Techniques for automatic analysis of two-dimensional electrophoresis gels by computer-aided image analysis are described. Original gels or photographic films are scanned using a laser scanner and the files are transferred to a microcomputer. The program package first performs a compression and preevaluation of the files. Spot identification and quantification is performed by the chain code algorithm after appropriate zooming and cutting. Labeling facilitates spot identification and quantification in numerical and graphical (pseudocolor) representation on peripheral devices for camera ready output. Interpolation between measured basepoints is performed by cubic spline algorithms which are automatically switched on and off, depending on the need by the program. High speed analysis and graphic representation is achieved using fast Assembler language routines rather than high level languages. One-dimensional gels can be analyzed using the same software. Spot matching between parallel two-dimensional gels has not yet been implemented.

IMMUNODETECTION IN FINE-NEEDLE ASPIRATES AND MULTIPARAMETRIC (SAMBA) image-ANALYSIS - RECEPTORS (MONOCLONAL ANTIESTROGEN AND ANTIPROGESTERONE) AND GROWTH FRACTION (MONOCLONAL KI67) EVALUATION IN BREAST CARCINOMAS

Immunocytochemical assays (ICA) using monoclonal antiestrogen receptors (ER, ICA), antiprogesterone receptors (PR ICA), and monoclonal antibody Ki67 (Ki67 ICA) were performed in 127 breast carcinomas. The immunostaining procedures were applied on frozen tissue sections, tumor imprints, and fine-needle aspirates in order to compare the variations in the distribution of the antigens detected in the three different types of preparations. Positive reactions detected with peroxidase-antiperoxidase and avidin-biotin-peroxidase, and alkaline phosphatase-antialkaline phosphatase complexes were evaluated through a computerized system of image analysis referred to as SAMBA 200 (SAMBA TITN, Grenoble, France). Application programs specifically developed for the analysis of tissue sections and of cytologic preparations were applied. This system allowed a multiparametric, accurate, reliable, reproducible and atomatized evaluation of the heterogeneity of the antigenic sites in tumors. For each markers positive cell surface (PS), and integrated and mean optical densities (IOD, MOD) and IOD histograms were compared. It was shown that (1) there was no significant variation in optical densities in cell imprints and aspirates whereas PS significantly (P < 0.01) differed in both preparations;(2) there were significant differences of the optical densities between tissue sections and cytological preparations, either imprints or aspirates, likely due to randomly cut nuclei in tissue sections;and (3) there was a significant difference between the PS of tissue sections and aspirates but no significant difference between tissue sections and imprints. It is concluded that fine-needle aspiration constitutes a convenient method for cell sampling, reliable for the diagnosis of malignancies. However, it may not reflect the heterogeneity of cell subpopulations in tissue.

Effects of image resolution on autoradiographic measurements of posterior cingulate activity in PDAPP mice: Implications for functional brain imaging studies of transgenic mouse models of Alzheimer's disease

Fluorodeoxyglucose positron emission tomography (PET) studies find that persons with Alzheimer's disease have preferential reductions in posterior cingulate activity. Using fluorodeoxyglucose autoradiography, we found that transgenic mice overexpressing a mutant form of the human amyloid precursor protein have preferentially reduced activity in the same region, providing a potential brain imaging indicator of Alzheimer's disease in these animals. In this study, we considered the feasibility of using in vivo imaging techniques, such as PET, to detect this reduction despite their limitations in spatial resolution. Autoradiographic measurements of posterior cingulate activity were remeasured in the previously studied PDAPP transgenic and littermate control mice after the images were filtered to lower spatial resolutions. We continued to detect significantly lower cingulate activity in the transgenic mice when the images were blurred to 0.50 mm, failed to detect significantly abnormal activity when the images were blurred to 0.75 mm, and, indeed, found significantly higher activity when the images were blurred to 1.0 mm. Reversal in direction of the abnormality appears attributable to a previously observed truncation in the corpus callosum in PDAPP mice. With the possible exception of future in vivo imaging techniques that have a spatial resolution greater than or equal to 0.50 mm and high sensitivity, noninvasive functional brain imaging techniques like PET may not be suitable for detecting declines in regional activity in PDAPP mice. It remains possible that these imaging techniques will prove useful in transgenic mouse lines that do not exhibit the same morphological abnormalities in neighboring white matter regions. A combination of energy-filtered electron microscopy (EFEM) and an image-analysis system (IBAS/2000) is used for a morphometric analysis of chemical reaction products in cells. Electron energy-loss spectroscopic element-distribution images are acquired from cytochemical reaction products in a variety of cellular objects: (1) colloidal thorium particles in extra-cellular coat material, (2) iron-containing ferritin particles in liver parenchymal cells, (3) barium-containing reaction products in endoplasmic reticulum stacks, (4) elements present in lysosomal cerium- and barium-containing precipitates connected with acid phosphatase (AcPase) or aryl sulphatase (AS) enzyme activity. Areas or area fractions are determined from such element-distribution images by application of an objective image segmentation method. By superposition of two or more element-distribution images, mutual element relations are qualitatively established in lysosomal cerium- and barium-containing precipitates connected with acid phosphatase (AcPase) or aryl sulphatase (AS) enzyme activity. By comparing electron spectroscopic images (ESI) with element-distribution images, the mutual contrast per element relations are quantitatively investigated. The obtained gain in resolution in such electron energy-loss spectroscopic element-distribution images will be explained and discussed.

MORPHOMETRIC STUDIES IN INTRADUCTAL BREAST-CARCINOMA USING computerIZED image-ANALYSIS

A morphometric study was performed using computerized image analysis on the histologic specimens of 26 patients with intraductal carcinoma of the breast and 26 normal control patients. A significant incidence of necrosis (56%) was observed in the breast ducts containing intraductal carcinoma. This was characterized by one or more areas of necrosis surrounded by a rim of viable tumor cells. The mean diameter of the ducts containing intraductal carcinoma was 349-mu-m compared with a mean diameter of 90-mu-m for the normal ducts. The mean diameter of the ducts containing necrosis was 470-mu-m compared with a mean diameter of 192-mu-m for the ducts containing a solid pattern of intraductal carcinoma. Central necrosis occurred in 94% of the ducts more than 180-mu-m in radius, whereas only 34% of the ducts less than 180-mu-m in size contained necrosis. The width of the viable neoplastic tissue was less than 180-mu-m in 91% of the ducts. These results are analogous to the findings of Thomlinson and Gray in human lung tumor cords and provide indirect evidence for the existence of a hypoxic compartment in intraductal carcinoma of the breast. This could be an explanation for the higher local failure rate after conservative surgery and irradiation for infiltrating cancers containing an extensive intraductal component.

image AND FLOW DNA CYTOMETRY OF SMALL CELL-CARCINOMA OF THE LUNG

Both image and flow DNA cytometry were performed in isolated nuclei from paraffin-embedded tumor tissue of patients with small cell carcinoma of the lung (SCCL). In 14 patients tissue was obtained by surgery from the primary tumor. From 14 patients tissue was taken by autopsy. From two patients tissue obtained by both surgery and later autopsy were available. From the autopsy patients tissue was taken only from the primary tumor (n = 6), from a metastasis (n = 1) and from the primary tumor and distant metastases (n = 7). Twleve of the tumors obtained by surgery were diploid, and two multiploid (two stem lines present). This was found both with image and flow cytometry. The group of patients could clearly be subdivided in short survivors (< 9 months, n = 6) and long survivors (> 16 months, n = 8);since in both groups one multiploid and the remainder diploid cases were present, ploidy did not seem to be a good prognosticator for survival. In most (n = 26) of the tissues measured from the autopsy patients, again, a good correlation between image and flow DNA cytometry was obtained, the histograms being either (near) diploid or multiploid. In six cases, however, flow cytometry showed multiploidy whereas image showed aneuploidy (one single peak clearly deviating from diploidy). This discrepancy is caused because normal diploid (noneoplastic) cells in the preparations could not be discarded from the flow cytometry measurments. Using the image cytometry data of the primary tumors, five diploid, three aneuploid, and four multiploid tumors were found. In five of the seven patients of whom tissue was obtained from the primary tumor and multiple metastases, differences between the histograms were found, mostly showing two malignant cell populations in one tissue and only one of them in another. Evaluation of endodontic files in digital radiographs before and after employing three image processing algorithms

Objectives: Two digital image processing algorithms, one aimed at correction for exponential attenuation and one at correction for visual response, have been developed. The aims of the present study were to test whether digital radiographs processed with these algorithms improve determination of the length of endodontic files and whether such processed radiographs are comparable with the radiographs processed with a default image processing method employed by one commercially available digital intraoral system. Methods: A dried human skull embedded in an acrylic compound was used for exposing radiographs of the upper and lower premolars and molars with endodontic files (Kerr files size 10 and size 15) positioned to the full length of the roots or 1.5 mm short of it. Radiographs were then processed in three sets. In one set, the radiographs were processed to compensate for exponential attenuation and the response of the human visual system. In the second, the radiographs were processed with the same compensation but with an additional shift in grey levels so that the output luminance in dentin at root tips corresponds to the mean of the luminance range of a computer monitor. In the third, the radiographs were processed with the default processing method in the Sidexis program. Ten viewers evaluated all radiographs. Receiver operating characteristic (ROC) curves were obtained and areas under the curves were calculated. Results: For file size 10, ROC curves for processed radiographs were higher than that for originals, while for file size 15, ROC curves for processed and original radiographs were close to each other. Significant differences were found between processed and original radiographs regarding areas under ROC curves for file size 10 but not between the differently processed radiographs. When denervated at the medium bud stage, limb blastemas of the newt, Pleurodeles waltlii Michah, stop growing. In order to better understand the role of nerves in the cell cycle in blastemas, we studied the distribution of mesenchymal cells in the G0-1, G1, S, G2 and M phases 48 and 96 h after denervation. The cell-cycle phases were determined by examining Feulgen-stained nuclei using a SAMBA 200 (System for Analytical Microscopy on Biological Applications) cell image processor. The cell nuclei were automatically analyzed by calculating 18 parameters related to the densitometry and texture of chromatin, and the shape of each nucleus. Cell-cycle phases were classified according to the unsupervised recognition method using a SAMBA 200 system as proposed by Moustafa and Brugal for cell-kinetics analysis. The classification obtained was tested against the results of stepwise linear discriminant analysis performed according to the method of Giroud. Our results show that, in blastemas 96 h after denervation, the percentage of cells in the S, G2, and M phases decrease significantly, while the percentage of G1 and G0-1 cells increases (+ 51% for G1 cells;+ 30% for G0-1 cells). Thus, it appears that denervation of medium-bud-stage limb blastemas promotes the lengthening of G1 and or premature exiting of cells from the cycle into the G0-1 phase. These results show that nerves (i.e, neurotrophic factor) regulate cell kinetics during newt limb regeneration by maintaining blastema mesenchymal cells in the cell-cycle.

Laparoscopic surface scanning and subsurface targeting: Implications for image-guided laparoscopic liver surgery

Segmental liver resection and locoregional ablative therapies are dependent upon accurate tumor localization to ensure safety as well as acceptable oncologic results. Because of the liver's limited external landmarks and complex internal anatomy, such tumor localization poses a. technical challenge. image guided therapies (IGT) address this problem by mapping the real-time, intraoperative position of surgical instruments onto preoperative tomogrophic imaging through a process called registration. Accuracy is critical to IGT and is a function of: 1) the registration technique, 2) the tissue characteristics, and 3) imaging techniques. The purpose of this study is to validate a novel method of registration using an endoscopic Laser Range Scanner (eLRS) and demonstrate its applicability to laparoscopic liver surgery. Six radiopaque targets were inserted into an ex-vivo bovine liver and a computed tomography (CT) scan was obtained. Using the eLRS, the liver surface was scanned and a surface-based registration was constructed to predict the position of the intraparenchymal targets. The target registration error (TRE) achieved using our surface-based registration was 2.4 +/- 1.0 mm. A comparable TRE using traditional fiducial-based registration was 2.6 +/- 1.7 mm. Compared to traditional fiducial-based registration, laparoscopic surface scanning is able to predict the location of intraparenchymal liver targets with similar accuracy and rate of data acquisition.

Purpose: We compared prostate tumor boundaries on magnetic resonance imaging and radical prostatectomy histological assessment using detailed software assisted co-registration to define an optimal treatment margin for achieving complete tumor destruction during image guided focal ablation. Materials and Methods: Included in study were 33 patients who underwent 3 Tesla magnetic resonance imaging before radical prostatectomy. A radiologist traced lesion borders on magnetic resonance imaging and assigned a suspicion score of 2 to 5. Three-dimensional reconstructions were created from high resolution digitalized slides of radical prostatectomy specimens and co-registered to imaging using advanced software. Tumors were compared between histology and imaging by the Hausdorff distance and stratified by the magnetic resonance imaging suspicion score, Gleason score and lesion diameter. Cylindrical volume estimates of treatment effects were used to define the optimal treatment margin. Results: Three-dimensional software based registration with magnetic resonance imaging was done in 46 histologically confirmed cancers. Imaging underestimated tumor size with a maximal discrepancy between imaging and histological boundaries for a given tumor of an average +/- SD of 1.99 +/- 3.1 mm, representing 18.5% of the diameter on imaging. Boundary underestimation was larger for lesions with an imaging suspicion score 4 or greater (mean 3.49 +/- 2.1 mm, p < 0.001) and a Gleason score of 7 or greater (mean 2.48 +/- 2.8 mm, p = 0.035). A simulated cylindrical treatment volume based on the imaging boundary missed an average 14.8% of tumor volume compared to that based on the histological boundary. A simulated treatment volume based on a 9 mm treatment margin achieved complete histological tumor destruction in 100% of patients.
