An fMRI study of processing novel metaphoric sentences

Due to inconsistent findings, the role of the two cerebral hemispheres in processing metaphoric language is controversial. The present study examined the possibility that these inconsistent findings may be due, at least partly, to differences in the type (i.e., words vs sentences) or the familiarity of the linguistic material. Previous research has shown that novel two-word metaphoric expressions showed stronger activation in the right homologue of Wernicke's area for the novel metaphors than for both literal expressions and unrelated word pairs. In the present study fMRI was used to identify the left (LH) and the right hemisphere (RH) neural networks associated with processing unfamiliar, novel metaphoric sentences taken from poetry, as compared to those involved in processing familiar literal sentences and unfamiliar nonsensical sentences. Across participants, several left lateralised brain regions showed stronger activation for novel metaphoric sentences than for the nonsensical sentences although both types of sentence represent unfamiliar linguistic expressions. Moreover, the metaphoric sentences elicited more activation in the left dorsolateral prefrontal cortex and the posterior middle temporal gyri than did both the literal sentences and the nonsensical sentences. The increased activation in these brain regions might reflect the enhanced demand on the episodic and semantic memory systems in order to generate de-novo verbal semantic associations. The involvement of the left posterior middle temporal gyri could reflect extra reliance on classical brain structures devoted to sentence comprehension.

Stereological measurement of rat＇s seminiferous tubule

Background The morphological measurements of seminiferous tubules are important in the studies of testis tissues. The purpose of this study was to evaluate the feasibility of using a stereological method to measure the geometric parameters of seminiferous tubule and to optimize the method. Methods A stereological image processing program was developed with Delphi for the stereological measurement. Fields of view were obtained from 15 healthy Wistar rats＇ testis tissues （n=247）. The diameter, area and volume of seminiferous tubule were estimated with the image processing program by two individual observers. The area results were compared with those obtained by the standard morphometric method of planimetry, Results Diameter measurements showed the diameters of different seminiferous tubules were almost the same and the mean value of about 50 tubules could be a good representation of the whole structure. Area measurements indicated there was no significant difference between stereology and planimetry （P 〉0.05）. But the stereological method required about 45% less time. Volume measurement showed the inter-observer variability was small （P 〉0.05） and the reproducibility of the stereological method was good. Conclusion The stereological technique was practical and efficient in the quantitative measurement of the rat＇s seminiferous tubule.

Mapping real-time images of high-speed AFM using multitouch control

Conventional AFM is highly restricted by its scan rate, a problem that has been overcome by the development of high-speed AFM systems. As the technology to produce higher scan rates has developed it has pushed forward the design of control software. However, the user interface has not evolved at the same rate, limiting the user to sequential control steps. In this paper we demonstrate the integration of HSAFM with a multitouch interface to produce a highly intuitive and responsive control environment. This enables nanometre resolution to be maintained whilst scanning the sample over tens of microns, and arbitrary paths to be traversed. We illustrate this by scanning around two chromosomes in water, before scanning on top of the chromosome, showing the surface structure.

Structure of the Anaphase-Promoting Complex/Cyclosome Interacting with a Mitotic Checkpoint Complex

Once all chromosomes are connected to the mitotic spindle (bioriented), anaphase is initiated by the protein ubiquitylation activity of the anaphase-promoting complex/cyclosome (APC/C) and its coactivator Cdc20 (APC/C-Cdc20). Before chromosome biorientation, anaphase is delayed by a mitotic checkpoint complex (MCC) that inhibits APC/C-Cdc20. We used single-particle electron microscopy to obtain three-dimensional models of human APC/C in various functional states: bound to MCC, to Cdc20, or to neither (apo-APC/C). These experiments revealed that MCC associates with the Cdc20 binding site on APC/C, locks the otherwise flexible APC/C in a "closed" state, and prevents binding and ubiquitylation of a wide range of different APC/C substrates. These observations clarify the structural basis for the inhibition of APC/C by spindle checkpoint proteins.

Nanografting versus Solution Self-Assembly of α,ω-Alkanedithiols on Au(111) Investigated by AFM

The solution self-assembly of alpha,omega-alkanedithiols onto Au(111) was investigated using atomic force microscopy (AFM). A heterogeneous surface morphology is apparent for 1,8-octanedithiol and for 1,9-nonanedithiol self-assembled monolayers (SAMs) prepared by solution immersion as compared to methyl-terminated n-alkanethiols. Local views from AFM images reveal a layer of mixed molecular orientations for alpha,omega-alkanedithiols, which evidence surface structures with heights corresponding to both lying-down and standing-up orientations. For dithiol SAMs prepared by solution self-assembly, the majority of alpha,omega-alkanedithiol molecules chemisorb with both thiol end groups bound to the Au(111) surface with the backbone of the alkane chain aligned parallel to the surface. However, AFM images disclose that there are also islands of standing molecules scattered throughout the surface. To measure the thickness of alpha,omega-alkanedithiol SAMs with angstrom sensitivity, methyl-terminated n-alkanethiols with known dimensions were used as molecular rulers. Under conditions of spatially constrained self-assembly, nanopattems of alpha,omega-alkanedithiols written by narografting formed monolayers with heights corresponding to an upright configuration.

Programmable fluidic production of microparticles with configurable anisotropy

We report a technique for continuous production of microparticles of variable size with new forms of anisotropy including alternating bond angles, configurable patchiness, and uniform roughness. The sequence and shape of the anisotropic particles are configured by exploiting a combination of confinement effects and microfluiclics to pack precursor colloids with different properties into a narrow, terminal channel. The width and length of the channel relative to the particle size fully specify the configuration of the anisotropic particle that will be produced. The precursor spheres packed in the production zone are then permanently bonded into particles by thermal fusing. The flow in the production zone is reversed to release the particles for collection and use. Particles produced have linear chain structure with precisely configured, repeatable bond angles. With software programmable microfluidics, sequence and shape anisotropy are combined to yield synthesized homogeneous (type "A"), surfactantlike (type "A-B") or triblock (type "A-B-A") internal sequences in a single device. By controlling the dimensions of the microfluidic production zone, triangular prisms and particles with controlled roughness and patchiness are produced. The fabrication method is performed with precursors spheres with diameter as small as 3.0 mu m.

Practical issues in imaging hydraulic conductivity through hydraulic tomography

Hydraulic tomography has been developed as an alternative to traditional geostatistical methods to delineate heterogeneity patterns in parameters such as hydraulic conductivity (K) and specific storage (S-s). During hydraulic tomography surveys, a large number of hydraulic head data are collected from a series of cross-hole tests in the subsurface. These head data are then used to interpret the spatial distribution of K and S-s using inverse modeling. Here, we use the Sequential Successive Linear Estimator (SSLE) of Yeh and Liu (2000) to interpret synthetic pumping test data created through numerical simulations and real data generated in a laboratory sandbox aquifer to obtain the K tomograms. Here, we define "K tomogram" as an image of K distribution of the subsurface (or the inverse results) obtained via hydraulic tomography. We examine the influence of signal-to-noise ratio and biases on results using inverse modeling of synthetic and real cross-hole pumping test data. To accomplish this, we first show that the pumping rate, which affects the signal-to-noise ratio, and the order of data included into the SSLE algorithm both have large impacts on the quality of the K tomograms. We then examine the role of conditioning on the K tomogram and find that conditioning can improve the quality of the K tomogram, but can also impair it, if the data are of poor quality and conditioning data have a larger support volume than the numerical grid used to conduct the inversion. Overall, these results show that the quality of the K tomogram depends on the design of pumping tests, their conduct, the order in which they are included in the inverse code, and the quality as well as the support volume of additional data that are used in its computation.

Sex differences in the serotonin 1A receptor and serotonin transporter binding in the human brain measured by PET

Women and men differ in serotonin associated psychiatric conditions, such as depression, anxiety and suicide. Despite this, very few studies focus on sex differences in the serotonin system. Of the biomarkers in the serotonin system, serotonin(1A) (5-HT1A) receptor is implicated in depression, and anxiety and serotonin transporter (5-HTT) is a target for selective serotonin reuptake inhibitors, psychotropic drugs used in the treatment of these disorders. The objective of the present study was to study sex related differences in the 5-11T(1A) receptor and 5-HTT binding potentials (BP(ND)s) in healthy humans, in vivo. Positron emission tomography and selective radioligands [C-11]WAY100635 and [C-11]MADAM were used to evaluate binding potentials for 5-11T(1A) receptors (14 women and 14 men) and 5-HTT (8 women and 10 men). The binding potentials were estimated both on the level of anatomical regions and voxel wise, derived by the simplified reference tissue model and wavelet/Logan plot parametric image techniques respectively. Compared to men, women had significantly higher 5-HT1A receptor and lower 5-HTT binding potentials in a wide array of cortical and subcortical brain regions. In women, there was a positive correlation between 5-11T(1A) receptor and 5-HTT binding potentials for the region of hippocampus. Sex differences in 5-HT1A receptor and 5-HTT BPND may reflect biological distinctions in the serotonin system contributing to sex differences in the prevalence of psychiatric disorders such as depression and anxiety. The result of the present study may help in understanding sex differences in drug treatment responses to drugs affecting the serotonin system. (c) 2007 Elsevier Inc. All rights reserved.
