INHBA-Associated Markers as Candidates for Stallion Fertility

Contents The inhibin beta A (INHBA) gene was chosen as candidate for stallion fertility and analysed for intragenic markers to find associations with pregnancy rate per oestrus. Intragenic single nucleotide polymorphisms (SNPs) were developed in order to perform an association and haplotype analysis using the least square means (LSM) of the pregnancy rate per oestrus for stallions as well as breeding values (BVs) for the embryonic and paternal component of the pregnancy rate per oestrus. The polymorphisms were genotyped in 161 Hanoverian warmblood stallions. Insemination records from approximately 20 000 Hanoverian warmblood mares were used to calculate LSM for stallions and to predict the paternal and embryonic component of BVs for the pregnancy rate per oestrus. We demonstrated significant associations of single markers and haplotypes with the LSM and the embryonic and paternal component of BVs for the pregnancy rate per oestrus. This is the first report on INHBA as an associated candidate gene with the LSM of stallions and the paternal and embryonic component of BVs for the pregnancy rate per oestrus.

The effect of the presence of an Fe-Cr alloy metallic interconnect on the performance and stability of La0.8Sr0.2MnO3 (LSM) oxygen electrodes is studied for the first time under solid oxide electrolysis cell (SOEC) operating conditions at 800 degrees C. The presence of the Fe-Cr interconnect accelerates the degradation and delamination processes of the LSM oxygen electrodes. The disintegration of LSM particles and the formation of nanoparticles at the electrode/electrolyte interface are much faster as compared to that in the absence of the interconnect. Cr deposition occurs in the bulk of the LSM oxygen electrode with a high intensity on the YSZ electrolyte surface and on the LSM electrode inner surface close to the electrode/electrolyte interface. SIMS, GI-XRD, EDS and XPS analyses clearly identify the deposition and formation of chromium oxides and strontium chromate on both the electrolyte surface and electrode inner surface. The anodic polarization promotes the surface segregation of SrO and depresses the generation of manganese species such as Mn2+. This is evidently supported by the observation of the deposition of SrCrO4, rather than (Cr, Mn)(3)O-4 spinels as in the case under the operating conditions of solid oxide fuel cells. The present results demonstrate that the Cr deposition is essentially a chemical process, initiated by the nucleation and grain growth reaction between the gaseous Cr species and segregated SrO on LSM oxygen electrodes under SOEC operating conditions.

Development of a non-invasive algorithm with transient elastography (Fibroscan) and serum test formula for advanced liver fibrosis in chronic hepatitis B

P>Background Non-invasive assessments of liver fibrosis in chronic hepatitis B were well established. Aim To develop a combined algorithm of liver stiffness measurement (LSM) and serum test formula to predict advanced liver fibrosis in chronic hepatitis B. Methods We reported an alanine aminotransferase (AST)-based LSM algorithm for liver fibrosis in 156 chronic hepatitis B patients, which formed the training cohort to evaluate the performance of APRI (AST-to-platelet-ratio-index), Forns index, FIB-4 and Fibroindex against liver histology. The best combined LSM-serum formula algorithm would be validated in another cohort of 82 chronic hepatitis B patients. Results In the training cohort, LSM has the best performance of diagnosing advanced (>= F3) fibrosis [area under the receiver operating characteristics curve (AUROC) 0.88, 95% confidence interval (CI) 0.85-0.91], while Forns index has the best performance among the various serum test formulae (AUROC 0.70, 95% CI 0.62-0.78). In the combined algorithm, low LSM or low Forns index could be used to exclude advanced fibrosis as both of them had high sensitivity (> 90%). To confirm advanced fibrosis, agreement between high LSM and high Forns index could improve the specificity (from 99% to 100% and from 87% to 98% in the training and validation cohorts respectively). Conclusion A combined LSM-Forns algorithm can improve the accuracy to predict advanced liver fibrosis in chronic hepatitis B.

Coincidental metabolic syndrome increases the risk of liver fibrosis progression in patients with chronic hepatitis B - a prospective cohort study with paired transient elastography examinations

Background Metabolic syndrome is a known risk factor of cirrhosis in chronic hepatitis B (CHB). Aim To investigate the effects of coincidental metabolic syndrome on liver fibrosis progression in treatment-naive CHB patients. Methods A total of 1466 CHB patients underwent liver stiffness measurement (LSM) by transient elastography in 2006-2008;663 patients remained treatment-naive and had second LSM in 2010-2012. Liver fibrosis progression was defined as an increase in LSM >= 30% at the second assessment. The impact of coincidental metabolic syndrome and its factors on liver fibrosis progression were evaluated after adjustment for viral load and hepatitis activity. Results At baseline, the mean age was 43 +/- 12years, 55% were males, serum alanine aminotransferase (ALT) was 44 +/- 40IU/L, HBV DNA was 4.0 +/- 2.0logIU/mL and LSM was 6.3 +/- 3.6kPa. Metabolic syndrome was diagnosed in 80 (12%) and 142 (21%) patients at baseline and follow-up visit, respectively;84 (13%) and 22 (3%) patients had coincidental and resolved metabolic syndrome respectively. After an interval of 44 +/- 7months, 107 (16%) patients developed liver fibrosis progression. Coincidental metabolic syndrome [adjusted odds ratio (aOR) 2.0, 95% confidence interval (CI) 1.1-3.5, P=0.015], central obesity (aOR 2.0, 95% CI 1.0-4.1, P=0.05) and low level of high-density lipoprotein cholesterol (aOR 1.9, 95% CI 1.0-3.7, P=0.04) were associated with liver fibrosis progression independent of change in viral load and ALT level. The effects of coincidental metabolic syndrome were most apparent in the immune-tolerant phase. Conclusion Coincidental metabolic syndrome increases the risk of liver fibrosis progression in patients with chronic hepatitis B infection, independent of viral load and hepatitis activity.

Reversible Decomposition of Secondary Phases in BaO Infiltrated LSM Electrodes-Polarization Effects

In operando Raman spectroscopy is used to study ceramic La0.85Sr0.15MnO3 +/-delta electrodes infiltrated with BaO. The aim of this work is to clarify why BaO infiltration reduces the polarization resistance in oxygen containing atmospheres. Prior to the in operando experiments, ex situ X-ray diffraction and Raman spectroscopy reveal the formation of a secondary phase, Ba3Mn2O8, on the electrode. During the in operando Raman investigation of the BaO-infiltrated La0.85Sr0.15MnO3 +/-delta electrodes, experiments are performed at 300 and 500 degrees C with oxygen partial pressure 0.1 atm and with -1 or +1 V applied potential. A changing electrode surface is observed during operation as the Ba3Mn2O8 secondary phase decomposes and manganese oxide accumulates on the electrode surface during cathodic polarization. The observed changes are reversible. These results suggest that the formation of Ba3Mn2O8 is responsible for the reduced polarization resistance observed at open circuit voltage (OCV) in an oxygen containing atmosphere. Furthermore, the results illustrate the dramatic differences between the electrode surface composition at OCV and during cathodic polarization. Overall, the results highlight the dynamic interactions between minor secondary phases and applied potential, a general effect that may be important for the high-performance frequently observed with ceramic electrodes prepared by infiltration.

Prediction of Recurrence after Curative Resection of Hepatocellular Carcinoma using Liver Stiffness Measurement (FibroScan^A®)

The purpose of this study was to investigate whether preoperative liver stiffness measurement (LSM) can predict recurrence after curative resection of hepatocellular carcinoma (HCC). LSM using FibroScan(A (R)) can assess the severity of liver fibrosis, which is significantly associated with recurrence after curative resection of HCC. Between February 2006 and March 2009, 133 patients who underwent preoperative LSM and curative resection for HCC were enrolled in this prospective study. LSM values were analyzed for association with recurrence, together with other clinical variables. The mean age of the patients (117 men and 16 women) was 57 years. During the follow-up period (median, 25.0 (range, 3.0-54.6) months), HCC recurred in 62 (46.6 %) patients. In multivariate analysis, together with satellite nodule and Edmonson-Steiner grade III-IV, LSM was selected as an independent predictor of recurrence (P < 0.05;hazard ratio, 1.034;95 % confidence interval, 1.007-1.061). When the study population was stratified into two groups using the optimal cutoff value (13.4 kPa) that maximized the sum of sensitivity (64.7 %) and specificity (76.1 %) from time-dependent receiver operating characteristic curves (area under the receiver operating characteristic curve = 0.676), patients with LSM values > 13.4 kPa were at a significantly greater risk for recurrence with a hazard ratio of 1.925 (P = 0.01;95 % confidence interval, 1.17-3.168) compared with those with LSM values a parts per thousand currency sign13.4 kPa. Our data suggest that LSM can be a useful predictor of recurrence after curative resection of HCC.

ObjectiveDyslipidaemia and central obesity are the major factors underlying the dramatic increase in metabolic syndrome (MS). We compared the effects of early combined therapy with pitavastatin and intensive lifestyle modification (LSM) on the amelioration of each component of MS with those of LSM only. Design/Participants/MeasurementsPROPIT (a PROspective comparative clinical study to evaluate the efficacy and safety of PITavastatin in patients with metabolic syndrome) was a prospective, randomized, multicenter open-label 48-week trial. We enrolled 187 patients with MS (central obesity and prediabetes) and randomized them into two treatment groups: 2mg pitavastatin daily+intensive LSM or intensive LSM only. The primary outcome was the improvements in the components of MS and in the percentage of non-MS converters. ResultsAfter 1year treatment, the improvement of MS score was significantly higher in the pitavastatin+LSM group (P=0039). However, non-MS converters (MS score 2) did not differ between the groups. The secondary outcomes, namely lipid profiles, the Apo B/A1 ratio, visceral fat/subcutaneous fat ratio and the Framingham risk score, were significantly improved in the pitavastatin group. There was no deterioration in glucose metabolism after treatment with pitavastatin for 1year. ConclusionsEarly statin treatment can be an effective option in obese patients with MS, prediabetes and mild dyslipidaemia with further improvement of cardiovascular risk factors. We could not observe the increase rate of glucose intolerance in statin group. Future longitudinal studies are needed to test the benefits of early statin treatment compared with LSM.

BIAS SPUTTER-DEPOSITION OF DENSE YTTRIA-STABILIZED ZIRCONIA FILMS ON POROUS SUBSTRATES

We demonstrate that completely dense yttria-stabilized zirconia (YSZ) electrolyte films <5 mu m thick can be reactively sputter deposited onto porous (similar to 0.5 mu m pore size) La0.8Sr0.2MnO3 substrate/electrodes. The two key factors in achieving the fully dense films were porous substrate preparation and the use of substrate bias during YSZ film deposition. Increasing the negative de substrate bias V-s from 0 to 300 V produced increasingly dense and flat-surfaced films: V-s = 75 V was chosen to yield high density without excessive film compressive stresses. Solid oxide fuel cells consisting of sputtered YSZ films (V-s = 75 V) and a Ni-YSZ fuel electrode on LSM exhibited open-circuit voltage values within 5% of the theoretical values for thicknesses down to 2 mu m. Cells deposited with V-s = 0 V short-circuited at thicknesses of 12 mu m or less.

Propulsion control of superconducting linear synchronous motor vehicle

The armature current of a superconducting Linear Synchronous Motor (LSM) for a maglev vehicle is controlled to produce a suitable propulsion force so that the vehicle follows the reference speed signal sent from a control station. Besides the power is supplied from some inverters to the LSM armature sections where the vehicle exists. This paper shows an exact mathematical modeling of the propulsion control system to treat the system analytically, which is used for, designing controllers and performance computer simulations. The calculated results include the simulations when the vehicle goes through power feeder section borders and tunnels that have a large aerodynamic drag force with taking account of an inverter failure.

Evaluation of MM5 mesoscale model at local scale for air quality applications over the Swedish west coast: Influence of PBL and LSM parameterizations

The performance of MM5 mesoscale model (Version 3.6.3) using different planetary boundary layer (PBL) and land surface model (LSM) parameterizations is evaluated and compared using high temporal and spatial resolution GOTE2001 campaign data at local scale (a few kilometers) over the Greater Goteborg area along the Swedish west coast during 7-20 May 2001. The focus is on impact of PBL and LSM parameterizations on simulated meteorological variables important for air quality applications such as global radiation, diurnal cycle of near-surface air temperature and wind, diurnal cycle intensity, near-surface vertical temperature gradient, nocturnal temperature inversion, boundary layer height, and low-level jet (LLJ). The model performance for daytime and nighttime and under different weather conditions is also discussed. The purpose is to examine the performance of the model using different PBL and LSM parameterizations at local scale in this area for its potential applications in air quality modeling. The results indicate that the influence of PBL and LSM parameterizations on simulated global radiation, diurnal cycle of near-surface air temperature and wind speed, diurnal cycle intensity, vertical temperature gradient, nocturnal temperature inversion and PBL heights, which are critical parameters for air quality applications, is evident. Moreover, the intensity and location of LLJ are simulated well by all schemes, but there also exist some differences between simulated results by using different PBL and LSM schemes. Therefore, the choice of PBL and LSM parameterizations is important for MM5 applications to air quality studies.
