD4 inhibitor I-BET151 and AURKA inhibitor alisertib. We show that, in vitro, the drugs act synergistically to inhibit viability in four models of high-risk neuroblastoma. We demonstrate that this synergy is driven, in part, by the ability of I-BET151 to mitigate reflexive upregulation of AURKA, MYC, and MYCN in response to AURKA inhibition. We then demonstrate that I-BET151 and alisertib are effective in prolonging survival in four xenograft neuroblastoma models in vivo, and this efficacy is augmented by the addition of the antitubule chemotherapeutic vincristine. These data suggest that epigenetic and posttranslational inhibition of MYC/MYCN-driven pathways may have significant clinical efficacy against neuroblastoma.

Jack Michael, Behavior Analyst

Jack Michael was the quintessential behavior analyst. Despite his reputation as a theoretician and expert in verbal behavior and on Skinner's book Verbal Behavior, Jack was actually an applied behavior analyst in the literal sense before anyone had coined the term. Prior to becoming one of Jack's doctoral students at Western Michigan University, I was a behavior-analytic neophyte. After I graduated and began my own teaching (modeled after Jack's behavior-analytic method of teaching) and thinking and writing about behavior analysis, I found myself talking and thinking like Jack. He had a similar behavior changing effect not only on many of his other students, but also on others who had the good fortune to know him.

Incidence, Risk factors, and Management of Blindness after Orbital Surgery

Purpose: Severe vision loss is a risk of orbital surgery which physicians should counsel patients about, but the overall risk rate is unknown. This research was conducted to determine the risk of severe vision loss related to orbital surgery. Design: Retrospective review. Participants: Patients who underwent orbital surgery at either of 2 academic medical centers between January 1994 and December 2014. Methods: A billing database search was conducted to identify all patients who had orbital surgery during the study period, cross-checked against diagnostic codes related to vision loss. Charts were screened to determine baseline demographic and medical history, surgical procedure, intraoperative and perioperative management, and visual acuity. Patients with preoperative visual acuity >= 20/200 that worsened <= 20/400 after orbital surgery were included for detailed review. Statistical analysis was conducted to identify factors posing particular risk or benefit to visual outcome in these patients. Main Outcome Measures: Visual acuity after orbital surgery. Results: A total of 1665 patients underwent orbital surgery during the inclusion period, with 14 patients sustaining severe vision loss ranging from counting fingers at 1 foot to no light perception (overall risk, 0.84%). The causes of vision loss included retrobulbar hemorrhage, malpositioned implant, optic nerve ischemia, or direct optic nerve insult. When stratified by surgical approach, the risk of a blinding surgical complication was significantly higher for patients undergoing orbital floor repair in the setting of multiple facial fractures (subgroup risk, 6.45%), bony decompression of the optic canal (subgroup risk, 15.6%), or intracranial approach to the orbital roof (subgroup risk, 18.2%). Seven of 8 patients with a potentially reversible cause of postoperative vision loss underwent urgent repeat surgery, and 2 regained substantial vision (20/20 and 20/25).
