The key proteolytic enzyme analysis of industrial Aspergillus oryzae at solid-state koji fermentation with a local database extension

The productivity and flavor quality of soy sauce are partly determined by the proteolytic enzymes secreted by the industrial microbe Aspergillus oryzae, which break down proteins into peptides and amino acids. Because of how complicated it is to classify proteolytic enzymes and understand how they work, not much is known about what is in the profiles of proteolytic enzymes and which enzymes are most important to A. oryzae during solid-state fermentation. In this study, we collected industrial A. oryzae from Chinese and Japanese soy sauce production, both of which have excellent protein hydrolysis capability but have numerous genetic differences. We systematically analyzed the in vitro enzyme profiles of two industrial A. oryzae based on 4D label-free proteome sequencing technology. In addition, we constructed a local database of proteolytic enzymes from the Aspergillus genus, which helps us analyze the proteomics data more precisely. We found that 96 proteolytic enzymes are excreted by industrial A. oryzae, of which 27 are endopeptidases and 39 are exopeptidases. Based on the relative protein expression and transcription quantity, as well as the catalytic characteristics, we identified 27 potential key proteolytic enzymes associated with high protein breakdown efficiency at solid-state fermentation. Meanwhile, we proposed eight proteolytic enzymes with potential hydrolytic activity. This study deepens our understanding of the proteolytic enzyme profiles excreted by A. oryzae at solid state and provides our knowledge of the candidate key proteolytic enzymes during koji fermentation.

Assembling and dietary application of a local trnL metabarcoding database for Cervus nippon kopschi in Taohongling Nature Reserve

The quality and completeness of the reference database have a direct impact on the accuracy of forage plant identification, thereby influencing the level of conservation and management of wildlife resources. In our research, target amplification was subjected to first-generation sequencing to assemble a local reference database using chloroplast trnL metabarcoding. We found that the primers c-h outperformed g-h as a universal DNA metabarcoding and 162 valid choloroplast trnL sequences were submitted (GenBank ID: PP081756- PP081917), which exhibited an obvious preference for A and T nucleotides (60.49%). The haplotype diversity (Hd), nucleotide diversity (Pi) and average number of nucleotide differences (K) of these trnL sequences were 0.978, 0.0484 and 4.743, respectively. To assess the availability of the local database in identifying the diet of South China sika deer ( Cervus nippon kopschi), high-throughput metabarcoding sequencing and BLAST analysis were performed. Ultimately, 25 forage plant species were identified, belonging to 19 families and 25 genera. Shrubs and herbaceous plants, such as Potentilla freyniana, Persicaria perfoliata, Rosa laevigata and Ardisia japonica etc, dominated the forage plants. This study established a local trnL reference database that holds immense value for the forage plant identification and nutritional evaluation for sika deer and other sympatric herbivores, as well as the conservation and management of biodiversity.

In this paper, a hot spot detection and monitoring system based on UAV is proposed. The system uses UAV to carry thermal imager and cruise automatically to collect images and transmit them back to the ground station control system. The real-time image processing of the ground station detects hot spots and saves the inspection results to the local database, and can encode and output the inspection video containing the hot spot location.

Hosts need to identify their location on the Internet. One way is to use an IP address. In IPV4 networks, each computer is identified by a unique 4-byte number, which is generally written as a four-point format, such as 199.1.35.90. When data are transmitted via the network, the header of the packet will contain the IP address of the destination host. Another method is to use the host name to identify themselves, such as ***, ***. Compared with the IP address, it is more memorable. However, the host name provides little or no information about the host's location in the network. In addition, since host names are usually composed of alphanumeric characters of varying lengths, routers are better at handling fixed-length, hierarchical IP addresses. Therefore, domain name system (DNS) has been developed for converting host names that are easy to be remembered by humans into digital Internet addresses. But the use of DNS sometimes reduces the efficiency of accessing the network because the network environment varies all the time, which results in the transmission rate of information between the client and the DNS server. If the package losses due to timeout, the respond of the DNS server cannot be received. As a result, this paper designs a model that can decrease the probability of these problems by introducing a local DNS database. © 2017 IEEE.

In recent studies on ultrasound-guided infraclavicular block (ICB), the authors have favoured a single injection posterior to the axillary artery rather than multiple injections;however, procedural complications and success rates associated with single-injection ultrasound-guided ICB are not well known. We undertook an observational study to evaluate the success rates of experienced and non-experienced operators performing ICBs and to identify the complications associated with ultrasound-guided single-injection ICB. We conducted an observational cohort study of all ultrasound-guided single-injection ICBs performed over a two-year period (2008-2010). We identified the subjects for our study using a local database and excluded patients younger than 18 yr and those who received a continuous ICB. Complications (non-neurological and neurological) and ICB success rates were the primary and secondary end points, respectively. We collected the following data from patients' charts: patient demographics, types of complications and their respective frequencies, and the experience of the clinician performing the ICBs, and we identified potential late complications by telephone interview. Using a seven-point Likert scale, two experts in regional anesthesia evaluated the likelihood of a relationship between the identified neurological signs or symptoms and the ICB. A neurologist then evaluated the complications identified as being potentially related to the ICB. Summary data were collated, and 95% confidence intervals (CI) were calculated. We reviewed 627 ICB procedures, and 496 (79%) patients received telephone interviews. Most patients were males who had undergone either plastic or orthopedic surgery. Mepivacaine 1.5% was used in 96% of cases with a median volume of 30 mL [interquartile range 30-38]. We identified 131 cases of neurological signs or symptoms. A Resource Discovery with Data Dissemination over Unstructured Mobile P2P Networks

Recently, along with the innovative development of wireless communication techniques and mobile devices, mobile P2P services in mobile wireless networks have gained a lot of attention. In this paper, we propose a resource discovery scheme with data dissemination over mobile P2P networks. In the proposed scheme, each peer manages a local information table, a resource index table, and a routing table in a local database to enhance the accuracy and cost of resource discovery. The local information table stores the status of a mobile peer, and the resource index table stores the resource information of the neighbor peers via the ranking function. The routing table is used to communicate with the neighbor peers. We use a timestamp message to determine whether or not the resource index table will be changed before the resource information is exchanged. Our ranking function considers the interest and mobility of the mobile peer and prioritizes the resource information transmitted from the neighbor peers. It is shown via various experiments that the proposed scheme outperforms the existing scheme.

This paper aims to improve the real-time patient monitoring systems with Web view by its extension in those systems which display the results on a mobile phone and send them to the central server system. The research is based on the telemonitoring system, emphasizing on patient data acquisition and storing into a local database, data transmission and graphical presentation to the physician. The telemonitoring system has been tested on several diabetic people during the clinical trials performed within the DIAdvisor FP7 research project (DIAdvisor Project, 2010). Then it was made an analysis of the system developed for iPhone and the application called Glooko Logbook launched by Apple, displaying on a terminal the blood glucose levels. The novelty of the research is to develop methods of transmitting data from the iPhone to a web server that stores the information and its electronic mark so that no entry can be repeated, nor lost. The server will send the feedback to the iPhone when streaming was done successfully or not. If not all information could be sent the server will test the marks that were saved to avoid duplication. These test methods of the flow of information between local databases, that mean iPhone database and the server database provides accurate monitoring of this system.

The paper presents a new system used for patient telemonitoring. The system is for ambulatory use and the physician has the possibility to monitor the patient state. The patient health state is measured by means of several dedicated wireless sensors, which values are saved into a local database. All new values that the local database stores in are periodically sent to the physician server and displayed into a graphical manner. Additional data analysis is performed in order to extract complex information regarding the patient health. Several alarms are also raised if some of the patient monitored parameters overpass the normal limits. The benefits are in terms of continuous supervision of a patient even he/she performs the undergoing treatment ambulatory.
