Casertana is an endangered autochthonous pig breed (raised in south-central Italy) that is considered to be the descendant of the influential Neapolitan pig population that was used to improve British breeds in the 19th century. Casertana pigs are characterized by a typical, almost complete, hairless phenotype, even though a few Casertana pigs are normal haired. In this work, using Illumina PorcineSNP60 BeadChip data, we carried out a genome-wide association study and an f-ST analysis with this breed by comparing animals showing the classical hairless phenotype (n=81) versus pigs classified as haired (n=15). Combining the results obtained with the two approaches, we identified two significant regions: one on porcine chromosome (SSC) 7 and one on SSC15. The SSC7 region contains the forkhead box N3 (fOXN3) gene, the most plausible candidate gene of this region, considering that mutations in another gene of the same family (forkhead box N1;foxn1 or fOXN1) are responsible for the nude locus in rodents and alopecia in humans. Another potential candidate gene, rho guanine nucleotide exchange factor 10 (ARHGEf10), is located in the SSC15 region. fOXN3 and ARHGEf10 have been detected as differentially expressed in androgenetic and senescent alopecia respectively. This study on an autochthonous pig breed contributes to shed some light on novel genes potentially involved in hair development and growth and demonstrates that local animal breeds can be valuable genetic resources for disclosing genetic factors affecting unique traits, taking advantage of phenotype variability segregating in small populations.

Glycosylation is a highly diverse set of co- and post-translational modifications of proteins. for mammalian glycoproteins, glycosylation is often site-, tissue-, and species-specific and diversified by microheterogeneity. Multitudinous biochemical, cellular, physiological, and organismic effects of their glycans have been revealed, either intrinsic to the carrier proteins or mediated by endogenous reader proteins with carbohydrate recognition domains. furthermore, glycans frequently form the first line of access by or defense from foreign invaders, and new roles for nucleocytoplasmic glycosylation are blossoming. We now know enough to conclude that the same general principles apply in invertebrate animals and unicellular eukaryotes-different branches of which spawned the plants or fungi and animals. The two major driving forces for exploring the glycomes of invertebrates and protists are (i) to understand the biochemical basis of glycan-driven biology in these organisms, especially of pathogens, and (ii) to uncover the evolutionary relationships between glycans, their biosynthetic enzyme genes, and biological functions for new glycobiological insights. With an emphasis on emerging areas of protist glycobiology, here we offer an overview of glycan diversity and evolution, to promote future access to this treasure trove of glycobiological processes.

Morphological Variability Among Broods of first-Stage Blue Crab (Callinectes sapidus) Zoeae

External morphology has been shown to influence predation and locomotion of decapod larvae and is, therefore, directly related to their ability to survive and disperse. The first goal of this study was to characterize first-stage blue crab zoeal morphology and its variability across larval broods to test whether inter-brood differences in morphology exist. The second was to identify possible correlations between maternal characteristics and zoeal morphology. The offspring of 21 individuals were hatched in the laboratory, photographed, and measured. Zoeae exhibited substantial variability, with all metrics showing significant inter-brood differences. The greatest variability was seen in the zoeal abdomen, rostrum, and dorsal spine length. A principal component analysis showed no distinct clustering of broods, with variation generally driven by larger zoeae. Using observed morphology, models of drag induced by swimming and sinking also showed significant inter-brood differences, with a maximum twofold difference across broods. In contrast to trends in other decapod taxa, maternal characteristics (female carapace width and mass and egg sponge volume and mass) are not significant predictors of zoeal morphology. These results suggest that brood effects are present across a wide range of morphological characteristics and that future experiments involving Callinectes sapidus morphology or its functionality should explicitly account for inter-brood variation. Additionally, inter-brood morphological differences may result in differential predation mortality and locomotory abilities among broods.

Luminescent metal-organic frameworks and coordination polymers as alternative phosphors for energy efficient lighting devices

The development of lower cost and higher performance phosphor materials for energy efficient lighting and other optoelectronic applications is both necessary and feasible. Luminescent metal-organic frameworks and coordination polymers (LMOfs and LCPs, respectively) are a class of materials that hold great promise for this application. Their luminescence is eminently tunable, and a myriad of structures with incredibly diverse properties have been reported, with emission wavelengths covering the entire visible spectrum, white light emission from a variety of mechanisms, and quantum yields approaching unity. This review will briefly describe the luminescence mechanisms commonly observed in these materials, discuss strategies for the rational design of LMOf/LCP phosphors, and present a number of representative examples of each mechanism and/or design strategy. Dynamic quantitative risk assessment of accidents induced by leakage on offshore platforms using DEMATEL-BN

On offshore platforms, oil and gas leaks are apt to be the initial events of major accidents that may result in significant loss of life and property damage. To prevent accidents induced by leakage, it is vital to perform a case-specific and accurate risk assessment. This paper presents an integrated method of Ddynamic Qquantitative Rrisk Aassessment (DQRA)—using the Decision Making Trial and Evaluation Laboratory (DEMATEL)-Bayesian Network (BN)—for evaluation of the system vulnerabilities and prediction of the occurrence probabilities of accidents induced by leakage. In the method, three-level indicators are established to identify factors, events, and subsystems that may lead to leakage, fire, and explosion. The critical indicators that directly influence the evolution of risk are identified using DEMATEL. Then, a sequential model is developed to describe the escalation of initial events using an Event Tree (ET), which is converted into a BN to calculate the posterior probabilities of indicators. Using the newly introduced accident precursor data, the failure probabilities of safety barriers and basic factors, and the occurrence probabilities of different consequences can be updated using the BN. The proposed method overcomes the limitations of traditional methods that cannot effectively utilize the operational data of platforms. This work shows trends of accident risks over time and provides useful information for risk control of floating marine platforms.

An antidepressant-related pharmacological signature for positive allosteric modulators of α2/3-containing GABA_A receptors

Data from transgenic animals and novel pharmacological agents has realigned scientific scrutiny on the therapeutic potential of positive allosteric modulators (PAMs) of alpha 2/3-containing GABA(A) receptors. Evidence for analgesic, anticonvulsant, and anxiolytic activity of alpha 2/3-selective PAMs has been presented along with the clinical potential for a milder motor-impacting profile compared to non-selective GABA(A) receptor PAMs. A new series of alpha 2/3-selective PAMs was recently introduced which has anxiolytic and anticonvulsant activity in rodent models. These molecules also produce efficacy against pain in multiple animal models. Additionally, co-morbid states of depression are prevalent among patients with pain and patients with anxiety. Compounds were shown to be selective for alpha 2 and alpha 3 constructs over alpha 1(except KRM-II-82), alpha 4, alpha 5, and alpha 6 proteins in electro-physiological assays in transfected HEK-293T cells. Utilizing the forced-swim assay in mice that detects conventional and novel antidepressant drugs, we demonstrate for the first time that alpha 2/3-selective PAMs are active in the forced-swim assay at anxiolytic-producing doses. In contrast, activity in a related model, the tail-suspension test, was not observed. Diazepam was not active in the forced-swim assay when given alone but produced an antidepressant-like effect in mice when given in conjunction with the alpha 1preferring antagonist, beta-CCT, that attenuated the motor-impairing effects of diazepam. We conclude that these alpha 2/3-selective PAMs deserve further scrutiny for their potential treatment of major depressive disorder. If effective, such a mechanism could add a beneficial antidepressant component to the anxiolytic, analgesic, and anticonvulsant spectrum of effects of these compounds.

Antifungal and plant growth promotion activity of volatile organic compounds produced by Bacillus amyloliquefaciens

fusarium wilt of watermelon, caused by f. oxysporum ***. niveum (fON), is a devastating disease that causes extensive losses throughout the world. five bacterial strains (L3, h, beta, b, and L) isolated from the watermelon rhizosphere showed antagonistic activity against fON during in vitro tests. Strain L3 produced diffusible and volatile organic compounds (VOCs) which showed the strongest antifungal activity. Arabidopsis thaliana plantlets exposed to VOCs produced by strain L3 showed a 2.39-fold increase in biomass, 1.40-fold increase in primary root length, and 5.05-fold increase in number of lateral roots. Confocal laser scanning microscope showed that the GfP-labeled strain L3 could colonize along the elongation and differentiation zones of watermelon roots. In greenhouse pot experiments, the biocontrol efficiency of strain L3 against fusarium wilt of watermelon was up to 68.4% in comparison with the control treatment. In addition, inoculation of the strain L3 resulted in a 23.4% increase in plant fresh weight. Based on 16S rDNA sequence analysis, the strain L3 was identified as Bacillus amyloliquefaciens L3. fourteen VOCs produced by strain L3 were identified through GC-MS analysis. Of nine VOCs tested, 2-nonanone and 2-heptanone were proved to have strong antifungal properties. Acetoin and 2,3-butanediol were found to promote plant growth. The results suggested B. amyloliquefaciens L3 was a potential biocontrol agent, and that VOCs produced by B. amyloliquefaciens L3 play important roles in the process of biocontrol and plant growth promotion.

Outbreak of tularaemia connected to a contaminated well in the Vastra Gotaland region in Sweden

Tularaemia, is a zoonotic disease caused by the bacterium francisella tularensis. This disease has been reported in Sweden since 1931 and its wide distribution in the country poses a challenge for understanding the transmission, ecology and epidemiology of the disease. In Sweden, the disease is usually transmitted by mosquitoes, but in this study we could show that consumption of well water was epidemiologically linked to the outbreak, by isolating *** from the water. In this article, we describe an outbreak of tularaemia in the region of Vastra Gotaland in the southwest of Sweden in spring of 2013.

Confirmation of Brain Death with Positron Emission Tomography

After recent advances regarding organ transplantation, accurate and timely diagnosis of brain death has gained importance. In the diagnosis of brain death, in addition to clinical findings, various ancillary tests are very crucial. In this study, the scintigraphic imaging of the brain death of an 8-year-old girl with both Tc-99m diethylenetriaminepentaacetic and 18f-fluorodeoxyglucose (fDG) has been presented. This case study shows that 18f-fDG positron emission tomography-computed tomography imaging can be a useful technique in evaluating brain death in patients.
